구척 에탄올 추출물이 Streptozotocin 유발 당뇨 흰쥐에 대한 항당뇨 및 항산화 작용에 미치는 효과 (Antidiabetic and Antioxidative Effects of Cibotium barometz in Streptozotocin-Induced Diabetic Rats)

  • Streptozotocin(STZ)으로 유도된 당뇨 흰쥐에게 구척 에탄올 추출물을 1일 1회 7일간 1,000 mg/kg.b.w의 용량으로 투여 후 glucose함량과 당대사에 관여하는 효소인 glucose-6-phosphatase(G-6-Pase), glucose-6-phosphate dehydrogenase(G-6-PDH)과 glucokinase(GK)활성과 glycogen 함량과 triglyceride(T.G), total cholesterol 등의 지질대사에 관여하는 물질들과 항산화작용에 관여하는 glutathione-s-transferase(GST),Catalase(CAT), glutathione peroxidase(GSH-Px)활성과 glutathione(GSH)함량을 측정한 결과 구척 에탄올 추출물 투여군이 glucose, T.G, total cholesterol 등의 함량과 G-6-Pase 활성의 유의적인 감소(p<0.05)를 나타내었으며 glycogen 함량과 G-6-PDH, GK의 활성이 유의적인 증가(p<0.05)를 나타내었다. 이와 같이 구척 에탄올 추출물이 항당뇨, 항산화작용의 개선효과를 갖는 유효성분을 함유하고 있음을 알 수 있었다.

  • Tryptophan을 계란에 투여하고 배양하였을 때의 초기 계배의 뇌형성에 미치는 영향을 대뇌피질세포의 전자현미경 관찰과 계배두부의 단백질 및 핵산과 serotonin의 정량 그리고 몇가지 기초대사에 관여하는 효소활성을 관찰하였다. 계란에게 tryptophan을 투여하고 $5\\sim10$일간 부란한 계배의 대뇌피질세포 전자현미경으로 관찰한 결과 정상에 비해 크게 손상되었으며 특히 핵막이 불규칙하고 핵응축현상이 심하며, 염색질이 핵막에 응집되어 있고 인이 심하게 분해되었고 핵막이 팽출되어 수포가 많이 생기고 확장된 Golgi체가 많으며 조면소포체가 확장되고 소포화왼 것들도 있었으나 lysosome은 드물게 보였으며 tryptophan 투여로 인한 기형현상이 뚜렷하였다. Tryptophan을 투여한 10일 계배의 DNA 함량은 크게 저하되지는 않았으나 RNA 함량과 단백질 함량 그리고 lactate dehydrogenase, succinate dehydrogenase, malate dehydrogenase, glucose 6-phosphate dehydrogenase와 같은 기초대사에 관여하는 효소들의 활성이 저하된 것으로 보아 tryptophan은 단백질 합성을 저하시킨 것으로 관찰되었다. 한편 10일 계배의 serotonin의 함량은 시험군에서 크게 늘고 있으며 serotonin의 증거가 intracellular yolk granule의 분해를 지연시켜 기형현상이 일어나는지는 분명치 않으나 tryptophan 투여로 인하여 단백질 합성이 저하되고 따라서 효소활성이 저하됨으로서 세포분화에 지장을 초래하는 것으로 생각된다.

Leucine의 섭취가 고지방 식이를 섭취한 흰쥐의 체중 감소에 미치는 영향 (Effect of Leucine Intake on Body Weight Reduction in Rats Fed High Fat Diet)

