We have isolated endophytic fungi from the Indian yew tree, Taxus baccata, and then screened for taxol production. Out of the 40 fungal cultures screened, one fungus Gliocladium sp. was found to produce taxol and 10-DABIII (10-deacetyl baccatin III). These compounds were purified by TLC and HPLC and characterized using UV-spectroscopy, ESI-MS, MS/MS, and proton NMR. One liter of Gliocladium sp. culture yielded $10\;{\mu}g$ of taxol and $65\;{\mu}g$ of 10-DABIII. The purified taxol from the fungus showed cytotoxicity towards cancer lines HL-60 (leukemia), A431 (epidermal carcinoma), and MCF-7 (breast cancer).

Secalonic acid D(SAD) was isolated as the major secondary metabolite of the marine lichen-derived fungus Gliocladium sp. T31. Its structure was established on the basic of physicochemical and spectroscopic data. This is the first report on the isolation of SAD from this fungus, as well as its inhibitory effect on K562 cell cycle and its cytotoxicity against several tumor cell lines in vitro.

Antagonistic microorganisms from rhizosphere soil were isolated, identified, and applied successfully as the biocontrol agents of damping-off caused by Rhizoctonia spp. Rhizosphere antagonists isolated from rhizosphere soil were identified as Trichoderma viride, T. harzianum, T. hamatum, T. polysporum, Gliocladium sp., Pseudomonas fluorescence, P. stutzeri, P. cepacia, Enterobacter sp., Serratia sp. and Erwinia herbicola. Of these, the most promising ones in vitro were T. virdie, T. harzianum, Gliocladium sp., Serratia sp., P. stutzeri, and P. cepacia. These above six antagonists were efficient in reducing disease incidence to $40{\sim}70%$ when the reselected rhizosphere antagonists preparations were applied to the soil at $10^6$ propagules per gram. Among six antagonists, T. viride was the most promising biocontrol agents against R. solani isolates in soil. The suppressive effect was more evident in steam-sterilized soil than in non-sterilized field soil.

Biocontrol agents, Gliocladium virens G872B and Pseudomonas putida Pf3, were compatible each other in colonizing cucumber rhizosphere, which contributed to a long-term inhibition of cucumber Fusarium wilt. G872B colonized successfully on the cucumber root system, irrespective of the introduction of Pf3. Pf3 also colonized well in the cucumber rhizosphere regardless of the presence of G872B. The individual strains effectively suppressed cucumber wilt up to 56 days after transplanting. The combined treatment of G872fB and Pf3 provided a long-term protection of about 80 days with the efficacy greater than that obtained by any individual strains under greenhouse conditions. These results suggest that the colonization of the biological control agents in the rhizosphere could be correlated directly to Fusarium wilt-suppressive potentials.

The endophytic fungal populations of different tissues of Taxus baccata grown at high altitudes in West Bengal, India were explored. These isolated fungal populations represented different genera, which were screened for taxol production using immunoassay technique. The culture AAT-TS-$4_1$ that produced taxol was identified as Gliocladium sp. based on its cultural, morphological characteristics, internal transcribed spacer, and 18S rRNA sequence analysis. Kinetics of taxol production as a function of culture growth were investigated.

Bacteria and fungi antagonistic to Fusarium oxysporum f. sp. cucumerinum Owen were effectively isolated with each of modified Triple Layer Agar (TLA) technique from rhizosphere soil where cucumber had been grown healthily in plastic film house. Three predominant bacterial isolates selected were identified as Pseudomonas fluorescens, and P. putida, Serratia sp. and three fungal isolates were Gliocladium sp. Trichoderma harzianum, and T. viride. Antagonistic bacteria inhibited $26-45\%$ of germination and $41-56\%$ of germ tube elongation of microconidia of F. oxysporum f. sp. cucumerinum on Water Agar (WA). P. fluorescens was the strongest inhibitor. Several my co parasitisms were observed on dual culture of WA between antagonistic fungi and F. oxysporum f. sp. cucumerinum such as coiling, penetration, overgrowing, and lysis. Mycelial lysis of the pathogen was the most severe at pH 4.6, followed by 3.6, 5.6 and 6.6 of the medium in decreasing order. At pH 6.6, mycelia of the pathogen were not conspicuously damaged, however, the antagonistic fungi formed abundant chlamydospores especially Gliocladium sp. T. harzianum revealed the most excellent antagonism in vitro.

