천연 항산화제로서 포도씨 분말 첨가가 육용계의 성장 및 항산화 작용에 미치는 영향 (Effect of Dietary Supplementation of Ground Grape Seed on Growth Performance and Antioxidant Status in the Intestine and Liver in Broiler Chickens)

  • 본 연구는 포도씨 분말 첨가 사료가 육계의 성장, 사료 이용성 및 체 조직내 항산화 효소 및 관련 생화학 지표 인자에 미치는 영향을 조사하기 위하여 수행되었다. 전체 50수 3일령 육계를 2주간 예비 실험 기간을 거친 다음 각 군당 10수씩 선발하여 대조구 및 포도씨 분말 1% 첨가구 등 2개 구를 설정하여 3주간 사양 시험을 실시하고 혈액 생화학 성분 및 체조직의 항산화 지표를 분석하였다. 그 결과는 다음과 같다. 1. 육계 사료내 1% 수준의 포도씨 분말 첨가는 체중, 일당 증체량, 사료 섭취량, 사료 요구율 및 장기 무게 등에 유의적인 영향을 미치지 않았다. 2. 혈액내 total antioxidant status(TAS)는 포도씨 첨가구에서 유의적(P<0.05)으로 증가하였으며, 혈중 생화학적 성분과 조직 손상을 나타내는 AST, ALT, LDH 등의 수준은 모든 구에서 비슷한 수준을 보였다. 3. 포도씨 분말 첨가는 소장 점막 세포내 SOD 활성도를 유의적으로(P<0.05) 증가시켰다. 그러나 간 조직내 GSHPx, SOD, GST 등과 같은 항산화 효소, 환원형 glutathione 및 지질 과산화(MDA) 수준은 대조구 및 포도씨 분말 급여구간에 차이는 없었다. 이상의 결과를 종합해 볼 때 천연 항산화제로서 포도씨 분말 1% 첨가 사료는 성장 및 사료 이용성에 특이적 영향을 미치지 않았으나, 혈액내 TAS 및 소장내 SOD활성도를 현저히 증가시키므로 육계에서 천연 항산화원으로서의 사용이 가능할 것으로 사료된다.

  • This study was designed to investigate the effects of mulberry (Morus alba L.) leaf extract (MLE) on oxygen radicals and their scavenger enzymes in liver membranes of rats. Sprague-Dawley (SD) male rats (160$\pm$10 g) were fed basic diet (control group), and experimental diets (MLE-100 and MLE-300 groups) added 100 and 300 mg/kg BW/day for 6 weeks. Hydroxyl radical (.OH) levels resulted in a significant decreases (15.2% and 18.1%, 5.6% and 8.0%, respectively) in liver mitochondria and microsomes could be not obtained. These are no significant differences in superoxide radical ($O_2$) levels of liver cytosol in MLE-100 and MLE-300 groups compared with control group. Lipid peroxide (LPO) levels were slightly decreased about 13.6% and 6.1% in liver mitochondria and microsomes of MLE-300 group compared with control group. Oxidized protein (OP) levels were remarkably decreased about 16.9% and 27.2% in liver microsomes only of MLE-100 and MLE-300 group compared with control group. Mn-SOD activities in liver mitochondria were remarkably increased (18.2% and 28.7%, respectively) in MLE-100 and MLE-300 groups, and Cu,Zn-SOD activities in liver cytosol were also significantly increased (11.3% and 20.2%, respectively) in MLE-100 and MLE-300 groups compared with control group. Mn-SOD activities in liver mitochondria were remarkably increased (18.2% and 28.7%, respectively) in MLE-100 and MLE-300 groups, and Cu,Zn-SOD activities in liver cytosol were also significantly increased (11.3% and 20.2%, respectively) in MLE-100 and MLE-300 groups compared with control group, but significant difference between GSHPx activities in liver cytosol could be not obtained. These results suggest that anti-aging effect of mulberry leaf extract (MLE) may play a pivotal role in attenuating a various age-related changes.

