Combination between Taxol-Encapsulated Liposomes and Eruca sativa Seed Extract Suppresses Mammary Tumors in Female Rats Induced by 7,12 Dimethylbenz(α)anthracene

  • Taxol (paclitaxel) is a powerful anti-cancer drug widely used against several types of malignant tumors. Because Taxol may exert several side effects, a variety of formulations have been developed. One of these features liposomes, regarded as one of the most promising drug carriers, biocompatible and best able to reduce drug toxicity without changing efficacy against tumor cells. Eruca sativa seed extract (SE) is considered a promising natural product from cruciferous vegetables against breast cancer, increasing chemotherapeutic and eliminating harmful side effects. The effects of Taxol-encapsulated liposomes (T) alone and in combination between Eruca sativa seed extract on nuclear factor kappa B (NF-${\kappa}B$), cyclooxygenase-2 (COX-2) and B-cell lymphoma-2 (Bcl-2) gene expression levels were investigated in rat mammary gland carcinogenesis induced by 7,12 dimethylbenz(${\alpha}$) anthracene (DMBA) using qRT-PCR. The results showed that DMBA increased NF-${\kappa}B$, COX-2 and Bcl-2 gene expression levels and lipid peroxidation (LP), while decreasing glutathione-S-transferase (GST) and superoxide dismutase (SOD) activities and total antioxidant concentration (TAC) compared to the control group. T and T-SE treatment reduced NF-${\kappa}B$, COX-2 and Bcl-2 gene expression levels and LP. Hence, T and T-SE treatment appeared to reduce inflammation and cell proliferation, while increasing apoptosis, GST and SOD activities and TAC.

Glutathione S-transferase M1 and T1 Polymorphisms, Cigarette Smoking and HPV Infection in Precancerous and Cancerous Lesions of the Uterine Cervix

  • Glutathione S-transferases (GSTs) play an important role in detoxification of carcinogenic electrophiles. The null genotypes in GSTM1 and GSTT1 have been implicated in carcinogenesis. Present study was planned to evaluate the influence of genetic polymorphisms of GSTM1 and GSTT1 gene loci in cervical carcinogenesis. The study was conducted in Lok Nayak hospital, New Delhi. DNA from clinical scrapes of 482 women with minor gynaecologic complaints attending Gynaecology OPD and tumor biopsies of 135 cervical cancer cases attending the cancer clinic was extracted. HPV DNA was detected by standard polymerase chain reaction (PCR) using L1 consensus primer pair. Polymorphisms of GSTM1 and GSTT1 were analysed by multiplex PCR procedures. Differences in proportions were tested using Pearson's Chi-square test with Odds ratio (OR) and 95% confidence interval (CI). The risk of cervical cancer was almost three times in women with GSTM1 homozygous null genotype (OR-2.62, 95%CI, 1.77-3.88; p<0.0001). No association of GSTM1 or GSTT1 homozygous null genotypes was observed in women with normal, precancerous and cervical cancerous lesions among ${\leq}35$ or >35 years of age groups. Smokers with null GSTT1 genotype had a higher risk of cervical cancer as compared to non-smokers (OR-3.01, 95% CI, 1.10-8.23; p=0.03). The results further showed that a significant increased risk of cervical cancer was observed in HPV positive smoker women with GSTT1 (OR-4.36, 95% CI, 1.27-15.03; p=0.02) and GSTM1T1 (OR-3.87, 95% CI, 1.05-14.23; p=0.04) homozygous null genotypes as compared to HPV positive non smokers. The results demonstrate that the GST null genotypes were alone not associated with the development of cervical cancer, but interacted with smoking and HPV to exert effects in our Delhi population.

