Study on biosynthesis of ginsenosides in the leaf of Panax ginseng by seasonal flux analysis

  • Seasonal ginsenoside flux in the leaves of 5-year-old Panax ginseng was analyzed from the field-grown ginseng, for the first time, to study possible biosynthesis and translocation of ginsenosides. The concentrations of nine major ginsenosides, Rg1, Re, Rh1, Rg2, R-Rh1, Rb1, Rc, Rb2, and Rd, were determined by UHPLC during the growth in between April and November. It was confirmed total ginsenoside content in the dried ginseng leaves was much higher than the roots by several folds whereas the composition of ginsenosides was different from the roots. The ginsenoside flux was affected by ginseng growth. It quickly increased to 10.99±0.15 (dry wt%) in April and dropped to 6.41±0.14% in May. Then, it slowly increased to 9.71±0.14% in August and maintained until October. Ginsenoside Re was most abundant in the leaf of P. ginseng, followed by Rd and Rg1. Ginsenosides Rf and Ro were not detected from the leaf. When compared to the previously reported root data, ginsenosides in the leaf appeared to be translocated to the root, especially in the early vegetative stage even though the metabolite translocated cannot be specified. The flux of ginsenoside R-Rh1 was similar to the other (20S)-PPT ginsenosides. When the compositional changes of each ginsenoside in the leaf was analyzed, complementary relationship was observed from ginsenoside Rg1 and Re, as well as from ginsenoside Rd and Rb1+Rc. Accordingly, ginsenoside Re in the leaf was proposed to be synthesized from ginsenoside Rg1. Similarly, ginsenosides Rb1 and Rc were proposed to be synthesized from Rd.

인삼의 근, 엽 및 경의 일반성분, 용매별 엑기스 및 사포닌 함량 비교 (Content Comparison of Proximate Compositions, Various Solvent Extracts and Saponins in Root, Leaf and Stem of Panax Ginseng)

  • 인삼의 재배과정중에 부산물로 산출되는 지상부위의 활용방안을 모색하기 위하여 엽과 경의 반성분, 용매별 엑기스 및 사포닌함량을 근의 함량과 대비 검토하여 다음과 같은 결과를 얻었다. 1. 엽은 총당의 함량이 21.5%로 근보다 낮았으나 조직유질, 조지방질 및 회분은 각각 9.41%, 3.43% 및 6.83%로 높았다. 경은 조직유질이 39.2%로 현저하게 많았고 총당과 조단백질은 22.7% 및 8.54%로 근보다 낮았다. 2. 용매별 추출물의 수율은 추출용매의 극성이 클수록 높았으며 근과 경에 비하여 엽은 methanol 및 ehanol 추출물의 수율이 35.9% 및 27.3%로 현저하게 많았고, acetone 및 ethyl acetate추출물도 5.64% 및 3.52%로 높았으며 그 외의 비극성 용매의 추출물의 수율도 대체로 높았다. 3. 총 조saponin의 수율은 근과 경은 4.78% 및 2.22%였으나 엽은 19.58%로 현저하게 높았다. HPLC에 의한 분석결과 엽에는 ginsenoside-Rg1(3.32%), -Re(3.24%), -Rd(2.32 %), -Rc(0.65%), -Rb2(0.92%), -Rbl(0.29%), and -Rf(0.11%)가 함유되었고, 경에는 ginsenoside-Rgl(0.28%), -Re(0.3%), -Rd(0.05%), -Rf(0.11%)외에 미량의 -Rbl, -Rb2, -Rc가 검출되었다. 특히 엽은 총 saponin과 ginsenoside-Rg1, -Re 및 -Rd외에도 -Rc와 -Rb2의 함량이 높아서 이들 성분의 분리용 시료로 적합함을 알 수 있었다.

PHYSIOLOGICAL RESPONSE OF PANAX GINSENG TO LIGHT

  • Physiological response of Panax ginseng var. atropurpureacaulo (purple stem variety, Pg) to light was reviewed through old literatures and recent experiments. Canopy structure, growth, pigment, leaf anatomy, disease occurence, transpiration, photosynthesis (PS), leaf saponin, photoperiodism and nutrient uptake were concerned. P. ginseng var. xanthocarpus (yellow berry variety, Px) and Panax quinquefolius(Pq) were compared with Pg if possible. Compensation point(Cp) increased with increase of light and ranged from 110 to 150 at $20^{\circ}C$ but from 140 to 220 at $30^{\circ}C$ with 4 to 15 Klux indicating occurence of light and temperature-dependent high photorespiration. Characteristics of Korea ginseng to hate high temperature was well accordance with an observation 2000 years ago. Korea ginseng showed lower Cp and appeared to be more tolerant to high light intensity and temperature than American sheng although the latter showed greater PS, stomata frequency and conductance, chlorophyll and carotenoids. Px showed lower PS than Pg probably due to higher Cp. Total leaf saponin was higher in leaves grown under high light. Ratio or diol saponin and triol saponin(PT/PD) decreased with increase of light intensity during growing mainly due to decrease of ginsenoside $Rg_1$ but increase of ginsenoside Rd. Leaves of Pg and Px had $Rg_1$ but no $Rb_3$ which was only found as much as $20\%$ of total in Pq leaves, and decreased with increase of light intensity. Re increased in Pg and Px but decreased in Pq with increase of light. PT/PD in leaf ranged 1.0-1.5 in Pg and Px but around 0.5 in Pq. Korea ginseng has Yang characteristics(tolerant to high light and temperature), cultured under Eum(shade) condition and long been used for Yang efficacy (to build up energy) while Pq was quite contrary. Traditional low light $intensity(3-8\%)$ for Korea ginseng culture appeared to be strongly related to historical unique quality. Effect of light quality and photoperiodism was not well known. Experiences are long but scientific knowledge is short for production and quality assessment of ginseng. Recent scientific knowledge of ginseng should learn wisdom from old experiences.