당뇨병 흰쥐에서 식이 제한 급여가 장기의 항산화효소 활성도 및 체중에 미치는 영향 (Effects of Dietary Restriction on the Body Weight and Antioxidant Enzymes in Various Organs of Diabetic Rats)

  • The effects of dietary restriction (DR) on antioxidant enzymes were studied in liver, lung and erythrocytes of diabetic rats. Experimental animals used Sprague-Dawley (SD; body weight 350$\pm$20g) male rats and Otsuka Long Evans Tokushima fatty (OLETE; body weight 5--$\pm$30g) male rats, as a model of type 2 diabetes mellitus. Type I diabetes was induced in SD rats by intramuscular injection of alloxan (80 mg/kg BW). Animals were randomly assigned either to continue the ad libitum diet or 40% DR (60% intake of ad libitum diet) groups. The body weight was measured at every 2 weeks to 4 months following DR. The activities of antioxidant enzymes (superoxide dismutase (SOD), catalase, glutathione peroxidase (GSHPx) were measured in liver, lung and erythrocytes and the concentration of TBARS as a marker of reactive oxygen species-induced tissue injry was also measured in rats after 4 months 40% DR. The body weight 4 months after 40% DR of control SD, alloxian-diabetid SD and OLETE rats were 80%, 98% and 75% of each control groups, respectively. The activities of SOD, catalase and GSHPx in lung and erythrocytes of rats were not change by 40% DR but in 4 month 40% DR rat liver, the activities of SOD and catalase were increased in control SD, alloxan-diabetic SD, and OLETF groups. The concentration of TBARS in lung and erythrocytes was also not changed by 40% DR, while liver TBARS concentration was decreased in OLETF and control SD rats compared to each non-DR control rats. These results suggested that the body weight changes in diabetic rats by DR was more prominent in type 2 diabetes and changes of antioxidant enzymes is most prominent in liver by DR either type 1 and 2 diabetic rats.

Lipid peroxide (LPO) levels were effectively inhibited (18.1% and 12.3%, respectively) in brain mitochondria and microsomes of MLE-300 groups compared with control group. Oxidized protein (OP) levels were significantly decreased (14.2%, and 10.9%, respectively) in brain mitochondria and microsomes of MLE-300 groups compared with control group. Mn-SOD activities in brain mitochondria were significantly increased (13.5% and 18.6%, respectively) in MLE-100 and MLE-300 groups, and Cu/Zn-SOD activities in brain cytosol were also effectively increased (about 17.7%) in MLE-300 groups compared with control group. GSHPx activities in brain cytosol were remarkably increased (17.2% and 23.9%, respectively) in MLE-100 and MLE-300 groups compared with control group. These results suggest that anti-aging effect of mulberry leaf extract (MLE) may play a pivotal role in attenuating a various age-related changes in brain.

  • This study was designed to investigate the effect of silk fibroin (Mw 500)power (SFP) on oxygen radicals and their scavenger enzymes in liver membranes of rats. Sprague-Dawley (SD) male rats (160$\pm$10 g) were fed basic diet (control group), and experimental diets (SFP-2.5 and SFP-5.0 groups) added 2.5 and 5.0 g/kg BW/day for 6 weeks. Hydroxyl radical (.OH) levels resulted in a considerable decreases (5.8% and 8.4%, 3.7% and 11.1%, respectively) in liver motochondria and micorsomes of SFP-2.5 and SFP-5.0 groups compared with control group, and $O_2$radical level was remarkably decreased about 15% and 20% in liver cytosol of SFP-2.5 and SFP-5.0 groups compared with control group. Lipid peroxide (LPO) levels were significantly decreased (8.3% and 18.0%, 13.4% and 18.4%, respectively) in liver mitochondria and microsomes of SFP-2.5 and SFP-5.0 groups compared with control group. Oxidized protein (OP) levels were remarkably decreased about 11.6% in liver mitochondria of SFP-5.0 group compared with control group. Mn-SOD activities were remarkably in creased (17.6% and 28.8%, respectively) in mitochondria of SFP-2.5 and SFP-5.0 groups, and Cu/Zn-SOD activities were also effectively in creased (about 14.4%) in liver cytosol of SFP-5.0 groups, but significant difference between GSHPx activity in liver cytosol of these two groups could be not obtained. These results suggest that anti-aging effect of silk fibroin may play an effective anti-aging role in a aattenuating a oxidative stress and increasing a scavenger enzyme activity in liver membranes.

