New hydroperoxylated and 20,24-epoxylated dammarane triterpenes from the rot roots of Panax notoginseng

  • Background: Root rot is a serious destructive disease of Panax notoginseng, a famous cultivated araliaceous herb called Sanqi or Tianqi in Southwest China. Methods: The chemical substances of Sanqi rot roots were explored by chromatographic techniques. MS, 1D/2D-NMR, and single crystal X-ray diffraction were applied to determine the structures. Murine macrophage RAW264.7 and five human cancer cell lines were used separately for evaluating the antiinflammatory and cytotoxic activities. Results and Conclusion: Thirty dammarane-type triterpenes and saponins were isolated from the rot roots of P. notoginseng. Among them, seven triterpenes, namely, 20(S)-dammar-25-ene-24(S)-hydroperoxyl-3β,6α,12β,20-tetrol (1), 20(S)-dammar-3-oxo-23-ene-25-hydroperoxyl-6α,12β,20-triol (2), 20(S)-dammar-12-oxo-23-ene-25-hydroperoxyl-3β,6α,20-triol (3), 20(S)-dammar-3-oxo-23-ene-25-hydroperoxyl-12β,20-diol (4), 20(S),24(R)-epoxy-3,4-seco-dammar-25-hydroxy-12-one-3-oic acid (5), 20(S),24(R)-epoxy-3,4-seco-dammar-25-hydroxy-12-one-3-oic acid methyl ester (6), and 6α-hydroxy-22,23,24,25,26,27-hexanordammar-3,12,20-trione (7), are new compounds. In addition, 12 known ones (12-16 and 19-25) were reported in Sanqi for the first time. The new Compound 1 showed comparable antiinflammatory activity on inhibition of NO production to the positive control, whereas the known compounds 9, 12, 13, and 16 displayed moderate cytotoxicities against five human cancer cell lines. The results will provide scientific basis for understanding the chemical constituents of Sanqi rot roots and new candidates for searching antiinflammatory and antitumor agents.

Pathogenesis strategies and regulation of ginsenosides by two species of Ilyonectria in Panax ginseng: power of speciation

  • Background: The valuable medicinal plant Panax ginseng has high pharmaceutical efficacy because it produces ginsenosides. However, its yields decline because of a root-rot disease caused by Ilyonectria mors-panacis. Because species within Ilyonectria showed variable aggressiveness by altering ginsenoside concentrations in inoculated plants, we investigated how such infections might regulate the biosynthesis of ginsenosides and their related signaling molecules. Methods: Two-year-old ginseng seedlings were treated with I. mors-panacis and I. robusta. Roots from infected and pathogen-free plants were harvested at 4 and 16 days after inoculation. We then examined levels or/and expression of genes of ginsenosides, salicylic acid (SA), jasmonic acid (JA), and reactive oxygen species (ROS). We also checked the susceptibility of those pathogens to ROS. Results: Ginsenoside biosynthesis was significantly suppressed and increased in response to infection by I. mors-panacis and I. robusta, respectively. Regulation of JA was significantly higher in I. robusta-infected roots, while levels of SA and ROS were significantly higher in I. mors-panacis-infected roots. Catalase activity was significantly higher in I. robusta-infected roots followed in order by mock roots and those infected by I. mors-panacis. Moreover, I. mors-panacis was resistant to ROS compared with I. robusta. Conclusion: Infection by the weakly aggressive I. robusta led to the upregulation of ginsenoside production and biosynthesis, probably because only a low level of ROS was induced. In contrast, the more aggressive I. mors-panacis suppressed ginsenoside biosynthesis, probably because of higher ROS levels and subsequent induction of programmed cell death pathways. Furthermore, I. mors-panacis may have increased its virulence by resisting the cytotoxicity of ROS.

