db/db 마우스에서 수풍순기환의 혈당강하 활성 및 기전연구 (Blood Glucose Lowering Activity and Mechanism of Supungsungihyan (SPSGH) in db/db Mouse)

  • Antidiabetic activity and mechanism of Supungsungihyan(SPSGH) were examined in db/db mice, which is a spontaneously hyperglycemic, hyperinsulinemic and obese animal model. SPSGH and acarbose were administered orally for 4 weeks. Fasting and non-fasting serum glucose, glycated hemoglobin and trig-lyceride of SPSGH treated group were all reduced when compared with those of db/db control group. At 12th week after birth, SPSGH increased an insulin secretion although statistic significance was not seen. Total activities of sucrose, maltase and lactase in SPSGH treated group were not significantly different from those in db/db control. On the other hand, sucrase and maltase activities in acarbose treated groups were increased. Effect of SPSGH on mRNA expression of glucose transporter(GLUT-4) was also examined by RT-PCR and in vitro transcription with co-amplification of rat $\beta$-actin gene as an internal standard. Muscular GLUT-4 mRNA expression in SPSGH treated group was increased significantly. These results may suggest that SPSGH lowered blood glucose ascribing to upregulation of muscular GLUT-4 mRNA expression.

Purification and Characterization of an α-D-Galactosidase from Grape Berry

  • Glycosidase activities were tested from the grape berries, Vitis labruscana B. Takasumi. Among various glycosidases, $\alpha$-D-galactosidase was found to be the most active in the flesh and other glycosidases were considerably active in the order of the following: $\alpha$-D-mannosidase>$\alpha$-D-glucosidase>$\beta$-D-glucosidase>$\beta$-D-galactosidase. In the seeds, $\alpha$-D-glucosidase activity was the highest and other glycosidases such as $\alpha$-D-galactosidase, $\beta$-D-glucosidase, and $\beta$-D-galactosidase were still significantly active. The $\alpha$-D-galactosidase in the grape flesh was purified over 83-folds through salting-out with $(NH_4)_2SO_4$ and a series of chromatographies employing Sephadex G-50, Octyl-Sepharose, Q-Sepha- rose, and Biogel P-100. The enzyme was a monomer of 45 kDs as determined through SDS-PAGE and Sephacryl S-200 chromatography. The purified enzyme showed a preference of $\alpha$-D-galactose to $\beta$-D-galactose as a substrate about 5.4 times. Sulfhydryl specific reagents such as N-ethylmaleimide and iodoacetamide significantly inhibited the enzyme activity to the extents of 48 and 52% of its initial activity, respectively. The optimumpH range of $\alpha$-D-galactosidase was around 6.5-7.0. The enzyme activity increased by 46% in the presence of 1mM $Fe^{2+}$.

잠재적인 생균제제 Bacillus subtilis CSY191에 의한 검정콩 청국장의 항산화 증진 효과 (Enhanced Antioxidant Effect of Black Soybean by Cheonggukjang with Potential Probiotic Bacillus subtilis CSY191)

  • 잠재적인 생균제제 Bacillus subtilis CSY191에 의해 제조된 국산 검정콩(서리태 및 서목태) 청국장 발효 중 ${\beta}$-glycosidase 활성, total phenolic와 isoflavone 함량 및 항산화 활성을 조사하였다. 청국장 발효 후 total phenolic 및 isoflavone-malonylgycoside와 -aglycone 함량은 증가하였고 이에 따라 ABTS 라디칼 소거활성 및 FRAP 활성은 증가하였으나 isoflavone-glycoside 함량은 감소하였다. 특히, $7^{\circ}C$에서 48시간 발효된 서리태 청국장은 서목태 원료 및 청국장보다 높은 활성을 나타내었다. 서리태 48시간 발효 후, daidzein, glycitein 및 genistein의 함량은 각각 253.0 ${\mu}g/g$ 72.5 ${\mu}g/g$ 및 114.1 ${\mu}g/g$을 나타내었다. 이 결과로부터, 검정콩 청국장의 높은 항산화 활성은 total phenolic 및 isoflavonemalonylgycoside와 -aglycone 함량 증가에 의한 것으로 추측할 수 있다.

흑마늘 와인의 발효 및 그 특성 (Fermentation of Black Garlic Wine and its Characteristics)

