Studies on Lipids in Fresh-Water Fishes 3. Distribution of Lipid Components in Various Tissues of Eel, Anguilla japonica (담수어의 지질에 관한 연구 3. 뱀장어(Anguilla japonica)의 부위별 지질성분의 분포)

  • The crude lipid contents in meat, skin and viscera of eel were 24.94, 20.00 and 14.96, respectively, and higher than the other fresh water fishes. The free lipid was consisted in order of neutral lipid(86.71~94.94), phospholipid(4.13~12.74) and glycolipid(0.63~1.22), and the bound lipid in order of phospholipid(51.74~75.21), neutral lipid(14.41~36.82) and glycolipid(5.12~7.51). The neutral lipid in free lipid was mainly consisted of TG(68.51~95.22), and that in bound lipid TG(36.54~39.94), ES & HC(17.20~18.01), and MG(15.81~18.11). The phospholipid in free lipid was mainly consisted of PC (32.40~56.08) and PE(18.71~31.09), while that in bound lipid PC (<42.51~67.90) and PS (23.37~34.49). The contents of major fatty acid of polar lipid in free and bound lipids were $C_{16:0}$(16.58, 14.10, $C_{18:0}$(12.44, 7.36), $C_{18:1}$(16.01, 15.78), $C_{20:4}$(10.01, 6.11) $C_{20:5}$(4.20, 8.67) and $C_{22:6}$(10.26, 18.32) and those of nonpolar lipid in free and bound lipids were $C_{16:0}$(22.31, 18.61), $C_{18:0}$(4.36, 5.69, $C_{18:1}$(36.30, 27.20) $C_{20:5}$(2.88, 2.38) $C_{22:5}$(6.38, 5.11) and $C_{22:6}$(1.20, 5.11). The total essential fatty acid(TEFA) content of polar lipid in meat was ranged from 17.33 to 22.88, and 2.0~3.0 times higher than that of nonpolar lipid, and the TEFA content of bound lipid in meat was ranged from 11.54 to 22.88, and 1.5~2.0 times higher than that of free lipid. The w3 highly unsaturated fatty acid(w3-HUFA) content of polar lipid in meat was ranged from 8.63 to 42.85, and higer than 11.48 to 14.42 of nonpolar lipid. Also the w3-HUFA content of bound lipid in meat was ranged from 13.35 to 42.85, and higher than 11.48 to 18.63 of free lipid. The w3-HUFA content in meat was higher than that in skin and/or viscera

Studios on Lipids in Fresh-Water Fishes 5. Distribution of Lipid Components in Various Tissues of Carp, Cyprynus carpio (담수어의 지질에 관한 연구 5. 잉어(Cyprynus carpio)의 부위별 지질성분의 분포)

  • Distribution of lipid components in the tissue of meat, skin and viscera from carp(Cyprynus carpio) was analyzed using the techniques of column chromatography, thin layer chromatography and gas liquid chromatography according to the previous report(Choi, et al., 1984). Lipid content was varied by the portion such as $3.88\%$ in meat (free lipid, $2.47\%$ ; bound lipid, $1.41\%$), $8.02\%$ in skin(free lipid, $5.65\%$ ; bound lipid, $2.37\%$) and $6.18\%$ in viscera (free lipid, $3.54\%$ ; bound lipid, $2.64\%$). In the all portions of the body, free lipid was composed of $68\%\;to\;92\%$ in neutral lipid, $3\%\;to\;6\%$ in glycolipid and $4\%\;to\;18\%$ in phospholipid whereas bound lipid was composed of $8\%\;to\;20\%$ in neutral lipid, $2\%\;to\;7\%$ in glycolipid and $47\%\;to\;62\%$ in phospholipid. The free lipids of the tissues on the each portion were mostly represented by triglycerides and some diglycerides, but free lipids in viscera contained considerable amounts of free fatty acids. The bound lipids, on the other hand, commonly comprised appreciable amounts of esterified sterol and hydrocarbon, and triglycerides. The phospholipid was mainly consisted of phosphatidyl choline, phosphatidyl ethanolamine and phosphatidyl serine in the both free and bound lipids, and much more phosphatidyl choline in the bound lipid. The predominant fatty acids of free and bound lipids were $C_{16:0},\;C_{18:0},\;C_{20:4},\;C_{22:6}\;and\;C_{18:2}$ acids in polar lipids, and $C_{16:0},\;C_{16:1},\;C_{18:0},\;C_{18:1}\;and\;C_{18:2}$ acids in non-polar lipids, whereas those of neutral lipids were $C_{14:0}(2.54{\sim}6.98\%),\;C_{16:0}(11.20{\sim}21.13\%)$ and $C_{18:0}(1.58{\sim}12.76\%)$ of saturated acids, $C_{16:1}(7.06{\sim}20.70\%),\;C_{18:1}(21.68{\sim}30.50\%)$ and $C_{20:1}(1.76{\sim}6.27\%)$ of monoenoic acids, and $C_{18:2}(4.50{\sim}6.89\%),\;C_{20:4}(1.52{\sim}4.29\%)$ and $C_{22:6}(0.73{\sim}6.62\%)$, respectively. In conclusion, the fatty acid compositions revealed apparent differences between the free lipid and bound lipids in the tissues of body.

