AKT1-targeted proapoptotic activity of compound K in human breast cancer cells

  • Background: Breast cancer is a severe disease and the second leading cause of cancer death in women worldwide. To surmount this, various diagnosis and treatment options for breast cancer have been developed. One of the most effective strategies for cancer treatment is to induce apoptosis using naturally occurring compounds. Compound K (CK) is a ginseng saponin metabolite generated by human intestinal bacteria. CK has been studied for its cardioprotective, antiinflammatory, and liver-protective effects; however, the role of CK in breast cancer is not fully understood. Methods: To investigate the anticancer effects of CK in SKBR3 and MDA-MB-231 cells, cell viability assays and flow cytometry analysis were used. In addition, the direct targets of CK anticancer activity were identified using immunoblotting analysis and overexpression experiments. Invasion, migration, and clonogenic assays were carried out to determine the effects of CK on cancer metastasis. Results: CK-induced cell apoptosis in SKBR3 cells as determined through 3-(4-5-dimethylthiazol-2-yl)-2-5-diphenyltetrazolium bromide assays, propidium iodide (PI) and annexin V staining, and morphological changes. CK increased the cleaved forms of caspase-7, caspase-8, and caspase-9, whereas the expression of Bcl-2 was reduced by CK. In assays probing the cell survival pathway, CK activated only AKT1 and not AKT2. Moreover, CK inhibited breast cancer cell invasion, migration, and colony formation. Through regulation of AKT1 activity, CK exerts anticancer effects by inducing apoptosis. Conclusion: Our results suggest that CK could be used as a therapeutic compound for breast cancer.

Gintonin-enriched fraction protects against sarcopenic obesity by promoting energy expenditure and attenuating skeletal muscle atrophy in high-fat diet-fed mice

  • Background: Gintonin-enriched fraction (GEF), a non-saponin fraction of ginseng, is a novel glycolipoprotein rich in hydrophobic amino acids. GEF has recently been shown to regulate lipid metabolism and browning in adipocytes; however, the mechanisms underlying its effects on energy metabolism and whether it affects sarcopenic obesity are unclear. We aimed to evaluate the effects of GEF on skeletal muscle atrophy in high-fat diet (HFD)-induced obese mice. Methods: To examine the effect of GEF on sarcopenic obesity, 4-week-old male ICR mice were used. The mice were divided into four groups: chow diet (CD), HFD, HFD supplemented with 50 mg/kg/day GEF, or 150 mg/kg/day GEF for 6 weeks. We analyzed body mass gain and grip strength, histological staining, western blot analysis, and immunofluorescence to quantify changes in sarcopenic obesity-related factors. Results: GEF inhibited body mass gain while HFD-fed mice gained 22.7 ± 2.0 g, whereas GEF-treated mice gained 14.3 ± 1.2 g for GEF50 and 11.8 ± 1.6 g for GEF150 by downregulating adipogenesis and inducing lipolysis and browning in white adipose tissue (WAT). GEF also enhanced mitochondrial biogenesis threefold in skeletal muscle. Furthermore, GEF-treated skeletal muscle exhibited decreased expression of muscle-specific atrophic genes, and promoted myogenic differentiation and increased muscle mass and strength in a dose-dependent manner (p < 0.05). Conclusion: These findings indicate that GEF may have potential uses in preventing sarcopenic obesity by promoting energy expenditure and attenuating skeletal muscle atrophy.

Gintonin-enriched fraction improves sarcopenia by maintaining immune homeostasis in 20- to 24-month-old C57BL/6J mice