  • 본 연구에서 와인발효의 최적 균주를 조사하였으며 최종 선발된 Saccharomyces sp. BCNU 6006의 발효 특성과 항산화 활성을 측정하였다. 먼저 효모는 막걸리, 과일 그리고 발효 식품에서 분리하였으며, 분리된 균주는 β-glucosidase, glycosidase와 protease 효소 활성과 에탄올 및 SO2 내성에 의해 선별하였고, 추가로 황화수소와 바이오제닉 아민을 생성하는 균주를 제외하여 총 9균주가 선별되었다, 한편 선별균주들은 계통적으로 K. sevazzii와 S. cerevisiae의 subcluster에 속하는 균주로 확인되었다. 최종 선발된 Saccharomyces sp. BCNU 6006균주의 흑마늘 와인 최적 발효조건은 26 brix, 28℃, 10일로 확인되었다. 발효가 완료된 흑마늘 와인의 성분은 에탄올 15.03%, 12 brix, pH 4.01로 측정되었고, 총 polyphenol, 총 flavonoid함량, tannin 그리고 5-HMF의 함량은 각각 3.85 mg/ml, 0.51 mg/ml, 5.90 mg/ml, 0.07 mg/ml으로 확인되었다. DPPH 라디칼 소거능, ABTS 라디칼 소거능 및 환원력은 각각 90.77%, 95.20% 그리고 1.261로 측정되었으며, 흑마늘 진액보다 낮았으나, Superoxide anion 라디칼 소거능은 94.42%로 흑마늘 진액보다 높았다. 효소활성, 발효특성 그리고 항산화 효능을 기초로 판단하면 Saccharomyces sp. BCNU 6006에 의해 제조된 흑마늘 와인은 산업적으로 잠재성이 높음이 확인되었다.

  • Gel filtration과 ion exchange chromatography, reversed-phase 및 ion exchange FPLC를 이용하여 고려인삼 뿌리에서 subunit 분자량 약 25 kDa의 주요단백질(이하 GMP)을 분리하였다. PAS 염색을 통하여 GMP는 carbohydrate moiety를 갖는 당단백질일 가능성을 보여주었고, 이는 glycosidase 처리후 protein band shift 실험으로 확인되었다. GMP는 native polyacrylamide 전기영동 및 gel permeation FPLC를 통해 native form은 약 63 kDa의 분자량을 갖는 dimer로 판단된다. GMP의 생리활성 측정결과, inflammation mediator의 기능탐색에 쓰이는 anticomplementary activity를 보여주었다.

Influence of Gibberellic Acid on α-D-Galactosidase Activity in the Grape Berry

  • Glycosidase activities in the grape flesh (Marguerite) were assayed, and the order of activity was marked as follows: ${\alpha}$-D-galactosidase>${\alpha}$-D-mannosidase>${\alpha}$-D-glucosidase>${\beta}$-D-galactosidase>${\beta}$-D-glucosidase. Of these glycosidases, ${\alpha}$- and ${\beta}$-D-galactosidases were prominently expressed by the treatment of gibberellic acid, resulting in 56 and 238% increase of activity, respectively. Most of ${\alpha}$-D-galactosidase was found in the flesh texture, and the activity increase by gibberellic acid occurred mostly in this tissue. The increase in ${\alpha}$-D-galactosidase activity was dependent on the concentration of gibberellic acid treated, showing a positive correlation. Gibberellic acid affected the content of total protein in the grape flesh, 49% increase by 75 ppm treatment. Above this concentration, higher gibberellic acid level did not influence the protein expression. Specific activity of the ${\alpha}$-D-galactosidase still increased, showing 24% increase in activity. Grape flesh subjected by gibberellic acid (100 ppm) resulted in the increased activity against a natural substrate, stachyose, showing 55% increase in activity from the grapes treated with 100 ppm of gibberellic acid. Other natural substrates, such as melibiose and raffinose, were also considerably hydrolyzed, and the extent was similar to that of stachyose hydrolysis. During postharvest storage, ${\alpha}$-D-galactosidase activity in the grape flesh increased by 51% after 20 days and then declined slowly.

Enzymatic Activities of Allergen Extracts from Three Species of Dust Mites and Cockroaches Commonly Found in Korean Home

  • Allergen extracts from dust mites and cockroaches commonly found in Korean homes were used to evaluate their enzymatic activity as they are believed to influence allergenicity. Allergen extracts were prepared from 3 dust mite species (Dermatophagoides farinae, D. pteronyssinus, and Tyrophagus putrescentiae) and 3 cockroach species (Blattella germanica, Periplaneta americana, and P. fuliginosa) maintained in the Korea National Arthropods of Medical Importance Resource Bank. Proteins were extracted in PBS after homogenization using liquid nitrogen. The activities of various enzymes were investigated using the API Zym system. No significant difference in phosphatase, lipase, or glycosidase activity was observed among the 6 allergen extracts, but much difference was observed in protease activity. Protease activity was assessed in more detail by gelatin zymography and the EnzChek assay. Extract from T. putrescentiae showed the highest protease activity, followed by those of the cockroach extracts. Extracts from D. farinae and D. pteronyssinus showed only weak protease activity. Gelatinolytic activity was detected mainly in a 30-kDa protein in D. farinae, a 28-kDa protein in D. pteronyssinus, a > 26-kDa protein in T. putrescentiae, a > 20-kDa protein in B. germanica, and a > 23-kDa protein in P. americana and P. fuliginosa. The information on various enzymatic activities obtained in this study may be useful for future studies. In particular, the strong protease activity found in cockroach extracts could contribute to sensitization to cockroach allergens, which is known to be associated with the development of asthma.

