Analysis of the Stoichiometry and the Domain for Interaction of Simian Virus 40 Small-t Antigen with Protein Phosphatase 2A

  • Simian virus 40 (SV40) small-t antigen (small-t) has been known to regulate the activity of a cellular enzyme, protein phosphatase 2A (PP2A), composed of A. B, and C subunits, via binding to the A subunit In the study presented here, the stoichiometry of the binding of small-t to PP2A was determined to be 1: 1. It was also shown that small-t binds to the AC form of PP2A with a higher apparent affinity than it binds to the free A subunit. We also characterized the interaction of PP2A with wild-type and various mutant small-ts. A single-point mutant (Val134Met) and a double-point mutant (Trp147Gly;Leu152 Pro) of small-t exhibited 3-fold and 5-fold lower potencies in inhibiting PP2A activity. respectively. This suggests that the region around amino acids between 134 and 152 of small-t might be important in regulating the enzyme activity of PP2A.

명태단백 Pronase 가수분해물의 제조 (Preparation of Pronase Hydrolysate from Alaska-pollack)

  • In order to enhance the utility of alaska-pollack, the optimum conditions for the preparation of pronase hydrolysate. The optimum temperature and pH for the hydrolysis of alaska-pollack by pronase were 4$0^{\circ}C$ and pH 7.0. The reaction time and enzyme concentration were 4 hr and 1,000 units per g of substrate. Under the above optimum conditions alaska-pollack was hydrolysed by pronase yielding a hydrolytic degree of about 89eye. The bitterness and hyrophobicity of pronase hydrolysate were decreased with increasing reaction time. Hydrophobic amino acids(Tyr, Met, Ala, flu, Leu, and Phe) were increased for 2 hr, but fur thor hydrolysis was showed decrease of hydrophobic amino acids content. Palatable amino acids (Asp, Glu, Pro, Ser, Thr and Gly) were increased with hydrolysis time.

호알카리성 Bacillus sp. MB 809의 알카리성 아밀라제의 말단 아미노산 서열과 그 상동성 (Terminal Amino Acid Sequences of Alkaline Amylase from Alkalophilic Bacillus sp. MB 809 and Their Homology)

  • Alkaline .alpha.-amylase expressed in the transformant, Baciollus subtills MB809, containing alkaline amylase gene cloned from alkalophilic Bacillus sp. AL-8, was purified through for step separation processes. The purified alkaline .alpha.-amylase had molecular weight of app[roximately 59, 000 daltons on SDS-PAGE and Sephaex G-100 gel filtration. Amino acid sequence of terminal portion of the enzyme was analyzed with pure amylase eluted form the SDS-PAGE gel. N-terminal amino acid sequence of .alpha.-amylase was determined by the Edman degradation method and resulted in $NH_{2}$-ser-thr-ala-pro-ser-(ile)-lys-ala-gly-thr-(ile)-leu. For C-terminal amino acid sequencing, purified .alpha.-amylase was digested with carboxypuptidase A and B, and reverse-phase HPLC gradient elution system resulted in -thr-trp-pro-lys-COOH.

Self-Assembly of Helical Pores from Nonpolar Dendritic Dipeptides

  • The synthesis of dendritic dipeptides $(4-3,4-3,5)12G2-CH_{2}-Boc-_{L}-Tyr-X-OMe\;where\;X\;=\;Gly,\;_{L}-Val,\;_{L}-Leu,\;_{L}-Ile,\;_{L}-Phe$, and L-Pro will be discussed. Their self-assembly in bulk and in solution and the structural and retrostructural analysis of their periodic assemblies will be compared to that of the previously reported and currently reinvestigated dendritic dipeptide with $X=_{L}-Ala$. All dendritic dipeptides containing as X nonpolar ${\alpha}-amino$ acids self-assemble into helical porous columns. The principles via which the aliphatic and aromatic substituents of X program the structure of the helical pores indicate synthetic pathways to helical pores with bioinspired functions based on artificial nonpolar ${\alpha}-amino$ acids will be discussed.