  • Background: Gintonin-enriched fraction (GEF) is a new non-saponin component glycolipoprotein isolated from ginseng root. This study examined the effect of GEF on age-related sarcopenia in old C57BL/6J mice. Methods: Young (3-6 months) and old (20-24 months) C57BL/6J mice received oral GEF (50 mg/kg/day or 150 mg/kg/day) daily for 5 weeks. During the oral administration period, body weight and grip strength were measured weekly. After sacrifice, muscles from the hindlimb were excised and used for hematoxylin and eosin staining and western blotting to determine the effects of GEF on sarcopenia. The thymus was photographed to compare size, and flow cytometry was performed to examine the effect of GEF on immune homeostasis in the thymus and spleen. Blood samples were collected, and the concentrations of pro-inflammatory cytokines and IGF-1 were measured. Results: GEF caused a significant increase in muscle strength, mass, and fiber size in old mice. GEF restored age-related disruption of immune homeostasis by maintaining T cell compartments and regulating inflammatory biomarkers. Thus, GEF reduced common low-grade chronic inflammatory parameters, which are the main cause of muscle loss. Conclusion: GEF maintained immune homeostasis and inhibited markers of chronic inflammation, resulting in anti-sarcopenia effects in aged C57BL/6J mice. Thus, GEF is a potential therapeutic agent that slows sarcopenia in the elderly.

Effect of wild ginseng on the laying performance, egg quality, cytokine expression, ginsenoside concentration, and microflora quantity of laying hens

  • The experiment was carried out to study the effect of Korean wild ginseng adventitious root supplementation on the laying performance, egg quality, cytokine expression, ginsenoside concentration, and microflora quantity of Institut de selection Animale (ISA) brown laying hens at 24 weeks old. A total of 90 laying hens were subjected to a completely randomized design at three treatments, five repetitions and six laying hens per replicate. The experiments were divided by diets into the basic feed (CON), basic feed + 0.1% wild ginseng (WG1), and basic feed + 0.5% wild ginseng (WG2). The feeding trial was carried out over a duration of 12 weeks after an initial acclimation period of 2 weeks. Feeds and water were administered ad libitum in mash form, and light was available for 16 hours per day. At the end of study, henday egg production (HDEP), average egg weight (AEW), and egg mass (EM) were increased (p <0.05) in WG2 at week 12. Feed conversion ratio (FCR) was decreased (p < 0.05) in WG2 at week 12. The ginsenoside content in egg yolk was increased (p <0.05) in laying hens in the WG2 treatment at week 12. Relative expression of tumor necrosis factor alpha (TNF-α) was reduced (p < 0.05) in the WG supplemented diets at week 12. The fecal microflora quantity of Lactobacillus was increased (p < 0.05) in WG2 at week 8 to week 12, and Escherichia coli (E. coli) was significantly decreased (p < 0.05) in the WG2 at week 12. We concluded that the result observed in the HDEP, AEW, EM and FCR was due to an increase in ginsenoside content, leading to an improvement in the TNF-α, and fecal microflora quantity such as Lactobacillus and E. coli in the WG2 supplemented diets. We therefore recommend the use of WG at application level 0.5% per basal diet for optimum laying performance in layer hens.

인삼이 이식편대숙주반응, 대식세포유주저지반응 및 Trichinella spiralis의 expulsion에 미치는 영향 (Effect of Panax ginseng on the Graft-versus-Host Reaction, Production of Leucocyte Migration Inhibitory Factor and Expulsion of Adult Trichinella spiralis in Mice)

