Effects of GA3 and Alternating Temperature on Breaking Bud Dormancy of Panax ginseng C. A. Meyer Seedling (인삼 묘삼 휴면 조기타파에 미치는 GA3 및 변온 처리 효과)

  • Background : Considerable time and effort is required to develop new Panax ginseng varieties. Ginseng breeders have been developing techniques to shorten the breeding cycle to resolve this problem. In this study, we investigated the effects of adding $GA_3$ and alternating temperature (AT, $2^{\circ}C{\rightarrow}-2^{\circ}C{\rightarrow}2^{\circ}C$) on breaking bud dormancy in the varieties (Chungsun and Sunun) of ginseng root. Methods and Results : The $GA_3$ soaking treatment and AT were applied to one year old roots, which greatly accelerated the emergence of new buds. In one year old roots, new buds emerged from the 4th day post transplanting and after breaking dormancy with $GA_3$ and AT treatments. The emergence of new buds was completed within two weeks. The rate of bud emergence for Chungsun was 60% - 98% over 15 - 60 days after the AT and $GA_3$ treatments. The emergence rate of Sunun was 46% - 92%. Normal growth of the ginseng seedling was observed in spite of the early breaking of bud dormancy by combined $GA_3$ and AT treatments. Conclusions : $GA_3$ and AT treatments shortened the dormancy period and facilitated the stable emergence of ginseng seedlings. However, some plants suffered deformities and early sprouting owing to the combined $GA_3$ and AT treatments. Early sprouting was free from dormancy after leaf fall from the of aerial part of the plant.

Studies on the Ginseng Plants(III) -Radioactive Sodium $Acetate-U-C^{14}$ Feeding Experiments- (인삼식물(人參植物)에 관한 연구(III) -동위원소화합물(洞位元素化合物) Sodium $Acetate-U-C^{14}$을 투여한 실험-)

  • The radioactive compound sodium $acetate-U-C^{14}\;(C^{14}-acetate)$ was administered to two- and four-year-old July and September American ginseng (Araliaceae, Panax quinquefolium L.) plants and cuttings. The $C^{14}-acetate$ uptake was approximately 99%. The autoradiochromatograms suggest that the saponins isolated by preparative thin-layer chromatography contained impurities, especially those isolated from the leaf and stem extracts. The root and fruit methanol extracts yielded relatively pure saponins. The large amounts of panaquilin B and its proximity to panaquilin C on preparative thin-layer plates resulted in some admixing. The average concentration (% plant dry weight) of semi-purified saponins were high in the leaves (13.8%), as compared to fruits (9.8%), stems (7.9%) and roots (6.3%). The average percentage of $C^{14}-acetate$ incorporation into panaquilins was 4.8%. The average percentage of $C^{14}-acetate$ incorporation into panaquilins B and C was higher (1.40% and 1.13%, respectively) than that into panaquilins C, (d), G-1 and G-2 (0.75%, 0.65%, 0.13% and 0.53%, respectively). Panaquilin synthesis may be depending upon the part, collection period and age of the plant. The average percentage of $C^{14}-acetate$ incorporation into panaquilin B is high in roots (0.58%) and stems (0.48%); that into panaquilins C and (d) high in leaves (0.40% and 0.45%, respectively); and that into panaquilin E high in roots and leaves (0.55% and 0.50%, respectively). Panaquilin G-2 was synthesized in all parts of plants. The panaquilins appear to be biosynthesized more actively in July than September (exception-panaquilin G-1). Panaquilins B, C and G-1 may be biosynthesized more actively in four-year-old plants and panaquilins (d) and E more actively in two-year-old plants. The results from expectance with cuttings suggest that the panaquilins are synthesized de novo in the above-ground parts of ginseng plants, and that panaquilin G-1 may be synthesized de novo in the leaf. It is known from the tissue culture studies that panaquilins are produced by leaf, stem and root callus tissues and cailus-root cultures of American and Korean ginseng plants. Panaquilins may actively be synthesized de novo in most any cell or organ of the ginseng plants. It was verified that $C^{14}-acetate$ was incorporated into the panaxadiol portions of the panaquilins of two-year-old plants (sp. act. 0.56 mmcCi/mg) and four-year-old plants $(sp.\;act.\;0.54\;m{\mu}Ci/mg)$.

  • Plant viruses of tobacco including tobacco mosaic virus (TMV) and potato virus Y (PVY) cause severe economic losses in leaf-tobacco production. Cultural practices do not provide sufficient control against the viruses. Use of valuable resistant cultivars is most recommendable for the control of the viruses. However, conventional breeding programs are not always proper for the development of virus-resistant plants mostly owing to the frequent lack of genetic sources and introduction of their unwanted properties. Therefore, we tried to develop virus-resistant tobacco plants by transforming commercial tobacco cultivars, NC 82 and Burley 21, with coat protein (CP) or replicase (Nlb) genes of TMV and PVY necrosis strain (PVY-VN) with or without untranslated region (UTR) and with or without mutation. Each cDNA was cloned and inserted in plant expression vectors with 1 or 2 CaMV 35S promotors, and introduced into tobacco leaf tissues by Agrobacterium tumefaciens LBA 4404. Plants were regenerated in kanamycin-containing MS media. Regenerated plants were tested for resistance to TMV and PVY In these studies, we could obtain a TMV-resistant transgenic line transformed with TMV CP and 6 genetic lines with PVY-VN cDNAs out of 8 CP and replicase genes. In this presentation, resistance rates, verification of gene introduction in resistant plants, stability of resistance through generations, characteristics of viral multiplication and translocation in resistant plants, and resistance responses relative to inoculum potential and to various PVY strains will be shown. Yield and quality of leaf tobacco of a promising resistant tobacco line will be presented.