Conformation of Substance P in Neutral Phospholipid Micelles

  • A linear undecapeptide, Substance P (SP) is involved in a wide variety of physiological processes such as pain, inflammation, salivation, and hypertension. Tertiary structure of SP in dodecylphosphocholine (DPC) micelles has been investigated by CD, NMR spectroscopy, and DGII calculation. CD spectrum of SP in the presence of 7.5 mM DPC micelles does not show any favorable secondary structure. The tertiary structure determined by NMR spectroscopy and DGII calculation shows that the Phe7-Phr8-Gly9-Leu10 region adopts a turn structure, while the N-terminal region is quite flexible. Both prolines in SP exist preferentially as the trans isoforms and the aromatic ring of Phe7 protrudes outward. Conformation of SP may be restrained by the contact of the Phe7 aromatic ring with the hydrophobic side chains of the DPC micelles and this interaction induces a turn structure. Structure of SP in aqueous solution in the presence of DPC micelles can represent a good model to study the conformation recognized by the receptor near neutral membrane.

  • The conformational study on cyclic Ac-Cys-Pro-Xaa-Cys-NHMe (Ac-CPXC-NHMe; X = Ala, Val, Leu, Aib, Gly, His, Phe, Tyr, Asn, and Ser) peptides has been carried out using the ECEPP/3 force field and the hydration shell model in the unhydrated and hydrated states. This work has been undertaken to investigate structural implications of the CPXC sequence as the chain reversal for the initiation of protein folding and as the motif for active site of disulfide oxidoreductases.(omitted)

  • A novel neuropeptide with a relaxing activity on the dorsal retractor muscle (DRM) was isolated from the whole body extract of the starfish, Asterina pectinifera. The peptide was purified by gel-filtration ion-exchange and $C_{18}$ reversed-phase HPLC. The complete amino acid sequence of this peptide, which was determined by automated Edman degradation and MALDI-TOF mass, was Phe-Gly-Lys-Gly-Gly-Ala-Tyr-Asp-Pro-Leu-Ser-Ala-Gly-Phe-Thr-Asp. A comparison of the amino acid sequence with those of other known neuropeptides revealed that the asteripectin was a novel neuropeptide with smooth muscle-relaxing activity on the starfish DRM. This peptide showed threshold response to relaxing activity on the DRM at $10^{-10}M$ and the maximal relaxing effect was $120{\pn}7.0\%$ at $10^{-5}M$. The relaxing activity of this peptide on the starfish DRM increased in a dose-dependent manner.

  • 젓갈에서 분리한 B. subtilis p-4와 B. licheniformis p-5 프로테아제의 기질(카제인)에 대한 친화도를 측정한 결과 각각 0.38mM, 0.18mM이었고 열변성에 의한 특성을 반응속도론적으로 검토한 결과, p-4 프로테아제는 $50^{\circ}C$에서 80분 열처리할 때 20%의 잔존 활성을 보였고, p-5 프로테아제는 거의 실활되었는데 일차반응식을 따랐다. 그리고 p4, p-5 프로테아제의 변성 속도 상수는 각각 $40^{\circ}C$에서 $12.2{\times}10^{-5}/sec,\;19.0{\times}10^{-5}/sec,\;50^{\circ}C$에서는 $35.7{\times}10^{-5}/sec,\;46.3{\times}10^{-5}/sec$였다. 또 활성화 에너지는 p-4프로테아제가 19.6Kca1/mole, p-5 프로테아제는 15.2Kcal/mole로 p-5 프로테아제가 열변성에 민감한 것을 알 수 있었다. 활성화 자유에너지는 p-4, p-5 프로테아제 모두 온도가 상승함에 따라 약간 증가하였는데 $40^{\circ}C$에서 각각 23.21, 22.93Kcal/mole, $50^{\circ}C$에서는 23.28, 23.11Kcal/mole이었다. 아미노산 조성은 p-4 프로테아제의 경우 151개의 아미노산 잔기를 가졌으며, 이중 glycine, glutamic acid, proline이 많았고, p-5 프로테아제는 247개의 잔기에 glycine, glutamic acid, aspartic acid가 많았다.

