생약제제의 이화학적 품질평가에 관한 연구 갈근탕(葛根湯)의 연구 (Studies on Quality Control of Crude Drugs Preparations, Chemical Analysis of 'Gal Gun Tang')

  • The prescription of Gal Gun Tang, which has been used for treatment of cold, fever, and muscular pain in Chinese herb medicine, is produced in the form of decoction However, the storage problem for this dosage form remains unsettled. Using HPLC and GC, we examined quantitative change of major constituents caused by time-progress and temperature-change. Nine major constituents, such as cinnamaldehyde, cinnamic acid, ephedrine, puerarin, paeoniflorin, daidzin, benzoic acid, glycyrrhizin and liquiritin, were selected as references. The content of cinnamaldehyde significantle decreased by the increase of temperature. While that of cinnamic acid increased. Benzoic acid showed the most significant change of the content in three months at $40^{\circ}C$. It is suggested that most of constituents are considerably stable when kept frozen.

  • The biologically active component of licoris(Glycyrrhizae Radix L.) is considered to be glycyrrhetinic acid, an aglycone of glycyrrhizin, on the basis of chemical and pharmacological studies. The present study was undertaken to investigate the effect of glycyrrhetinic acid on the hepatic morphine-6-dehydrogenase activity, which catalize morphine to morphinone. Morphine-6-dehydrogenase was further purified by centrifugation, $(NH_4)_2SO_4$ fractionation, sephadex G-100, hydroxyapatite column. Hepatic morphine-6-dehydrogenase activity was significantly decreased by the treatment of glycyrrhetinic acid. When effect of glycyrrhetinic acid on the hepatic morphine-6-dehydrogenase was investigated in vitro, it was powerfully inhibited the enzyme activity with dose-dependent manner. From the above results, glycyrrhetinic acid inhibits hepatic morphine-6-dehydrogenase activity and decreases the morphine induced harmful side effects.

Determination of Total Glycyrrhetic Acid in Glycyrrhizae Radix by Second Derivative UV Spectrometry

  • Second derivative (D2) spectrometry using ion-pair extraction technique was development for the determination of total glycyrrhetic acid (GA) in Glycyrrhizae Radix, Glycyr-rhizin (G) obtained from Glycyrrhizae Radix was hydrolyzed into GA in 2 N-HCI and methanol (1:1) and extracted from aqueous phase in the form of an ion-pair complex with tetrapentylammonium bromide (TPA) as a counter ion. Maximum D2 amplitude (Z value) was obtained when 1000-fold or greater molar ratio of TPA was used at pH 11. Reaction an effective extraction solvent of the ion-pair complex. The linearity of standard curve of ion-pair GA was obtained in the range of 4.120 $\mu$g/ml as GA. Assayed contents of GA in dry powder by D2 UV spectrometry and HPLC method were 5.31 $\pm$ 0.04% and 5.20 $\pm$ 0.008%, respectively.

  • A high performance liquid chromatographic method was developed for the determination of glycyrrhetinic acid contained in licorice powder. Glycyrrhetinic acid which is hydrolysate of glycyrrhizin extracted from licorice powder, was determined with good result by HPLC using 2-bromoacetyltriphenylene labeling reagent. The glycyrrhetinic acids were labeled with 2-bromoacetyltriphenylene in acetonitrile using 18-crown-6-ether and KOH as a catalyst. Derivatized glycyrrhetinic acids were separated from the extracted licorice powder on a reversed-phase column (chemopak $C_{18}$) using 100% acetonitrile as a mobile phase and monitored by an UV-detector at 268nm. Linearity of calibration curve was obtained between 5 ng and 20 ng, and the lower limit of detection was 2 ng. The recovery of glycyrrhetinic acid to licorice powder was about 99.3%. This method was sensitive, reliable and useful for, determination of glycyrrhetinic acid.

