Glutathione S-Transferase Activities of S-Type and L-Type Thioltransferases from Arabidopsis thaliana

  • The glutathione S-transferase (GST) activities of S-type and L-type thioltransferases (TTases), which are purified from the seeds and leaves of Arabidopsis thaliana, respectively, were identified and compared. The S-type and L-type TTases showed $K_m$ values of 9.72 mM and 3.18mM on 1-chloro-2,4-dinitrobenzene (CDNB), respectively, indicating the L-type TTase has higher affinity for CDNB. The GST activity of the L-type TTase was rapidly inactivated after being heated at $70^{\circ}C$ or higher. The GST activity of the S-type TTase remains active in a range of $30-90^{\circ}C$. $Hg^{2+}$ inhibited the GST activity of the S-type TTase, whereas $Ca^{2+}$ and $Cd^{2+}$ inhibited the GST activity of the L-type TTase. Our results suggest that the GST activities of two TTases of Arabidopsis thaliana may have different catalytic mechanisms. The importance of the co-existence of TTAse and GST activities in one protein remains to be elucidated.

Glutathione-S-Transferase T1 Polymorphism is Associated with Esophageal Cancer Risk in Chinese Han Population

  • Background: Glutathione-S-Transferase T1 (GSTT1) gene has been shown to be involved in the development of esophageal cancer. However, the results have been inconsistent. In this study, the authors performed a meta-analysis to clarify the association between GSTT1 polymorphism and esophageal cancer risk among Chinese Han population. Methods: Published literature from PubMed, the China National Knowledge Infrastructure and Wanfang Data were searched. Pooled odds ratio (OR) and 95% confidence interval (95%CI) was calculated using a fixed- or random-effects model. Results: Eleven studies with a total of 2779 individuals were included in the meta-analysis. The results showed that GSTT1 null genotype was significantly associated with esophageal cancer risk in Chinese (OR = 1.31, 95%CI 1.12 to 1.53, p = 0.001). Further sensitivity analyses confirmed the significant association. The cumulative meta-analysis showed a trend of an obvious association between GSTT1 null genotype and esophageal cancer risk as information accumulated by year. Conclusions: This meta-analysis suggests a significant association of GSTT1 null genotype with esophageal cancer risk in the Chinese Han population.

Resistance of Saccharomyces cerevisiae to Fungicide Chlorothalonil

  • The toxicity of chlorothalonil on the growth of yeasts was investigated using several yeast strains. An alcohol tolerant yeast, Saccharomyces cerevisiae F38-1, was the most chlorothalonil-tolerant. The glutathione content and the glutathione S-transferase activity were related to the chlorothalonil-tolerant phenotype. Several thiol compounds affect the dissipation of chlorothalonil. However, there was no significant difference on the effects of chlorothalonil dissipation among the thiol compounds tested. The growth of yeast cells was arrested by chlorothalonil. It took about 13 h to dissipate 1 mg/l of chlorothalonil, and the growth was restored as the chlorothalonil content decreased. The glutathione content and glutathione S-transferase are suggested to be among the most important factors of yeast resistance to chlorothalonil.

In Vitro Cancer Chemopreventive Activities of Polysaccharides from Soybeans Fermented with Phellinus igniarius or Agrocybe cylindracea

  • Chemopreventive activities of polysaccharides from soybeans fermented with either Phellinus igniarius or Agrocybe cylindracea were investigated by measuring the induction of quinone reductase (QR), glutathione S-transferase (GST) activities, and glutathione (GSH) levels in the cell culture along with inhibition of polyamine biosynthesis. The polysaccharides from soybeans fermented with P. igniarius strongly (p<0.005) induced QR activity at all concentrations tested. The extract not only induced GST activity in a dose-dependent manner in the concentration range of 0.1-1.0 mg, but significantly induced GSH revels in cultured Hepa 1c1c7 cells with a maximal 1.4-fold increase at 0.1 mg. The polysaccharides from soybeans fermented with A. cylindracea were effective in inhibiting polyamine metabolism. These results suggest that polysaccharides from soybeans fermented with P. igniarius or A. cylindracea have cancer chemopreventive activities in in vitro models and, therefore, could be considered as potential agents for cancer chemoprevention.

  • This study was undertaken to examine the effect of Cheongahwan(CAH), being known to reinforce Kidney-yang, on the activities of endogenous antioxidant enzymes and the production of oxygen free radicals in the kidney tissues. Alterations in enzyme activities were observed after in vivo treatment in rats. CAH caused a significant increase in the activities of superoxide dismutase (SOD), glutathione peroxidase and glutathione S-transferase. But catalase activity was not significantly altered by CAH. Treatment in vitro of CAH decreased the production of oxygen free radicals in a dose-dependent fashion. These results suggest that CAH stimulate the activities of antioxidant enzymes and inhibit directly the production of oxygen free radicals. These effects of CAH may contribute to prevent the oxygen free radical-induced impairment of cell function.

  • Our previous reports on the effect of dietary protein on methanethiol, ethacrynic acid, bromobenzene and carbon tetrachloride metabolism were overall reviewed. The methanethiol, ethacrynicacid and bromobenzene treated rats showed the more severe liver damage in those fed a low protein diet than those fed a standard protein diet. These xenobiotics treated rats showed the lower content of hepatic glutathione and its conjugated enzyme, glutathione S-transferase activities in those fed a low protein diet than those fed a standard protein diet. In case of carbon tetrachloride treated rats, the liver damage was more reduced in rats fed a low protein diet than those fed a standard protein diet. Concomitantly the hepatic cytochrome P-450 content, and its decreasing rate to the control were lower in rats fed a low protein diet than those fed a standard protein diet.

AMPLIFICATION OF MERCURY TOXICITY BY GLUTATHIONE DEPLETION IN V79 CELLS

  • The treatmene of V79 cells with diethyl maleate (DEM) led to decrease in glutathione (GSH) level as increasing DEM concentration. Mercuric chloride, treated for 6 hrs with 2ng/ml, affected the GSH metabolizing enzymes glutathione S-transferase (GST) and glutathione peroxidase (GSP), dropping their activities to 60% and 75%, respectively, though not so much in GSH level(80%). However, the toxic effects of mercuric chloride on those enzymes and GSH level were both amplified when the Hg2+ treatment was combined with the preceding DEM treatment.

아세트아미노펜 유도 흰쥐에서 수산생물자원 추출물의 in vivo 간보호작용 (Protective Effect of Marine Natural Products on the Hepatic Lipid Peroxidation in Acetaminophen-treated Rats)

  • The study was initiated to elucidate the protective mechanism by examining in vivo effect of some marine natural products, Styela plicata, Ecklonia stolonifera and Pachymeniopsis elliptica on acetaminophen-induced lipid peroxidation. The methanol extract of S. plicata prevented acetaminophen (800mg/kg, i.p.)-induced hepatotoxicity in rats as evidenced by the decreased formation of lipid peroxide. But the methanol extracts of E. stolonifera and P. elliptica were not affected on the formation of lipid peroxidation. The activities of cytochrome P-450, animopyrine N-demethylase and aniline hydroxylase were not changed by the treatment with S. plicata in comparison with acetaminophen-teated group. In acetaminophen-treated control rats, the glutathione S-transferase activity was decreased markably. However. in S. plicata pretreated group, the effect caused by acetaminophen was markably reduced. A-cetaminophen decreased the level of hepatic, glutathione, which was restored to same degree by S. plicata pretreatment. And activity of ${\gamma}$-glutamylcystein synthetase was not changed by S. plicata pretreatment, but the activity of glutathione reductase was increased significantly.
