식물공장에서 생산된 새싹인삼의 크기에 따른 진세노사이드 함량 및 항산화 활성 비교 (Comparison of ginsenoside contents and antioxidant activity according to the size of ginseng sprout has produced in a plant factory)

  • 본 연구에서는 식물공장을 통해 생산된 새싹인삼을 크기에 따라 분류하고 이를 지상부와 지하부로 나누어 진세노사이드 함량과 항산화 활성을 비교하였다. 지상부의 경우 총 phenolic 함량은 중간 크기인 M에서 5.16 GAE mg/g로 가장 높았으며 가장 큰 크기인 L에서 2.23 GAE mg/g으로 가장 낮은 함량을 보였다. 지하부 역시 M 크기에서 가장 높은 함량을 보였으나, 큰 차이를 보이지 않았다. 한편, 총 flavonoid 함량 역시 지상부(5.16 RE mg/g) 와 지하부(1.28 RE mg/g) 모두 M 크기에서 높은 함량을 보였다. 지상부의 주요 진세노사이드는 Re (20.33-24.15 mg/g) > Rd (11.36-27.42 mg/g) > Rg1 (4.48-5.54 mg/g) 순 있었고 지하부는 Rb1 (5.09-8.61 mg/g) > Re (4.48-5.54 mg/g) > Rc (3.11-4.11 mg/g) 순 있었다. M 크기의 경우 Re와 Rd는 각각 지상부에서 24.15 mg/g과 27.42 mg/g 및 지하부에서 5.20 mg/g과 1.43 mg으로 약 4배와 19배 높은 함량을 보였다. 지상부에는 F3 및 Rh1이 검출되었으나, 지하부에서는 검출되지 않았다. DPPH (74.95%)와 ABTS (94.47%), hydroxyl (70.39%) 라디칼 소거 활성 및 FRAP (2.169) 활성은 다른 크기들보다 M 크기에서 가장 높은 활성을 보였다.

홍삼음료의 함량 및 안전성 실태조사 (A Survey on the Content and Safety of Red Ginseng Products)

  • 대장균군은 유통형, 맞춤형 및 건강기능형 홍삼음료 모두에서 음성이었다. 일반세균은 유통형 홍삼음료 및 건강기능형 홍삼음료에서는 검출되지 않았으나, 맞춤형 홍삼음료에서는 58건 중 13건(22.4%)이 검출되었다. 총 100건의 제품 중 5건의 제품에서 안식향산이 검출되었다. 함량은 19.2-543.0 mg/kg이며 이 중 유통형 홍삼음료에서 검출된 1건의 제품(19.2 mg/kg)과 건강기능형 홍삼음료 1건(31.4 mg/kg)의 함량은 원료에서 유래된 것으로 판단되었다. 유통형 홍삼음료의 납 평균 함량은 62.3 ${\mu}g/kg$, 맞춤형 홍삼음료는 74.6 ${\mu}g/kg$, 건강기능형 홍삼음료는 28.8 ${\mu}g/kg$으로 나타났다. 카드뮴의 평균 함량은 유통형 1.15 ${\mu}g/kg$, 맞춤형 1.62 ${\mu}g/kg$, 건강기능형 홍삼음료는 4.18 ${\mu}g/kg$으로 조사되었다. 수은의 평균 함량은 0.10-0.18 ${\mu}g/kg$으로 조사되었다. 홍삼 건강기능식품의 지표성분인 Rg1과 Rb1함량은 유통형 및 맞춤형 홍삼음료가 0.1-23.4 mg/90 mL으로 함유되어 있었다. 하루에 3회 섭취한다고 가정하고 건강기능식품의 일일 섭취량 3-80 mg/day와 비교해보면, 이번에 조사된 37건의 유통형 홍삼음료 중에서 22건(59%), 맞춤형 홍삼음료 58건에서는 26건(45%)이 건강기능식품에 해당하는 지표성분(Rg1과 Rb1)을 함유하고 있는 것을 알 수 있었다. 그러나 홍삼음료들 간에 서로 홍삼성분의 함량차이가 크고 건강기능식품 섭취기준(3-80 mg/day) 양에 미치지 못한 47건 중 1회 분량 90 mL 기준 홍삼성분이 0.2 mg 이하로 함유되어 있는 제품도 20 건이나 되는 것으로 나타났다.

홍삼 Ext 수용액의 살균과 저장 중 성분의 변화와 생성된 침전물의 이화학적 특성 (Changes in Chemical Components of Red Ginseng Extract Solution and Physicochemical Properties of Precipitates Formed During Pterilization and Storage)

  • Red Ginseng extracts sol'n was sterilized at 85f for 20 mins and/or stored at 4$0^{\circ}C$ for 6 months and centrifuged for 20 mins at 8,500xg in order to investigate the changes in chemical components of supernatants and the properties of precipitates. Contents of crude saponin and ginsenoside-$Rb_1$, -$Rg_1$, -Re were partially decreased during heating and storage. Starch contents were decreased from 26.81% in red ginseng extracts to 17.50-8.81% in supernatants, whereas free sugar contents were increased from 15.50% to 20.29~21.35% by heating and storage. The contents of protein and minerals in supernatants were decreased, but acidic polysaccharides and polyphenol compounds were not changed. pH values of supernatants and precipitates were decreased. The absorbances of brown color precursor and brown pigment in precipitates, detected at 285 nm and 440 nm were remarkably increased. The Overa11 data suggest that precipitates in red ginseng extracts sol'n formed during steilization and storage are provably the brown pigments resulting from Maillard reaction of amino compounds with reducing sugar which could be released from starch and protein matrix and $Cu^+$, $Ca^{2+}$ and $Fe^{3+}$ ions are implicated with the reaction incorporated.