  • We studied the chloroplast rearrangement, short-term regulation depending on the light conditions in plants, and the characteristic of photosynthic rate as affected by in Panax ginseng C.A. Meyer. The chloroplast rearrangement of ginseng mesophyll cell was induced with the irradiation of blue light (400~500 nm) and through this process the rate of leaf transmittance increased 5~7.5%. The time to reach the maximum value of photosynthesis was shorter above 20 minutes with the blue light irradiation than that of the red light.

  • This study was conducted to investigate the application possibility of leaf and stem extract(LSE) from the mixture of leaf and stem of ginseng radix(Panax Ginseng CA Meyer). This study measured antifatigue effects by rota-rod test and swimming test. Also we examined the cell toxicity in normal liver and kidney cells, and acute toxicity in mice. Dropping times of LSE treated group decreased about 39-83% compared to the control group according to supplementation intake levels. Swimming time in LSE treated group increased compared to the control group at short and long supplementation. Negative effects were not found from the results of the cell toxicity. Also acute toxicity not shown. These results imply that the leaf and stem of ginseng radix could be used as possible food resources and functional food material and feed stuff.

인삼잎과 줄기 혼합 추출액의 첨가급식이 흰쥐의 성장, 영양효과 및 혈청 지질에 미치는 영향 (A Study on the Growth Rate, Nutritional Effects and Serum Lipid Contents of Rats by Feeding with Leaf and Stem Extract of the Ginseng Radix)

  • This study was conducted to investigate the application possibility of leaf and stem extract(LSE) extracted from the mixture of leaf and stem of ginseng radix(Panax ginseng CA. Meyer). This study measured the intake levels and efficiency ratio, growth rate, absorption ratio of carbohydrate, lipid and protein of rat by feeding with LSE during 6 weeks. We analyse the hematological and serum metabolic variables, serum lipid concentrations. Total diet and protein intake levels were low, but efficiency ratios were significantly high in LSE administered groups than the control group. Weight gain, liver and kidney weight of LSE groups were significantly higher than the control group. Blood RBC, Hct, Hb, total protein and albumin concentrations were reasonable levels in LSE administered groups compared to the basal diet group. Also serum total cholesterol, LDL-cholesterol, triglyceride contents of LSE groups were low, but HDL-cholesterol level was higher than the basal diet group. These results imply that leaf and stem of ginseng radix could be used as possible food resources, functional food material and feed stuff.

고려인삼에 대한 제초제 2, 4-D의 반응연구 (Study on the Response of Korean Ginseng (Panax ginseng C.A. Meyer) to the Herbicide 2,4-D Application)

  • Various rates of 2,4-D were sprayed on 2 and 3 year old ginseng plants as foliar spray to define the critical concentration. No apparent plant injury was noticable for those ginseng plants when application concentration of 2,4-D doubled the recommended dosage (70 ml/10a). Neither abnormal foliar change occurred nor any inhibition in leaf and stem growth was resulted for the plants treated with 2,4-D concentrated two times of the recommended dosage. When the rates of 2,4-D application were increased greater than this level, injury ratings increased linearly with the rates of 2,4-D application and plant you was inhibited. Ethylene gas was not produced from the ginseng plant treated with 2 times concentrated 2,4-D, however the ginseng plants produced 0.03 to 0.09 ppm ethylene gas when the rate of application were increased 3 and 4 times, respectively. On the other hand the soybean treated with the recommended amount of 21-D produced ethylene gas of 10-20 times higher compared with ginseng plants and died. Photosynthesis ability of the ginseng leaf was significantly decreased by 2,4-D foliar application but it was recovered 4 weeks after 2,4-D foliar treatment. The herbicide 2,4-D was applicated to 2,3 and 4 years old ginseng plants as foliar spray with the rates of 0.5, 1.0, 1.5 and 2.0 times of the recommended dosage to define the effects of 2,4-D on the plant growth and root yield of the ginseng. There were no significant differences in the leaf and stem growth between untreated and 2,4-D treated plant. Berry maturing of 3 and 4 year old ginseng was not influenced by 2,4-D. The root weight of 4 years old ginseng plant was not reduced by application of 2,4-D concenrated 2 times of the recommended dosage, Application time of the herbicide 2,4-D had no effects on the leaf or stem growth of 2,3 and 4 year old year old ginseng plants. When the ginseng seedling was treated with 2,4-D, detrimental phenomena as stem bending and docoration of seedling leaf margin occurred, but stem bending was recovered in a few day s. 인삼 1차 부산물의 생산량 및 기능성 성분 특성 (Yield and Quality Characteristics of Ginseng's First Byproducts)

