The responses of whole body protein and glucose kinetics and of nitrogen (N) metabolism to non-protein energy intake (NPEI) were determined using an isotope dilution approach and measurement of N balance in three adult male goats. The diets containing 1.0, 1.5 and 2.0 times ME maintenance requirement, with fixed intake of CP (1.5 times maintenance) and percentage of hay (33%), were fed twice daily for each 21 d experimental period. After an adaptation period of 11 d, N balance was determined over 3 d. On day 17, whole body protein synthesis (WBPS) and glucose irreversible loss rate (ILR) were determined during the absorptive state by a primed-continuous infusion of [$^2H_5$]phenylalanine, [$^2H_2$]tyrosine, [$^2H_4$]tyrosine and [$^{13}C_6$]glucose, with simultaneous measurements of plasma concentrations of metabolites and insulin. Ruminal characteristics were also measured at 6 h after feeding over 3 d. Nitrogen retention tended to increase (p<0.10) with increasing NPEI, although digestible N decreased linearly (p<0.05). Increasing NPEI decreased (p<0.01) ammonia N concentration, but increased acetate (p<0.05) and propionate (p<0.05) concentrations in the rumen. Despite decreased plasma urea N concentration (p<0.01), increased plasma tyrosine concentration (p<0.05), and trends toward increased plasma total amino N (p<0.10) and phenylalanine concentrations (p<0.10) were found in response to increasing NPEI. Increasing NPEI increased ILR of both glucose (p<0.01) and phenylalanine (p<0.05), but did not affect ($p{\geq}0.10$) that of tyrosine. Whole body protein synthesis increased (p<0.05) in response to increasing NPEI, resulting from increased utilization rate for protein synthesis (p<0.05) and unchanged hydroxylation rate of phenylalanine ($p{\geq}0.10$). These results suggest that increasing NPEI may enhance WBPS and glucose turnover at the absorptive state and improve the efficiency of digestible N retention in goats, with possibly decreased ammonia and increased amino acid absorption. In addition, simultaneous increases in WBPS and glucose ILR suggest stimulatory effect of glucose availability on WBPS, especially when sufficient amino acid is supplied.

This study is a performance evaluation of a blood sugar monitoring system that combines a PPG sensor, which is an evaluation device for blood glucose monitoring, and a DNN algorithm when monitoring capillary blood glucose. The study is a researcher-led clinical trial conducted on participants from September 2023 to November 2023. PPG-BGMS compared predicted blood sugar levels for evaluation using 1-minute heart rate and heart rate variability information and the DNN prediction algorithm with capillary blood glucose levels measured with a blood glucose meter of the standard personal blood sugar management system. Of the 100 participants, 50 had type 2 diabetes (T2DM), and the average age was 67 years (range, 28 to 89 years). It was found that 100% of the predicted blood sugar level of PPG-BGMS was distributed in the A+B area of the Clarke error grid and Parker(Consensus) error grid. The MARD value of PPG-BGMS predicted blood glucose is 5.3 ± 4.0%. Consequentially, the non-blood-based PPG-BGMS was found to be non-inferior to the instantaneous blood sugar level of the clinical standard blood-based personal blood glucose measurement system.
Poly(3-hydroxybutyrate)(P(3HB)) accumulation under nutrient-rich condition with various amounts of $(NH_4)_2 SO_4$ was systematically investigated. The results of the electron-microscopy and the solvent extraction showed that the P(3HB) accumulation is unavoidable even under nutrient-rich condition. This indicates that in a two-step culture of Alcaligenes eutrophus H16, the researches should be careful in interpreting the data of polyhydroxyalkanoates(PHAs) accumulation in terms of the carbon-source fed in the second step because the two-step culture product contains the P(3HB) produced under nutrient-rich condition. The polyester production capability in a two-step batch culture of A. eutrophus H16(ATCC 17699) was also investigated using various saccharides and their derivatives such as glucose, fructose, gluconic acid, glucaric acid, sorbitol, lactose, galactose, and mannose. The polyesters synthesized were characterized by 500 MHz$^{1}H-NMR$ spectroscopy, intrinsic viscosity$[\eta]$ measurement in chloroform and differential scanning calorimetry(DSC). 500 MHz $^{1}H-NMR$ analysis showed that all polyesters synthesized generally contained 1~2 mol% of 3HV. Another finding is that the glucose utilization can be increased by changing the autoclaving procedure of the substrate to enhance the P(3HB) production yield up to 46 wt% of P(3HB) in dry cells.
Objectives : The purpose of this study was to investigate inhibitory effects of steamed Polygonatum odoratum extract (POE) on insulin resistance in rat skeletal muscle cells, L6 cells. Methods : Polygonatum odoratum (P. odoratum) extract was extracted with ethyl acetate. Activity of α-glucosidase in POE was measured for blood glucose regulation. MTT assay was examined for cell toxicity. Western blot analysis for measurement of adiponectine, peroxisome proliferator-activated receptorγ (PPARγ), insulin receptor substrate (IRS), glucose transporter 4 (Glut-4) and phosphorylation of serine/threonine-specific protein kinase (Akt) expressions were performed. Akt signaling pathway were analyzed with LY294002, which is a specific PI3K/Akt inhibitor. Results : The results revealed that POE inhibited α-glucosidase activity. Treatment of POE in L6 cells inhibited the differentiation of L6 cells compared to those of vehicl control. Additionally, protein expressions of adiponectine, PPARγ, IRS and Glut-4 were significantly regulated compared to those of vehicle control (p < 0.05), respectively. Futhermore, phosphorylation of Akt was increased in L6 cells treated with POE compared to that of vehicle control (p < 0.05). pAkt expression was significantly accentuated with Akt inhibitor (LY294002).Conclusions : These results suggest that POE may have potential as a natural agent for prevention/improvement of diabetes, especially, regulation of blood glucose. Therefore, further additional study should be conducted to elucidate in depth the pharmaceutical efficacy of these.