수산물 가공부산물의 이용에 관한 연구 II-가공방법에 따른 가다랭이 내장 silage의 저장 중 성분변화 (Studies on the Utilization of Wastes from Fish Processing II -Changes of Chemical Properties of Skipjack Tuna Viscera Silage during Storage by the Processing Method)

  • 가다랭이 통조림 제조시 부산물로 대량 생산되는 내장을 유효하게 이용할 목적으로 유산균을 이용한 방법과 산첨가 방법에 따라 silage를 제조하고, 저장기간 중 성분변화를 검토한 결과, 산첨가 silage는 수분, 조지방, 조단백질 및 무기질의 함량이 유산균 발효 silage보다 높은 반면에, 탄수화물 및 다가불포화지방산 함량은 낮았다. 특히 저장기간의 경과에 따라 유산균 발효 silage의 n-3 지방산 함량의 증가가 두드러졌다. 산첨가 및 유산균 발효 silage의 주요 아미노산은 Glu, Asp, Leu, Gly 및 Ala 이었다. 산첨가 silage는 필수아미노산인 트립토판의 함량이 원료 내장에 비하여 $30\%$ 감소하였으나, 유산균 발효 silage는 $5\%$ 감소하였다. 또한 저장 기간의 경과에 따라 유산균 발효 silage의 비타민 $B_1$$B_2$ 의 함량은 증가하였으나, 산첨가 silage는 $B_1$ 함량만 미미하게 증가하였고, $B_2$는 감소하는 경향을 나타내었다. 산첨가 silage는 회갈색을 띄고 비린내 및 젓갈냄새가 났으나, 유산균 발효 silage는 다갈색을 띄고 생성된 유기산에 의하여 산미를 나타내었다.

치아 씨앗으로부터 분리한 단백다당체의 성분분석과 보습 효과 (Moisturizing Effects and Composition Analysis of Proteoglycan Isolated from Chia (Salvia hispanica) Seed)

  • 본 논문에서는 치아 씨앗으로부터 분리한 단백다당체(proteoglycan)의 성분분석과 분자량 측정 및 피부보습 효과를 연구하였다. 분리된 단백다당체의 성분분석 결과 치아 씨앗 단백다당체를 이루고 있는 당 성분(%)은 galactose (46.8), glucuronic acid (27.1), rhamnose (8.7), xylose (7.6), glucose (4.9), fructose (2.3), mannose (1.8), arabinose (0.9)로 확인되었으며, 함유된 단백질의 함량은 31.3 mg/g으로 이를 구성하는 아미노산 성분(mg/g)은 Asp (1.9), Glu (3.6), Ser (0.9), Gly (3.6), Thr (0.8), Arg (1.0), Ala (2.0), Tyr (0.4), Cys (4.8), Val (1.1), Phe (0.5), Ile (0.6), Leu (0.9)로 확인 되었다. Gel permeation chromatography (GPC)에 의해 측정한 치아 씨앗 단백다당체의 분자량 범위는 100,000~250,000 Da로 분자량 분포에 의한 평균 분자량은 170,000 Da이었다. 또한, 사람을 이용한 피부수분 보유 효과 및 피부 수분 손실량을 평가한 결과, 피부의 수분함량을 유지하는 효과가 우수하였으며 표피 수분 손실량 또한 적어서 피부 보습제로서 매우 우수하였다. 결론적으로 치아 씨앗의 다당체로부터 분리한 단백다당체를 화장품에 응용하면 우수한 보습효과를 가지는 화장품을 개발 할 수 있을 것으로 사료된다.