  • This study was undertaken to assess the effect of ginseng administration on T lymphocyte induced local xenogenic graft-versus-host(GVM) reactions which were induced with thymocyte, spleen cell and lymph node cell of ICR mice. Mice received daily 10mg of 70% alcohol ginseng extract oral1y for 100days and control mice remained untreated for the same period of time. The cells from donor mice were injected intradermally into the closely shaven abdominal skin of Sprague-Dawley rats for GVH tests. The thymocyte from control(ginseng-untreated) mice showed a negative local GVH reaction, whereas thymocyte from experimental(ginseng-treated) mice showed a positive reaction with the rate of 17.4%. When spleen cells were injected, the incidence of positive local GVH reaction was 66.7% among ginseng-treated mice, as opposed to incidence of 45.5% of positive local GVH reaction among control mice. The incidence of positive local GVH reaction of the lymph node cells when injected into a recipient was 71.4% among ginseng-treated mice as compared with that of 18.9% among control mice. The relationship between spleen cell inoculum and intensity of the local GVH reaction was assessed in ginseng-untreated mice. The intensity of GVH reaction clearly appears to be dose related. In ginseng-treated mice, a minimum of $1{\times}10^7$ spleen cell was required for production of positive local GVH reaction with almost linear relationship up to an inoculum of $5{\times}10^8$ cells. In control mice, however, a minimum of $1{\times}10^8$ spleen cells was required for positive GVH reaction. These results strongly suggest that the ginseng administration augments significantly the local xenogenic GVH reaction which was used to assess T lymphocyte function and immunocompetence of mice and in addition to this, these results appear to support previous suggestions that the local GVH reaction consitutes a qualitative test of the functional activity of T lymphocytes. These results may be the first to induce local GVH reaction, employing rats as recipient and mice as donor. This study was also desingned to investigate some of the effects of ginseng extract on lymphocyte-macrophage interactions. This was accomplished by in vitro quantification of 1) migratory inhibitory factor(MIF) synthetic capacity of splenic lymphocytes in mice previously primed with ginseng 2) MIF responsiveness of mouse peritoneal macrophages or chicken peripheral leucocytes under the presence of ginseng extract 3) migration ability of chicken peripheral leucocytes by direct stimulation of ginseng extract or ginseng saponin and 4) immunosuppressive effects of immunosuppressants such as cyclophosphamide, cyclosporin A or dexamethasone. Mice divided equally into the ginseng and the saline groups, which received intraperitoneally daily 0.2ml of ginseng absolute alcohol-extract(5mg/ml) and same amount of saline for 15 days, respectively. The cellular immune responsiveness of these mice was assayed 15 days after ginseng pretreatment. Splenic lymphocytes of mice treated with ginseng, when stimulated with sensitized specific-antigen such as sheep red blood cells or toxoplasmin, or with polyclonal activator concanavalin A, produced significantly more MIF than those of control saline group. MIF responsiveness of normal mouse macrophages was significantly augmented when assayed under the presence of ginseng extract (1mg/ml). The migratory ability of normal chicken leucocytes in the absence of MIF was significantly decreased by the stimulation of ginseng extract alone. MIF response was significantly decreased by immunosuppressants and this impaired response was not restored by ginseng pretreatment. This study was additionally performed to evaluate the effect of ginseng on the expulsion of adult Trichinella spiralis in mice. ICR mice were infected experimentally by esophageal incubation of 300 T. spiralis infective muscle larvae prepared by acid-pepsin digestion of infected mice. and received oral administration of 70% alcohol ginseng extract(10mg/mouse/day) for the indicated days plus 4 days before infection. At various times after infection, the number of adult T. spiralis worms in small intestines was determined. Interestingly, ginseng-treatment was accompanied by accelerated expulson of T. spiralis. These results led to the conclusion that Panax ginseng caused some enhancing effect on GVH reaction, macrophage migration inhibition reaction and expulsion of T. spiralis. In addition these results suggested that the mechanisms responsible for this enhancement of ginseng may be chiefly or partially due to nonspecific stimulation of cell-mediated immune response.

반복 스트레스에 의한 흰쥐 해마조직내 신경전구세포의 생성과 brain-derived neurotrophic factor (BDNF) mRNA 발현 변동에 미치는 고려홍삼 사포닌의 반복 투여 효과 (Effects of Korea Red Ginseng Total Saponin on Repeated Unpredictable Stress-induced Changes of Proliferation of Neural Progenitor Cells and BDNF mRNA Expression in Adult Rat Hippocampus)

  • 본 연구 결과를 통하여 홍삼 성분인 고려홍삼 사포닌을 반복 투여시 흰쥐 해마 SGZ 부위의 신경전구세포 생성이 유의하게 증가되었으며, 이와 같은 경향은 반복 스트레스에 노출되어도 유지되었다. 또한 스트레스를 가하지 않은 흰쥐에서 고려홍삼 사포닌 반복 투여시 해마 CA3와 CA1 부위에서 BDNF mRNA의 발현이 증가되었으나, 반복 스트레스를 가한 흰쥐의 CA3와 CA1부위에서 BDNF mRNA의 감소를 차단하지는 못하였다. 따라서 고려홍삼 사포닌 반복 처치에 의한 해마 신경전구세포의 생성에 BDNF 보다는 다른 요인이 관여할 가능성이 클 것으로 추정된다.

