C2C12 골격근 세포에서 갈근황금황련탕의 당 대사 및 에너지 조절 효과 (The Effects of Galgunhwanggumhwangryun-tang on Glucose and Energy Metabolism in C2C12 Myotubes)

  • Objectives: This study aimed to observe the anti-diabetic effect and underlying mechanisms of Galgunhwanggumhwangryun-tang (GHH; Gegen-Qinlian-decoction) in the C2C12 myotubes. Methods: GHH (1.0 mg/ml) or metformin (0.75 mM) or insulin (100 nM) were treated in C2C12 myotubes after 4 days differentiation. The glucose uptake was assessed by 2-[N-(7-160 nitrobenz-2-oxa-1,3-diazol-4-yl)amino]-2-deoxy-d-glucose uptake by C2C12 cells. The expression of adenosine monophosphate-activated protein kinase (AMPK) and phosphorylation AMPK (pAMPK) were measured by western blot. We also evaluated gene expression of glucose transporter type 4 (Slc2a4, formerly known as GLUT4), glucokinase (Gk), carnitine palmitoyltransferase IA (Cpt1a), nuclear respiratory factors 1 (Nrf1), mitochondrial transcription factor A (Tfam), and peroxisome proliferator-activated receptor γ coactivator 1α (Ppargc1a) by quantitative real-time polymerase chain reaction. Results: GHH promoted glucose uptake in C2C12 myotubes. The expression of AMPK protein, which plays an essential role in glucose metabolism, was increased by treatment with GHH. GHH treatment tended to increase gene expression of Slc2a4, Gk, and Nrf1 but was not statistically significant. However, GHH significantly improved Tfam and Ppargc1a gene expression in C2C12 myotubes. Conclusions: In summary, GHH treatment promoted glucose uptake in C2C12 myotubes. We suggest that these effects are associated with increased gene expression involved in mitochondrial biosynthesis and oxidative phosphorylation, such as Tfam and Ppargc1a, and increased expression of AMPK protein.

단기간의 당섭취 증가가 인슐린 감수성에 미치는 영향 (Effect of Acutely Increased Glucose Uptake on Insulin Sensitivity in Rats)

  • 인슐린 저항성이란 인슐린의 생물학적 효과가 감소하는 상태를 말하며 당뇨병의 중요한 병리기전이다. 인슐린 저항성은 매우 다양한 특성을 가지고 있으며 유전적인 요인이나 후천적인 요인에 의하여 발생한다. 이제까지 가장 잘 알려진 인슐린 저항성의 유발인자로는 고혈당과 고인슐린증이 있다. 장기간의 고혈당이나 고인슐린증은 가장 중요한 인슐린의 작용조직인 골격근에서 인슐린 수용체의 결합능을 감소시키고 당섭취에 필요한 제 4형 당수송체의 발현을 억제하는 것으로 알려져 있다. 그러나 이들 인자는 모두 골격근에서 당섭취를 증가시키는 것으로서 인슐린 저항성은 말초조직이 세포내로의 과도한 영양소 섭취를 방지하기 위하여 작동시키는 조절기전일 가능성을 생각할 수 있다. 본 연구에서는 단기간의 당섭취 증가가 더 이상의 당섭취를 억제할 수 있는지를 평가하고 그 기전을 규명하고자 흰쥐에서 2시간 동안 다양한 조건으로 당섭취를 유도하고 1 시간의 휴식기간을 가진 후 인슐린 예민도를 측정하고 골격근을 분리하여 인슐린 수용체와 제 4 형 당수송체를 분석하여 당섭취량과 인슐린 감수성과의 상관관계를 규명하고 인슐린 수용체 결합능과 제 4형 당수송체량을 비교하였다. 실험군은 hyperinsulinemic glucose clamp 전에 시행한 당섭취의 조건으로써 5개 군으로 구분하였다. 제 I 군은 대조군으로 생리식염수만 주입하여 기초상태의 인슐린 농도와 혈당을 유지하였으며, 제 II 군은 somatostatin과 포도당을 주입하여 정상 인슐린농도와 고혈당을 유지하였으며, 제 III 군은 포도당을 주입하여 고인슐린증과 고혈당을 유지하였고, 제 IV 군은 인슐린(100 mU/kg/min)과 포도당을 주입하여 초고인슐린증(${\sim}4000{\mu}U/ml$)과 정상혈당(~100 mg/dl)으로 유지하였으며, 그리고 제 V 군은 인슐린과 포도당을 주입하여 초고인슐린증과 고혈당(~200 mg/dl)을 유지하였다. 체중, 공복시 혈당 및 인슐린 농도는 각 군 간에 유의한 차이가 없었다. Hyperinsulinemic euglycemic clamp 전에 주입한 포도당의 총량(gm/kg)은 제 II 군이 $1.88{\pm}0.151$, 제 III 군이 $2.69{\pm}0.239$, 제 IV 군이 $3.54{\pm}0.198$, 그리고 제 V 군이 $4.32{\pm}0.621$이었다. Hyperinsulinemic euglycemic clamp의 평형상태 당제거율(mg/kg min)은 제 I 군이 $16.9{\pm}1.74$, 제 II 군이 $13.5{\pm}0.53$, 제 III 군이 $11.2{\pm}0.52$, 제 IV 군이 $13.2{\pm}0.92$, 그리고 제 V 군이 $10.4{\pm}0.41$로 전체 군 간에 유의한 차이가 있었으며(p<0.001) 각 군간의 비교에서 제 II, III, IV, V 군의 값이 제 I 군에 비하여 유의하게 낮았다 (p<0.05). 전체 실험동물에서 포도당 주입량과 당제거율 사이에 역상관관계 (r=-0.701, p<0.001)를 보였다. 인슐린 수용체 결합과 Western blot으로 분석한 제 4형 당수송체량은 각 군 간에 유의한 차이가 없었다. 이상의 결과로 미루어 단기간의 당섭취 증가가 더이상의 당섭취를 억제할 수 있으나 인슐린 수용체 결합과 제 4형 당수송체의 숫적 감소없이 발생하였다.

