Biocatalytic Production of Chiral Epoxides (생촉매를 이용한 광학활성 에폭사이드 생산)

  • Chiral epoxides are key intermediates for the production of chiral pharmaceuticals, agrochemicals, and functional food additives. Chiral epoxides can be produced by either chemical or biological method. In biocatalytic production routes, chiral epoxides can be produced via epoxidations of prochiral alkenes by monooxygenase or peroxidase. Kinetic resolution of racemic epoxides using whole cells of bacteria or fungi might be commercially useful, since it is possible to obtain chiral epoxides with high optical purities from relatively cheap and readily avaiable racemic epoxides. Some bioprocesses already are commercially developed: the biocatalytic production of chiral epichlorohydrin via microbial stereospecific dehalogenation, and lipase-catalyzed enantioselective hydrolysis in a hollow fiber membrane bioreactor for the production of chiral methyl trans-3-(4-methoxyphenyl)glycidate. the intermediate for calcium antagonist diltiazem. The importance of biocatalytic production of chiral epoxides with several examples from literature are presented.

Synthesis and Surface Active Properties of Diglycerol N-Acyl Glutamic Acid Esters (디글리세롤 N-아실 글루탐산 에스테르류의 합성 및 계면성)

  • Nonionic surfactants, diglycerol N-acyl glutamates were prepared through the esterification of N-acryl glutamic acids and glycidol in the presence of quarternary ammonium salts. Their cmc evaluated by the surface tension method was $0.15{\sim}0.75mol/{\ell}$, and the surface tension of aqueous solution was decreased to 28~35dyne/cm. L-DGLG with acyl group of carbon number of 12 showed good foaming power and emulsifying power, and the emulsifying power in toluene was stronger than in soybean oil. Dispersion effect of L-DGPG and L-DGSG with acyl group of carbon number of 16 and 18 in carbon black and ferric oxide were higher than L-DGLG. All of them showed good biodegradability at $20^{\circ}C$.

Fusion Protein Cleavage by Urokinase Covalentley Immobilized to Activated Sepharose Gels (활성화된 Sepharose Gels에 공유결합으로 고정화된 Urokinase를 이용한 융합단백질 절단반응)

  • Urokinase (UK), a thrombolytic enzyme used to clear catheters obstructed by blood clots, can be also used industrially in the recombinant protein purification system to cleave a fusion protein linked with a certain fragment of GST. We have immobilized UK by covalent attachment to activated Sepharose 6B-Cl gels and evaluated its performance to cleave a fusion protein of hGH and GST. The Sepharose gels were activated by etherification with glycidol (2,3-epoxypropanol) and further oxidized with periodate resulting in glyceryl-Sepharose gels. After the activation treatment, surface density of the aldehyde groups was 7-30 $\mu$mol-aldehde/mL-gel. Immobilization yield was higher than 99% at high pH (10.5), and the immobilized UK maintained ca. 80% specific activity of the soluble UK. In a column reaction the cleavage yield heavily depended on the feed rate, and it was nearly 86% of that from soluble UK. And the immobilized UK was successfully regenerated by unfolding and refolding with 6M GuHCl. After cleavaging reaction, the monomeric hGH was purified by using expanded bed adsorption chromatography.

Determination of Enantiopurity of Chiral Epoxides by Vibrational Circular Dichroism Spectroscopy (진동 원편광 이색성 분광기를 사용한 키랄 에폭사이드의 광학순도 분석)

  • In this work, vibrational circular dichroism (VCD) technique was applied for the determination of %EE of chiral compounds. It may provide an easy way to determine the %EE with a proper accuracy within 2% error ranges as well as the absolute configuration of enantiomers. We demonstrated herein a flow cell VCD (FT-VCD) technique for time-dependent %EE measurements. The simultaneous monitoring of the mole fraction and %EE for two chiral species (epichlorohydrin and glycidol mixture) in the mixture was shown to be successful without any further separation steps. Thus, we demonstrate that FT-VCD is an appropriate analytical tool to monitor the kinetics of reactions involving chiral molecules. FT-VCD also provides a convenient nondestructive approach for the time dependent determination of the optical purity of individual components in a reaction mixture containing chiral molecules.

Synthesis and Characterization of Interfacial Properties of Glycerol Surfactant (글리세롤계 계면활성제 합성 및 계면 특성에 관한 연구)

  • The CMCs of LA and LA3 nonionic surfactants obtained from the reaction between glycidol and lauryl alcohol were found to be $0.97{\times}10^{-3}mol/L$ and $1.02{\times}10^{-3}mol/L$ respectively and the surface tensions for 1 wt% surfactant were 26.99 and 27.48 mN/m respectively. Dynamic surface tension measurements using a maximum bubble pressure tensiometer showed that the adsorption rate of surfactant molecules at the interface between the air and the surfactant solution was found to be relatively fast in both surfactant systems, presumably due to the high mobility of surfactant molecules. The contact angles of LA and LA3 nonionic surfactants were 27.8 and $20.9^{\circ}$ respectively and the dynamic interfacial tension measurement by a spinning drop tensiometer showed that interfacial tensions at equilibrium condition in both systems were almost the same. Also both surfactant systems reached equilibrium in 2~3 min. Both surfactant solutions showed high stability when evaluated by conductometric method and the LA nonionic surfactant system provided the higher foam stability than the LA3 nonionic surfactant system. The phase behavior experiments showed a lower phase or oil in water (O/W) microemulsion in equilibrium with an excess oil phase at all temperatures studied. No three-phase region was observed including a middle-phase microemulsion or a lamellar liquid crystalline phase.