20(S)-Protopanaxadiol 및 20(S)-Protopanaxatriol이 활성화된 비만세포로부터의 염증 매개체 유리에 미치는 영향 (Effects of 20(S)-Protopanaxadiol and 20(S)-Protopanaxatriol on the Inflammatory Mediators Release from the Activated Mast Cells)

  • 인삼 사포닌은 면역계에 다양한 약리 효과를 발휘한다. 20(S)-프로토파낙사다이올 (PPD) 및 20(S)- 프로토파낙사트리올 (PPT)은 장내 세균에 의하여 생성되는 인삼 대사체의 일종이며 생체 내 투여 시 순환계에서 탐지된다. 활성화된 비만 세포로부터의 염증 매개체 유리에 미치는 20(S)-프로토파낙사다이올 (PPD) 및 20(S)-프로토파낙사트리올 (PPT)의 영향을 평가하였다. 인삼 사포닌 대사체를 처치 후, 히스타민 유리는 활성화된 해명 폐 비만세포에서 평가하였으며, 인터루킨-4, 인터루킨-8, 및 종양괴사인자-알파 유리는 HMC-1 비만세포에서 평가하였다. 결과는 다음과 같다. PPT는 최고 $100\;{\mu}M$ 농도에서 PMA에 의하여 자극된 HMC-1 세포로부터의 인터루킨-4 유리를 완전히 차단하였다. 또한, 이는 HMC-1 세포로부터의 인터루킨-8의 유리를, PMA와 DMSO동시 처치 시얻어진 수치를 기준으로 대략 40-50% 정도 억제하였다. PPD는 최고 $100\;{\mu}M$ 농도에서 해명 폐 비만세포로부터의 히스타민 유리를 초래하였으나 통계적 유의성은 없었다. PPD는 HMC-1 세포에 PMA와 DMSO 동시 처치 시 얻어진 수치를 기준으로 할 때, 인터루킨-4의 유리를 대략 89% 정도 억제하였으나, 인터루킨-8의 유리에는 유의적인 효과를 초래하지 않았다. 그러나 PPD 및 PPT 모두, PMA에 의하여 자극된 HMC-1 세포로부터의 종양괴사 인자-알파의 유리에는 전혀 효과를 나타내지 않았다. 그러므로 본 연구 결과는 PPD와 PPT가 경구로 투여된 인삼 추출물의 면역조절 작용을 담당하는 장내 인삼 대사체 중의 한 종류임을 제시한다.

홍삼박 효소분해물을 이용하여 제조한 홍삼주의 발효특성 변화 (Changes in Fermentative Characteristics of Red Ginseng Wine using Enzymatically Hydrolyzed Red Ginseng Marc)

  • 복합 탄수화물 분해효소와 단백질 분해효소로 처리하여 제조한 홍삼박 효소분해물에 효모를 발효시킨 홍삼주의 발효특성을 조사하였다. 초기 가용성 고형분 함량을 $20^{\circ}Bx$$30^{\circ}Bx$로 조정한 후 효모를 접종하고 $25^{\circ}C$에서 발효시험을 실시한 결과, 발효 9일 후 가용성 고형분 함량은 $7.5^{\circ}Bx$$13.1^{\circ}Bx$로, 환원당 함량은 18 g/L와 24 g/L까지 감소하였다. 에탄올 함량은 당 소비량과 비례하여 최종 에탄올 함량은 각각 65 g/L와 106 g/L까지 증가하였다. pH는 모든 실험구에서 발효 2일째까지 급격하게 감소하여 pH 4.73에서 pH 3.99로 낮아졌으며, 그 이후 9일째까지 pH의 변화는 미미하였다. 적정산도는 pH 변화와 비례하여 증가하였다. 7일간 발효시킨 후 홍삼주의 조사포닌 함량은 효소 처리하지 않은 대조구에 비하여 100~200% 향상되었다. 그러므로 홍삼박 효소분해물은 홍삼 발효주의 원료로 이용 가능함을 확인하였다.