The isolation of Bacillus subtilis KYS-10 with antifungal activity against plant pathogens (식물 병원균에 대한 항진균 활성을 갖는 Bacillus subtilis KYS-10의 분리)

  • This study was investigated for the purpose of the isolation and identification of antagonistic bacteria with antifungal activity against plant pathogens. This bacteria denominated Bacillus subtilis KYS-10 and the optimum growth condition were 4% sucrose, 1% yeast extract, 0.2% $K_2HPO_4$, pH 7, 150 rpm, $30^{\circ}C$, 8 day. The antifungal activities against nine plant pathogens determined inhibition zone size by diffusion methods. The results, G. zeae (scab) 70 mm and P. grisea KACC 40439 (blast), P. capsici KACC 40177 (phytophthora blight) and C. destructans KACC 41077 (root rot of ginseng) 40~43 mm, and C. gloeosporioides KACC 43520 (ripe rot), C. gloesporioides KACC 40003 (anthracnose), S. shiraiana KACC 41065 (stem rot) and S. shiraiana (mulberry sclerotial disease) 35~39 mm and F. Oxysporum KACC 44452 (bulb rot of ginseng) 28 mm. From these experiment results, author suggest that Bacillus subtilis KYS-10 would be developed as a biological control agent thorough the field experimet in the near future.

Effect of Seed Density, Number of Seeds Sown Per Hole and Thinning Treatment on Growth Characteristics and Disease Occurrence in Greenhouse-Cultivated Ginseng (인삼 하우스 직파재배시 파종밀도, 혈당 파종립수, 솎음처리가 생육과 병 발생에 미치는 영향)

  • This study was conducted to investigate the effects of sowing density, number of seeds sown per hole, and thinning treatment on growth characteristics and disease occurrence in Panax ginseng under direct sowing cultivation in a blue plastic greenhouse. Seedling were grown from 2 or 3 seeds sown, and the healthiest was only retained, while the rest were thinned out at the foliation stage. $NO_3$-N, $P_2O_5$, and organic matter content differed significantly between growth conditions in the plastic greenhouse and in conventional shade in the soil. Disease also tended to be higher in the conventional shade than in the plastic greenhouse. Plant height and stem length showed an increasing trend with increasing sowing density and number of seeds sown per hole. However, these measures noticeably decreased when thinning treatment was conducted. Growth of the subterranean part of ginseng was not markedly influenced by sowing density, the number of seeds sown per hole, or thinning treatment. Root weight, which is an important factor in yield, was significantly affected by the number of seeds sown and thinning treatment. Interestingly, root weight tended to be higher in the thinning treatment plot than the untreated control plot. Damping-off and root rot increased noticeably as the number of seeds sown increased. Disease also tended to be substantially higher in the thinning treatment plot than the untreated control. However, physiological disorder of the plants did not vary with sowing density, the number of seeds sown, or thinning treatment.

Antifungal Mechanism of Pseudomonas stutzeri YPL-l for Biocontrol of Fusarium solani causing Plant Root Rot (식물근부균 Fusarium solani에 대한 Pseudomonas stutzeri YPL-1의 생물학적 방제기작)

  • For the selection of powerful antagonistic bacterium for biological control of soilborne Fusarium solani causing root rot of many important crops, the best YPL-1 strain was selected among 300 strains of bacteria isolated from rhizosphere in ginseng root rot-suppressive soil. The strain was identified to be a species to Pseudomonas stutzeri. With in vitro fungal inhibition tests, antagonistic substance of P. stutzeri YPL-1 against F. solani was presumed to be heat unstable, macromolecular substances such as protein. Also, it was shown that antifungal activity of P. stutzeri YPL-1 increased in proportion to its chitinase production. P. stutzeri YPL-M122 (chi-, lam -) which was deprived of the productivity of chitinase and laminarinase by NTG mutagenesis had lost antifungal activity, completely. And P. stutzeri YPL-MI53 (chi-) had only 4.1% of its antifungal activity. P. stutzeri YPL-1 was not able to produce any extracellular siderophore in iron-deficent minimal medium. It is confident that the antifungal mechanism of P. stutzeri YPL-1 for biocontrol of F. solani depends on lysis rather than antibiosis :the mechanism of lysis appears to involve enzymatic degradation of the cell will components of F. solani by hydrolytic enzymes of more chitinase and less laminarinase.

