산양의 과배란 처리에 따른 발정발현과 단발정주기의 발생 및 발정기의 혈중 estradiol-17$\beta$의 농도변화 (Behavioural Estrous, Short Estrous Cycles and Serum Level of Estradiol-17$\beta$ durig Peri-Estrus following Superovulation Treatments in Korean Native Goats)

  • Behavioural estrus and short estrous cycles were observed and serum concentrations of estradiol-17$\beta$(E2) before and after of estrous were measured following superovulation treatments in 30 pluriparous Korean native goats. The goats were divided into 2 groups. Fifteen goats were injected IM with 1,000IU PMSG on Day 12 of the estrous cycle followed by 10mg PGF2$\alpha$ 48h later(P4+PMSG), and the other 15 goats were injected IM with 10mg progesterone(P4)in oil once daily for 10d beginning at any days of estrous cycle followed by 1,000IU PMSG and 10mg PGF2$\alpha$ at the 8th day of progesterone treatment(P4+PMSG group). After injection of PGF2$\alpha$, onset of standing estrus occurred in 12 of 15 goats(80.0%) at 50.0$\pm$7.7h and in 11 of 15 goats(73.3%) at 135.6$\pm$10.1h in PMSG and group and P4+PMSG group, respectively. The mean interval from PGF2$\alpha$ injection to first estrus was significantly(P<0.01) earlier in PMSG group than in P4+PMSG group. This result indicate that the delayed infusion of P4 in P4+PMSG group caused the later exhibition of their estrous behaviors. However, duration fo frist estrus(31.5$\pm$2.6h vs 26.2$\pm$2.3h), length of estrous cycle(14.1$\pm$3.3d vs 16.6$\pm$3.8d) and percentage of short estrous cycle(50.0% vs 45.5%) were not different between PMSG and P4+PMSG group. The mean concentration of serum E2 in 4 goats showing normal estrous cycle in P4+PMSG group(PP-NEC) was higher than in 6 goats showing normal(P-NEC) or in 6 goats showing short estrous cycle(P-SEC) in PMSG group. The peak level of serum E2 was observed at the time of onset of standing estrus in PP-NEC(67.6pg/ml), 6h earlier in P-NEC(53.1pg/ml) and 6h later in P-SEC(52.3pg/ml) than the onset of standing estrus. The profiles of serum concentration of E2 during the period of peri-estrus was similar in the goats of PMSG or P4+PMSG and also in the goats showing the subsequent estrous cycle of normal or short length.

Evaluation of carcass traits, meat quality and the expression of lipid metabolism-related genes in different slaughter ages and muscles of Taihang black goats

  • Objective: This study was conducted to investigate the effect of slaughter age on carcass traits, meat quality, and the relative mRNA levels of lipid metabolism-related genes in different muscles of Taihang black goats. Methods: In this study, the triceps brachii (TB), longissimus dorsi (LD) and gluteus (GL) muscles of 15 grazing Taihang black goats slaughtered at the age of 2, 3, and 4 (designated as 2-year-old, 3-year-old, and 4-year-old, respectively) were collected. The differences in carcass shape, meat quality, amino acid composition and lipid metabolism gene expression among Taihang black goats of different ages and from different plant parts were compared. Results: Compared with goats at other ages, goats slaughtered at the age of 4 had greater live and carcass weights, meat weights, bone weights and skin areas (p<0.05). LD in the 4-years-old had the lowest cooking loss and moisture content. The crude protein content in the LD of 2-year-old was significantly greater than that in the other age group, and at the age of 2, the LD had the highest crude protein content than TB and GL. The highest fat content was in LD, followed by TB, for goats slaughtered at the age of 4. Eight out of 9 essential amino acids had higher content in the TB compared with other muscles, regardless of age. The total essential amino acid content was highest in the 4-year-old and lowest in the GL muscle at the age of 3. The sterol regulatory element-binding protein-1c (SREBP-1c) and adipose triglyceride lipase (ATGL) genes were significantly more abundant in the TB muscle than in the other muscles for goats slaughtered at the age of 2. At the age of 4, the ATGL and peroxisome proliferator-activated receptor γ (PPARγ) genes were significantly more abundant in the GL than in the LD, while the fatty acid synthase (FAS) genes were significantly less abundant in the GL than in the other muscles. Similarly, compared with those in goats of other ages, the relative mRNA expression levels of the FAS and heart-type fatty acid binding protein (H-FABP) genes in goats slaughtered at the age of 4 were the highest, and the relative mRNA expression of the PPARγ gene was the lowest (p<0.05). The relative mRNA expression of the H-FABP and FAS genes was positively correlated with the intramuscular fat (IMF) content, while the relative mRNA expression levels of the PPARγ and ATGL genes was negatively correlated with the IMF content. Conclusion: Overall, a better nutritional value was obtained for TB from 4-year-old goats, in which the total essential amino acid and fat contents were greater than those of other muscles. The comprehensive action of lipid metabolism genes was consistent with that of the IMF content, among which the FAS, H-FABP, PPARγ, and ATGL genes had positive and negative effects on the process of IMF deposition in Taihang black goats.