인삼부산물 급여 수준에 따른 돈육의 저장특성 (Characteristic of Pork Quality during Storage Fed with Ginseng By-Products)

  • 인삼성분이 함유된 돼지고기 생산을 위하여 축산연구소에서 사육된 100 kg 내외의 랜드레이스 30두를 이용하여 인삼부산물을 14일간 첨가수준별로 급여하여 생산돈 돈육의 저장중 육질특성을 분석한 결과는 다음과 같았다. 저장기간 동안의 육색 L값의 변화는 대조구보다 인삼껍질 3% 급여구에서 5일과 10일차에 높게 나타났으며(p<0.05), 또한 전반적으로 처리구가 대조구에 비하여 높은 경향을 보였다. 전단력은 대조구와 인삼껍질 급여구간에는 저장 15일까지는 유의적인 차이가 나타나지 않았다. 저장 20일차에는 인삼껍질 6% 급여구가 대조구에 비하여 높게 나타났으며(p<0.05), 가열감량은 인삼껍질 9% 급여구가 대조구보다 낮은 경향을 보였으며, 저장 15일차에서는 인삼껍질 3%와 9% 급여구에서 낮은 경향을 보였다(p<0.05). 보수력은 처리간에 차이를 보이지 않았으나, 저장 20일차에 잎줄기 추출물 5.5% 급여구가 가장 감소가 적어 보수력이 가장 좋은 것으로 나타났다. 지방산패도(TBA)에서는 저장 5일부터 대조구에 비하여 인삼부산물을 급여한 시험구에서 유의적으로(p<0.05) 낮은 결과를 보였으며, 단백질 변패도(VBN)은 저장 5일차와 20일차에서 인삼껍질 9% 급여구와 인삼 잎줄기추출물 5.5% 급여구에서 유의적으로 낮은 결과를 보였다(p<0.05). 이상의 결과로 인삼의 사포닌 성분이 돈육내 축적 가능성을 보여주는, 인삼부산물 급여구의 전단력, TBA 및 VBN의 분석결과 숙성지연 및 저장 중 식육의 변패지연 효과가 있는 것으로 나타나 돈육으로서 저장에 따른 상품으로 이용이 가능한 것으로 나타났다.

수삼의 저장조건에 따른 홍삼의 이화학적 특성 (Physicochemical Properties of Red Ginseng on Storage Condition of the Fresh Ginseng)

  • 본 연구는 수삼의 저장기간 (무저장, 2일, 6일, 8일, 10일) 및 저장온도 $(20^{\circ}C,\;34^{\circ}C,\;-10^{\circ}C)$로 구분하여 제조된 홍삼의 품질 및 이화학적 특성변화를 비교 조사하였다. 수삼의 수분함량은 저장일이 길어질수록 감소하는 경향이었다. 수삼을 10일동안의 단기간 저장온도는 $34^{\circ}C$의 냉장저장이 적합하다고 생각한다. 전질소 함량은 저장일 10일로 갈수록 무저장구에 비해 다소 증가하는 경향이었다. 저장온도별로는 실온, 냉동, 냉장저장 순으로 무저장구보다 전질소 함량이 증가함을 보였다. 각각의 저장구별 페놀성 화합물의 함량은 무저장구에 비하여 저장기간이 길수록 증가하는 경향이었다. 저장 온도별로는 냉장 저장구가 실온, 냉동 저장구에 비하여 다소 페놀성 화합물의 함량이 높았다. 저장조건별 홍삼의 사포닌 Rf 에 대한 8종의 $ginsenoside\;Rb_1,\;Rb_2,\;Rc,\;Rd,\;Re,\;$Rg_3,\;Rg_1,\;Rg_2$의 상대적 함량 (%)비 변화를 분석한 결과, 각각 저장조건에서 Rf 대한 $Rb_1,\;Rg_1$의 상대적 함량비가 저장기간이 늘어날수록 감소하는 경향이었다. 기타 ginsenoside 함량변화는 시료간의 차이에서 나타난 것으로 판단되며 저장조건에 따른 변화 경향은 뚜렷하지 않았다. 관능적 성질에서 각각 저장구별로 홍삼의 품질, 맛과 향을 종합적으로 평가하여 볼 때, $34^{\circ}C$에서 냉장저장 운반하는 것이 홍삼 향미의 안정적 유지에 가장 적합한 것으로 평가되었다.