Sprague-Dawley(SD) male rats(160$\pm$10g) were fed basic diet (control group), and experimental diets(SWP-200 and SWP-400 groups) added 200 and 400 mg/kg BW/day for 6 weeks. Hydroxyl radical (.OH) levels resulted in a consistent decreases (4.0% and 7.2%, 5.0% and 14.1%, respectively) in liver mitochondria and microsomes of SWP-200 SWP-400 groups compared with control group, and O2 radical level was significantly decreased about 12% in liver cytosol of SWP-400 group compared with control group. Lipid peroxide(LPO) levels were significantly decreased (14.4% and 9.1%, respectively) in liver mitochondria and microsomes of SWP-400 group only compared with control group. Oxidized protein (OP) levels were remarkably decreased about 12.7% and 16.3% in liver microsomes only of SWP-200 and SWP-400 groups, but significant difference between liver mitochondria could not obtained. Mn-SOD activities were remarkably increased (15.8% and 25.2%, respectively) in mitochondria of SWP-200 and SWP-400 groups, but significant difference between Cu, Zn-SOD activities in these group could be not obtained. GSHPx activity was significantly increased in liver cytosol of SWP-400 group compared with control group. These results suggest that silkworm powder may play an effective role in a attenuating a oxidative stress and increasing a scavenger enzyme activity in liver membranes.

갈근 에탄올 추출물이 흰쥐의 항산화계에 미치는 영향 (Effects of Supplementation of Puerariae Radix Ethanol Extract on the Antioxidative Defense System in Rats)

  • This study was performed to investigate the effect of Puerariae radix-ethanol extracts rich in isoflavone on the antio-xidative system of rats. For this purpose, first, Puerariae radix was extracted with ethanol, and its total isoflavone and puerarin contents were analysed. Second, female Sprague Dawley rats were fed for 6 weeks with four diets which were based on AIN96G diet and supplemented with Puerariae radix-ethanol extracts to contain isoflavone. The isoflavone contents of four experimental diets were 0 mg, 500 mg, 1,000 mg, 2,000 mg per kg diet, respectively (control, P0.05%,P0.1%, P0.2%). Liver and erythrocyte activities of antioxidative enzyme such as superoxide dismutase (SOD), catalase,glutathione peroxidase (GSHpx) were measured. Also, plasma and liver malondialdehyde (MDA) concentrations, liver glutathione (GSH) and oxidized glutathione (GSSG) concentrations were measured. The total isoflavone content of Puerariae radix-ethanol extract was 3067.6 mg per 100 g extract and the content of puerarin was 2557.4 mg per 100 g extract. The erythrocyte activities of GSH-Px and catalase were higher in group P0.1% and P0.2%. But SOD activity of erythocyte did not show any difference by the Puerariae radix-ethanol extract supplementation in diet. The activity of SOD in liver increased significantly by the supplementation of extract, showing highest level in P0.1% group. The liver GSH concentration increased significantly in group of P0.05%, P0.1%, and P0.2% compared with control group (p <0.05). The GSSG concentration in liver showed no difference by the supplementation of Puerariae radix extract from the control group, except P0.2% group. The plasma MDA concentration did not show any significant differences by the extract supplementation. But the liver MDA concentration decreased by the extract supplementation, showing the lowest level in P0.1 % diet group. These results suggest that the supplementation of Puerariae radix-ethanol extract can inhibit lipid peroxidation in liver and enhance the antioxidative defense competence of rats.

