Insecticidal and Repellent Activities of Crude Saponin from the Starfish Asterias Amurensis

  • Crude saponin, extracted from the starfish Asterias amurensis, was assessed for its capacity as a biological pesticide. As part of this analysis, its insecticidal and repellent activities, in addition to its acute and chronic toxicities were tested. In comparison with the control group, insecticidal activity of saponin against tobacco cutworm, Spodoptera litura, in kale, Brassica loeracea was low at 36.4%. Repellent activities of the extracted saponin against green peach aphid, Myzus persicae, and S. litura, on soybean leaf, Glycine max and kale were 65.6% and 35.0% at $1^{st}$ day, and 54.5% and 30.0% at $3^{rd}$ day, respectively. Acute and chronic toxicity analysis was carried out using acute immobilizatioin test and reproduction impairment test, respectively. The saponin had 48 h-$EC_{50}$ of $65.21{\mu}g/mL$. Twenty-one day accumulative reproduction after treatment was lower in Daphnia magna at $7{\mu}g/mL$ saponin (78 youngs), compared with the control group (129 youngs). These results indicate that the extracted saponin exhibited some toxicity and has potential as a repellent against insects.

Effect of Intercropped Corn and Soybean Silage on Nutritive Values, in vitro Ruminal Fermentation, and Milk Production of Holstein Dairy Cows

  • This study was conducted to examine the effect of corn (Zea mays L.) - soybean (Glycine max L.) silage prepared by intercropping method on the nutritive value of the silage, in vitro rumen fermentation characteristics, dry matter degradability, as well as milk yield and milk composition of dairy cows. In a couple of experiments intercropped corn-soybean silage (CSBS) was compared with corn silage (CS) and/or Italian ryegrass hay (IRG). Numerically, CSBS had higher crude protein, ether extract, and lactic acid contents compared to CS. In vitro rumen fermentation analysis demonstrated that up to a 24-h incubation period, both CS and CSBS showed higher total gas production, ammonia N concentration, and dry matter degradability compared to IRG (p<0.05). The investigation on animals was conducted in a commercial dairy farm located in Gyeongju, South Korea, employing 42 Holstein cows that were divided into 2 group treatments: CS and CSBS in a completely randomized design. Although no significant difference was observed in milk yield, animals fed on CSBS showed significantly higher milk protein (p<0.05) and milk fat content (p<0.01), compared to animals fed on CS. Taken together, our findings indicate that corn-soybean silage that is cultivated, harvested, and prepared through intercropping can improve the protein content of the silage, and can also enhance in vitro rumen fermentation, dry matter degradability, and performance of dairy cattle.

Beneficial Effects of Phyto-Extract Complex (CME) on Degenerative Arthritis (식물추출복합물(CME)의 퇴행성관절염 개선효과)

  • Objectives : Degenerative arthritis arises from several physiological factors. The purpose of this study is to investigate the beneficial effects of Phyto-extract Complex (CME) on degenerative arthritis. Methods : CME is composed of extracts of mulberry (Morus alba L.) fruit, mulberry leaves and black beans (Glycine max (L.) Merr.). To measure the toxicity of CME, we performed the single-dose toxicity study. For the evaluation of its effects on degenerative arthritis, we examined the inhibition of cyclooxygenase-2 (COX-2) activity, using in vitro enzyme activity assay, the reduction of protein expression of COX-2, 5-lipoxygenase (5-LO), and inducible nitric oxide synthase (iNOS) in RAW264.7 cells which were stimulated by lipopolysaccharide (LPS). We also examined the serum level of prostaglandins (PGs) and injury of the knee joint cartilage, using animal model of degenerative arthritis induced by mono-sodium iodoacetate (MIA). Results : CME did not have any toxicity in single-dose toxicity study. The CME inhibited the activity of COX-2 and could reduce the protein expression of COX-2, 5-LO and iNOS in RAW264.7 cells. The CME also reduced the serum level of PGs and prevented from the cartilage injury of knee joint in animal model of degenerative arthritis induced by MIA. Conclusions : Taken altogether, the CME could be useful for the improvement of degenerative arthritis through its various anti-inflammatory activities and prevention from the cartilage injury of knee joint.