재래종과 Saanen 종 염소의 갑상선 기능과 증체율의 비교 (Thyroid Activity and Body Gain in Native and Saanen Goats)

  • In native and Saanen goats, the thyroidal iodine uptake rate and its correlation between thyroidal activity and body gain were studied. The results were as follows: 1. The thyroidal iodine-131 uptake rate in native and Saanen goats were 10.46% and 21.26%, respectively, and the difference was statistically significant (P<0.01). 2. The correlation coefficients between monthly body gain and thyroidal iodine-131 uptake rate were -0.32 and -0.46 in native and Saanen goats, respectively. The correlations were negative and not significant between the rates.

The Effects of Skin Pigmentation on Physiological Factors of Thermoregulation and Grazing Behaviour of Dairy Goats in a Hot and Humid Climate

  • The aim of this study was to understand the effects of skin pigmentation on physiological parameters of thermoregulation and grazing behaviour of dairy goats in a hot and humid climate. The study used 26 crossbred Saanen yearling goats (95% Saanen+5% Local Hair Breed). The animals were raised at semi-intensive private farms in Adana (36 59'N, 35 18'E). Groups were selected 2 d before the start of observations. Goats were categorized as predominantly pigmented (P) skin and unpigmented (UP) skin. All observations and measurements were collected on grassland during the grazing period of June and July 2007 (60 d). Air temperature and relative humidity were recorded at 10 min intervals by a portable data logger. The physiological data (rectal temperature, respiration and pulse rate, and skin temperatures from head and udder) were recorded twice weekly in the morning (07:00-08:00); at midday (13:00-14:00); and in the evening (18:00-19:00). Additionally, the activity of the animals was observed and classified (eating, ruminating, drinking, standing, walking, lying) for 12 h during the day twice weekly, using a portable camera system linked directly to a computer. Panting behaviour was also observed. According to the THI values, the experimental goats were subjected to stressful conditions. The pigmented goats had significantly lower rectal temperatures (39.68 vs. $29.89^{\circ}C$), pulse rate (74.08 vs. 84.10 beat/min) and respiration rate (65.65 vs. 88.23 breath/min.) compared with unpigmented goats at midday when the THI exceeded 92. The rectal, head and udder temperatures, pulse and respiration rates of the non-pigmented group exceeded $40^{\circ}C$, $37^{\circ}C$, $37.5^{\circ}C$, 84 beats/min and 78 breaths/min, respectively. Higher activity was observed among pigmented compared with unpigmented goats. Unpigmented goats grazed (4.3 vs. 5.6 h), ruminated (2.0 vs. 2.4 h), and stood (0.8 vs. 1.2 h) less, but lay down (2.2 vs. 1.8 h) more than pigmented goats. The data obtained in this experiment support the hypothesis that unpigmented goats are more adversely affected by climatic stress, likely due to their decreased activity and increased water consumption, as demonstrated by previous studies.