  • Background : Anthracofibrosis, a descriptive term for multiple black pigmentation with fibrosis on bronchoscopic examination, has a close relationship with active tuberculosis (TB). However, TB activity is determined in the later stage by the TB culture results in some cases of anthracofibrosis. Therefore, it is necessary to identify early markers of TB activity in anthracofibrosis. There have been several reports investigating the serum levels of IL-2 $sR{\alpha}$, IFN-${\gamma}$ and TBGL antibody for the evaluation of TB activity. In the present study, we tried to measure the above mentioned serologic markers for the evaluation of TB activity in patients with anthracofibrosis. Methods : Anthracofibrosis was defined when there was deep pigmentation (in more than two lobar bronchi) and fibrotic stenosis of the bronchi on bronchoscopic examination. The serum of patients with anthracofibrosis was collected and stored under refrigeration before the start of anti-TB medication. The serum of healthy volunteers (N=16), patients with active TB prior to (N=22), and after (N=13), 6 month-medication was also collected and stored. Serum IL-2 $sR{\alpha}$, IFN-${\gamma}$ were measured with ELISA kit (R&D system, USA) and serum TBGL antibody was measured with TBGL EIA kit (Kyowa Inc, Japan). Results : Serum levels of IL-2 $sR{\alpha}$ in healthy volunteers, active TB patients before and after medication, and patients with anthracofibrosis were $640{\pm}174$, $1,611{\pm}2,423$, $953{\pm}562$, and $863{\pm}401$ pg/ml, respectively. The Serum IFN-${\gamma}$ levels were 0, $8.16{\pm}17.34$, $0.70{\pm}2.53$, and $2.33{\pm}6.67$ pg/ml, and TBGL antibody levels were $0.83{\pm}0.80$, $5.91{\pm}6.71$, $6.86{\pm}6.85$, and $3.22{\pm}2.59$ U/ml, respectively. The serum level of TBGL antibody was lower than of other groups (p<0.05). There was no significant difference of serum IL-2 $sR{\alpha}$ and IFN-${\gamma}$ levels among the four groups. Conclusion : The serum levels of IL-2 $sR{\alpha}$, IFN-${\gamma}$ and TBGL antibody were not useful in the evaluation of TB activity in patients with anthracofibrosis. More useful ways need to be developed for the differentiation of active TB in patients with anthracofibrosis.