  • The principal objective of this study was to determine the effects of leucine on body weight reduction in high fat diet-induced overweight rats. To induce overweight, six-month-old male Sprague-Dawley rats (n = 80) were divided into 8 groups; one group of 10 rats was fed on a normal fat diet and the remaining 70 rats were fed on a high-fat diet (40% of energy as fat) for 14 weeks. Then, 10 rats fed on the normal fat diet and another 10 rats fed on the high fat diet were sacrificed to identify overweight induction. The remaining 60 rats were divided randomly into 6 groups according to body weight and fed on one of the diets with different dietary fat levels (9.6% or 40% of energy as fat) and leucine levels (0, 0.6 or 1.2 g/kg BW) for the following 5 weeks of experiments. The body weight loss in the Leu-administered groups (0.6 g, 1.2 g/kg BW) was significantly higher than those of Leu non-administered groups. The perirenal fat pad weights in the Leu-administered groups were significantly lower than those of the Leu non-administered groups. Of the hepatic enzymes, glucose-6-phosphate dehydrogenase (G6PDH) activities were reduced significantly in the Leu-administered groups than in the Leu non-administered groups. With the oral glucose tolerance test (OGTT), the incremental areas under the curve of the glucose response (IAUC) of the Leu-administered groups were significantly lower than those of the Leu non-administered groups. The fasting glucose concentration and HOMA-IR of the Leu-administered groups were significantly lower than those of the Leu non-administered groups. In conclusion, the results of this study suggest that one of the possible mechanisms of leucine in the observed body weight reduction might involve the inhibition of lipogenic enzyme activities such as glucose-6-phosphate dehydrogenase, rather than the activation of lipolysis enzymes. Additionally, leucine adminstration resulted in improved glucose metabolism.

Reciprocal Effect of DHEA and Rietary Fat on Glutathione Utilizing Detoxifying System in Rat Liver Tissue

  • This study was intended to examine whether dehydroepiandrosterone (DHEA) and dietary fat level or source could modulate glutathione utilizing detoxifying system activity and the cytosolic NADPH generation in rat liver. Male Sprague-Dawley rats were fed semipurifed diet containing either 2%(w/w) corn oil (low level of corn oil diet: 5 ca% of fat) 15% corn oil (high level of corn oil diet: 31 cal% of fat) or 13% sardine oil plus 2% corn oil(high level of fish oil diet: 31 cal% of fat) for 9 weeks. Half of the rats in each diet group were fed a diet supplemented with 0.2% DHEA (w/w). DHEA administration increased plasma total cholesterol level in low corn oil diet-fed rats. The high fish oil diet significantly decreased plasma total cholesterol level compared to the high corn oil diet. Plasma triglyceride level was not significantly changed by DHEA administration and dietary fat level and source. Fasting plasma glucose level was increased by DHEA administration and fish oil diet. Glucose 6-phosphate dehydrogenase activity in liver tissue was significantly increased by DHEA administration and high fat diet, especially fish oil diet. Malic enzyme activity in liver tissue was significantly increased by DHEA administration and high fat diet, especially fish oil diet. Malic enzyme activity in liver tissue was significantly increased by DHEA administration. DHEA suppressed the glutathione peroxidase, glutathione-dependent enzymes compared to the low corn oil diet, while fish oil diet elevated the activity of glutathione peroxidase and glutathione reductase compared to corn oil diet. These results suggest that DHEA administration and high level of corn oil diet may suppress the cellular detoxifying system activity through reduction of glutathione utilization, while the fish oil diet did not show these effects.

Hypoglycemic effect of Rehmannie Radix Preparata (Sookjihwang) extract in streptozotocin-induced diabetic rats

  • Rhemannie Radix Preparata (RRP) has been previously employed in traditional oriental medicine as a treatment for diabetic thirst and improving blood flow. The aim of this study was to evaluate its hypoglycemic control by assaying the activities of key enzymes of carbohydrate metabolism in streptozotocin-(STZ)-induced diabetic rats. Further, RRP extracts were prepared in water (RRPW), in 50% ethanol (RRP50), and in 100% ethanol (RRP100), respectively, and compared for their actions in diabetic rats. The oral treatment of RRP (5 mg/kg b.w./d) to diabetic rats for 21 days resulted in a significant decline in blood glucose by 67% compared to diabetic control rats (P < 0.05). The altered activities of glucokinase, glucose-6-phosphate dehydrogenase (G6PD), 6-phosphogluconate dehydrogenase (6PGD), and acetyl CoA carboxylase (ACC) in the livers of diabetic rats were reversed significantly to near-normal levels by the administration of RRP (P < 0.05). Among the three RRP extracts, RRP100 was the most effective in terms of hypoglycemic action. However, the administration of RRP to diabetic rats did not improve insulin production. The modulatory effects of RRP100 on the attenuation of carbohydrate enzyme activities appear to hold promise for widespread use for the treatment of diabetes in the future.