Evaluation of Some Agri-industrial By-products Available in Samoa for Goats

  • Nutritional evaluation of some agro-industrial byproducts available in Samoa [dry brewers' grains (DBG), cocoa shell (CS), cocoa dust (CD) and desiccated coconut waste meal (DCWM)] available in Samoa was carried out using both the in vivo and in vitro techniques. In the in vivo study 24 Anglo-nubian goats were offered by-products with other feed ingredients to compound four different diets. The goats were randomly allocated to 4 diets on the basis of liveweight (18.7-0.3kg). The ADF content of the byproducts followed a similar trend to NDF. The byproducts have a high content of organic matter (91.0-95.4%). Gross energy (GE) content was higher in DCWM (25.1 MJ/kg DM), closely followed by CD (23.2 MJ/kg DM). Concentrate intake was significantly different (p<0.05) among the goats. Average daily live weight gains were 105, 92, 88 and 97 g/goat/day for DBG, CS, CD and DCWM, respectively. Daily live weight gains were higher (p<0.05) in the goats that received DBG, while the least gain was obtained in the goats that received CS byproduct diet. DM digestibility was significantly higher (p<0.05) in the goats on DBG diet than in the other goats. The least DM digestibility was obtained in the goats that received CD diet (p>0.05). CP digestibility followed a similar pattern to DM digestibility. The digestibility of NDF and ADF was influenced by the nature of the diets. The digestibility of OM and GE were best (p<0.05) in the goats that received DBG, DCWM and CS byproduct diets than in CD. Significant differences (p<0.05) among the byproducts were recorded for net gas production. Potential gas production (a+b) ranged from 7.064 to 42.17 ml. Organic matter digested (OMD) from gas production value at 24 h was higher in DBG (47.6 g/kg DM) and this was followed by DCWM (42.5 g/kg DM). The least OMD was obtained in CD (17.9 g/kg DM). A significant difference (p<0.05) in DM disappearance after 4, 8, 16, 24, 48 and 72 h was recorded. The potential and effective degradability varied significantly (p<0.05) from 85.95-99.6 g/kg DM and from 39.9-65.8%, respectively. The digestibility of the byproducts in both the in vivo and in in vitro techniques demonstrated that they are potential source of feed ingredients for ruminant livestock in Samoa and possibly in the other small Pacific Island countries. On the basis of their potential degradability the byproducts could be ranked in the following order:DCWM>DBG>CD>CS. In conclusion, the results obtained suggest that all the byproducts can contribute to ruminant livestock diets without adverse effects on feed intake, growth rate and apparent nutrient digestibility coefficients.

Effect of Supplemental Selenomethionine on Growth Performance and Serum Antioxidant Status in Taihang Black Goats

  • An experiment was conducted to evaluate the effect of different levels of supplemental selenomethionine (Se-Met) on growth performance and serum antioxidant status in Taihang Black goats. Fifty 16-week-old goats with an average body weight of 12.5${\pm}$0.5 kg were randomly assigned to five treatments fed a basal diet (0.049 mg Se/kg DM) supplemented with 0 (control), 0.10, 0.30, 0.50 and 1.00 mg of Se/kg DM (form Se-Met) for 80 days. Average daily gain and feed efficiency were higher (p<0.05) in the groups supplemented with 0.30 to 0.50 mg Se/kg DM compared with the control group. However, Se-Met supplementation had no influence on average daily feed intake (p>0.05). Se-Met supplementation significantly increased (p<0.01) the activity of glutathione peroxidase enzymes (GSH-Px) and superoxide dismutase (SOD) in serum. The group supplemented with 0.50 mg Se/kg DM had the highest activity of GSH-Px compared with other groups (p<0.05). Serum SOD activity was higher (p<0.05) in goats supplemented with both 0.30 and 0.50 mg Se/kg DM than in control goats and goats supplemented with 1.00 mg Se/kg DM. Serum glutathione-S-transferase (GST) activity and malondialdehyde (MDA) concentration were significantly decreased (p<0.05) in goats supplemented with 0.30, 0.50 and 1.00 mg Se/kg DM compared with control values. These results indicated that Se-Met supplementation markedly improved the antioxidant status in goats. Blood Se concentration increased linearly (p<0.001) and quadratically (p<0.001) as the level of supplemental Se-Met increased. The concentration of Se in the control diet (0.049 mg Se/kg DM) did not satisfy the Se requirement in goats as indicated by reduced growth rate, feed efficiency, activities of GSH-Px and SOD in serum, and blood Se concentrations. In conclusion, it is recommended that 0.30 to 0.50 mg of Se/kg DM from Se-Met (total diet Se of 0.349 to 0.549 mg/kg DM) be supplied in the diet of Taihang Black goats to enhance growth performance and improve antioxidant status.