Fifteen PGPF (plant growth promoting fungi) isolates were selected from 728 fungal isolates collected from rhizosphere of zoysiagrass in Korea. Identification of the 15 isolates was based on their morphological characteristics. They were classified as Gliocladium sp. (n=1), Penicillium sp. (n=5), Trichoderma sp. (n=3), Fusarium sp. (n=3), and unidentifed species (n=3). Of the 15 isolates, six (PF-31, PF-136, PF-238, PF-255, PF-395, PF-420) significantly promoted the growth of tomato seedlings, and three (PF-31, PF-101, PF-255) also promoted the growth of hot pepper and two (PF-31, PF-225) also promoted the growth of cucumber, The 15 PGPF isolates were divided into 4 groups based on root colonizing ability. Isolates PF-17, PF-101 and PF-225 were included in the group 1, which had high root colonizing ability.

토양에 존재하는 진균인 Gliocladium virens는 식물병을 감소 또는 방제할 수 있는 생물학적인 특성에 의하여 G. virens는 지난 수십년간 실용가능성이 큰 생물학적 방제균(또는 길항균)으로 집중적으로 연구되었다. 이 균이 식물병의 발생을 감소시키는 생물적 방제효과는 항생작용, 중복기생, 근권에서의 생존과 집단번식, 뿌리표면에서의 정착 등에 의한 것으로 분류되고 있다. 특히, 항생물질인 gliotoxin, gliovirin, viridin 등은 Rhizoctonia solani 및/또는 Pythium spp. 등에 항생효과가 뚜렷하고, 식물병의 발생과 직접적인 상관관계를 나타내고 있어 G. virens의 식물병의 방제에 관련된 중요한 작용기작으로 제시되어 있다. 또한, 근권에서 이균의 생존과 집단증식 및 뿌리표면에서의 정착은 식물병의 방제와 상관관계를 나타낼 수 있는 중요한 작용기작으로 제시되고 있다. 그러나 이균이 R. solani 등에 기생하는 현상은 식물병의 생물적 방제의 직접적인 연관관계를 나타내고 있지 않다. G. virens을 이용의 생물적 방제효과를 증진시키기 위한 방법으로 다음과 같은 두 가지 방법을 들 수 있다. 첫째, 길항효과가 높은 G. virens 균주를 선발하기 위하여 여러 종류의 토양에서 길항력이 높은 G. virens의 선발이 지난 수십년간 진행되고 있다. 또한, 특정 길항효과를 발현하는 유전자를 G. virens의 염색체에 도입하고 이를 발현시킴으로써 생물적 방제효과를 증진시키는 것으로 이러한 방법은 1980년 후반부터 진행되고 있다. 둘째, G. virens의 길항효과가 최대의 효율로 발현될 수 있도록 최적의 미세환경을 갖추고 있으며 농민이 편리하게 사용할 수 있는 G. virens의 운송매체의 개발이 중요하다. 운송매체의 개발에 의한 'Glioguard'는 G. virens의 포자를 alginate 입자에 제형화한 것으로서 미국에서 시판되고 있다. Aldicarb, metalaxyl, atrazine 등의 농약을 분해할 수 있는 능력은 G. virens의 다른 생물적 특성중의 하나이다. 특히, parathion을 분해할 수 있는 Flavobacterium sp.의 유전자(opd)가 G. virens의 염색체에 도입되여 발현될 수 있는 방법이 제시되었으며, 이는 G. virens을 이용한 토양에서의 특정한 농약의 분해효율을 증진시킬수 있는 가능성을 제시한 것이다. 그러나, G. virens를 이용한 농약의 생물적 분해에 관한 연구는 기초단계로 평가되고 있으며, 포장에서 이를 실용화하기 위해서는 향후 지속적인 연구가 필요하다.

This study was undertaken to find a new formulation of soil amendment, and selection of antogonists and to effectively control brown and large patch of turfgrasses caused by Rhizoctoniz solani AG1-1 and AG 2-2. Fourteen inorgainc chemicals (1%, w/w) were added individually in vitro, and some chemicals showed suppressiveness to R. solani. Alum suppressed effectively mycelial growth of R. solani in the range of 17 to 77% as compared with control. The four chemicals such as Al2(SO4)3, alum, CaO, and NH4NO3 were finally selected. Out of three organic compounds, composted pine bark (CPB) showed prominent suppressive effect as compared with milled alfalfa and pine leaves. After inoculation of R. solani isolates AG-1 and AG2-2 on the turf seedlings, water soaked lesions and blight symptoms were developed on the whole seedlings. According to inhibition zone method, mycelial growth of the fungus were greatly suppressed by culture filterates of the antagonists, Gliocladium virens (Gl1-) and Pseudomonas sp. (P713). CPB soil amendment mixed with antagonists (1% w/w) controlled not only brown and large patch of turfgrasses, but also promote the good growth of the seedlings. In addition, the controlling effect was maintained more than 30 days. Especially, the controlling effect of two antagonists was similar to Cㅖㅠ soil amendment with the antagonists and also stimulated a favorable growth of the seedlings. Therefore, its is expected that continuous control of Rhizoctonia blight of turfgrasses can be obtained in field by subsequent applications of the antagonists.