식이 Vitamin E가 급성 카드뮴중독 흰쥐 간조직의 항산화계에 미치는 영향 (Effects of Vitamin E on Antioxidative Defense System of Liver in Acute Cadmium-Poisoned Rats)

  • The purpose of this study was to investigate the effects of vitamin E on antioxidative defense system of liver in acute cadmium poisoned rats. Sprague-Dawley male rats weighing 100$\pm$10gm were randomly assigned to one control and three cadmium injected groups. Cadmium injected groups were fed vitamin E free diet(OE-Cd group), 40mg vitamin E per kg diet(40E-Cd group) or 400 mg vitamin E per kg diet(400E-Cd group). Vitamin E level of normal group was 40mg per kg diet. Animals were injected intraperitoneally with 2.0mg Cd$^2$$\^$+//kg bw for 4 days after the rats were fed diets with three different levels of vitamin E for 2 and 4weeks. Activities of superoxide dismutase(SOD), glutathione peroxidase(GSHPx) and glutathione S-transferase(GST) were decreased in cadmium injected groups but those were significantly improved by dietary vitamin I supplementations. Vitamin E contents reduced glutathione(GSH) in the live were decreased in cadmium injected groups, but we., not significantly different among three groups with different levels of vitamin E supplementations. Contents of liver thiobarbituric acid reactive substance (TBARS) of 0E-Cd group were higher than those of 400E-Cd and 400E-Cd groups, but those were markedly alleviated according to vitamin E supplementations. These results indicate that cadmium poisoning in rats causes decreasing antioxidative defense system and increasing peroxidative damage in liver, however can be restored by vitamin E supplements. 미역 (Undaria pinnatifida) 국수의 투유가 랫트 간장중의 활성산소 및 제거효소에 미치는 영향 (Effect of Brown Algae (Undaria pinnatifida)-Noodle on Oxygen Radicals and Their Scavenger Enzymes in Liver of Sprague-Dawley Rats)