Concentrations of Progesterone and Estradiol in Peripheral Plasma during the Estrous Cycle and after Ovariectomy in Huanghuai Goats of High or Poor Prolificacy

  • The objective of this study was to characterize the litter sizes in Huanghuai goats with high prolificacy (HP, five or more kids born per litter on at least one occasion), and to compare their peripheral blood concentrations of progesterone and estradiol with those of goats with poor prolificacy (PP, up to three kids born per litter on any occasion). The circulating concentrations of progesterone and estradiol were measured by radioimmunoassay from daily blood samples taken during natural estrus cycles and at 1-5 days after ovariectomy. Estrus was synchronized using two doses of a prostaglandin analog. Litter size for the HP goats increased up to a parity of five and decreased thereafter. The percentage of goats with litter sizes of $\geq$4 from parities 3 to 6 ranged from 44.5% to 58.3%. Although small differences in litter size were obtained for goats of parities three, four and six relative to five, parity five does had the highest mean litter size. Progesterone concentrations began to rise earlier and were higher in the HP than in the PP goats on most days of the luteal phase, but not during the follicular phase of the cycle or after ovariectomy. There was a significant difference between the two groups (p<0.05) in the magnitude of the progesterone plateau. Mean estradiol concentrations in the HP group remained significantly higher than in the PP group (p<0.05) during the estrus cycle. There were two estradiol peaks in the HP goats during the early luteal phase, but only one in the PP group. Measurements of individual corpora lutea (CL) in vitro showed that there was a greater prevalence of small CL (<6 mm in diameter) in the HP group than in the PP group (p<0.05). After ovariectomy, the estradiol level on day 1 was significantly higher than at the nadir during the estrus cycle in both the HP (p<0.01) and PP (p<0.05) goats, while levels decreased by 12.3% and 26.2% respectively compared with the mid-luteal period in HP and PP goats (p<0.05). The overall mean estradiol concentrations in HP goats were lower than in the PP group, but no significant differences were found between groups at 1-5 days after ovariectomy.

Rumen Parameters and Urea Kinetics in Goats and Sheep

  • The effects of animal species and supplements on rumen fluid characteristics, plasma urea-N (PUN) concentration, plasma urea-N pool size, urea-N degradation in the gut and urea-N net flux (urea-N synthesis rate) were studied in goats and sheep, with some minor differences detected. The animals were fed either chopped rice straw ad libitum+200 g soybean meal (SBM), or chopped rice straw ad libitum+190 g soybean meal+300 g sago meal (SBM+SM) for 14 days. The supplements were isonitrogenous (80 g crude protein/animal/d). [$^{14}C$]-urea was used as the marker for urea metabolism studies. Two animals from each species were fed either supplement in a cross-over design in two periods. The results showed that rumen pH was significantly (p<0.001) lower in animals fed SBM+SM than those fed SBM supplement. The ammonia concentrations of rumen fluid were significantly (p<0.01) higher in sheep (382.9 mg N/L) than goats (363.1 mg N/L) when fed SBM supplement but lower (282.5 mg N/L) than that of goats (311.0 mg N/L) when fed SBM+SM supplement. Total VFA concentrations were significantly (p<0.05) higher in animals fed SBM+SM supplement than those fed SBM supplement. Goats had significantly (p<0.01) higher molar proportions of acetate (79.1, 77.7%, respectively) than sheep (75.8, 74.0%, respectively) in both supplements. The molar proportion of acetate was significantly (p<0.05) higher, while that of butyrate lower in animals fed SBM supplement than those fed SBM+SM supplement. In animals fed SBM supplement, the molar proportion of propionate was significantly (p<0.01) higher in sheep (18.0%) than in goats (15.6%), but in animals fed SBM+SM, the molar proportion of butyrate was significantly (p<0.01) higher (9.6%) in sheep than in goats (7.2%). Plasma urea-N concentration, plasma urea-N pool size, urea-N degradation in the gut, urea-N net flux and the fraction of urea-C from the blood entering the rumen were not significantly different between goats and sheep fed either supplement. However, PUN concentration was significantly (p<0.05) lower in animals fed SBM+SM supplement (average of 13.8 mg N/100 ml) than in those fed SBM supplement (average of 16.5 mg N/100 ml). The urea net flux was significantly (p<0.05) higher in goats (average of 14.5 g N/d) than sheep (average of 12.9 g N/d), and animals fed SBM supplement showed higher (average of 14.9 g N/d) urea net flux than animals fed SBM+SM supplement (average of 12.9 g N/d). A significant (p<0.05) positive correlation was observed between urea-N net flux and urea-N degradation; urea-N net flux and pool size; urea-N net flux and urea excretion in the urine; and PUN and rumen ammonia in goats. While in sheep, significant (p<0.05) positive correlation was observed between urea-N net flux and urea excretion in the urine; and PUN and rumen ammonia.