Heritabilities, Genetic Correlations and Path-coefficient Analysis of Some Economic Characters among Dihaploids from Anther Cultured (Nicotiana tabaccum L.) (담배 약배양에 의한 반수체배가계통의 특성에 관한 연구 - 유전력, 유전상관, 경로계수를 중심으로 -)

  • Dihaploid lines derived from the another of the $F_1$ of single cross, SC72 $\times$ Hicks (Nicotiana tabacum L.) were estimated the variation, heritablities, genetic correlations and path-co-efficients on agronomic characters and chemical compositions. Dinaploids exhibited reduced vigor and growth when comparecd with the mid-parent value but had increased nicotine contents, and the range of dihaploids characters were deviated more than that of their parents. Heritabilities of days to flowering, leaves per plant, nicotine. contents, leaf size and plant height appeared high value, and the yield, value per kg. and reducing sugar contents were low. There were positive genetic correlations between yield, and value per kg., leaf width, leaves per plant and reducing sugar contents and negative genetic correlations appeared between nicotine contents and the other characters except for index of leaf shape. Path-coefficient analysis of genotypic correlation coefficients showed that leaves per plant and leaf width lave a high direct effect, and plant height and days to flowering have a high indirect effect on yield, respectively.

Analysis of Growth Characteristics and Physiological Disorder of Korean Ginseng Affected by Application of Manure in Paddy-Converted Field (축분퇴비 시용 수준에 따른 논전환밭 인삼의 생육특성 및 생리장해 분석)

  • This study was carried out to investigate the difference of the content of soil chemical components and growth characteristics in five years old ginseng affected by application of manure in paddy-converted field. As all livestock manure regardless of kinds increased along with the whole soil chemical component, including the pH and EC in 2008. Change in the EC of control plot was slightly increased but not exceeded 1 ds/m over the years. However, the changes in the EC of livestock manure regardless of kinds and amounts were highly increased and irregularly exceeded 1.5 ds/m in 2012. The 5 years old ginseng root fresh weight, treatment of fertilizing pig manure compost 4 ton per 10 areas (PMC 4t on/10a) and fowl manure compost 4 ton per 10 areas (FMC 4 ton/10a), were superior to the others. But there were no difference between PMC 4 ton/10a, FMC 4 ton/10a and control. The standing crop rate 39.6%, treatment of fertilizing cattle manure compost 4 ton per 10 areas (CMC 4 ton/10a), was best in all livestock manure. However that was relatively lower than control. Physiological disorder occurrence rates of livestock manure related with leaf and root of ginseng were also higher than that of control. If excessively using non-decomposed livestock manure, It would be caused physiological disorder in many ways. It is a big problem to be producing the quality ginseng. More research is needed to find out the economic and effective fertilizer.

Survey on Pesticide Use by Ginseng Growers at Gangwon Farmland in Korea (강원도 인삼재배지 농약 사용 실태)

  • This study aims to investigate if the properly use of pesticide in Ginseng farmland at Gangwon-do, Korea. Questionnaire included 36 questions such as control method for plant diseases, pests and weeds and pattern of pesticide use was answered by 271 Ginseng farmers lived in 4 cities and 8 counties at Gangwon-do. Ginseng farmers have noxious plant diseases, insects and weeds such as leaf spot, anthracnose, snail, stem-mining fly, horseweed and hairy crabgrass. To control of these, 35% of farmers relied on various type of pesticides and 31.5% of farmers used pesticides mingling with natural enemy, microorganism and organic materials. Farmers selected the pesticide based on the their own experiences or by recommendation of market dealers and neighbors, resulting in the use of inappropriate pesticides in the Ginseng farmland(78.8 % ). They followed standard dosage of the pesticide from Handbook of Pesticide Application(96.3%). They, however, used the same pesticides from 2 to 5 years(44.1 %). This consecutive use of pesticide could be induced resistance. This survey resulted that study for pesticide resistance and systematic educational program for proper use and selection of pesticide to Gingseng farmers should be conducted in farmland at Gangwon-do, Korea.