Oxyfluorfen에 대한 내성(耐性) 및 감수성(感受性) 벼품종(品種)의 생리활성(生理活性) 기구(機構) - V. 항산화제(抗酸化劑) 함량(含量) 및 GST 활성(活性) (Different Physiological Activity of Selected Rice Cultivars to Diphenylether Herbicide, Oxyfluorfen - V. Different Content of Antioxidant and GST Activity)

  • DPE계 제초제(除草劑)인 oxyfluorfen의 생리활성(生理活性)을 내성(耐性)으로 선발되었던 벼 3품종(品種)과 감수성(感受性) 벼 4품종(品種)및 피를 공시하여 oxyfluorfen을 처리 후 항산화제(抗酸化劑) 함량과 GST 활성(活性)으로 조사하였다. 1. 식물자체가 보유하는 vitamin C 함량은 내성(耐性) 벼품종(品種)들이 감수성(感受性) 벼품종(品種)들이나 피보다 많았다. Vitamin E, carotenoid 및 glutathione(GSH, GSSG, total glutathione) 함량은 내성(耐性)과 감수성(感受性)식물들간에 양적인 차이를 볼 수 없었다. 2. Oxyfluorfen 처리에 따른 vitamin C 함량은 내성(耐性) 벼품종(品種) Hawon과 Baru에서 감수성(感受性) 벼품종(品種)들과 피보다 감소가 적었다. Oxyfluorfen 처리후 vitamin E 및 GSH 함량은 내성(耐性) 벼품종(品種)들이 감수성(感受性) 벼품종(品種)들이나 피보다 높았다. 그러나 carotenoid 함량은 내성(耐性) 벼품종(品種)들이 감수성(感受性) 벼품종(品種)들보다 높았으나, 감수성(感受性)인 피와는 차이가 없었다. 그리고 GSSG 함량은 내성(耐性)과 감수성종(感受性種)들간 차이가 없었다. 3. CDNB 또는 oxyfluorfen을 기질로 사용했을 때 식물 자체가 보유한 GST 활성(活性)은 내성(耐性) 벼품종(品種)들이 감수성(感受性) 벼품종(品種)들이나 피보다 높았다. 4. Oxyfluorfen 처리후 GST 활성(活性)은 벼에서 유도되지 않았으나, 오히려 피에서는 유도되었다. 또한, 벼의 GST 활성(活性)은 acifluorfen, bifenox 및 oxadiazon 처리에서도 유도되지 않았다.

넙치 카텝신 B의 분자생물학적 분석 및 효소학적 특성 연구 (Molecular Analysis and Enzymatic Characterization of Cathepsin B from Olive Flounder (Paralichthys olivaceus))

  • Papain family중 하나인 cysteine protease는 근골격계 질환 치료를 위한target molecule로 인식 되어왔으며 Cathepsin B 는 단백질 분해의 초기과정에 관여하는 cysteine proteases 중 하나이다. 본 연구는 넙치의 cathepsin B 유전자의 발현 양상과 넙치 cathepsin B(PoCtB)의 클로닝, 발현 및 효소특성을 분석하였다. cDNA Library Screening을 이용하여 넙치의 cDNA를 클로닝하였다. 넙치의 동정된 cathepsin B 유전자는 993bp의 open reading frame과 330개의 아미노산으로 이루어져있다. Cathepsin B의 propeptide region 내에 GNFD motif와 occluding loop 가 존재함으로써 이것이 명백하게 cathepsin B group이라는 것을 보여주며, 계통 유전학적 분석 결과 다른 종의 cathepsin B에 비해 초창기에 분화되어 나온 것으로 사료된다. mature enzyme인 maPoCtB은 fusion protein인 glutathione S-transferase를 포함하는 pGEX-4T-1 vector에 삽입하여 E.coli 균주인 $DH5{\alpha}$ 내에 발현시켰다. 재조합 단백질인 PoCtB을 과발현 시킨 결과 53kDa의 분자량을 가진다. 넙치 cathepsin B 활성은 Z-Arg-Arg-AMC와 같은 fluorogenic 펩타이드 기질을 이용하여 측정되었고 적정 pH는 pH.7.5 이다.

