Objective: This study was performed to examine whether the porcine glutamic-oxaloacetic transaminase 1 (GOT1) gene has important functions in regulating adipocyte differentiation. Methods: Porcine GOT1 knockout and overexpression vectors were constructed and transfected into the mouse adipogenic 3T3-L1 cells. Lipid droplets levels were measured after 8 days of differentiation. The mechanisms through which GOT1 participated in lipid deposition were examined by measuring the expression of malate dehydrogenase 1 (MDH1) and malic enzyme (ME1) and the cellular nicotinamide adenine dinucleotide phosphate (NADPH) content. Results: GOT1 knockout significantly decreased lipid deposition in the 3T3-L1 cells (p<0.01), whereas GOT1 overexpression significantly increased lipid accumulation (p<0.01). At the same time, GOT1 knockout significantly decreased the NADPH content and the expression of MDH1 and ME1 in the 3T3-L1 cells. Overexpression of GOT1 significantly increased the NADPH content and the expression of MDH1 and ME1, suggesting that GOT1 regulated adipocyte differentiation by altering the NADPH content. Conclusion: The results preliminarily revealed the effector mechanisms of GOT1 in regulating adipose differentiation. Thus, a theoretical basis is provided for improving the quality of pork and studies on diseases associated with lipid metabolism.
Sera from male Spague-Dawley rats, exposed to $30\\pm 0.5^\\circ C$ for 240 hours or $33\\pm 0.5^\\circ C$ for 64 hours, were assayed for the activities of serum glutamic oxaloacetic transaminase(SGOT) and serum glutamic pyruvic transaminase(SGPT) at various time during the heat exposure. 1. When compared to control animals maintained at $23\\pm 1^\\circ C$, the animals exposed to $30\\pm 0.5^\\circ C$ ro $33\\pm 0.5^\\circ C$ showed a significant increase in SGOT and SGPT activities, 2. The SGOT activity incressed at 16 and 72 hours after the exposure to $30^\\circ C$, and at 30 and 64 hours after the exposure to $33^\\circ C$. After 72 hours, the activity returned to the initial value in case of $30^\\circ C$ exposure. 3. The SGPT activity increased significantly as early as 4 hours after the exposure to $30^\\circ C$ or $33^\\circ C$. It was also high at 16 hours after the exposure. The activity was also high at 72 hours and at 64 hours after the exposure to $30^\\circ C$ and $33^\\circ C$ respectively. After 144 hours, SGPT level increased slightly in the case of $30^\\circ C$ exposure. 4. The activities of SGOT and SGPT were significantly higher in rats exposed to $33^\\circ C$ at 16, 30, and 64 hours than those exposed to $30^\\circ C$. 5. It may be inferred from above data that the prolonged heat exposed rat has the abnormal metabolism of transamination.
In order to determine the influence of low atmospheric pressure on serum glutamic oxaloacetic transaminase (SGOT) and serum lactic dehydrogenase (SLDH) activities of rats, blood samples were collected from laboratory-conditioned male rats of the Sprague-Dawley strain which were randomly grouped into control and the experimental subjected to a series of one hour-exposure a day to low atmospheric pressure of 500 and 380mmHg up to the the time of 15 day. Results obtained indicated that decompression caused marked alterations in SGOT and SLDH levels when compared to that of the control. The trend of increases or decreases in these enzyme levels were similar in both 500 and 380mmHg exposed rats although the changes were greater in the latter group. Thus, generally all the experimental rats showed temporary steady state to low atmospheric pressure. Changes in enzymatic contents depended on the intensity and extent of the environmental stress under study. The lower the atmospheric pressure the greater is the effect on these serum enzyme levels.
Eight cattle and ten sheep were administrated various degree of PMA-B and EPN, and S-GOT, S-GPT and serum cholinesterase activity were tested. Serum cholinesterase activity showed no typical tendency, however, S-GOT/S-GPT ratios were decreased by the administration of PMA-B. It was suggested some degree of liver damage by the administration of the chemicals was recognized.
This study was performed to investigate the effect of Mori Folium and Crataegus pinnatifida leave extracts on liver damage induced by CC1$_4$ in rats. 30% and 60% methanol extracts of Mori Folium and Crataegus pinnatifida leaves were administered orally at the dose of 10 mg/kg on every day for 6 days and liver damage was induced by intraperitoneal injection of $CCl_4$ (0.4 ml/kg) on 6th day. 30% Methanol extracts of Mori Folium treated group showed significant protective effect on hepatotoxicity with the lowest Glutamic Oxaloacetic Transaminase (GOT) and Glutamic Pyruvic Transaminase (GPT). The low density lipoprotein (LDL) levels of 30% methanol extracts of Mori Folium and Crataegus pinnatifida leave and 60% methanol extracts of Mori Folium treated groups were significantly lower than normal group, respectively. In particular, LDL level of 30% Mori Folium extracts treated group was significantly decreased compared to $CCl_4$ treated group. These results support that Mori Folium and Crataegus pinnatifida leave extracts are hepatoprotective effects against hepatotoxicity induced by $CCl_4$.