For the screening of antidiabetic effect, this study was carried out to some natural products by measurements of glucose level. MeOH and $CH_2Cl_2$ extracts of natural products were investigated for the effect on glucose level of $N_2$-STZ diabeted Rat. Among the natural products, Dioscorea Rhizoma, Trichosanthes Radix, Nelumbis Semem and Lycii Fructus were shown a remarkable antidiabetic effect.
Purpose : The purpose of this study was to analyze the reaction of recovery for the fatigue by sports massage treatment after Taekwondo training and then to verify the effectiveness. Subject : The subjects of this study were all the 20 high school Taekwondo players divided into two groups (rest recovery group and sports massage group) selected by systematic random assignment sampling. Method : The variables were the level of hemoglobin, lactic acid and glucose, and measured at rest, immediately-after exercise and 30-minute recovery. The data from this study were analyzed by using one-way ANOVA, independent sample t-test. Results : The results are as follows ; 1. The measurement of the hemoglobin, there was not the significant differences between experiment group and control group. 2. The lactate showed lower in the experiment group than in the control group in 30-minute recovery (p<.001). 3. The glucose showed lower in the experiment group than in the control group in 30-minute recovery (p<.01). So I think the sports massage treatment is more effective in the relief of fatigue than static rest method during the recovery after sports. However the massage treatment influence on the activation of metabolism according to the intensity and the duration of sports, and vary the effect of treatment according to the method and execution time. Therefore the longitudinal investigation of the variation by the long time massage through the more detailed program must be requisite for us.

The objective of the study was to observe the anti-diabetic effect of Solani nigri Herba(SH). The following experimental results were derived from the measurement of the levels of glucose, GOT and GPT in the serum and weight from a group of experimental rats by way of the injection of streptozotocin as well as the oral administration of Solani nigri Herba water extracts. The glucose level in the serum significantly decreased in the Solani nigri Herba-dosed group(SH2,12.6 mg/100 g rat weight) on the 10th day. The GOT level of SHI(4.2 mg/100 g Solani nigri Herba) and SH2 in the serum slightly decreased, but no significance to compare with control group. The GPT level of SH1 and SH2 in the serum slightly decreased, but no significance to compare with control group. The weight of rat of SH1 and SH2 in the serum slightly increased, but no significance to compare with control group. In conclusion, Solani nigri Herba appeared to have significance inhibitory effects on the rise of blood sugar level in the experimentally induced diabetics.
The physicochemical and in vitro physiological properties of soluble dietary fiber (SDF) from wax gourd (Benincasa hispida) pulp and peel were investigated. The pulp was composed of 11.4% SDF and 24.3% insoluble dietary fiber (IDF), while the peel contained 3.2% SDF and 43.3% IDE The predominant sugar in the SDF of the wax gourd pulp and peel was uronic acid, followed by galactose and rhamnose. The SDFs from the wax gourd pulp and peel gave similar elution patterns, with 4 main neutral sugar and uronic acid peaks eluted by 0.4, 0.5, 1, and 2 M ammonium acetate buffer. The pulp SDF had a much higher glucose retardation index (GRI) than the peel SDF for all measurement times. The pulp SDF showed strong growth-inhibiting activity against Escherichia coli and Clostridium perfringens, whereas the peel SDF produced strong growth-promoting activity against Bifidobacterium longum, Bifidobacterium infantis, and Lactobacillus brevis when compared to glucose.
A new method for enzyme immobilization has been developed to remove interference by potential interferents in body fluids. Instead of using electron mediators, we chose direct hydrogen peroxide measurement route. Extremely hydrogen peroxide-selective polymer was coated as an inner membrane to exclude interferents and then glucose oxidase(GOx) was entrapped by electropolymerization of inert monomers. There was no solvent casting step throughout the whole fabrication procedure but all membranes on Pt-Ir electrode were formed by electropolymerization. Thus, membrane thickness, quantity of enzyme loaded and can be controlled by electrochemical parameters. As a result, reproducibility of biosensor characteristics becomes remarkably improved in terms of mass production.
A dipstick-type immunochemical biosensor for the detection of the organophosphorus insecticide fenthion was developed using a screen-printed electrode system as an amperometric transducer with polyclonal antibodies against fenthion as a bioreceptor. The assay of the biosensor involved competition between the pesticide in the sample and pesticide-glucose oxidase conjugate for binding to the antibody immobilized on the membrane. This was followed by measurement of the activity of the bound enzyme by the supply of the enzyme substrate (glucose) and amperometric determination of the enzyme reaction product ($H_2O_2$). The activity of the bound enzyme was inversely proportional to the concentration of pesticide. The optimized sensor system showed a linear response against the logarithm of the pesticide concentration ranging from $10^{-2}$ to $10^3\;{\mu}g/L$.