Studies on the Lipid Composition in Three Species of Shellfish (3종(種) 패류(貝類)의 지질조성(脂質組成)에 관한 연구(硏究))

  • In this study, the lipid components of three species of shellfish included oyster(Crassostrea gigas), top shell(Turbo cornutus) representing salt water shellfish and corb shell(Corbicula fluminea producta) representing flesh water shellfish were analysed and nutriontional significances were discussed. Analysed the total lipid composition, and the fatty acid and sterol composition of total lipid were determined. The lipid was fractionated into three lipid classes neutral, glyco and phospholipid by column chromatography. The fatty acid composition of each lipid class and sterols were determined by gas liquid chromatography. The lipid components of total lipid and neutral lipid were estimated by thin layer chromatography and TLC scanner. The results were as follows: Total lipid contents of shellfish were 1.8% in oyster, 0.4% in top shell and 4.0% in corb shell. The contents of total fatty acid in total lipid were 80.7, 71.2 and 73.2%; and the contents of unsaponifiable matters were 15.4, 18.1 and 23.1% respectively. Total lipids were mainly composed of triglycerides, polar lipid-pigments and sterols as major component, and hydrocarbon-esterified sterols were determined in each sample. The major fatty acids in total lipid were palmitic(37.0%), eicosapentaenoic(13.5%) and linoleic acid(11.2%) in oyster, Octadecatetraenoic(15.8%), palmitic(11.2%), oleic(8.6%) and linoleic acid(8.1%) in top shell, but palmitic(34.0%), linoleic(12.3%) and paimitoleic acid(9.8%) in corb shell. Particularly, the contents of eicosapentaenoic acid of oyster and top shell were higher than those of corb shell. Sterol composition from three species of shellfish were mainly consisted of cholesterol (42.7~64.0%), brassicasterol(15.6~24.7%) and 24-methylenecholesterol (4.7~21.9%). But sitosterol (5.3%) was detected only in oyster and 22-dehydrocholesterol(12.9%) was only in top shell. The contents of fractionated neutral lipid was commonly higher than that of polar lipid in each sample. Glycolipid and phospholipid in polar lipid showed similar in quantity. The neutral lipids were composed of triglycerides(33.0~36.7%), free sterols(25.7~31.2%), esterified sterol(12.4~23.7%) and free fatty acids(5.1~11.7%). The contents of triglycerides and free sterols were higher than those of free fatty acids and esterified sterols. The major fatty acids in neutral lipid were palmitic(28.4~26.4%) eicosapentaenoic(18.6~21.9%) and linoleic acid(9.0~5.4%) in oyster and corb shell but octadecatetraenoic(14.5%), eicosapentaenoic (13.5%) and palmitic acid(12.3%) in top shell. The major fatty acids in glycolipid were eicosenoic(10.2%), palmitic(12.1%) and linolenic acid (10.2%) in oyster, Eicosenoic(26.0%), octadecatetraenoic(14.6.%) and eicosadienoic acid(12.9%) in top shell. But eicosadienoic(21.4%) stearic(14.6%), octadecatetraenoic(8.5%) and eicosenoic acid(8.5%) in corb shell. The major fatty acids in phospholipid were myristic(16.0%), stearic(10.6%), eicosenoic(10.5%) and palmitic acid(10.3%) in oyster, Oleic(22.2%), stearic(20.7%) and linolenic acid (11.8%) in top shell but eicosapentaenoic(25.1%), myristic(8.7%) and arachidonic acid(8.3%) in corb shell.