The effects of dichloromethane (DCM) on carbon tetrachloride (CT) toxicity were examined in adult male rats. A concomitant treatment of rats with DCM (0.3, 0.6, 1.2 g/kg, po) significantly potentiated the hepatotoxicity of CT (1.0 g/kg, po) as determined by increase in serum GPT (glutamic pyruvic transaminase), GOT (glutamic oxaloacetic transaminase), and SDH (sorbitol dehydrogenase) activity 24 hr following the treatments. Serum LDH (lactate dehydrogenase) activity was increased by either DCM or CT treatment.

They were administered for 5 weeks. We measured body weight, serum growth hormone, insulin-like growth factor and thyroid stimulating hormone, RBC, concentration of Hb and PCV ratio, total WBC and its composition, the values of plasma glutamic oxaloacetic transaminase(GOT) and glutamic pyruvic transaminase(GPT) activities. Results 1. In body weight, Kyungohkgo 0.1%, Kyungohkgo Ga Nokyong 0.1%, 0.2% groups were showed sighnificantly different than control group. But That groups were not showed significantly different than each others. 2. In serum growth hormone, insulin-like growth factor and thyroid stimulating hormone, we obtained the results of tendency to increase in Kyungohkgo Ga Nokyong group, however these values showed no significantly different. 3. In the counts of RBC, Kyungohkgo group and Kyungohkgo Ga Nokyong group were showed significantly different than control group. But these values showed no significantly different. In the concentration of Hb, Kyungohkgo Ga Nokyong 0.2%group was showed significantly different than control group. Kyungohkgo Ga Nokyong 0.2% group and Kyungohkgo 0.2% group were showed significantly different than control group. 4. The counts of total WBC and its composition showed no significantly different in all treatment groups. 5. The values of plasma glutamic oxaloacetic transaminase(GOT) and glutamic pyruvic transaminase(GPT) activities showed no significantly different in all treatment groups. Conclusions So Kyungohkgo Ga Nokyong and Kyungohkgo have an effect of promoting growth of rats. And We know that the effect of Kyungohkgo Ga Nokyong is better than Kyungohkgo.
The present investigation involves a comparative study of enzymatic activities in various animals. The levels of the liver protein of rat (22.0$\pm$0.01mg/ml and pigeon (22.0$\pm$ 0.16) are twice as high as that of crucian carp (13.0$\pm$ 0.09)(p < 0.01). Generally, the specific acitivity (3.77 $\pm$0.18 unit/mg) of rat glutamic pyruvic transaminase (GPT) is highest, pigeon intermediate (1.93 $\pm$0.01), and crucian carp lowest (0.71$\pm$0.07). On the other hand, the specific activity (8.23$\pm$0.09 unit/mg)of rat glutamic oxaloacetic transaminase (GOT) is highest, pigeon intermediate (3.95$\pm$0.09), and crucian carp lowest (0.92$\pm$0.01) (p < 0.01). Ratios of GOT activity to GPT activity appear no remarkable difference from the levels of various animals. Specific activity of glutamic dehydrogenase(GDH) in pigeon tissue exceeds those of rat and crucian carp. In liver, rat GOT specific activity is greater than crucian carp and pigeon. On the other hand, pigeon GDH specific activity is greater than those of rat and crucian carp. This wouls seem to be in accord with protein metabolic intensity. The patterns for GDH isozyme were remarkably appeared in various animals. Glutamic dehydrogenase isozymes gave different electrophoretic mobilities in various animals. It is interesting that crucian carp, pigeon, and rat would show this difference, which may be indicative of an evolutional pattern. The fact that livers in various animals show quite different enzyme activities would suggest the existence of such a general phylogenetic relationship.
Serum glutamic oxaloacetic transaminase (GOT), latic dehydrgenase (LDH), and alkaline phosphatase (ALP) activities were determined in 5 Korean native cattle to obtain the base line of blood enzyme activities in relation to the stage of gestation. Serum GOT activities showed an increasing tendency with the course of gestation period, serum LDH levels were within normal range through all the stage of pregnancy, and the slight increase in serum ALP activities was acknowledged at the 2nd and 3rd stage of gestation.
The cultured mycelia of Ganoderma lucidum were extracted and the extract was separated into six fractions by organic solvent fractionation. The antihepatotoxic activity of all the fractions was evaluated by measuring activities of glutamic pyruvic transaminase (GPT) and glutamic oxaloacetic transaminase (GOT). Among the fractions tested, the high-polarity fractions such as aqueous and n-butanol fractions significantly reduced activities of GPT and GOT in CCl4- and galactosamine-intoxicated rat primary hepatocytes.