제초제(除草劑) bensulfuron과 imazaquin에 대한 1, 8-naphthalic anhydride(NA)의 옥수수와 콩에 대한 약해경감작용기구(藥害輕減作用機構) (Safening Mode of Action of 1, 8-Naphthalic Anhydride on Corn and Soybean Against Herbicide Bensulfuron and Imazaquin)

NA의 bensulfuron 및 imazaquino에 대한 약해경감효과(藥害輕減效果)는 옥수수에 대해서 약해경감효과지수(藥害輕減效果指數)가 각각 10.2, 5.0이였으나, 콩에 대해서는 두 약제(藥劑) 모두 약해경감효과지수(藥害輕減效果指數)가 1.3 정도(程度)로서 미약(微弱)한 효과(效果)를 보여 주었다. 2. 옥수수와 콩에 있어서 NA 처리시(處理時) 내생(內生) GST가 각가(各各) 1.8배(倍), 1.3배(倍) 증가(增加) 되었으나, 증가(增加)된 GST가 관여(關與)하는 GSH-CDNB conjugation 반응(反應)에 대한 bensulfuron 및 imazaquin의 억제활성(柳制活性)은 나타나지 않았다. 3. NA 처리시(處理時) 옥수수에서만 ALS가 증가(增加)되었고, 유출(抽出)된 ALS에 대한 bensulfuron 및 imazaquin의 억제활성(抑制活性)은 낮은 농도(濃度)에서는 차이(差異)를 보였으나, 높은 농도(濃度)에서는 NA처리(處理) 유무에 관계(關係)없이 동일(同一)한, 경향(傾向)이었다. 콩에서는 ALS의 증가(增加)도 없었고, 두 약제(藥劑)의 억제활성(抑制活性)도 NA 처리(處理)에 관계(關係)없이 동일(同一)하였다. 4. NA 처리시(處理時) 옥수수에서는 ACCase가 3.2배(倍) 증가(曾加)되었으나, 콩에서는 거의 증가(增加)가 없었고, 효소작용(酵素作用)에 대한 두 제초제(除草劑)의 영향(影響)은 아주 미약(微弱)하였다.

Consumption of Oxidized Soybean Oil Increased Intestinal Oxidative Stress and Affected Intestinal Immune Variables in Yellow-feathered Broilers

  • This study investigated the effect of oxidized soybean oil in the diet of young chickens on growth performance and intestinal oxidative stress, and indices of intestinal immune function. Corn-soybean-based diets containing 2% mixtures of fresh and oxidized soybean oil provided 6 levels (0.15, 1.01, 3.14, 4.95, 7.05, and $8.97meqO_2/kg$) of peroxide value (POV) in the diets. Each dietary treatment, fed for 22 d, had 6 replicates, each containing 30 birds (n = 1,080). Increasing POV levels reduced average daily feed intake (ADFI) of the broilers during d 1 to 10, body weight and average daily gain at d 22 but did not affect overall ADFI. Concentrations of malondialdehyde (MDA) increased in plasma and jejunum as POV increased but total antioxidative capacity (T-AOC) declined in plasma and jejunum. Catalase (CAT) activity declined in plasma and jejunum as did plasma glutathione S-transferase (GST). Effects were apparent at POV exceeding $3.14meqO_2/kg$ for early ADFI and MDA in jejunum, and POV exceeding $1.01meqO_2/kg$ for CAT in plasma and jejunum, GST in plasma and T-AOC in jejunum. Relative jejunal abundance of nuclear factor kappa B ($NF-{\kappa}B$) P50 and $NF-{\kappa}B$ P65 increased as dietary POV increased. Increasing POV levels reduced the jejunal concentrations of secretory immunoglobulin A and cluster of differentiation (CD) 4 and CD8 molecules with differences from controls apparent at dietary POV of 3.14 to $4.95meqO_2/kg$. These findings indicated that growth performance, feed intake, and the local immune system of the small intestine were compromised by oxidative stress when young broilers were fed moderately oxidized soybean oil.

Ginsenoside F1 attenuates pirarubicin-induced cardiotoxicity by modulating Nrf2 and AKT/Bcl-2 signaling pathways

