Growth Performance and Field Evaluation of Herbicide-Resistant Transgenic Creeping Bentgrass

  • This study examined the growth performance and field evaluation of the dual herbicide-resistant transgenic creeping bentgrass plants. The effect of glyphosate treatment on the herbicide resistance of the transgenic creeping bentgrass plants was determined, and the non-transgenic control plant withered at the concentration $11{\mu}g/mL$ or higher whereas the transgenic creeping bentgrass plants survived the treatment at the concentration of $3,000{\mu}g/mL$, and the increase of the plant length was repressed as the glyphosate treatment concentration was increased. At field evaluation, glufosinate-ammonium and glyphosate were simultaneously treated to investigate the weed control effect. The results showed that more than 90% of the weeds withered four week after herbicide treatment, while the transgenic creeping bentgrass plants continued to grow normally. Therefore, the dual herbicide-resistant creeping bentgrass plants may be able to greatly contribute to the efficiency of weed control and to the economic feasibility of mowing in places such as golf courses.

Evaluation of Crossability, Seed Dormancy and Overwintering Ability in Glufosinate Ammonium-Resistant GM Rice and Their Hybrids with Non-GM and Weedy Rice

  • This study was conducted to investigate the crossability, seed dormancy and overwintering ability of rice plant in GM (glufosinate ammonium-resistant lines. Iksan 483 and Milyang 204) and non-GM (their parents) or red rice (Andongaengmi). Seed-setting rate was not significantly different between GM and non-GM rice varieties. Iksan 483 and Milyang 204 showed the similar level of seed germination rate from 30 to 50 days after heading as compared to non-GM rice varieties. After overwintering in paddy field, seed germination rate of GM and non-GM rice varieties ranged from 14.3 % to 57.6 % in dry soil condition, but there was no germination in wet-soil except red rice. The result in wet-soil condition may help to set up a strategy for reducing the risk of gene flow of transgene via dispersal of seeds of GM plants. The crossability, seed dormancy and seed overwintering of Iksan 483 and Milyang 204, herbicide resistant GM rice varieties, were not significantly different compared to non-GM rice varieties. The results might be helpful to reduce the risk of transgene dispersal from GM crop via seeds and pollens.

A simple model for selection and rapid advancement of transgenic progeny in sorghum

  • To select agronomically useful transgenic plants, a large number of transgenic events are initially produced, gene transfer confirmed, and advanced to obtain homozygous lines for testing in field trials. Direct in planta assays for identifying the transgene carriers in the segregating progeny are based on the activity of selectable marker gene and are easy, simple and inexpensive. For this purpose, expression of bar gene as measured by tolerance to damage by glufosinate ammonium, the active ingredient in the herbicide BASTA, was investigated. Dose damage curves were generated by leaf paint tests with BASTA on four genotypes of sorghum. Transgenic plants were characterized in terms of sensitivity to the concentration of glufosinate ammonium. In transgenics, symptoms of BASTA swab tests at different growth stages and PCR analysis for cry1B were carried out and correlated. Germination tests could not be employed for large scale evaluation of transgenic progeny because of mortality of tolerant seedlings after transplantation to soil. Based on the above findings, a simple, inexpensive, time-saving, two-step scheme for effective evaluation of transgenics and their progeny containing bar gene as selection marker using BASTA swab tests is described.

세계 잡초연구 동향 및 전망 (Current Status and Perspectives of Weed Science in the World)

