Cytotoxicity of Compound K (IH-901) and Ginsenoside Rh2, Main Biotransformants of Ginseng Saponins by Bifidobacteria, against Some Tumor Cells

  • When ginsenoside $R_{*}$b1/ and $R_{b2}$ were anaerobically incubated with human fecal microflora, these ginsenosides were metabolized to compound K (IH-901). When ginsenoside $R_{g3}$ was anaerobically incubated with human fecal microflora, the ginsenoside $R_{g3}$ was metabolized it to ginsenoside $R_{h2}$. Among ginsenosides, IH-901 and 20(S)-ginsenoside $R_{h2}$ exhibited the most potent cyotoxicity against tumor cells: 50% cytotoxic concentrations of IH-901 in the media with and without fetal bovine serum (FBS) were 27.1-31.6 $\mu$M and 0.1-0.61 $\mu$M, and those of 20(S)-ginsenoside $R_{h2}$ were 37.5->50 and 0.7-7.1 $\mu$M, respectively. The cytotoxic potency of ginsenosides was IH-901>20(S)-ginsenoside R $h_{h2}$》20(S)-ginsenoside $R_{g3}$>ginsenoside $R_{b1}$(equation omitted) $R_{b2}$.EX>$R_{b2}$./.

Protective Effect of Ginsenoside R0 on Anoxic and Oxidative Damage In vitro

  • To examine the neuroprotective effects of ginsenoside R0, we investigated the effects of ginsenoside R0 in PC12 cells under an anoxic or oxidative environment with Edaravone as a control. PC12 neuroendocrine cells were used as a model target. Anoxic damage or oxidative damage in PC12 cells were induced by adding sodium dithionite or hydrogen peroxide respectively in cultured medium. Survival ratios of different groups were detected by an AlamarBlue assay. At the same time, the apoptosis of PC12 cells were determined with flow cytometry. The putative neuroprotective effects of ginsenoside R0 is thought to be exerted through enhancing the activity of antioxidant enzymes Superoxide dismutases (SOD). The activity of SOD and the level of malondialdehyde (MDA) and intracellular reactive oxygen species (ROS), were measured to evaluate the protective and therapeutic effects of ginsenoside R0. Ginsenoside R0 treated cells had a higher SOD activity, lower MDA level and lower ROS, and their survival ratio was higher with a lower apoptosis rate. It is suggested that ginsenoside R0 has a protective effect in the cultured PC12 cells, and the protection efficiency is higher than Edaravone. The protective mechanisms of these two are different. The prevent ability of ginsenoside R0 is higher than its repair ability in neuroprotection in vitro.

백삼 및 홍삼 농축액의 사포닌 분석 (Analysis of Ginsenosides of White and Red Ginseng Concentrates)

  • 백삼 가공품과 홍삼 가공품의 사포닌 분포 내용과 함량을 비교하기 위하여 시판되고 있는 백삼 농축액(WGC)과 홍삼 농축액(RGC)을 각각 1종 선정하여 조 사포닌의 함량과 개별 ginsenoside의 함량분포를 조사하였다. Shibata의 방법과 우리나라 식품공전에 따라 측정한 조 사포닌의 양은 WGC가 각각 10.65와 21.77%이었으며 RGC는 5.80와 10.94%이였고, HPLC에 의한 총 사포닌의 양은 WGC가 7.40와 10.64%, RGC는 3.31와 3.13%로서 백삼 농축액의 사포닌 함량이 홍삼 농축액의 경우 보다 전반적으로 높았다. HPLC로 분석한 인삼 사포닌, ginsenoside $Rb_1,\;Rb_2,\;Rc,\;Rd,\;Re,\;Rf,\;Rg_1,\;20(S)\;Rg_3,\;20(R)Rg_3,\;20(S)\;Rh_1$ 그리고 $20(R)\;Rh_1$ 이었으며 대부분 홍삼농축액 보다는 백삼농축액의 함량이 높았으며, 특히 ginsenoside $Rb_1,\;Rg_1$ 그리고 $Rb_2$은 백삼 농축액에 3배 이상 더 함유되어 있었다. 또한 protopanaxadiol group과 protopanaxatriol group의 비율(PD/PT)에 있어서는 농축액간의 차이는 크지 않았다. 홍삼의 특유 사포닌으로 알려진 20(S)- 및 20(R)-ginsenoside $Rg_3$가 WGC와 RGC에 비슷하게 분포하는 것으로 확인되었다. 20(S)-ginsenoside $Rg_3$의 조 사포닌 조제법에 따라 RGC에서 0.48과 0.47% WGC에 0.40와 0.53%, 20(R)-ginsenoside $Rg_3$도 RGC에 0.10과 0.11%, WGC에 0.14와 0.22%이었다.