  • The synthesis of cis- and trans-1,6-dihydroxypyrrolizidine starting from trans-4-hydroxy-L-proline and their evaluation as glycosidase inhibitors are reported. The cis-isomer was found to be a potent inhibitor against .alpha.-glucosidase and showed weak inhibitory effect against other glycosidases. The trans-isomer exhibited weak inhibitions of b-glucosidase and amylo-glucosidase and poor inhibition of other glycosidases.

db/db 마우스에서 상백피의 혈당강하효과 (Anti-hyperglycemic Effect of Cortex Mori radicis in db/db Mice)

  • 8주령의 C57BL/KsJ db/db마우스에 상백피 냉침추출물을 투여하면서 5주간 사육하여 식이섭취량, 체중변화, 혈중 포도당 농도를 관찰하고, 소장내 ${\alpha}-glycosidase$ 활성변화와 혈중 당화헤모글로빈 농도, 중성지방 농도, 인슐린 농도를 측정하였다. 상백피추출물 500 mg/kg을 공급하는 Rad 500군과 1000 mg/kg을 공급하는 Rad 1000군, db/db 대조군과 lean군으로 나누고 대조약물로는 acarbose 5 mg/kg을 투여하였다. Rad 1000군은 식이섭취량에서 db/db 대조군보다 낮은 수준이었으며 그에 따라 체중증가량도 유의적으로 낮았다. 또한 공복시 혈중포도당 농도는 Rad 500, Rad 1000, acarbose군 모두 db/db 대조군보다 낮았다. 혈중 당화헤모글로빈 농도와 중성지방 함량 측정결과는 농도의존적으로 시료투여군이 db/db대조군보다 낮게 측정되었다. 혈중 인슐린 농도 측정결과 lean군을 제외한 나머지군간에 유의적인 차이는 없었고 Rad 500군에서 약간 높은 수준이 관찰되었다. 소장을 proximal, middle, distal 세부위로 나누어 각각에 대해 maltase, sucrase, lactase의 활성을 측정해본 결과 db/db군은 lean군에 비해 세효소의 활성이 매우 높은 활성을 나타내었으며, 시료투여군은 sucrase의 경우 proximal, middle 부위는 db/db군 보다 낮고 distal 부위는 활성이 높았다. Maltase, lactase의 활성은 모든부위에서 시료투여군이 db/db군 보다 낮아 lean군 수준이었다. 이상의 결과들로 미루어 상백피는 db/db마우스에서 식이섭취량을 줄여주어 체중증가를 억제시키고 소장내 glycosidase활성을 전반적으로 낮춰 혈중포도당 농도를 낮춰주고 혈중 당화헤모글로빈 농도와 중성지방 농도를 낮춰주는 등의 항당뇨효과가 있음을 알 수 있었다.

  • 아가리쿠스 버섯(동결 건조, 온풍 건조)에서 추출한 ${\beta}-glucan$을 7주령의 db/db마우스에 5주간 음용수로 제공하고 식이섭취량 및 체중의 변화를 관찰하고 혈액내의 포도당, 인슐린, 당화 헤모글로빈, 소장의 glycosidase의 활성을 측정하고 중성지방농도, 혈중 지질조성을 측정하였으며 결과는 다음과 같다. db/db 마우스에서 Agari.F군과 Agari.H군은 식이섭취량에서 Control군보다 유의적으로 낮았고, 그에 따라 체중증가량도 낮았으며, 혈중 인슐린 농도와 당화헤모글로빈 농도는 Control군에 비해 Agari.F군, Agari.H군, Acarbose군이 유의적으로 낮은 수준이었다. 소장의 maltase, lactase, sucrase의 활성은 Control군에서 Lean군에 비해 세 효소의 활성이 매우 높게 관찰되었고, 시료투여군인 Agari.F군, Agari.H군은 maltase의 경우 proximal, middle 부위는 대조군보다 낮았고 distal 부위에서는 Agari.F군의 활성이 가장 높았다. Sucrase, lactase의 활성은 모든 부위에서 시료투여군이 Control군보다 낮은 수준이었다. 혈중 중성지방 농도는 Control군에 비해 Agari.F군, Agari.H군, Acarbose군은 유의적으로 낮은 수준이었다. 혈중 total cholesterol의 농도의 경우 Control군과 Agari.H군은 유의적인 차이가 없었지만 Agari.F군과 Acarbose군은 유의적으로 낮았다. 혈중 VLDL cholesterol의 경우는 Control군에 비해 Agari.F군, Agari.H군은 유의적으로 낮았고 Acarbose군은 유의적인 차이가 없었다. 혈중 LDL cholesterol의 농도는 Control군이 유의적으로 높았으며, Acarbose군이 가장 낮았다. 혈중 HDL cholesterol의 농도는 Agari.H군이 유의적으로 가장 높았고, Control군이 가장 낮았다.