  • 세계 잡초연구자들이 세계잡초학회에서 발표한 최근 연구동향을 파악하여 이를 토대로 향후 잡초연구 발전방향과 전망을 제시하고자 하였다. 제 7차 세계잡초학회는 우리나라를 포함한 59개국 520여명이 참석하였으며, 9개 분야 625편이 발표되었다. 주요 연구분야로는 제초제 저항성잡초, 잡초생태, 농경지 및 비농경지의 잡초관리, 제초제 살포기술, 비화학적 잡초방제 등이었다. 이중 제초제 저항성잡초 연구분야는 전체 30%를 점유하여 향후에도 이들 연구가 지속될 것으로 판단된다. 특히 설포닐우레아계 보다는 비선택성 제초제인 glyphosate, glufosinate-ammonium 저항성 잡초에 대한 발표가 많았다. 신규 제초제 개발은 synthetic auxin 작용기작의 제초제인 arylex, HPPD 저해제로 개발 중인 3가지 물질을 safener와의 합제 연구 등이 있었다. 그리고 세계 주곡작물인 옥수수, 밀, 벼 재배지에서 발생되는 제초제 저항성잡초에 대한 연구가 집중되었다. 특히 잡초의 non-target site에 대한 저항성 연구가 집중적이었다. 또한 우리나라에는 없는 기생잡초에 대한 연구 폭이 넓어지고 있으며, 예년에는 취급하지 않았던 비화학적 잡초방제 부문도 많았다. 향후에는 제초제 저항성 메카니즘 구명, 기생잡초 연구, 제초제 살포기술, 비화학적 잡초방제 등의 연구가 폭넓게 진행될 것으로 전망되었다.

애기수영의 화학적 방제를 위한 제초제 선발 및 선발 제초제의 살초효과 (Response of Red Sorrel (Rumex acetosella L.) to Several Soil- and Foliar-Applied Herbicides)

  • 본 실험의 목적은 강원도 고냉지 목초지에 크게 발생하고 있는 외래잡초 애기수영을 방제할 수 있는 토양처리 및 경엽처리 제초제를 실내실험을 실시하여 선발하고, 포장실험을 통하여 그 약효를 검정하는데 있다. 토양처리 제초제인 pendimethalin, simazine, alachlor, metolachlor, ethalfluralin은 실내실험시 애기수영에 대한 방제효과가 거의 없었다(P=0.05). 그러나 경엽처리 제초제인 glufosinate, paraquat, glyphosate, glyphosate + 2,4-D, dicamba, mecoprop, 2,4-D, bentazone은 처리 2주 후 애기수영에 대해 60% 이상의 살초효과를 보였다. 이들 중 살초효과가 좋았던 옥신타입 제초제, dicamba와 2,4-D의 처리시기를 알기 위하여 엽기가 다른 애기수영에 처리한 결과, 두 옥신타입 제초제의 살초효과는 엽기와 관련이 없었다. 강원도 고냉지에 위치한 풍전목장에서 경엽처리 제초제들의 살초효과를 검정한 결과, 애기수영은 glufosinate 1.44 kg a.i. $ha^{-1}$, glyphosate 3.28 kg a.i. $ha^{-1}$, dicamba 0.96 kg a.i. $ha^{-1}$, mecoprop 2.50 kg a.i. $ha^{-1}$ 살포시 처리 4주 후 80% 이상 방제되었다. 본 실험의 결과들은 비선택성 제초제인 glufosinate와 glyphosate, 그리고 선택성 제초제인 dicamba와 mecoprop을 이용하여 강원도 고냉지 목초지에서 문제가 되고 있는 애기수영을 방제할 수 있다는 점을 보여주고 있다.

제초제가 줄지렁이 치사에 미치는 영향평가 (Assessment of the Effects of Some Herbicides on Mortality of Earthworm (Eisenia fetida))

  • 농경지에 과거에 사용했거나 사용 중인 9종의 제초제가 농경지에 서식하는 지렁이에게 악영향이 있는지 알기위하여 추천사용량, 2배량 및 4배량을 기준으로 토양표토처리법, 침지법, 여지접촉법으로 실험을 수행하였다. 그 결과 토양표토처리법에서 처리양과 제초제 종류와 관계없이 무처리와 비교하여 통계적인 유의성이 없었다. 침지법의 4배량에서 phamid WP는 34%, alra EC는 64%의 치사율을 나타냈고, 여지접촉법의 추천사용량에서 phamid WP는 33%, 2배량에서 phamid WP는 37%, alra EC는 80%, triclopyr EC는 10%, 4배량에서 phamid WP는 96%, alra EC는 80%, triclopyr EC는 37%, glyphosate SL는 47%, Paraco SL는 37%, pyributicarb WP는 10%, Glufosinate-ammonium SL는 33%의 지렁이 치사율을 나타냈다.