  • The famous prescription of So Cheong Ryong Tang (小靑龍湯)in Chinese herb medicine, which has been used for the treatment of common cold, influenza, asthmatic bronchitis, and bronchial asthma and, is being commercially produced in the form of decoction. However, the storage problem for this dosage form remains unsettled. The quantitative changes of the major constituents, at different temperature and time course, were examined employing HPLC and GC. Cinnamaldehyde, cinnamic acid, ephedrine, albiflorine, paeoniflorin, benzoic acid, glycyrrhizin, and liquiritin were choosen as phytochemical markers. The content of cinnamaidehyde significantly decreased when stored at increased temperature, while that of benzoic acid has increased. By overall consideration of the present experimental data, it was suggested that most constituents are considerably stable when preserved below freezing temperature.

Conditions for Preparing Glycyrrhiza uralensis Extract for Inhibiting Biofilm Formation of Streptococcus mutans

  • Licorice, which has an extensive history of use as an herbal medicine, has been suggested to have oral health benefits. However, to date, no systematic study has been conducted on the preparation method of licorice extracts for oral health. In this study, licorice extracts prepared using water and ethanol were investigated for its ability to inhibit the biofilm formation of Streptococcus mutans. The licorice extract prepared with around 60% ethanol effectively inhibited the biofilm formation of S. mutans. Licorice extracted with 50% ethanol almost completely inhibited the biofilm formation at 1.5 g/L of licorice extract. This inhibitory activity was confirmed in a microplate assay and a flow cell system. Glycyrrhetic acid was extracted from licorice effectively with 60% ethanol concentration. The strong inhibitory activity of glycyrrhetic acid and the synergistic inhibition with glycyrrhizin on biofilm formation were suggested as major reasons for a concentration-specific extraction. These results suggest that licorice extract prepared using around 60% ethanol effectively inhibits the biofilm formation of S. mutans.

  • Glucocorticoids are used most widely for the treatment of inflammatory bowel disease (IBD). For the efficient treatment and reduction of side effects, colon-specific delivery of a glucocorticoid is highly desirable. Previously, we synthesized prednisolone 21-sulfate sodium (PDS) as a colon-specific prod rug of prednisolone (PD) expecting that it might be stable and nonabsorbable in the upper intestine and hydrolyze in the colon to release PD. 오배자 성분의 항산화 및 간보호 효과 (Antioxidative and Hepatoprotective Effect of Compounds Isolated from Galla Rhois(Rhus javanica Linne))

  • 활성산소에 기인하는 다양한 성인병을 예방 치료할 수 있는 안전하고 우수한 항산화 물질을 천연물로 부터 개발하기 연구의 일환으로 오배자의 주성분에 대한 항산화효과 및 간보호효과를 연구하여 다음과 같은 결과를 얻었다. 1. Ferric-Thiocyanate 법에 의하여 과산화반응 억제에 따른 항산화 활성을 측정한 결과 모두 tocopherol보다 강한 과산화반응 억제 활성을 보였으며 gallic acid methyl ester, protocatechuic acid와 syringic acid는 tocopherol 보다는 강한 활성을 보였으며 BHA와는 유사 또는 그 이상의 활성을 보였다. 특히 1, 2, 3, 4, $6-penta-O-galloyl-{\beta}-D- glucose$는 tocopherol과 BHA보다 매우 강한 과산 화반응억제 활성을 보였다. 2. TBA 법에 의한 과산화지질의 생성의 지표인 MDA 억제효과에 의한 항산화효과를 측정한 결과 gallic acid 는 $1.6{\times}10^{-4}$의 농도에서 BHA 보다 높은 억제율을, protocatechuic acid는 $1.6{\times}10^{-6}$의 농도에서 BHA 보다 높은 억제율을 보였다. gallic acid methyl ester와 1, 2, 3, 4, $6-penta-O-galloyl-{\beta}-D-glucose$는 농도가 증가함에 따라 우수한 MDA의 억제활성을 나타내었다. 3. Glycyrrhizin을 비교군으로 하여 간 보호 실험을 한 결과 1, 2, 3, 4, $6-penta-O-galloyl-{\beta}-D-glucose$의 0.5mg/ml첨가군에서는 GOT의 강한 감소 효과를 보였으며 GPT 실험에서는 각 시료 0.5mg/ml 첨가군에서 감소하는 경향을 나타내었다.