Grapefruit 종자추출물이 Enterobacter pyrinus의 생리기능에 미치는 영향 (The Inhibitory Effect of Grapefruit Seed Extracts on the Physiological Function of Enterobacter pyrinus)

  • Grapefruit 종자추출물(GFSE)에는 박테리아와 곰팡이류들에 대하여 항균 효과를 갖는 성분이 있다고 알려져 있다. 배나무에 병반에서 분리한 Enterobacter pyrinus도 GFSE를 가하여 준 배지에서 생육이 억제되었다. 대조군이 stationary phase로 접어든 배양 20시간까지 GFSE의 최종 농도가 0.01%를 함유한 배지에서 E. pyrinus는 전혀 성장하지 못하였다. 항균 성분을 분리하기 위하여 GFSE를 용매추출하여 친수성 분획과 소수성 분획으로 나누었을 때 억제 효과는 소수성 분획에서 관찰되었다. GFSE가 에너지 대사에 관여하는 몇가지 효소에 미치는 영향을 조사하여 본 결과, hexokinase, glucose-6-phosphate dehydrogenase, malate dehydrogenase와 succinate dehydrogenase 등에는 별다른 효과를 나타내지 않았다. 세포막에 미치는 영향을 알아보기 위하여 ${\beta}-galactosidase$의 인위적 기질인 ONPG의 분해 여부를 조사한 결과, GFSE는 세포막에 영향을 끼침을 관찰하였다. GFSE의 항균 성분을 분리 확인하기 위해서 HPLC $C_{18}$ reverse phase column chromatography를 하여 5개의 peak를 얻었으며, 이중 1개의 분획에서 강한 항균 활성을 검출하였다. 이 분획을 GC-Mass로 사용하여 분석한 결과 1-chloro-2-methyl-benzene (o-toluene), N,N-dimethyl-benzenemethaneamine, 1-[2-(2-ethylethoxy)ethoxy]-4- (1,1,3,3-tetramethyl)-bezene 이외에 분획의 대부분을 차지하는 1개의 물질을 포함하여, 확인하지 못한 3개의 물질이 있음을 알았으며, GFSE의 항균활성을 보이는 성분은 새로운 물질일 가능성을 시사하였다.

  • A rapid, inexpensive enzymatic method is proposed for indirect water quality testing in terms of heavy metal toxicity. The activity of glucose-6-phosphate dehydrogenase was applied for heavy metal toxicity test as an effective criterion in water quality. The toxicity of Pb (lead) and Cd (cadmium) for water flea, Moina macrocopa, were evaluated for $2{\sim}8\;days$ with variables of mobilization ability. And the reproduction impairment of Moina macrocopa were investigated as the parameter of chronic toxicity twst for Pb and Cd. As a result, the $EC_{50}$ for immobilization of Moina macrocopa were Pb and Cd were 1.6749 and 0.4683, respectively. The values of reproducive impairment to Moina macrocopa for Pb and Cd were 9.5938 and 8.3264 in $EC_{50}$. A significant alteration of G6PDH (Glucose-6-phosphate dehydrogenase) activity of Moina macrocopa was observed when Cd and Pb were treated in media. The results obtained indicate that G6PDH activity of Moina macrocopa can be used as an indicative parameter in aquatic toxicity tests for heavy metals.

Alterations in the Activities of Antioxidant Enzymes of Human Dermal Microvascular Endothelial Cells Infected with Orientia tsutsugamushi

  • Changes in the Activities of several antioxidant enzymes in transformed human dermal microvascular endothelial Cells (HMEC-1) by infection with the obligate intracellular bacterium Orientia tsutsugamushi, the causative agent of scrub typhus, were investigated. The activities of glucose-6-phosphate dehydrogenase, catalase, and glutathione peroxidase were significantly decreased in HMEC-1 cells infected with Ο. tsutsugamushi. However, the level of superoxide dismutase increased slightly. Furthermore, Increased levels of intracellular peroxide was observed in HMEC-1 during infection. These results support the hypothesis that cells infected by this intracellular bacterium experience oxidant-mediated injury that may eventually contribute to cell death.