  • This study was carried out to utilize the byproducts (flower, immature and mature berry, leaf and stem) of ginseng. Yield of byproducts were $32.7{\pm}9.8g$ in flower, $68.2{\pm}2.2g$ in immature berry, $48.5{\pm}4.3g$ in mature berry, $316.2{\pm}20.5g$ in leaf, and $296.6{\pm}15.4g$ in stem per $3.3m^2$ ($180{\times}90cm$, ginseng root $675.5{\pm}35.7g$/drybasis. The total saponin contents of ginseng byproducts and root are $52.36{\pm}1.24$, $68.71{\pm}1.98$, $168.89{\pm}0.57$, $68.26{\pm}1.32$, $7.85{\pm}0.61$ and $35.08{\pm}0.96$ mg/g, respectively. The main ginsenoside of all byproducts was Re and the highest content was $132.23{\pm}1.56$ mg/g in mature berry. But flower and berry was not detected Rf and Rh1, respectively. Total polyphenolic compound content on mature berry was the highest, $2.242{\pm}0.140%$, after, immature berry > leaf > flower > root > stem order. The DPPH radical scavenging activity on mature berry was the highest, $0.115{\pm}0.004$ mg/mL($IC_{50}$), and the others were the same order of polyphenolic compound and ginsenoside content on byproducts.

Spatial protein expression of Panax ginseng by in-depth proteomic analysis for ginsenoside biosynthesis and transportation

  • Background: Panax ginseng, as one of the most widely used herbal medicines worldwide, has been studied comprehensively in terms of the chemical components and pharmacology. The proteins from ginseng are also of great importance for both nutrition value and the mechanism of secondary metabolites. However, the proteomic studies are less reported in the absence of the genome information. With the completion of ginseng genome sequencing, the proteome profiling has become available for the functional study of ginseng protein components. Methods: We optimized the protein extraction process systematically by using SDS-PAGE and one-dimensional liquid chromatography mass spectrometry. The extracted proteins were then analyzed by two-dimensional chromatography separation and cutting-edge mass spectrometry technique. Results: A total of 2,732 and 3,608 proteins were identified from ginseng root and cauline leaf, respectively, which was the largest data set reported so far. Only around 50% protein overlapped between the cauline leaf and root tissue parts because of the function assignment for plant growing. Further gene ontology and KEGG pathway revealed the distinguish difference between ginseng root and leaf, which accounts for the photosynthesis and metabolic process. With in-deep analysis of functional proteins related to ginsenoside synthesis, we interestingly found the cytochrome P450 and UDP-glycosyltransferase expression extensively in cauline leaf but not in the root, indicating that the post glucoside synthesis of ginsenosides might be carried out when growing and then transported to the root at withering. Conclusion: The systematically proteome analysis of Panax ginseng will provide us comprehensive understanding of ginsenoside synthesis and guidance for artificial cultivation.

인삼엽 단백질의 추출 및 영양가 (Protein Concentrate from Ginseng Leaf and its Nutritive Value)

  • 인삼엽 단백질의 식량화를 위하여 엽단백질 농축물(LPC)을 조제하여 그 아미노산 조성과 인공소화율을 검토하였다. 인삼엽을 $-20^{\circ}C$ 냉아세톤으로 처리하고 0.2% NaOH(0.5% SDS, 0.5% 2-ME 함유)로 단백질을 추출하며 인삼 LPC를 얻었다. 인삼 LPC의 일반성분으로는 지방과 회분이 1% 미만이었고 단백질은 75% 내외로 많았으며 총 당은 약 5% 이었다. 총 사포닌 함량은 약 1.2%이고 panaxadiol과 panaxatriol의 비는 1.67이었다. 아미노산 조성은 aspartic acid의 함량이 약 13.4%로 가장 많았으며 methionine의 함량은 매우 낮았다. 필수아미노산토 고루 들어 있었고 총 아미노산에 대한 필수아미노산은 54.5%이였고 E/T ratio는 3.02이었다. LPC의 제 1제한아미노산은 함황아미노산이고, 그 amino acid score는 43.1이었다. Pepsin및 trypsin을 사용한 인삼 LPC의 소화율은 비교단백질인 milk casein에 비하며 낮았다.