Application of Near Spectroscopy for Nondestructive Evaluation of Ginsenoside Content

  • The effective component Ginsenoside in Ginseng has been widely used to cure some hypochondriasis and be as supplementary medicines. There are many chemical analysis methods to measure the contents of Ginsenoside in Ginseng; however, all these methods have some shortcomings such as long time, environmental pollution and damaging the samples. In this paper, it is possible to use near infrared spectroscopy to measure the content of Ginsenoside in Ginseng without destruction. As the results, Rg1, Rb1, Re and T-Saponin of Ginsenoside can be measured with the accuracy of R(0.81) SEP (1.704 mg/g), R(0.74) SEP (1.211 mg/g), R (0.78) SEP (1.049 mg/g) and R(0.84), SEP(4.537 mg/g).

  • 인삼 saponin 중에서 조성 비율이 가장 큰 ginsenoside R $b_1$을 약효면에서 훨씬 우수한 ginsenoside-Rd로 선택적 전환할 수 있는 전환효소를 Rhizopus japonicus 배양물로부터 순수하게 정제한 후 그 효소학적 특성을 조사하였다. 본 효소는 pH 4.8~5.0, 45$^{\circ}C$에서 그 활성도가 가장 높았으며 pH4~9의 범위에서 안정하였고, pH5.0에서 6$0^{\circ}C$ 열처리할 경우 50% 실활하는데 2시간이 소요되었다. M $n^{++}$, F $e^{++}$에 의해 효소활성이 촉진되었으며 C $u^{++}$, A $g^+$에 의해서는 저해되었다. 효소 저해제중에는 EDTA, o-phenanthroline 등에 의해 저해되기도 했다. 본 효소는 당류중에는 gentiobiose, cellulose만을, glycoside 중에서는 amygdalin, purnasin, salicin 등을 분해할 수 있었으며 ginsenoside R $b_1$, 이외의 다른 ginsenoside는 전혀 분해할 수 없었다. 본 효소의 Km치는 total saponin 기질 및 ginsenoside Rb group saponin 기질은 ginsenoside R $b_1$으로서 5.0mM 이었으며, gentiobiose는 4.8mM, amygdalin은 3.7mM 이었다.

Effects of Panax notoginseng, ginsenoside Rb1, and notoginsenoside R1 on proliferation of human breast carcinoma MCF-7 cells

  • In this study, we evaluated the antiproliferative effects of Panax notoginseng, ginsenoside Rb1, and notoginsenoside R1 in the human breast carcinoma MCF-7 cell line. Our results indicated that both Panax notoginseng radix extract (NRE) and Panax notoginseng rhizoma extract (NRhE) possess significant antiproliferative activities in MCF-7 cells. Compared to control group (100%), at the concentrations of 0.05, 0.5, and 1.0 mg/ml NRE, cell growth was concentration-dependently reduced to 81.0 ${\pm}$ 6.1 (P < 0.01), 34.2 ${\pm}$ 4.8 (P < 0.001), and 19.3 ${\pm}$ 1.9 (P < 0.001), respectively. Similar results with NRhE at concentrations of 0.5 and 1.0 mg/ml were obtained in these MCF-7 cells. To identify the responsible chemical constituent, we tested the antiproliferation effects of two representative saponins, ginsenoside Rb1 and notoginsenoside R1, on the MCF-7 cells. The data showed that ginsenoside Rb1 was endowed with antiproliferative properties, while notoginsenoside R1 did not have an inhibitory effect in the concentrations tested. Our studies provided evidence that Panax notoginseng extracts and ginsenoside Rb1 may be beneficial, as adjuvants, in the treatment of human breast carcinoma.

Highly Efficient Biotransformation of Notoginsenoside R1 into Ginsenoside Rg1 by Dictyoglomus thermophilum β-xylosidase Xln-DT