Nutrient Components in the Siphon of the Surf Clam Tresus keenae

  • We evaluated the nutritional composition of the siphon of the surf clam Tresus keenae in regard to the presence of nitrogenous [amino acids, nucleotides and their related compounds, total creatinine, betaine, trimethylamine oxide (TMAO), and trimethylamine (TMA)] and non­nitrogenous compounds (sugars and organic acids), lipid fatty-acid composition, and occurrence of minerals. The content of total free amino acids was 660.27 $\pm$ 7.94 mg/100 g, and the predominant amino acids were arginine, alanine, sarcosine, glycine, and glutamic acid. These amino acids accounted for $71\;\%$ of the total free amino acids. Among the nucleotides and their related compounds, inosine was the major component and comprised 40.38 $\pm$ 0.02 mg/100 g. Free amino acids were the largest contributor to total extracted nitrogen, comprising $49.94\%$, followed by total creatinine, betaine, nucleotides, and ammonia; the contribution of TMAO and TMA was small. For the non-nitrogenous compounds, malic acid, propionic acid, and succinic acid comprised the major portion of the ten kinds of organic acids detected, and the sugars found were glucose, maltose, and arabinose, which were estimated to be $147.0\pm7.15,\;34.45\pm1.09,\;and\;1.21\pm0.02\;mg/100\;g,$ respectively. The predominant minerals were Na and K, which comprised $11.43\pm1.06\;and\;9.46\pm1.02\;mg/100\;g,$ respectively. The major fatty acids were C22:6, C20:5, C23:0, C18:3, and C16:0 in the lipid fractions. The 23:0 level of glycolipid (GL) was the highest of any other lipid fraction. The amount of total polyunsaturated fatty acids (PUFA) in the lipid fractions was higher, ranging from $58.22\%\;in\;GL\;to\;77.1\%$ in phospholipid (PL), compared to the saturated and monounsaturated fatty acids. Of the n-3 fatty acids, C20:5 and C22:6 contributed $35.30-64.44\%$ of PUFA in the lipid fractions. The ratios of n-3 to n-6 PUFA in total lipid (TL), neutral lipid (NL), PL, and GL were 4.35, 4.26, 6.69, and 2.04, respectively.

Fatty Acid Composition of Different tissues of Spodoptera exigua Larvae and a Role of Cellular Phospholipase A2 (파밤나방 유충의 조직별 지방산 구성과 세포성 인지질분해효소의 역할)

  • Eicosanoids are a group of C20 oxygenated polyunsaturated fatty acids (PUFAs). To monitor biosynthetic precursors of these PUFAs, this study extracted fatty acids from different tissues of the beet armyworm, Spodoptera exigua, and assessed their compositions using GC/MS. Fifth instar larvae were dissected to isolate different tissues of gut, fat body, hemocytes, and integument. From each tissue, total lipids were extracted and fractionated into neutral lipid (NL), glycolipid (GL), and phospholipid (PL). Most tissues contained palmitic acid (16:0), stearic acid (18:0), oleic acid (18:1), linoleic acid (18:2), and linolenic acid (18:3). However, their compositions were different among tissues and lipid types. Fat body and hemocytes possessed other type of fatty acids such as myristic acid (14:0) and three unknown fatty acids. Among lipid types, PL contained relatively high levels of linolenic acid than NL and GL, while it had lower saturated fatty acids. Total unsaturated fatty acid composition was varied among tissues and lipid types. PL was rich in unsaturated fatty acids in fat body, gut, and hemocytes. There was a significant influence of calcium-independent phospholipase $A_2$ ($iPLA_2$) on maintaining fatty acid composition because RNA interference of $iPLA_2$ expression significantly modified fatty acid compositions in NL and PL. However, this study did not detect arachidonic acid, a main eicosanoid biosynthesis precursor, in all tissues. This suggests an alternative biosynthesis of eicosanoids in insects, which is distinct from the biosynthetic pathway of mammals.

The Comparison of Chemical Compounds in Korean and Chinese Sesame Seeds" (한국산 및 중국산 참깨의 화학성분 비교)

  • Sesame samples used in this study were Korean variety cultivated in Chochiwon City of Korea (KvKc), Chinese variety cultivated in Jilin Province of China(CvCc), Chinese variety cultivated in Chochiwon city of Korea(CvKc) and Korean variety cultivated in Jilin Province of China(KvCc). The extraction yields of sesame oils from KvKc, CvCc, CvKc and KvCc were 47.8%, 48.1%, 48.6% and 49.3% respectively. The color value (L value) of sesame oil from KvKc was lower than that of CvCc and KvKc sesame oil showed dark brown color. The composition of neutral lipid, glycolipid and phospholipid of free lipids from KvKc and U sesame oils were 93.1%, 6.6%, 0.2% and 94.7%, 4.6%, 0.8% respectively. Major fatty acids of KvKc and CvCc sesame were usaturated fatty acids, such as oleic and linoleic acid. Total composition of the two major fatty acids of KvKc was 84.6% which was almost same with that of CvCc. However the composition of oleic and linoleic acid of KvKc were 47.1% and 37.5%, while the composition of CvCc were 40.5% and 44.3%. When cross-cultivated, the composition of oleic and linoleic acid of CvKc were 41.0% and 43.5%, while the composition of KvCc were 42.0% and 43.7%. The contents of total amino acids and essential amino acids of KvKc were 713 mg% and 309 mg%, which were much higher than those of CvCc. When cross-cultivated, the content of total amino acids of KvCc was 44% lower than that of KvKc while the content of total amino acids of CvKc was 36% higher than that of CvCc.