  • Background: Pirarubicin (THP) is an anthracycline antibiotic used to treat various malignancies in humans. The clinical usefulness of THP is unfortunately limited by its dose-related cardiotoxicity. Ginsenoside F1 (GF1) is a metabolite formed when the ginsenosides Re and Rg1 are hydrolyzed. However, the protective effects and underlying mechanisms of GF1 on THP-induced cardiotoxicity remain unclear. Methods: We investigated the anti-apoptotic and anti-oxidative stress effects of GF1 on an in vitro model, using H9c2 cells stimulated by THP, plus trigonelline or AKT inhibitor imidazoquinoxaline (IMQ), as well as an in vivo model using THP-induced cardiotoxicity in rats. Using an enzyme-linked immunosorbent test, the levels of malondialdehyde (MDA), brain natriuretic peptide (BNP), creatine kinase (CK-MB), cardiac troponin (c-TnT), lactate dehydrogenase (LDH), superoxide dismutase (SOD) and glutathione (GSH) were determined. Nuclear factor (erythroid-derived2)-like 2 (Nrf2) and the expression of Nrf2 target genes, including heme oxygenase-1 (HO-1), glutathione-S-transferase (Gst), glutamate-cysteine ligase modifier subunit (GCLM), and expression levels of AKT/Bcl-2 signaling pathway proteins were detected using Western blot analysis. Results: THP-induced myocardial histopathological damage, electrocardiogram (ECG) abnormalities, and cardiac dysfunction were reduced in vivo by GF1. GF1 also decreased MDA, BNP, CK-MB, c-TnT, and LDH levels in the serum, while raising SOD and GSH levels. GF1 boosted Nrf2 nuclear translocation and Nrf2 target gene expression, including HO-1, Gst, and GCLM. Furthermore, GF1 regulated apoptosis by activating AKT/Bcl-2 signaling pathways. Employing Nrf2 inhibitor trigonelline and AKT inhibitor IMQ revealed that GF1 lacked antioxidant and anti-apoptotic effects. Conclusion: In conclusion, GF1 was found to alleviate THP-induced cardiotoxicity via modulating Nrf2 and AKT/Bcl-2 signaling pathways, ultimately alleviating myocardial oxidative stress and apoptosis.

항산화 방어 시스템에 의한 유기용매 내성세균 Pseudomonas sp. 균주에서의 톨루엔 내성 (Toluene Tolerance in Solvent Tolerant Pseudomonas sp. Strains By Antioxidant Defense Systems)

  • 유기용매 내성세균들이 항산화 방어시스템에 의하여 독성 유기용매로부터 자신을 보호하는지를 밝히기 위해 두 균주의 유기용매 내성 세균에서 항산화 효소 활성과 총 항산화능(T-AOC)을 조사하였다. 톨루엔 농도 100 mg/l에서 유기용매 내성세균 BCNU 106의 슈퍼옥사이드 디스무타아제(SOD) 레벨은 상대적으로 증가하였으며, 유기용매 내성세균 BCNU 171의 경우에는 톨루엔 농도 200 mg/l에서 상대적으로 증가하였다. 톨루엔 농도 200와 300 mg/l에서 유기용매 내성세균 BCNU 106에서 카타라제(CAT) 레벨이 3배 이상 증가함이 확인되었고, BCNU 171에서는 CAT 레벨이 2배 이상 증가함이 확인되었다. 또한 유기용매 내성세균에서 글루타치온 S- 전이효소(GST) 레벨이 높은 것으로 조사되었다. 총 항산화능도 유기용매에 대하여 내성을 보이지 않는 일반 세균인 KACC 10266보다 유기용매 내성세균들에서 높은 것으로 조사되었다. 한편 톨루엔 농도 200 mg/l 존재 하에서 BCNU 171 균주의 SOD 레벨은 1시간 후 정점에 도달하였다. BCNU 106에서 1시간과 14시간 후 CAT 레벨이 정점에 도달하였고, BCNU 171균주에서는 3시간 후 정점에 도달하였다. T-AOC 레벨의 정점은 BCNU 106에서는 톨루엔에 노출 9시간 후에 나타났으며, BCNU 171에서는 1시간과 9시간 후 두 번 T-AOC 레벨의 정점이 나타났다. 모든 유기용매 내성 세균은 왕성한 항산화 방어 반응을 보였으며, 이러한 항산화 방어 시스템을 통하여 유기용매 내성 세균이 유기용매를 비롯한 가혹한 환경하에서 생존할 수 있는 것으로 판단된다.