Meat Quality Characteristics of Small East African Goats and Norwegian Crosses Finished under Small Scale Farming Conditions

  • The aim of the experiment was to study the effect of feeding system on meat quality characteristics of Small East African (SEA) goats and their crosses with Norwegian ($SEA{\times}N$) goats finished under small scale farming conditions. Twenty four castrated goats at the age of 18 months with live body weight of $16.7{\pm}0.54kg$ from each breed (SEA and $SEA{\times}N$) were distributed in a completely randomized design in a $2{\times}3$ factorial arrangement (two breed, and three dietary treatments). The dietary treatments were; no access to concentrate (T0), 66% access to ad libitum concentrate allowance (T66) and 100% access to ad libitum concentrate allowance with 20% refusal (T100) and the experimental period was for 84 days. In addition, all goats were allowed to graze for 2 hours daily and later fed grass hay on ad libitum basis. Daily feed intakes were recorded for all 84-days of experiment after which the animals were slaughtered. Feed intake of T100 animals was 536 g/d, which was 183 g/d higher than that of T66 group. Supplemented goats had significantly (p<0.05) better feed conversion efficiency. The SEA had higher (p<0.05) hot carcass weight (8.2 vs 7.9 kg), true dressing percentage (54.5 vs 53.3) and commercial dressing percentage (43.3 vs 41.6) compared to $SEA{\times}N$. There was no significant difference (p>0.05) for dressing percentage and carcass conformation among supplemented goats except fatness score, total fat depots and carcass fat which increased (p<0.05) with increasing concentrate levels in the diet. Increasing level of concentrate on offer increased meat dry matter with subsequent increase of fat in the meat. Muscle pH of goats fed concentrate declined rapidly and reached below 6 at 6 h post-mortem but temperature remained at $28^{\circ}C$. Cooking loss and meat tenderness improved (p<0.05) and thawing loss increased (p<0.05) with ageing period. Similarly, meat tenderness improved (p<0.05) with concentrate supplementation. Shear force of muscles varied from 36 to 66, the high values been associated with Semimembranosus and Gluteobiceps muscles. The present study demonstrates that there are differences in meat quality characteristics of meat from SEA goats and their crosses with Norwegian breeds finished under small scale farming conditions in rural areas. Therefore, concentrate supplementation of goats of both breeds improves meat quality attributes.

한국재래염소의 mtDNA 다양성 및 계통유전학적 분석 (mtDNA Diversity and Phylogenetic Analysis of Korean Native Goats)

  • 한국재래염소는 흑모색의 특징을 나타내며, 유일한 염소 품종으로서 오랫동안 한반도에서 사육되어 왔다. 하지만 이들에 대한 유전적 다양성, 계통유전학적 분석 등을 통한 기원 추정 등에 대한 연구는 미비한 실정이다. 본 연구에서 한국재래염소 5개 집단, 60두를 대상으로 mtDNA D-loop 영역 중 HVI 영역의 서열을 이용하여 유전적 다양성 및 계통유전학적 분석을 실시하였다. 한국재래염소는 다른 나라 염소들에 비해서 haplotype 다양성 지수가 낮게 나타났다. 또한 본 연구에서 분류된 한국재래염소 10개 haplotype 중 현재까지 보고되지 않은 6개의 새로운 haplotype이 확인되었다. 계통유전학적 분석 결과, 분석에 사용된 모든 한국재래염소는 mtDNA 모계혈통 A에 속하였다. 10개의 haplotype 중 8개는 베트남, 일부 중국 염소와 함께 subgroup을 형성하였다. 그러나 나머지 2개 haplotype은 각각 서로 독립적인 계통유전학적 위치를 보였다. 이런 결과들을 토대로 한국재래염소는 상대적으로 높은 근친상황으로 외부 유전자 유입이 적었을 것이라고 추정된다. 한국재래염소의 새로운 mtDNA haplotype의 발견 및 유전자원 보존 및 평가를 위해서 더 많은 분석집단 및 개체를 수집하고, MS 마커를 이용한 추가분석이 필요하다고 사료된다.