  • Notoginsenoside R1 and ginsenoside Rg1 are the main active ingredients of Panax notoginseng, exhibiting anti-fatigue, anti-tumor, anti-inflammatory, and other activities. In a previous study, a GH39 β-xylosidase Xln-DT was responsible for the bioconversion of saponin, a natural active substance with a xylose group, with high selectivity for cleaving the outer xylose moiety of notoginsenoside R1 at the C-6 position, producing ginsenoside Rg1 with potent anti-fatigue activity. The optimal bioconversion temperature, pH, and enzyme dosage were obtained by optimizing the transformation conditions. Under optimal conditions (pH 6.0, 75℃, enzyme dosage 1.0 U/ml), 1.0 g/l of notoginsenoside R1 was converted into 0.86 g/l of ginsenoside Rg1 within 30 min, with a molar conversion rate of approximately 100%. Furthermore, the in vivo anti-fatigue activity of notoginsenoside R1 and ginsenoside Rg1 were compared using a suitable rat model. Compared with the control group, the forced swimming time to exhaustion was prolonged in mice by 17.3% in the Rg1 high group (20 mg/kg·d). Additionally, the levels of hepatic glycogen (69.9-83.3% increase) and muscle glycogen (36.9-93.6% increase) were increased. In the Rg1 group, hemoglobin levels were also distinctly increased by treatment concentrations. Our findings indicate that treatment with ginsenoside Rg1 enhances the anti-fatigue effects. In this study, we reveal a GH39 β-xylosidase displaying excellent hydrolytic activity to produce ginsenoside Rg1 in the pharmaceutical and food industries.

Effects of Ginsenoside Rg3 Epimers on Swine Coronary Artery Contractions

  • The previous reports demonstrated that ginseng saponins, active ingredient of Panax ginseng, inhibited blood vessel contraction induced by various hormones or high $K^+$. Recently, we demonstrated that 20(R)- and 20(S)-ginsenoside $Rg_3$. regulate ion channel activities with differential manners. The aim of this study was to examine whether ginsenoside $Rg_3$ isomers also show differential effects on swine coronary artery contractionresponses induced by high $K^+$, serotonin (5-HT) or acetylcholine. Treatment of 20(S)- but not 20(R)-ginsenoside $Rg_3$ caused a concentration-dependent relaxation of coronary artery contracted by 25mM KCI. 20(S)- and 20(R)-ginsenoside $Rg_3$ induced significant relaxations of coronary artery contraction induced by 5-HT $(3{\mu}M)$ in the presence of endothelium with concentration-dependent manner and, also in the absence of endothelium only 20(S)-ginsenoside $Rg_3$ induced a strong Inhibition of coronary artery contraction induced by 5-HT in a concentration-dependent manner. 20(S)-ginsenoside $Rg_3$ caused relaxation of coronary artery in the absence and presence of endothelium. In contrast, treatment of 20(S)- and 20(R)-ginsenoside $Rg_3\;(100{\mu}M)$ did not show significant inhibition of coronary artery contraction induced by acetylcholine $(0.01\;to\;30{\mu}M)$ in the presence of endothelium, whereas both isomers caused significant inhibition of coronary artery contraction induced by acetylcholine $(0.01\;to\;30{\mu}M)$ in the absence of endothelium in a concentration-dependent manner. These findings indicate that 20(S)-or 20(R)-ginsenoside $Rg_3$ exhibits differential relaxation eff3cts of swine coronary artery contractions caused by high $K^+$, acetylcholine, and 5-HT treatment and that this differential vasorelaxing effects of ginsenoside $Rg_3$ isomers also might be dependent on endothelium.

Production of the Rare Ginsenoside Rh2-MIX (20(S)-Rh2, 20(R)-Rh2, Rk2, and Rh3) by Enzymatic Conversion Combined with Acid Treatment and Evaluation of Its Anti-Cancer Activity

  • The ginsenoside Rh2 has strong anti-cancer, anti-inflammatory, and anti-diabetic effects. However, the application of ginsenoside Rh2 is restricted because of the small amounts found in Korean white and red ginsengs. To enhance the production of ginsenoside Rh2-MIX (comprising 20(S)-Rh2, 20(R)-Rh2, Rk2, and Rh3 as a 10-g unit) with high specificity, yield, and purity, a new combination of enzymatic conversion using the commercial enzyme Viscozyme L followed by acid treatment was developed. Viscozyme L treatment at pH 5.0 and $50^{\circ}C$ was used initially to transform the major ginsenosides Rb1, Rb2, Rc, and Rd into ginsenoside F2, followed by acid-heat treatment using citric acid 2% (w/v) at pH 2.0 and $121^{\circ}C$ for 15 min. Scale-up production in a 10-L jar fermenter, using 60 g of the protopanaxadiol-type ginsenoside mixture from ginseng roots, produced 24 g of ginsenoside Rh2-MIX. Using 2 g of Rh2-MIX, 131 mg of 20(S)-Rh2, 58 mg of 20(R)-Rh2, 47 mg of Rk2, and 26 mg of Rh3 were obtained at over 98% chromatographic purity. Then, the anti-cancer effect of the four purified ginsenosides was investigated on B16F10, MDA-MB-231, and HuH-7 cell lines. As a result, these four rare ginsenosides markedly inhibited the growth of the cancer cell lines. These results suggested that rare ginsenoside Rh2-MIX could be exploited to prepare an anti-cancer supplement in the functional food and pharmaceutical industries.