융합단백질로 발현된 톡소포자충의 주요막단백질(p30) 절편의 항원성 (Analysis of antigenic domain of GST fused major surface protein (p30) fragments of Toxoplasma gondii)

  • 톡소포자충(Toxoplosma gondii) 주요막단백질의 하나인 30 kDa 단백질(p30)의 항원부위를 결정하고자 p30의 아미노산 분석에 따른 친수성 부위 및 혐수성 부위에 맞게 유전자를 증폭하고 발현시켜 항원성을 검토하였다 p30의 절편으로는 p30 전체 p30의 N-말단 Signal Sequence와 C- 탈단의 혐수성 부위를 제거한 S28. S28의 N-말단 2/3부위인 Al9. S28의 C-말단 2/3부위인 Pl9. 528의 N-탈난 1/3부위인 X9 중앙 1/3부위인 Y10 및 C-말단 1/5부위인 Z9로 구성하였다. 각절편에 대한 primer에는 EcoR I의 clampsequence를 포함시켜 중합효소반응으로 증폭시켰으며 G57를 발현하는 pGEX-4T-1 vector에 삽입시킨 후 Eschericha coli(.JM105 strain)에 형질변형시키고 IgG로 각 절편이 GST와 융합단백질로 발현되도록 하였다 SDS-PAGE상에서 p30은 63 kDa. S28는 54 kDa Al9과 Pl9은 각각 45 kDa. X9은 35 kDa. Y10은 36 kDa 및 29은 35 kDa 단백질로 발현되었다. 각각의 단백질은 westemblot상에서 GSTdetectionkit와 잘 반응하여 융합단백질임을 확인하였다. 톡소포자충증 환자 혈청과 westem blot에서 p30. S28 및 Al9은 반응하여 항원성이 인정되었으나 Pl9 . X9, Y10 및 Z9는 반응하지 않았다 따라서. p30의 중간 1/3 부위의 존재하에 N-말단 1/3부위가 항원성을 나타내는 구조적 항원이거나. 첫 1/3부위와 중 간 1/3부위의 경계에 위치한 polypeptide가 항원성을 발현하는 것으로 추정되었다.

신문혈 자침이 어린 백서 해마의 단백질 발현에 미치는 영향 (Proteomic Changes by Acupuncture Stimulation at HT7 in the Hippocampus of Rat Pups)

  • Objectives : Hippocampus, a region of temporal lobe, plays an important role in the pathogenic mechanisms of brain diseases such as Alzheimer's disease, depression and temporal lobe epilepsy. This research is designed to investigate hippocampal changes after acupuncture stimulation at Shinmun(HT7) using 2-dimensional gel electrophoresis(2-DE). Methods : On postnatal-day 15, rat pups were randomly devided into Normal(NOR) or HT7 group. All of Pups kept with their mothers for 7 days, but pups in HT7 group received acupuncture stimulation at HT7 daily. On postnatal-day 21, hippocampus of each rat pup was dissceted 30 minutes after last acupuncture stimulation and the protein expressions were investigated using 2-DE. Results : After acupuncture stimulation at HT7, expression of 20 proteins were significantly increased. Succinate semialdehyde dehydrogenase, glyceraldehyde-3-phosphate dehydrogenase-like, transketolase, aconitate hydratase and phosphoglucomutase-1 were related to glucose methabolism. Eukaryotic initiation factor(eIF) 4A-II, eIF 4A-III, mitochondrial Tu translation elongation factor and chain A of crystal structure of the 70-Kda heat shock cognate protein involve in the protein synthesis in ribosome. Tubulin ${\beta}$-4 chain, tubulin T ${\beta}$-15 and tubulin ${\alpha}$-1B chain comprise cytoskeleton. Glutathione S-transferase(GST) ${\omega}$-1, GST P and GST Yb-3 can reduce oxidative stress. ${\beta}$-soluble N-ethylmaleimide-sensitive fusion protein attachment protein is required for vesicular transport between the endoplasmic reticulum and the Golgi apparatus, glycerol-3-phosphate dehydrogenase plays a major role in lipid biosynthesis, creatine kinase U-type catalyses the conversion of creatine and consumes adenosine triphosphate to create phosphocreatine and adenosine diphosphate. Platelet-activating factor acetylhydrolase IB subunit alpha and voltage depedent anion-selective channel protein 2 were also increased. Conclusions : The results suggest that acupuncture stimulation at HT7 may enhance glucose and lipid metabolism, protein synthesis, cytoskeletal substance and anti-oxidative stress in hippocampus.