Effect of Bisphenol A on Insulin-Mediated Glucose Metabolism In Vivo and In Vitro

  • Bisphenol A (BPA), an environmental endocrine disrupter, enters the human body continuously in food and drink. Young children are likely to be more vulnerable than adults to chemical exposure due to the immaturities of their organ systems, rapid physical development, and higher ventilation, metabolic rates, and activity levels. The direct effect of BPA on peripheral tissue might also be of importance to the development of insulin resistance. However, the influence that BPA has on insulin signaling molecules in skeletal muscle has not been previously investigated. In this study, we examined the effect of BPA on fasting blood glucose (FBG) in post-weaned Wistar rats and on insulin signaling proteins in C2C12 skeletal muscle cells. Subsequently, we investigated the effects of BPA on insulin-mediated Akt phosphorylation in C2C12 myotubes. In rats, BPA treatment (0.1-1,000 ng/mL for 24 hours) resulted in the increase of FBG and plasma insulin levels, and reduced insulin-mediated Akt phosphorylation. Furthermore, the mRNA expression of insulin receptor (IR) was decreased after 24 hours of BPA treatment in C2C12 cells in a dose-dependent manner, whereas the mRNA levels of other insulin signaling proteins, including insulin receptor substrate-1 (IRS-1) and 5'-AMP-dependent protein kinase (AMPK), were unaffected. Treatment with BPA increased GLUT4 expression and protein tyrosine phosphatase 1B (PTP1B) activity in C2C12 myotubes, but not in protein levels. We conclude that exposure to BPA can induce insulin resistance by decreasing IR gene expression, which is followed by a decrease in insulin- mediated Akt activation and increased PTP1B activity.

The effect of multi-strain probiotics as feed additives on performance, immunity, expression of nutrient transporter genes and gut morphometry in broiler chickens

  • Objective: This study was conducted to investigate the effects of dietary multi-strain probiotic (MSP) (Bacillus coagulans Unique IS2 + Bacillus subtillis UBBS14 + Saccharomyces boulardii Unique 28) on performance, gut morphology and expression of nutrient transporter related genes in broiler chickens. Methods: A total of 256 (4×8×8) day-old CARIBRO Vishal commercial broiler chicks of uniform body weight were randomly distributed into four treatments with 8 replicates each and having eight chicks in each replicate. Four dietary treatments were T1 (negative control-basal diet), T2 (positive control-antibiotic bacitracin methylene disalicylate at 20 mg/kg diet), T3 (MSP at 107 colony-forming unit [CFU]/g feed), and T4 (MSP at 108 CFU/g feed). Results: During 3 to 6 weeks and 0 to 6 weeks, the body weight gain increased significantly (p<0.05) in T3 and T4 groups. The feed intake significantly (p<0.05) reduced from T1 to T3 during 0 to 3 weeks and the feed conversion ratio also significantly (p<0.05) improved in T3 and T4 during 0 to 6 weeks. The humoral and cell mediated immune response and the weight of immune organs were also significantly (p<0.05) improved in T3 and T4. However, significant (p<0.05) dietary effects were observed on intestinal histo-morphometry of ileum in T3 followed by T4 and T2. At 14 d post hatch, the relative gene expression of glucose transporter (GLUT5), sodium-dependent glucose transporter (SGLT1) and peptide transporter (PepT1) showed a significant (p<0.05) up-regulating pattern in T2, T3, and T4. Whereas, at 21 d post hatch, the gene expression of SGLT1 and PepT1 was significantly (p<0.05) downregulated in MSP supplemented treatments T3 and T4. Conclusion: The supplementation of MSP at 107 CFU/g diet showed significant effects with improved performance, immune response, gut morphology and expression of nutrient transporter genes. Thus, the MSP could be a suitable alternative to antibiotic growth promoters in chicken diets.