Genetic Transformation of Biocontrol Agent Bacillus sp, YBL-7 by Plasmid pE194 (생물방제균 Bacillus sp. YBL-7의 형질전환조건)

  • Bacillus sp. YBL-7 which had been isolated from ginseng root-rot suppressive soil was able to antagonize Fusarium solani causing ginseng root-rot by their antibiotic substance. In order to develop multifunctional antagonist on Bacillus sp. YBL-7 as a biocontrol agent against Fusarium salam', optimal conditions for protoplast transformation system of Bacillus sp.YBL-7 by the vector plasmid pE194 were investigated. The protoplasts of Bacillus sp. YBL-7 were obtained at best efficiency by treatment with 200${\mu}g$/ml of lysozyme in the pH 7.0 of SMM buffer for 90 minutes at $40^{\circ}C$. The cell wall of the protoplast was regenerated on the agar plate containing 1.2% agar and 0.7 M mannitol. Under the best condition for protoplast formation and regeneration, the optimal transformation was achieved with 40% polyethylene glycol (M.W. 4000) treatment for 10minutes. The vector plasmid pE194 showed the best transformation frequency at 5$\mu$g/ml of final concentration. The pE194 was very stable over 80% in the transformants.

  • Ginseng fields abolished by potato rot nematode (Ditylenchus destructor) (heavily infested with the nematode) were treated with selected nematicides after preplanting treatment with fumigants or non fumigant chemicals in the fall of 1982. Suppression of the nematode populations was noted in the fields fumigated by cylon (Cy) and treated with ethoprop (E) or triazophos (T). Also in these treatments, high percentages of shoot sprouting and low nematode damages were observed. In the fields fumigated by Cy, subsequent treatments with nonfumigant nematicides such as E, aldicarb (A), phenamiphos (Ph), oxamyl (O) and carbofuran (C) during cultivation had no effect on the significant decrease of the nematode, compared with Cy alone. No differences in shoot sprouting and symptom appearance were observed; however, survivalities of plants and root weights of 4-year old ginseng were high in A and E or E following preplanting treatment with E. Correlation coefficients of the nematode densities to host responses such as sprouting of shoots, symptom appearance, survivality of plants and weight of 4-year old ginseng roots were significant at and mostly before and after the time periods of survey.

  • Some interactions in various soil conditions, numbers of microbial populations, root rot disease development and rates of spore germiation in three different location of soils were investigated. The calcium and magnesium contents were higher in replanted fields of ginseng (Panax ginseng) at Goesan. Potassium contents were high in replanted field at Poonggi and textural class of the soils was silt loam except for silt clay loam in first cultured field of ginseng at Goesan. For the germination process of Fusarium solani, F. moniliforme, F. oxysporum, and Alternaria panax, the percentage germination of fungal spores was high in double distilled water and Pfeffer's solution as media, whereas the lower rate of germination of spores was observed in soil extracts. Numbers of bacteria were high in replanted field soil at Gumsan, and propagules of fungi in replanted fields at Gumsan and Poonggi were higher than other soils, but higher numbers of actinomycetes were found in the first cultured field of ginseng at Goesan and Poonggi. Fungistasis was induced by higher microbial populations present in soil that was initiated when amended with garlic stalk, crushed bean and ginseng leaves. On the other hand, there was no fungistasis in soil amended with wheat and barley straw, and this tendency was a little difference on the soil sample.

Studies on the Root Rot of Ginseng(III) (인삼근부병에 관한 연구 3)

  • Around and in the area of Wolgot-Muon, Gimpo-Gun, Kyunggi province, I examined total bacteria, general Pseudomonas spp., fluorescent Pseudomonas spp., in soil layers and also in different kinds of soil of respective diseased, uncultivated, and healthy areas, and found the followings. 1. In the diseased and uncultivated areas, the content of moisture and silt was greater than in the healthy area. 2. Contrary to the above, the healthy area contained a greater amount of inorganic elements such as $P_2O_5$, K, Ca and of soil particle such as Cs and Fs. The degree of pH and content of Mg were even in three types of soils. 3. Total bacteria were found in abundance in the healthy soil. It was observed that in all types of areas, bacteria reside in abundance in the rhizosphere, i.e., 10-15 cm layers and that the closer the surface, the greater the numbers of the bacteria. 4. General Pseudomonas spp. were also found to the greater in number on the surface of the soil, especially so in the rhizosphere, with the numbers decreasing as the soil layers increase. Numbers of this bacteria in all types of area were nearly uniform. 5. A great number of fluorescent Pseudomonas spp. were found in the diseased area, especially so in the rhizosphere.