The Chracterization of Critical Ranges of Soil Physico-chemical Properties of Ginseng Field and Nutrient Contents of Ginseng Leaves in Gyeonggi Province (경기지역 인삼재배지의 토양 및 엽중 적정양분함량 검정)

  • Ginseng growth is largely affected by characteristics of soil in Ginseng field. In this study, we determined the critical ranges of physico-chemical properties of soil for optimization of ginseng growth by analyzing the soils from Anseong and Pocheon regions in Gyeonggi province. Fresh weight of ginseng was 2 to 5 fold higher in good growth field compared to poor growth field within Anseong region. In the case of Pocheon region, 1.5 to 2 fold differences of fresh weight of ginseng was observed between good and poor growth field. These results indicate the difference of ginseng growth even in the same region. Based on these results, critical ranges of physico-chemical properties of soils were determined by comparing the good and poor growth field of each regions, which are follows; more than 50% of soil porosity, 2.0~2.8 g/kg of total nitrogen, 500~900 mg/kg for Av. $P_2O_5$, 2.3~3.5 $cmol_c\;kg^{-1}$ for Exch. Ca in Anseong; less than 13% of liquid phase, 400~650 mg/kg for Av. $P_2O_5$, 4.0~4.7 $cmol_c\;kg^{-1}$ for Exch. Ca, less than 0.8 and 0.5 $cmol_c\;kg^{-1}$ for Exch. Mg and K, respectively, in Pocheon. Interestingly, we found that ginseng growth was affected by exchangeable base ratio (Ca:Mg:K) especially in Anseong region, which were 6:2:1 in good growth field while 4:2:1 in poor growth field. Critical ranges for nutrient contents of ginseng leaves were also characterized, which are less than 0.2% and 0.22% of each P and Mg, respectively, in Anseong, while less than 1.8% and 0.18% of each N and P, respecively, and 1.5~3.0% of K in Pocheon. In addition, we determined critical ranges for inorganic nutrient contents in the current study.

Mycological Characteristics of Fusarium solani f. sp. pisi Isolated from Pea, Ginseng and Soybean in Korea

  • Fungal isolates belong to Fusarium solani (perfect stage : Nectria haematococca) were isolated form pea, ginseng roots and soybean during 1995 and 1996 in Korea. Thirty-five isolates were identified as F. solani f. sp. pisi based on the morphological characteristics and their pathogenicity. Microconidia formed on the long conidiophore were ovoid or oblong sizing 5~14 x 2.5~5.0 $\mu\textrm{m}$. Macroconidia were formed on carnation leaf agar media with 4.8~5.3 x 32.0~40.7 $\mu\textrm{m}$ in size. They belonged to $\beta$-type since their 3-septate macroconidia were smaller than $5\mu\textrm{m}$ in width. Chlamydospores were smooth- or rough- walled and formed in terminal or lateral branches of hyphae, intercalary, or in chains. Most isolates were highly virulent to pea seedlings, producing dark brown or rot lesions. This is the first report of f. solani f. sp. pisi being pathogenic to pea in Korea.

Effect of Lime Application and Fertilization Level on Prevention of Grey Tobacco Leaves (토양산도 및 시비량 조절에 의한 연초의 Grey엽 발생방지 효과)

  • This study was carried out to get agronomic information about the tobacco culture in paddy soil where incidence of grey tobacco leaves used to appearly uptake of iron, manganese and chlorine due to soil acidity and high water level. Application of lime to paddy soil(pH5.4-5.6) reduced content of iron, manganese known as proceeding materials giving rise to variegated grey tobacco after curing, compared with non-treatment. Grey leaves were found mainly at lower and middle stalk positions, and incidence of grey tobacco was lowered by application of lime in the well drained field but was not affected by level of fertilizer application. Amendment of soil acidity by lime tended to decrease chlorine and manganese content in leaves. Nicotine and mangenese content of leaves were lowered by reduction of one-quarter fertilizer level. In case of lime treatment, increase of yield reached to 4-6% comparing with those of non-treatment but price per kg was not affected. Reduction of N fertilizer level to three-quarters had the equal yield but high quality of leaf comparing with standard fertilization in paddy field.

Characteristics on the Field Growth of Plantlets Regenerated from Leaf Segment Cultures of Boxthorn (Lycium chinense Mill.) (구기자나무의 엽절편체에서 유도된 재분화식물체의 포장생육 특성)

  • 194 plantlets regenerated from leaf explants of boxthorn 'Cheonyang' were cultivated to investigate their morphological characteristics in the field for 2 years. Based on the morphology of leaves, 66.1% of them had elliptical type leaf, the same as that of mother plants, while 22.2% in oval type, 7.2% in obovate type, 2.6% in long-obvate type and 2.1% in lanceolate type. They were classified to 4 groups; group A was selected with both high fruit size and fruit yield, group B with only high fruit size, group C with larger or thicken leaf, and group D with multiple brenches. In comparision of production efficiency between the selected groups and mother plants, group A (99741, 99781, 99854, 99870 and 99886) were longer (2.1 to 2.7 mm) in length of fruits and higher in fruit production (15 to 30%) as compared to mother plants. Croup D (99797 and 99892) was higher in leaf production (7.2%) as compared to boxthorn 'CL1-48', which is the highest in leaf production among boxthorn veriaties.