A Study on the Composition of Polar Lipid in Adlay (율무의 극성지질 조성에 관한 연구)

  • The compositional characteristics of polar lipids in the brown and polished adlay were studied. Total lipids of brown and polished adlay were extracted, purified and fractionated into neutral and polar lipids by silicic acid column chromatography. The glycolipid(GL) and phospholipid(PL) of polar lipids were seperated by thin layer chromatography and quantitated by scanner. The fatty acid composition in polar lipids were determined by gas chromatographr. The contents of GL and PL in brown adlay were 5.67% and 1.83%, and their contents on polished adlay were 5.49% and 1.78%, respectively. Of the GL in the brown and polished adlay, monogalactosyl diglycerides+esterified steryl glycosides and digalactosyl diglycerides were the major components, but cerebrosides+steryl glycosides were also found as the minor component of GLs. Of the PLs, phosphatidyl choline and phosphatidyl ethanolamine were the major components. Phosphatidyl inositol, lysophosphatidyl choline and phosphatidyl serine were also present in the PLs. The major fatty acids of GLs were linoleic, oleic and palmitic acids. The fatty acid composition in the PLs were similar to the pattern in the GLs, but PLs contained the lower percentage of linoleic acids than the GLs.

Comparison of Lipid Components in Wild and Cultured Bastard (천연 및 양식산 넙치의 지방질성분)

  • The lipid components of cultured bastard(Paralichthys olivaceus) were analyzed and compared with those of wild one and flounder(Pleuronichthys cornutus). Total lipid(TL) content of dorsal muscle in wild and cultured bastard, flounder was 2.0%, 1.6%, and 1.9%, respectively. Wild bastard and flounder were higher in neutral lipid(NL) and glycolipid(GL) contents, while lower in phospholipid(PL) content than those of cultured one. The NL was mainly consisted of triglyceride(70.7-72.6%), and PL was mainly occupied by phosphatidylcholine(53.6-58.3%), phosphatidyl-ethanolamine(25.9-29.5%). In fatty acid composition of TL, cultured bastard showed higher content in polyenes such as docosahexaenoic acid. and lower in monoenes such as palmitoleic, oleic acid than those of wild one. Flounder was higher in Percentage of arachidonic, eicosapentaenoic acid compard with bastard. The major fatty acids of these fishes were palmitic, docosahexaenoic, oleic, eicosapentaenoic and palmitoleic acid in order. These fatty acid composition of NL, GL and PL fractions were not significantly different among wild and cultured bastard, flounder.

  • Daenjang, a traditional Korean fermented soybean paste, was prepared by the fermentation with Aspergillus oryzae for 3 days, and then ripened with salt addition(13%) for 45 days. The changes of composition and fraction pattern of lipid in Daenjang during ripening period were studied. The total lipid of initial Daenjang(0 day ripening) consisted of 91.8% of neutral lipid, 7.1% of phospholipid and 1.1% of glycolipid, and minor changes of the composition were observed during the ripening. The contents of free fatty acid and esterified sterol in nonpolar lipid fraction were increased, however, no significant changes in polar lipid fraction were observed from the Daenjang in ripening. Linoleic, oleic, palmitic acids were the major fatty acyl moiety in the fractions of total lipid, triglyceride and free fatty acid, respectively. No significant changes in fatty acid compositions of the fractions were noted, although the saturated fatty acyl moiety of the fraction were increased a little during Daenjang ripening.