인삼 근절편 배양시 Peroxidase Isozyme에 관한 연구 (Peroxidase Isozyme in Root Differentiation from Cultured Ginseng Root Explants)

  • In order to pursue some physiological studies on organogenesis in ginseng tissue culture, ginseng root explants were cultured on a modified MS medium containing NAA and kinetin. The activities of peroxidase and some enzymes were investigated and their isoenzyme patterns were also observed. The activity of peroxidase decreased by 20% in one week's culture and increased thereafter by 80% in culturing for 7 weeks compared with the control group. Glucose-6-phosphate dehydrogenase activity increased by 400% after culturing for 5 weeks and increased during the days preceeding root formation. The activities of glutamate dehydrogenase and acid phosphatase also increased during the culture. After 3 weeks' culture, new peroxidase isozyme (pH 7.6) appeared and 7 weeks' culture, another new peroxidase isozyme (pH unidentified) appeared. These patterns were also identified by using FPLC. After 7 weeks' culture, a new esterase isozyme of pH 8.5 appeared and isozyme patterns of acid phosphatase were quite changed compared with the isozyme patterns of tissue cultured for 5 weeks. In so far as these new isoenzymes appear distinctively after 7 weeks' culture, root differentiation is supposed to be induced after 7 weeks' culture.

매실추출물이 변패미생물의 생리기능에 미치는 영향 (Inhibitory Effect of Prunus mune Extracts on Physiological Function of Food Spoilage microorganisms)

  • 본 연구는 PME 처리가 미생물의 생리변화에 미치는 영향을 중심으로 PME의 항균작용을 규명하기 위해서 소수성 분획과 친수성 분획으로 분리하고, 공시균주를 사용하여 미생물의 성장 및 에너지대사 관련효소의 활성에 미치는 영향을 조사하였다. 매실추출물 분획별 항균활성은 변패미 생물인 C. fragariae를 대상으로 paper disk법으로 관찰하였다. 총 추출물(PME)의 소수성 분획은 C. fragariae의 성장을 저해하기는 하나 그 효과가 총 추출물 보다 약한 반면, 친수성 분획은 총 추출물보다 더 뚜렷한 저해 효과를 보였다. 이와 같은 매실추출물의 분획별 미생물의 생육억제효과는 다른 식품부패미생물 군에서도 동일한 패턴을 보여 주었다. B. cereus, E, coli 및 Fusarium sp.에 대해서는 PME의 친수성 분획은 뚜렷한 항균효과를 보여 주었으며, 총 추출물 또한, 항균력을 나타내었으나, 소수성 분획은 낮은 항균력을 보여 본 실험의 공시균주인 C. fragariae에 대한 매실추출물의 분획별 항균효과를 확인할 수 있었다. 매실 추출물첨가 배지 상에서 C. fragariae의 생육이 크게 억제된다는 실험결과를 토대로 매실추출물의 항균활성물질이 미생물 세포내의 효소계의 효소활성을 저해에 기인한 것인가를 알아보기 위하여, 에너지생성대사에 관여하는 몇 가지 효소 활성(glucose-6-phosphate dehydrogenase, succinate dehydrogenase, malate dehydrogenase, hexokinase)에 미치는 매실추출물의 영향을 살펴보았다. 이 결과로 매실추출물은 총 추출물 및 친수성 분획이 일부 에너지 생성대사 효소계의 효소활성에 영향을 미치는 것으로 나타나(12-40%), 매실 추출물의 항균물질이 세포내로 침투되어 membrane에 존재하는 효소의 활성을 억제하는 것으로 추정할 수 있었다. 본 연구결과를 토대로 미루어, 매실추출물의 항균 작용은 세포내 에너지 생성에 관여하는 효소의 활성을 감소시키는 현상에 기인하는 것으로 추정되었다.