단일 응급의료기관에서 경험한 글루포시네이트 암모니움 포함 제초제 중독 후 중증도 예측에 관한 연구 (A Study of Predicting the Severity Following Glufosinate Ammonium Containing Herbicide Poisoning Experienced in Single Emergency Medical Institution)

  • Purpose: Owing to the increased agricultural use of the herbicide glufosinate ammonium (GLA), the incidence of GLA poisoning has recently increased. Therefore, we investigated the possible predictive factors associated with severe complications following GLA poisoning. Methods: A retrospective analysis of medical records was conducted based on 76 patients who had visited our regional emergency medical center with GLA poisoning from 2006 to 2017. Severe complications were defined as respiratory failure requiring intubation, systolic blood pressure less than 90 mmHg, Glasgow Coma Scale (GCS) less than 8, and presence of seizure. Results: Age, ingested amount and ingested amount per weight were significantly greater in the severe group (p<0.001). PSS grade 2 or higher was more common in the severe group (p<0.001), and In addition, the APACHE II score was significantly higher in the severe group (p<0.001), as were the SOFA scores (p=0.002). Serum ammonia levels were significantly higher in the severe group (p=0.007), while MDRD-GFR was smaller in the severe group (p=0.002). The spot urine protein levels were significantly higher in the severe group (p=0.005), as was the urine protein to creatinine ratio (p=0.001). Upon multivariate analysis, the amount ingested per weight and PSS grade 2 or higher were identified as significant predictors. Conclusion: Our study showed that MDRD-GFR was significantly lower in the severe group after GLA poisoning. PSS grade 2 or higher and ingested amount per weight may be useful to evaluate the severity of complications after GLA poisoning.

Agrobacterium을 이용한 형질전환에서 sonication과 vir 유전자들의 효과 (Effect of Sonication and vir Genes on Transient Gene Expression in Agrobacterium-Mediated Transformation)

  • Sonication tremendously improves the efficiency of Agrobacterium infection by introducing small and uniform fissures and channels throughout the targeted tissue. Using shoot tips of cotton as explants, the effect of sonication treatment and virulence genes in Agrobacterium tumefaciens on transformation efficiency was investigated. The pat gene which encodes resistance to the herbicide, glufosinate, was used as a selectable marker. Transformation efficiency was evaluated on th basis of survival rates of cocultivated shoot tips on selection medium containing 2.5 mg/l gulfosinate-ammonium(ppt) adn 25. mg/l Clavamax. Sonication from 5 to 15 second has a positive effect on shoop tip survival. However, whil virE as well as virG or vir GN54D showed an enhancement in transformation efficiency, virE,. virG resulted in the most significant enhancement. Overall, the combination of additional virG/virE gene and sonication treatment resulted in the most significant increase in transformation efficiency.

복숭아 과원에서 잡초방제방법이 신초 생육 및 수량에 미치는 영향 (Effects on the Shoot Growth and Yield Related to Weed Control Methods at the Peach Orchard)

  • 본 시험은 복숭아 과원 잡초방제 방법별 신초 생육과 수량에 미치는 영향을 비교하고자 수행하였다. 1차 처리 후 10일 간격 3회 조사한 결과, 제초제(glufosinate ammonium) 2회 및 3회 처리구의 잡초방제 효과는 10일째 98.2~98.6%, 20일째 77.5~78.2%, 30일째 40.7~41.4%로 나타났고 부직포 처리구는 100%, 100%, 97.5%이었으며, 기계예초 처리구는 92.1%, 56.4%, 36.4%이었다. 수확 전과 후의 신초 생육량을 비교한 결과, 제초제 2회 및 3회 처리구, 부직포 피복구, 기계예초 처리구 모두 무처리보다 전반적으로 생육이 좋았다. 평균 정형과수는 제초제 2회 처리구와 기계예초 처리구가 무처리와 비교하여 통계적으로 차이가 인정되었다. 따라서, 제초제 2회 또는 3회 살포가 복숭아 과원에서 조사된 14종의 모든 초종에 탁월한 방제효과와 더불어 복숭아나무의 수체 생육과 정형과수 확보에 있어 지장이 없기 때문에 향후 활용가치가 높다고 판단한다.