Effect of Grain Size and Drying Temperature on Drying Characteristics of Soybean (Glycine max) Using Hot Air Drying (열풍건조 시의 건조 온도와 입경에 따른 콩(Glycine max)의 건조 특성)

  • The effects of drying temperature on drying characteristics of soybeans with different grain sizes [6.0 (S), 7.5 (M), and 9.0 mm (L) (${\pm}0.2$)] with 25.0% (${\pm}0.8$) initial moisture content were studied. Drying temperatures varied at 25, 35, and $45^{\circ}C$, with a constant air velocity (13.2 m/s). Thin-layer drying models were applied to describe the drying process of soybeans. The Midilli-Kucuk model showed the best fit ($R^2$ >0.99). Based on the model parameters, drying time to achieve the target moisture content (10%) was successfully estimated. Drying time was strongly dependent on the size of soybeans and the drying temperature. The effective moisture diffusivity ($D_{eff}$) was estimated by the diffusion model based on Fick's second law. $D_{eff}$ values increased as grain size and drying temperature increased due to the combined effect of high temperatures and high drying rates, which promote compact tissue. Deff values of S, M, and L estimated were in the range of $0.83{\times}10^{-10}$ to $1.51{\times}10^{-10}m^2/s$, $1.17{\times}10^{-10}$ to $2.17{\times}10^{-10}m^2/s$, and $1.53{\times}10^{-10}$ to $2.95{\times}10^{-10}m^2/s$, respectively, whereas activation energy ($E_a$) based on drying temperature showed no significant differences in the size of soybeans.

Effect of Phosphorus Stress on Photosynthesis and Nitrogen Fixation of Soybean Plant under $CO_2$ Enrichment (대기 $CO_2$ 상승시 인산공급이 식물체의 광합성 및 질소고정에 미치는 영향)

  • The objective of this study was to examine the effect of phosphorus deficiency on nitrogen fixation and photosynthesis of nitrogen fixing soybean plant under $CO_2$ enrichment condition. The soybean plants(Glycine max [L.] Merr.) inoculated with Bradyrhizobium japonicum MN 110 were grown with P-stressed(0.05 mM-P) and control(1 mM-P) treatment under control$(400\;{\mu}l/L\;CO_2)$ and enrichment$(800\;{\mu}l/L\;CO_2)$ enviromental condition in the phytotron equipped with high density lamp$(1000\;{\mu}Em^{-2}S^{-1})$ and $28/22^{\circ}C$ temperature cycle for 35 days after transplanting(DAT). At 35 DAT, phosphorus deficiency decreased total dry mass by 64% in $CO_2$ enrichment condition, and 51% in control $CO_2$ condition. Total leaf area was reduced significantly by phosphorus deficiency in control and enriched $CO_2$ condition but specific leaf weight was increased by P deficiency. Phosphorus deficiency significantly reduced photosynthetic rate(carbon exchange rate) and internal $CO_2$ concentration in leaf in both $CO_2$ treatments, but the degree of stress was more severe under $CO_2$ enrichment condition than under control $CO_2$ environmental condition. In phosphorus sufficient plants, $CO_2$ enrichment increased nodule fresh weight and total nitrogenase activity(acetylene reduction) of nodule by 30% and 41% respectively, but specific nitrogenase activity of nodule and nodule fresh weight was not affected by $CO_2$ enrichment in phosphorus deficient plant at 35 DAT. Total nitrogen concentrations in stem, root and nodule tissue were significantly higher in phosphorus sufficient plant grown under $CO_2$ enrichment, but nitrogen concentration in leaf was reduced by 30% under $CO_2$ enrichment. These results indicate that increasing $CO_2$ concentration does not affect plant growth under phosphorus deficient condition and phosphorus stress might inhibit carbohydrate utilization in whole plant and that $CO_2$ enrichment could not increase nodule formation and functioning under phosphorus deficient conditions and phosphorus has more important roles in nodule growth and functioning under $CO_2$ enrichment environments than under ambient condition.