플라보노이드 처리된 체세포 핵이식 배아의 체외 발달 및 제주흑우 복제 소 생산 (In Vitro Development of Somatic Cell Nuclear Transfer Embryo Treated with Flavonoid and Production of Cloned Jeju Black Cattle)

  • This study was to investigate the effect of flavonoid treatment on in vitro development of bovine somatic cell nuclear transfer (SCNT) embryos, and their pregnancy and delivery rate after embryo transfer into recipient. In experiment 1, to optimize the flavonoid concentration, parthenogenetic day 2 ($\geq$ 2-cell) embryos were cultured in 0 (control), 1, 10 and $20\;{\mu}M$ flavonoid for 6 days. In the results, in vitro development rate was the highest in $10\;{\mu}M$ flavonoid group (57.1%) among treatment groups (control, 49.5%; $1\;{\mu}M$, 54.2%; $20\;{\mu}M$, 37.5%), and numbers of total and ICM cells were significantly (p<0.05) higher in $10\;{\mu}M$ flavonoid group than other groups. We found that $10\;{\mu}M$ flavonoid treatment can significantly (p<0.05) decrease the apoptotic index and derive high expression of anti-oxidant, anti-apoptotic, cell growth and development marker genes such as Mn-SOD, Survivin, Bax inhibitor, Glut-5, In-tau, compared to control group. In experiment 2, to produce the cloned Jeju Black Cattle, beef quality index grade 1 bull somatic cells were transferred into enucleated bovine MII oocytes and reconstructed embryos were cultured in $10\;{\mu}M$ flavonoid added medium. When the in vitro produced day 7 or 8 SCNT blastocysts were transferred into a number of recipients, $10\;{\mu}M$ flavonoid treatment group presented higher pregnancy rate (10.2%, 6/59) than control group (5.9%, 2/34). Total three cloned Jeju Black calves were born. Also, two cloned calves in $10\;{\mu}M$ flavonoid group were born and both were all healthy at present, while the one cloned calf born in control group was dead one month after birth. In addition, when the result of short tandem repeat marker analysis of each cloned calf was investigated, microsatellite loci of 11 numbers matched genotype between donor cell and cloned calf tissue. These results demonstrated that the flavonoid addition in culture medium may have beneficial effects on in vitro and in vivo developmental capacity of SCNT embryos and pregnancy rate.

Anti-diabetic effect of purple corn extract on C57BL/KsJ db/db mice

  • BACKGROUND/OBJECTIVES: Recently, anthocyanins have been reported to have various biological activities. Furthermore, anthocyanin-rich purple corn extract (PCE) ameliorated insulin resistance and reduced diabetes-associated mesanginal fibrosis and inflammation, suggesting that it may have benefits for the prevention of diabetes and diabetes complications. In this study, we determined the anthocyanins and non-anthocyanin component of PCE by HPLC-ESI-MS and investigated its anti-diabetic activity and mechanisms using C57BL/KsJ db/db mice. MATERIALS/METHODS: The db/db mice were divided into four groups: diabetic control group (DC), 10 or 50 mg/kg PCE (PCE 10 or PCE 50), or 10 mg/kg pinitol (pinitol 10) and treated with drugs once per day for 8 weeks. During the experiment, body weight and blood glucose levels were measured every week. At the end of treatment, we measured several diabetic parameters. RESULTS: Compared to the DC group, Fasting blood glucose levels were 68% lower in PCE 50 group and 51% lower in the pinitol 10 group. Furthermore, the PCE 50 group showed 2-fold increased C-peptide and adiponectin levels and 20% decreased HbA1c levels, than in the DC group. In pancreatic islets morphology, the PCE- or pinitol-treated mice showed significant prevention of pancreatic ${\beta}$-cell damage and higher insulin content. Microarray analyses results indicating that gene and protein expressions associated with glycolysis and fatty acid metabolism in liver and fat tissues. In addition, purple corn extract increased the phosphorylation of AMP-activated protein kinase (AMPK) and decreased phosphoenolpyruvate carboxykinase (PEPCK), glucose 6-phosphatase (G6pase) genes in liver, and also increased glucose transporter 4 (GLUT4) expressions in skeletal muscle. CONCLUSIONS: Our results suggested that PCE exerted anti-diabetic effects through protection of pancreatic ${\beta}$-cells, increase of insulin secretion and AMPK activation in the liver of C57BL/KsJ db/db mice.

