Synthesis of 68Ga-labeled gold nanoparticles for tumor targeted positron emission tomography imaging

  • Herein we present the synthesis of $^{68}Ga$-labeled gold nanoparticles for in vivo PET imaging. A novel chelator DTPA-Cys was easily prepared from diethylenetriaminepentaacetic dianhydride in high yield. The ${\alpha}_v{\beta}_3$ integrin receptor targeted gold nanoparticle probe was synthesized by using DTPA-Cys, polyethylene glycol and cRGD peptide. $^{68}Ga$ labeling of cRGD conjugated gold nanoparticle was carried out at $40^{\circ}C$ for 30 min. Observed radiochemical yield was more than 75% as determined by radio-TLC and the probe was purified by centrifugation. In vitro stability test showed that 90% of $^{68}Ga$-labeled gold nanoparticle probe was stable in FBS for 1 h. Those results demonstrated that $^{68}Ga$-labeled gold nanoparticle could be used as a potentially useful probe for specific tumor imaging.

Use of Gold Nanoparticle Fertilizer Enhances the Ginsenoside Contents and Anti-Inflammatory Effects of Red Ginseng

  • Red ginseng, a steamed and sun-dried ginseng, is a popular health-promoting food in Korea and other Asian countries. We introduced nanofertilizer technology using gold nanoparticles in an effort to develop red ginseng with an elevated level of ginsenosides, the main active compounds of ginseng. Shoots of 6-year-old ginseng plants were fertilized three times with colloidal gold nanoparticle sprays. Red ginseng extract was prepared from the main roots. The concentrations of gold and ginsenosides were measured following gold nanoparticle treatment. To evaluate the anti-inflammatory effects, mouse peritoneal macrophages of male BALB/c mouse were stimulated with lipopolysaccharide plus interferon-γ in the presence of extracts from red ginseng with or without gold nanoparticle treatment. The content of ginsenosides, such as Rg1, Re, Rf, and Rb1, increased in ginseng treated with gold nanofertilizer whereas the steaming process increased only the levels of Rd and Rg3. The levels of nitric oxide, inducible nitric oxide synthase, and interleukin-6, but not tumor necrosis factor-α, were more suppressed in macrophages treated with extract from gold nanoparticle-treated red ginseng. Our results show that the use of a colloidal gold nanoparticle fertilizer improved the synthesis of ginsenosides in ginseng and enhanced the anti-inflammatory effects of red ginseng. Further research is required to elucidate the causal factors for the gold-induced change in ginsenoside synthesis and to determine the in vivo effect of gold nanoparticle-treated ginseng.

Synthesis of Gold Nanoparticles by Chemical Reduction Method for Direct Ink Writing

  • Aqueous gold nanoparticle dispersion was synthesized by chemical reduction method using diethanolamine as reducing agent and polyethyleneimine as dispersion stabilizer. The synthesis conditions for the stable dispersion of the gold nanoparticle suspension were determined by changing the amount of the reducing agent and dispersant during the wet chemical synthesis procedures. The face centered cubic lattice structure of the gold nanoparticles was confirmed by using X-ray diffraction and the morphologies of the nanoparticles were observed by transmission electron microscope. The synthesized gold nanoparticle dispersion was concentrated by evaporating the dispersion medium at room temperature followed by the addition of ethyleneglycol as humectant for the increase of the elastic properties to obtain gold nanoparticle inks for direct ink writing process. The line patterns were obtained with the gold nanoparticle inks during the writing procedures and the morphologies of the fine patterns were observed by scanning electron microscope.

  • 반도체 집적회로의 고집적화 및 고성능화를 위한 기본 소자(MOSFET)의 미세화 및 단위공정의 물리적 한계를 극복하기 위해 기존의 Top-down 방식에서 buttom-up 방식의 공정에 대한 연구가 진행되고 있다. 그 중 nanoparticles를 이용한 나노소자 제작 연구가 이루어지고 있다. 하지만 이러한 nanoparticles를 이용한 나노소자의 제작에 있어서 원하는 위치에 nanoparticles를 배열하고 정렬하는데 어려움을 겪고 있다. 이 문제를 해결하기 위해서 자기조립 특성을 가지고 있는 DNA분자와 기능화를 통하여 표면에 positive charge를 띄고있는 Gold nanoparticles를 상호결합 시키는 실험을 하였다. Au-DNA nanowire는 backbone에 있는 phosphate부분에서 negative charge를 띠고 있는 DNA와 positive charge를 띠고 있는 Gold nanoparticles가 결합하는 원리로 형성된다. 그렇지만 Gold particles를 표면이 아닌 DNA에만 붙이는 것은 아직 해결해야 할 부분으로 남아있다. 본 연구에서는 이 문제를 해결하기 위하여 pH 조절을 통하여 기능화된 Gold particles의 charge의 변화를 주고 이를 Zeta potential 측정기로 측정한 후에 이 particles와 DNA를 결합시켜서 FE-SEM과 AFM 으로 확인하는 실험을 하였다.