추출방법(抽出方法)에 따른 내소화중탕(內消和中湯)의 효능(效能)에 대(對)한 연구(硏究) (An Experimental Study on the Component Variation of Naesowhajung-tang by the Three Types of Extraction Method and the Effects of Each Type on the Gastrointestinal Tract)

  • When I evaluated the component variation of Naesowhajung-tang by the three types of extraction method, and each type's effects on the gastrointestinal tract, I got the following results. 1. The output ratio of extracts made out of Naesowhajung-tang were not significantly different among 13.8% of water extract(Sample-I), 13.5% of 50% ethanol extract(Sample-II), and 15.6% of water extract by spray dryer(Sample-III). 2. magnolol, honokiol, hesperidin, naringin, poncirin and glycyrrhizin Sample II had the largest amount of the following contents: magnolol, honokiol, hesperidin, naringin, poncirin and glycyrrhizin. 3. All the extracts of Naesowhajung-tang showed the inhibitory effect on the smooth muscle contraction of the isolated ileum in mice and fundus-strip in rats induced by acetylcholine chloride and barium chloride. 4. High concentration Sample-II was recognized to be effective in preventing gastric ulcers in Shay's rats. but not in the other rat group. 5. All the extracts of Naesowhajung-tang were recognized to be effective in preventing gastric ulcers induced by Ethanol-HCl in rats. 6. The increase of transport ability in the small intestine was recognized only when the concentration of all the samples was doubled, but not in the other concentrations. 7. The increase of transport ability in the large intestine was recognized only when the concentration of Sample-II was doubled, but not in other concentration. Using the results mentioned above, I suggest that Sample-II has more significant effects on the gastrointestinal tract than the others.

한약박에서 재배한 느타리버섯의 성장 및 저장 중 품질 특성의 비교 (The Comparison of Growth and Quality Characteristics during the Storage of Pleurotus ostreatus Cultivated in the Remnants of Medicinal Herb Extracts)

  • 일반적으로 폐기 처분하거나 비료로 재활용하는 한약을 추출하고 남는 한약박을 발효하여 영양분으로 첨가하는 팰릿 대신 폐면과 섞어 균상배지로 사용하여 느타리버섯을 재배할 때 수확량과 저장성에 대한 연구를 하였다. 또한, 한약박은 불특정 한약재들이 섞여 있으므로 이로 인해 문제가 나타날 수도 있으므로 한약박의 성분이 느타리버섯으로 전이되는지 여부를 조사하였다. 한약박 70%나 100%에서 느타리버섯을 재배할 때 질병이 생겨 고사되는 버섯의 수도 적었고, 성장속도도 빨라서 수확량도 많았다. 폐면과 펠릿을 이용한 대조군과 한약박을 배지에서 재배한 처리군들의 HPLC chromatogram에 차이를 전혀 나타내지 않았고 지표물질로 사용한 glycyrrhizin이 전혀 검출되지 않았다. 즉, 한약박의 성분 자체가 그대로 전이되지는 않았고, 한약박에 함유된 성분을 영양소로 이용하여 버섯의 품질을 향상시켰다. 한약박을 70%나 100%를 사용한 배지에서 재배된 느타리버섯의 저장성이 향상되어 저장기간 동안 중량 감소가 가장 적었고, 갓과 자루의 경도의 저하도 가장 적었다. 그러나 외관의 품질을 나타내 주는 색도의 변화는 배지 종류에 영향이 적었다. 결론적으로 한약박을 70% 이상을 배지로 사용하였을 때 한약성분의 직접적인 전이는 없어서 버섯의 상품성을 저하시키지 않았으며 성장률이 빠르며 수확률도 많았고, 조직이 치밀하여 저장성도 향상시켰다. 그러므로 한약박을 느타리버섯뿐만 아니라 여러 종류의 버섯 재배에 활용하는 것이 필요하겠다.