Identification of Quantitative Trait Loci Associated with Leaf Length. Width and Length/width Ratio in Two Recombinant Inbred Lines of Soybean (Glycine max L.) (두 집단의 재조합 근친교잡 계통 (RIL) 콩에서 엽장과 엽폭 및 장폭비와 관련된 양적헝질 유전자좌 분석)

  • The increasing apparent photosynthetic rate per leaf area may improve seed yield in soybean. Leaf area, length and width are related to the photosynthetic capability of the plant. In this study, two populations derived from the cross of Keunolkong, Shinpaldalkong and Iksanl0 were evaluated with simple sequence repeat (SSR) markers to identify length, width and length/width ratio of leaf. Leaf length/width ratio were significantly negative correlation with leaf width in K/S and K/I populations. In the K/S population, two minor QTLs for leaf length (LL) were found on LG Dlb+W and 1. Two QTLs on LG J and L were related to LL in K/I population. Two and three minor QTLs were identified in leaf width with total phenotypic variation of 13% and 18.04 in K/S and K/I populations, respectively. The leaf length/width ratio, two QTLs on LG I and L, and three QTLs on LG Cl, E and L were related to K/S and K/I populations, respectively. Thus it is assumed that the leaf traits are very much dependent on the genotype used and different breeding approach should be considered for the selection of favorite leaf traits in soybean breeding programs.

Response of Growth and photosynthesis to NaCl stress in Soybean(Glysine max L.) Seedlings (NaCl stress에 의한 몇가지 콩 품종들의 생육과 광합성 반응)

  • This study was conducted in to elucidate the changes of growth characteristics and photosynthesis in three soybean (Glycine max L. cv. Danwonkong, Hwangkeumkong and Kwangankong) 30 day old seedlings to 100mM NaCl concentration containing 1/2 Hoagland's nutrient solution in sand culture. The main stem height and number of main stem node were decreased. thus, leaf area and dry matter were decreased with 100mM NaCl. Growth reduction was less little in Hwangkeumkong than other cultivars. The stem growth rate was affected less than other parts as root or leaf, by NaCl treatment. The specific leaf area (SLA), shoot : root ratio and leaf : root ratio decreased with NaCl solution except for those of Hwangkeumkong. There is no general tendency in leaf thickness by leaf position of three cultivars. The chlorophyll content (SPAD) of the primary and 2nd leaf slightly decreased under NaCl solution but rapidly increased in non-NaCl solution at 15 days after treatments. The photosynthesis, stomatal conductance and transpiration of 2nd leaf positions reduced by NaCl and there were a sigificant correlation between photosynthesis and stomatal conductance or transpiration.

Effects of Pre-cropping with Rice (Oryza sativa L.) Alternative Crops on Grain Yield and Flour Quality of Winter Wheat (Triticum aestivum L. 'Jokyung') on the Paddy Fields (논에서 벼 대체작물의 전작 재배가 조경밀의 곡물 생산성과 밀가루의 품질에 미치는 영향)

  • The grain yield and flour quality of winter wheat (Triticum aestivum L. 'Jokyung') were investigated in the paddy fields in which the double-cropping of wheat linked to rice (Oryza sativa L.) and its alternative crops [black soybean (Glycine max (L.) Merr.), sesame (Sesamum indicum L.), and perilla (Perilla frutescens (L.) Britton)] was applied. In the soils in which black soybean, sesame, and perilla as pre-crops were cultivated, the soil pH was higher and the electrical conductivity was lower than in the rice pre-cropped soil. In addition, the available phosphate (Av. P2O5), and contents of Ca2+ and Mg2+ were higher than in the rice pre-cropped soil. Winter wheat growth characteristics such as culm length, spike length, number of spikes and grains were generally favorable in the black soybean pre-cropped soil. However, the grain yield of winter wheat increased in the black soybean, sesame, and perilla pre-cropped soils by 100 kg/10a or more than in the rice pre-cropped soil. Furthermore, protein content and SDS-sedimentation value of the flour were higher, while amylose content was slightly lower, in the black soybean, sesame, and perilla pre-cropped soils than in rice pre-cropped soil. These results suggest that cultivation of rice alternative crops such as sesame, black soybean, and perilla as pre-crops in paddy soil could improve the physical and chemical properties of the soil and contribute to producing high-quality wheat flour more advantageous for the baking process.