  • 간장중의 활성산소 및 제거효소의 활성에 미치는 영향을 평가하기 위하여 미역의 건조분말을 $10{\%},\;20{\%},\;40{\%}$-첨가 조제한 미역국수 ($10{\%},\;20{\%},\;40{\%}\;FBA-noodle$)를 4주동안 SD계 흰쥐에 투여하여 미역국수의 생리작용을 평가하였다. $10{\%},\;20{\%},\;40{\%}$ FBA-noodle의 투여그룹의 mitochondria의 ${\cdot}OH$ 생성은 대조그룹 대비 각각 $20{\%},\;25{\%},\;35{\%}$의 현저한 OH 생성 억제효과가 인정되었다. 또한 $10{\%},\;20{\%},\;40{\%}$ FBA-noodle의 투여그룹의 microsome의 ${\cdot}OH$ 생성도 대조그룹 대비 $12{\~}20{\%}$의 유의적인 ${\cdot}OH$ 생성 억제효과가 인정되었다. $10{\%},\;20{\%},\;40{\%}$ FBA-noodle의 microsome의 $H_2O_2$의 생성은 유의적인 억제효과를 인정할 수 없었지만, cytosol중의 $O_2^({\cdot}-)$ 생성은 $20{\%},\;40{\%}$ FBA-noodle에서 $10{\%}$$O_2^({\cdot}-)$의 생성 억제효과가 인정되었다. $10{\%},\;20{\%},\;40{\%}$ FBA-noodle 투여그룹의 간장 mitochondria의 Mn-SOD 활성은 대조그룹 대비 $10{\~}15{\%}$의 Mn-SOD 활성의 증가효과가 인정되었다. 간장 microsome의 Mn-SOD 활성은 $10{\%} 및 20{\%}$ FBA-noodle 투여그룹은 Mn-SOD 활성의 유의적인 증가 효과를 인정할 수 없었지만, $40{\%}$ FBA-noodle 투여그룹은 $12{\%}$의 유의적인 Mn-SOD 활성 증가효과가 인정되었다. 또한 $10{\%},\;20{\%},\;40{\%}$ FBA-noodle의 투여그룹의 간장 cytosol의 Cu, Zn-SOD 활성은 대조그룹 대비 $10{\~}20{\%}$의 Cu, Zn-SOD 활성의 유의적인 증가효과가 인정되었다. 한편 간장 cytosol의 글루타치온 퍼옥시다아제 (GSHPx) 활성을 평가하여 보면 $10{\%}, 20{\%}, 40{\%}$ FBA-noodle 투여그룹의 cytosol의 GSHPx 활성은 대조그룹 대비 $20{\~}40{\%}$의 매우 효과적인 GSHPx활성의 증가효과가 인정되었다. $10{\%}, 20{\%}, 40{\%}$ FBA-noodle 투여그룹의 mitochondria의 LPO의 함량을 비교하여 보면 $10{\%}$ FBA-noodle은 유의적인 LPO의 생성 억제효과를 인정할 수 없었지만, $20{\%},\;40{\%}$ FBA-noodle은 $10{\%}$의 유의적인 LPO 생성 억제효과가 인정되었다. $10{\%},\;20{\%},\;40{\%}$ FBA-noodle 투여그룹의 microsome의 LPO의 함량은 $10{\%}$ FBA-noodle은 유의적인 LPO의 생성 억제효과를 인정할 수 없었지만, $20{\%} 및 40{\%}$ FBA-noodle은 약 $10{\~}12{\%}$의 유의적인 LPO 생성 억제효과가 인정되었다.

키토산올리고당이 카드뮴을 투여한 생쥐의 간독성에 미치는 효과 (Effects of Chitosanoligosaccharide on the Hepatotoxicity in Cadmium-treated Mice)

  • 본 연구는 카드뮴으로 유발된 생쥐 간중독에 대한 키토산올리고당의 효과를 알아보기 위하여 시도되었다. Mouse를 대상으로 카드뮴 (5.0 mg/kg; i.p.) 단독투여군(group Cd), 카드뮴과 키토산올리고당 (0.5% solution) 동시투여군 (group Cd+Chi) 그리고 키토산올리고당 1주일 전처치군 (group Chi7+Cd)으로 구분한 후 간손상 억제효과를 알아보기 위해 혈청중 효소활성도와 glutathione peroxidase (GSH-Px)의 활성도 그리고 glutathione reductase (GR)의 활성도를 비교측정하였다. 또한 간조직의 형태학적 손상을 확인하기 위해 조직학적 관찰을 실시하였다. 혈청 효소활성결과, AST, ALT 그리고 LDH의 활성도는 키토산올리고당을 처치한 Cd+Chi군과 Chi7+Cd군이 Cd군보다 감소되었다. 그러나 TP와 Alb의 활성도는 키토산올리고당을 처치한 Cd+Chi군과 Chi7+Cd군이 Cd군보다 증가되었다. GSH-Px활성은 Cd군에 비해 Cd+Chi군과 Chi7+Cd군이 현저하게 증가되었다. GR활성은 Cd군에 비해 Cd+Chi군과 Chi7+Cd군이 현저하게 감소하였다. 광학현미경적 관찰 결과, Cd군은 카드뮴 독성에 의한 간세포의 괴사가 관찰되었다. Cd+Chi군과 Chi7+Cd군은 정상군과 유사한 소견을 보였다. 이상과 같은 결과로, 키토산올리고당이 생쥐 간조직에 미치는 카드뮴의 독성을 감소시킴을 알 수 있었다.