세리신잠(Nd-s)의 발육특성과 혈액아미노산 조성 (The Development Properties of Sericin Jam and Composition of Amino Acid in Hemolymph)

  • 세리신 잠의 발육, 실샘 성장, 세리신 량, 혈액 아미노산 조성 등 세리신 잠의 특성을 알아보기 위하여 잠 120 과 같은 조건으로 사육하여 얻은 결과는 다음과 같다. 1. 부화율에 있어서 세리신 잠은 85%이며 잠 120 은 95%이었다. 특히 세리신 잠은 부화가 고르지 않았다. 2. 유충 기간에 있어서 세리신은 20 일 23 시간이었고, 잠 120 은 21 일 22시간이었다. 3. 실샘에 있어서 세리신 잠은 중부실샘이 대부분이며 후부 실샘의 길이는 약 2~3cm 이었고 굴곡은 없었다. 4. 세리신 잠에 있어서 상족후 나방까지의 기간은 약 12 일이었으며, 화용후 화아까지는 7 일로 대단히 짧은 특성을 보였다. 5. 고치에 있어서 세리신 잠은 이주 얇고 가벼웠으며 견충중은 2.7cg 이며 견충 비율은 3.65%이였다. 6. 세리신 (sericin protein)양에 있어서, 세리신 잠의 견충이 sericin 만 으로 구성되어 있고 잠 120 은 28%가 sericin 이었다. 세리신 잠의 견충 당 sericin 율은 잠 120 의 34.6%정도였다. 7. 혈액 아미노산의 양에 있어서 세리신 잠은 histidine, lysine, glutamic acid 가 많았으며 5 령 발육에 따른 조성의 변화에 있어서 대부분의 혈액 아미노산들은 증가하였다.

A Research Program for Modeling Strategic Aspects of International Container Port Competition

  • As national economies globalize, demand for intercontinental container shipping services is growing rapidly, providing a potential economic boon for the countries and communities that provide port services. On the promise of profits, many governments are investing heavily in port infrastructure, leading to a possible glut in port capacity, driving down prices for port services and eliminating profits as ports compete for business. Further, existing ports are making strategic investments to protect their market share, increasing the chance new ports will be overcapitalized and unprofitable. Governments and port researchers need a tool for understanding how local competition in their region will affect demand for port services at their location, and thus better assess the profitability of a prospective port. We propose to develop such a tool by extending our existing simulation model of global container traffic to incorporate demand-side shipper preferences and supply-side strategic responses by incumbent ports to changes in the global port network, including building new ports, scaling up existing ports, and unexpected port closures. We will estimate shipper preferences over routes, port attributes and port services based on US and international shipping data, and redesign the simulation model to maximize the shipper's revealed preference functions rather than simply minimize costs. As demand shifts, competing ports will adjust their pricing (short term) and infrastructure (long term) to remain competitive or defend market share, a reaction we will capture with a game theoretic model of local monopoly that will predict changes in port characteristics. The model's hypotheses will be tested in a controlled laboratory experiment tailored to local port competition in Asia, which will also serve to demonstrate the subtle game theoretic concepts of imperfect competition to a policy and industry audience. We will apply the simulation model to analyze changes in global container traffic in three scenarios: addition of a new large port in the US, extended closure of an existing large port in the US, and cooperative and competitive port infrastructure development among Korean partner countries in Asia.

Altitude training as a powerful corrective intervention in correctin insulin resistance