금 미세 입자에 의한 그래핀 광학 특성 변화 (Gold Nanoparticle Optical Effect on Graphene)

  • Graphene and Graphene oxide have intense interest in fields such as physics, chemistry, and materials science, among others. They are the promising material for solving the current limitation that organics have barely luminescence. We observed variation of photoluminescence on graphene oxide based solution with Gold nanoparticle. Gold nanoparticles lead to shift the peak wavelength of graphene oxide and to enhance the photoluminescence intensity totally. This shows the possibility that control the luminescence property of graphene oxide by adding gold nanoparticle.

Heat-induced coarsening of layer-by-layer assembled mixed Au and Pd nanoparticles

  • This article shows the coarsening behavior of nanoparticle multilayers during heat treatments which produce larger metallic nanostructures with varying shapes and sizes on glass slides. Nanoparticle multilayer films are initially constructed via the layer-by-layer self-assembly of small and monodispersed gold and/or palladium nanoparticles with different compositions (gold only, palladium only, or both gold and palladium) and assembly orders (compounding layers of gold layers over palladium layers or vice versa). Upon heating the slides at $600^{\circ}C$, the surface nanoparticles undergo coalescence becoming larger nanostructured metallic films. UV-Vis results show a clear reliance of the layering sequence on the optical properties of these metal films, which demonstrates an importance of the outmost (top) layers in each nanoparticle multilayer films. Topographic surface features show that the heat treatments of nanoparticle multilayer films result in the nucleation of nanoparticles and the formation of metallic cluster structures. The results confirm that different composition and layering sequence of nanoparticle multilayer films clearly affect the coalescence behavior of nanoparticles during heat treatments.

Fabrication and Characterization of Polystyrene/Gold Nanoparticle Composite Nanofibers

  • Polystyrene/gold nanoparticle (PS/AuNP) composite fibers were fabricated using an electrospinning technique. Transmission electron microscopy (TEM) showed that the diameters of the naphthalenethiol-capped gold nanoparticles (prior to incorporation into the PS fibers) ranged from 2 to 5 nm. UV-vis spectroscopy revealed the surface plasmon peaks of the gold nanoparticles centered at approximately 512 nm, indicating that nano-sized Au particles are well-dispersed in solution. This was consistent with the TEM observations. The electrospun nanofibers of PS/AuNP composites were approximately 60-3,000 nm in diameter. The surface morphology of the PS/AuNP composite and the dispersability of the Au nanoparticles inside of PS after electrospinning process were investigated by SEM and TEM. The thermal behavior of the pure PS and PS/AuNP nanocomposites and fibers were examined by DSC.

Preparation of Gold Nanoisland Arrays from Layer-by-Layer Assembled Nanoparticle Multilayer Films

  • This article introduces a facile nanoparticle self-assembly/annealing method for the preparation of nanoisland films. First, nanoparticle-polymer multilayer films are prepared with layer-by-layer assembly. Nanoparticle multilayer films are then annealed at $~500^{\circ}C$ in air to evaporate organic matters from the films. During the annealing process, the nanoparticles on the solid surface undergo nucleation and coalescence, resulting in the formation of nanostructured gold island arrays. By controlling the overall thickness (number of layers) of nanoparticle multilayer films, nanoisland films with various island density and different average sizes are obtained. The surface property of gold nanoisland films is further controlled by the self-assembly of alkanethiols, which results in an increased surface hydrophobicity of the films. The structure and characteristics of these nanoisland film arrays are found to be quite comparable to those of nanoisland films prepared by vacuum evaporation method. However, this self-assembly/annealing protocol is simple and requires only common laboratory supplies and equipment for the entire preparation process.

Sonochemical Synthesis of Closed [5,6]-bridged Aziridino[70]fullerene Derivative and Self-assembled Multilayer Films

  • This cycloaddition of [70]fullerene with methyl azidoacetate in benzene under ultrasonic irradiated condition afforded the closed [5,6]-bridged aziridino[70]fullerene derivative, which was unusual product of cycloaddition to the 5,6-junction of fullerene. Its structure was determined by FAB-MS, UV-vis, $^1H-$ and $^{13}C$-NMR spectral data. The closed [5,6]-bridged aziridino[70]fullerene-functionalized gold nanoparticle films were self-assembled using the layer-by-layer method on the reactive of glass slides functionalized with 3-mercaptopropyl trimethoxysilane. The functionalized glass slides were alternately soaked in the solution containing closed the [5,6]-bridged aziridino[70]fullerene and 4-aminothiophenoxide/hexanethiolate-protected gold nanoparticles. The closed [5,6]-bridged aziridino[70]fullerene-functionalized gold nanoparticle films have grown up to 5 layers depending on the immersion time. The self-assembled nanoparticle multilayer films were characterized using UV-vis spectroscopy showed that the surface plasmon band of gold at 527 nm gradually became more evident as successive layers were added to the films.