세포배양에 의한 고려인삼 성분의 생산연구 (The Production of Ginseng Saponins with the Cell Culture of Korean Ginseng Plant)

  • Panax ginseng root has been widely used as an important drug for thousands years in China, Korea and Japan. The main effective components of ginseng have been believed to be saponins. However, ginseng cultivation is very difficult and needs many years for growth. It has already been shown that Panax ginseng callus produces a considerable amount of the same kinds of saponins as in intact plants. Various culture conditions were examined for increased production of ginseng saponins by cell culture. The saponin contents and the growth rates in two cell lines of ginseng callus were compared in static and suspension cultures, rotary and reciprocal shaking cultures. It was shown that the growth rate in rotary shaking cultures of D5-B2K-B2K callus was the highest and ginseng saponin production was most effective in reciprocal cultures of D5-B2K-B2K callus. The saponin content per fresh weight of the culture was 1.03 times higher than that of the fresh ginseng root.

흑삼 농축액 첨가 수준에 따른 흑삼 젤리의 품질 특성 (Quality Characteristics of Black Ginseng Jelly)

  • The principal objective of this study was to evaluate the quality characteristics of black ginseng jelly prepared with different 5 levels(0, 0.5, 1.0, 1.5, and 2.0%) of black ginseng extract. We assessed the ginsenosides level of white and black ginseng for comparison between white and black ginseng. And we conducted the pH, sugar content, Hunter's color values, the mechanical characteristics and sensory evaluation of black ginseng jelly samples. The levels of ginsenoside $Rg_3,\;Rh_1$, and $Rh_2$ of black ginseng were higher than those of white ginseng. The more black ginseng extract was increased, the sugar contents of black ginseng jelly were significantly increased(p<0.05). We noted that the luminance and Hunter's b values of jelly samples were decreased according to black ginseng extract was increased, but in Hunter's a values 0.5% black ginseng jelly was the highest of the all. With regard to the mechanical properties of the black ginseng jelly samples, the score of hardness, gumminess and chewiness were significantly increased. In color, taste and overall quality, the score of jelly with 1.0% black ginseng extract was significantly increased than those of the all.

A Structure-Function Relationship Exists for Ginsenosides in Reducing Cell Proliferation and Inducing Apoptosis in THP-1 Cells

  • Ginsenosides of 20(S)-protopanaxadiol and 20(S)-protopanaxatriol classification including the aglycones, PD, PI and ginsenosides Rh2, Rhl were shown to posses characteristic effects on proliferation of THP-l human leukaemia cells. A similar result was not apparent for ginsenoside Rg3 or dexamathasone. The concentration to inhibit $50\%$ of cells $(LC_{50})$ for PD, Rh2, PI and Rhl were 13 ${\mu}g/mL,\;15{\mu}g/mL,\;19{\mu}g/mL\;and\;210\;{\mu}g/mL$ respectively. Cell cycle analysis showed apoptosis with PD and PI treatment of THP-1 cells resulting in a build up of sub-G1 cells after 24, 48 and 72 hours of treatment. Rh2, and dexamathasone treatments also increased apoptotic cells after 24 hours, where as Rhl did not. After 48 and 72 hours Rh2, Rhl and dexamathasone similarly increased apoptosis, but these effects were significantly (P<0.05) lower than observed for both PD and PI treatments. Furthermore, treatments that produced the largest build up of apoptotic cells were also found to have the largest release of lactate dehydrogenase (LDH). It can be concluded from these studies that the presence of sugars to PD and PI aglycone structure reduces the potency to induce apoptosis, and alternately alter membrane integrity. These cytotoxic effects to THP-l cells were different from dexamethasone.

  • Panax ginseng Meyer is a traditional Chinese medicine that is widely used as tonic in Asia. The main pharmacologically active components of ginseng are the dammarane-type ginsenosides, which have been shown to have anti-cancer, anti-inflammatory, immunoregulatory, neuroprotective, and metabolic regulatory activities. Moreover, some of ginsenosides (eg, Rh2 and Rg3) have been developed into nutraceuticals. However, the utilization of ginsenosides in clinic is restrictive due to poor permeability in cells and low bioavailability in human body. Obviously, the dammarane skeleton and glycosyls of ginsenosides are responsible for these limitations. Therefore, improving the oral bioavailability of ginsenosides has become a pressing issue. Here, based on the structures of ginsenosides, we summarized the understanding of the factors affecting the oral bioavailability of ginsenosides, introduced the methods to enhance the oral bioavailability and proposed the future perspectives on improving the oral bioavailability of ginsenosides.

  • Korean Red Ginseng (KRG) has long been used not only as a food supplement but also as a treatment for various diseases. Ginseng originated in South Korea, which later spread to China and Japan, has a wide range of pharmacological activities including immune, endocrine, cardiovascular, and central nervous system effects. KRG is produced by repetitions of steaming and drying of ginseng to extend preservation. During this steaming process, the components of ginseng undergo physio-chemical changes forming a variety of potential active constituents including ginsenoside-Rg3, a unique compound in KRG. Pandemic Coronavirus disease 2019 (COVID-19), has affected both men and women differentially. In particular, women were more vulnerable to COVID-related distress which in turn could aggravate menopause-related disturbances. Complementary and alternative medicinal plants could have aided middle-aged women for several menopause-related symptoms during and post COVID-19 pandemic. This review aimed to explore the beneficial effects of KRG on menopausal symptoms and gynecological cancer.