  • Oxygen is the final acceptor of electron transport from fat and carbohydrate oxidation, which is the rate-limiting factor for cellular ATP production. Under altitude hypoxia condition, energy reliance on anaerobic glycolysis increases to compensate for the shortfall caused by reduced fatty acid oxidation [1]. Therefore, training at altitude is expected to strongly influence the human metabolic system, and has the potential to be designed as a non-pharmacological or recreational intervention regimen for correcting diabetes or related metabolic problems. However, most people cannot accommodate high altitude exposure above 4500 M due to acute mountain sickness (AMS) and insulin resistance corresponding to a increased levels of the stress hormones cortisol and catecholamine [2]. Thus, less stringent conditions were evaluated to determine whether glucose tolerance and insulin sensitivity could be improved by moderate altitude exposure (below 4000 M). In 2003, we and another group in Austria reported that short-term moderate altitude exposure plus endurance-related physical activity significantly improves glucose tolerance (not fasting glucose) in humans [3,4], which is associated with the improvement in the whole-body insulin sensitivity [5]. With daily hiking at an altitude of approximately 4000 M, glucose tolerance can still be improved but fasting glucose was slightly elevated. Individuals vary widely in their response to altitude challenge. In particular, the improvement in glucose tolerance and insulin sensitivity by prolonged altitude hiking activity is not apparent in those individuals with low baseline DHEA-S concentration [6]. In addition, hematopoietic adaptation against altitude hypoxia can also be impaired in individuals with low DHEA-S. In short-lived mammals like rodents, the DHEA-S level is barely detectable since their adrenal cortex does not appear to produce this steroid [7]. In this model, exercise training recovery under prolonged hypoxia exposure (14-15% oxygen, 8 h per day for 6 weeks) can still improve insulin sensitivity, secondary to an effective suppression of adiposity [8]. Genetically obese rats exhibit hyperinsulinemia (sign of insulin resistance) with up-regulated baseline levels of AMP-activated protein kinase and AS160 phosphorylation in skeletal muscle compared to lean rats. After prolonged hypoxia training, this abnormality can be reversed concomitant with an approximately 50% increase in GLUT4 protein expression. Additionally, prolonged moderate hypoxia training results in decreased diffusion distance of muscle fiber (reduced cross-sectional area) without affecting muscle weight. In humans, moderate hypoxia increases postprandial blood distribution towards skeletal muscle during a training recovery. This physiological response plays a role in the redistribution of fuel storage among important energy storage sites and may explain its potent effect on changing body composition. Conclusion: Prolonged moderate altitude hypoxia (rangingfrom 1700 to 2400 M), but not acute high attitude hypoxia (above 4000 M), can effectively improve insulin sensitivity and glucose tolerance for humans and antagonizes the obese phenotype in animals with a genetic defect. In humans, the magnitude of the improvementvaries widely and correlates with baseline plasma DHEA-S levels. Compared to training at sea-level, training at altitude effectively decreases fat mass in parallel with increased muscle mass. This change may be associated with increased perfusion of insulin and fuel towards skeletal muscle that favors muscle competing postprandial fuel in circulation against adipose tissues.

HMGB1/Snail cascade에 의한 epithelial-mesenchymal transition 및 glycolytic switch, mitochondrial repression 유도 (High-mobility Group Box 1 Induces the Epithelial-mesenchymal Transition, Glycolytic Switch, and Mitochondrial Repression via Snail Activation)

  • 암세포는 epithelial mesenchymal transition (EMT)를 통해 tumor invasion과 metastasis가 일어나며, 또한 정상세포와 다른 oncogenic metabolic phenotypes 획득 즉, glycolytic switch 등이 암 발생과 진행에 깊이 연관되어 있음이 잘 알려져 있다. High-mobility group box 1 (HMGB1)은 chromatin-associated nuclear protein으로 알려져 있으나, dying cells 또는 immune cells로부터 방출되기도 한다. 방출된 HMGB1은 damage-associated molecular pattern (DAMP)로서 작용하여 EMT 및 invasion, metastasis를 유도함으로서 tumor progression에 기여한다고 알려졌다. 본 연구에서 HMGB1에 의해 EMT와 glycolytic switch 유도되며, 이 과정은 Snail 의존적임을 확인하였다. 또한 HMGB1/Snail cascade는 COX subunits인 COXVIIa와 COXVIIc의 발현 억제를 통해 mitochondrial repression과 cytochrome c oxidase (COX) inhibition을 유도하였다. HMGB1은 Snail를 통해 glycolytic switch의 주요 효소인 hexokinase 2 (HK2), phosphofructokinase-2/fructose-2,6-bisphosphatase 2 (PFKFB2), phosphoglycerate mutase 1 (PGAM1)의 발현을 증가시켰다. 이들 효소는 glycolytic switch에 중요하게 관여하는 것으로 알려져 있다. 이들 해당과정의 효소들을 knockdown한 결과 HMGB1에 의한 EMT를 억제함으로써 glycolysis와 HMGB1-induced EMT가 밀접하게 연관되어 있을 제시하였다. 이상의 연구 결과들은 HMGB1/Snail cascade가 EMT 및 glycolytic switch, mitochondrial repression에 중요하게 작용할 것임을 시사한다.