Evaluation of External Quality of Brand Soybeans (콩 시판 브랜드 제품의 외관 품질 평가)

  • Although high nutritional values and continuous identification of important functional substances of soybean [Glycine max (L.) Merrill.] promote consumption of soybean products worldwide, informations on quality of brand soybean is not enough for consumers. Total of 100 brand soybeans [32 for soypaste and source, 45 black testa (lage), and 17 black testa (small) or medicinal soybean and beansprout soybean] were collected at supermarkets and several external quality factors were analyzed. Brand soybeans were marked with the environmental friendly and intimating words along with soybean (white or yellow), black soybean (black-, frost-, late frost-, green or inner-green-), medicinal soybean and beansprout soybean. Among 100 brand soybeans 30% was 1 kg package and 59% was 500 g package, difference between printed and actual weights of 70% brand soybeans was ${\pm}1%$ and weights of 2/3 of brand soybeans were higher than printed weight. Range of 100 seed weights of soypaste and source, black testa (large) and black testa (small) and beansprout soybeans were $23.7{\sim}47.8g$, $21.9{\sim}44.5g$ and $9.5{\sim}15.0g$, respectively. Although ranges of 100 seed weights of soypaste and source and black testa (large) soybeans were similar, 63% of soypaste and source were less than 29 g, while 78% of black testa (large) soybeans were higher than 30 g. Although average and highest percentages of seeds separated with 6.7 mm sieve were similar with 87.4% and 99.9% for soypaste and source soybean and 86.5% and 99.5% for black testa (large) soybean, respectively, the lowest percentages were 70.7% for soypaste and source soybean and 14.4% for black testa (large) soybean. When 100 seed weight was greater than 35 g, 90% of seeds were remained on 6.7 mm sieve. On the other hand 100 g weight and percentage of seeds remained on 6.7 mm sieve showed significantly positive correlations [r=0.7488** for soypaste and source soybean and r=0.7874** for black testa (large) soybean when 100 seed weight was $20{\sim}30g$. Based on hilum color and/or appearance, 76% of brand soybeans collected (more than 90% in yellow testa soybeans) were found to be mixed more than 10% with other cultivars or landraces. Foreign materials such as sand, piece of clothe, wood piece, dead insects, other soybeans were found in 20% of brand soybeans. Average test weight of brand soybeans was 762g $L^{-1}$ with a range of $645{\sim}820g\;L^{-1}$. Soybeans from local markets were as good as brand soybeans in 100 seed weight, uniformity of seeds, weight of foreign materials and test weight.

Efficient Transformation Method of Soybean Using Meristematic Tissues of Germinating Seeds (발아종자의 분열조직을 이용한 효율적인 콩 형질전환 방법)

  • An efficient transformation method for soybean [Glycine max (L.) Merr.] using meristematic tissues of germinating seeds has been established. The embryonic axes were excised from germinating seeds of Korean soybean cultivar, Iksannamulkong and 0.5-2 cm long segment containing meristematic tissues were prepared by cutting hypocotyl region. The explants were inoculated with Agrobacterium tumefaciens strain LBA4404 harboring a binary vector with the bar gene as a selectable marker gene and a ${\beta}-glucuronidase$ (GUSINT) reporter gene, and then co-cultured for 7 days on co-cultivation medium (CCM). The meristematic tissues were cultured on shoot induction medium (SIMP6) supplemented with 0.4 mg/l $N_6-benzylaminopurine$ (BAP) and 0.1 mg/l indolebutyric acid (IBA) in the presence of 6 mg/l L-phosphinotricin (PPT) for 2 weeks and the surviving explants were transferred to shoot elongation medium (SEMP6). Transformation was confirmed by Southern blot analysis and the transformation efficiencies ranged from 1.48 to 2.07%. The new modified transformation method was successfully implemented for obtaining several transgenic lines with SMV-CP gene. It is expected that this method could efficiently be used for the transformation of recalcitrant soybean cultivars.
