Genetic Analysis of 5 Mountain Cultivated Ginseng and Wild Ginseng in Korea (국내 5개 지역의 장뇌삼과 산삼의 유전 분석)

  • ISSR PCR technique was applied to investigate genetic relationship among 5 Mountain cultivated ginseng populations (Jinan, Hongcheon, Punggi, Andong and Yeongju) and cDNA libraries of wild ginseng roots were constructed and analyzed functional genes related to morphogenesis via EST. Twenty four ISSR markers tested produced 127 polymorphic loci from 5 regional Mountain cultivated ginseng. Among the regional samples, Yeongju was made 18 polymorphic loci that were the highest level of variations among the cultivated regions. The range of similarity coefficient was 0.46~0.58 and the regional samples of Punggi and Hongcheon, Jinan and Andong were classified to similar groups respectively, whereas Yeongju was shown to be separate group with high level of genetic variation in UPGMA cluster analysis. As a result, there was no relationship according to geographical distance and genetic similarity. Eleven cDNA clones were consisted of 9 known genes and 2 unknown genes analyzed by BLAST program of NCBI. To recognize expression pattern of Homeodomain transcription factor related genes, Northern Blot analysis was performed for wild ginseng's leaf and root. As a result, the gene was only expressed by Mountain wild ginseng root.

Growth Characteristics and Ginsenoside contents of Korean Ginseng (Panax ginseng C.A. Meyer) by Green Manure Crops (녹비작물 재배에 따른 인삼의 생육과 진세노사이드 함량)

  • This study investigated ginseng growth and ginsenoside contents after control a reserved ginseng cultivation land using various green manure crops for stable ginseng cultivation. Followings are results obtained from this research. After cultivate the green manure crops, microbial flora in soil was diversified, organic matter and total-N content increased, but salt content decreased. The highest output obtained from the wheat cultivated area among various green manure corps. Growth of shoot and root of two years old ginseng increased significantly at the green manure crop cultivated area. Specially, the wheat cultivated area was the most effective in growth. Also, the rate of the leaf discoloration at the aerial part and the rusty root at the root was the least at the wheat cultivation area. Meanwhile, the ginsenoside content was the most at the wheat cultivation area. Thus, the reserved ginseng cultivation land could be managed by cultivating wheat for effective ginseng growth.

Microbiological Hazard Analysis of Ginseng Farms at the Cultivation Stage to Develop a Good Agricultural Practices (GAP) Model (인삼의 GAP 실천모델 개발을 위한 재배단계의 미생물학적 위해도 평가)

  • This study validated microbiological hazards of ginseng farms at the cultivation stage and suggested recommendations to develop a good agricultural practices (GAP) model. A total of 96 samples were collected from cultivation environments (soil, irrigation water, and atmosphere), plants (ginseng and its leaf), personnel hygiene (glove, cloth, and hand) of 3 ginseng farms (A, B, and C) and were tested to analyze sanitary indicator bacteria (aerobic plate count, coliforms and Escherichia coli), major foodborne pathogens (E. coli O157:H7, Listeria monocytogenes, Salmonella spp., Staphylococcus aureus, and Bacillus cereus), and fungi. Total bacteria, coliform, and fungi in the 3 ginseng farms were detected at the level of 1.3~6.0, 0.1~5.0, and 0.4~4.9 v/g (or mL, hand, and $100cm^2$), respectively. Only irrigation water collected from one ginseng farm was confirmed to be E. coli positive. In case of pathogenic bacteria, B. cereus was detected at levels of 0.1~5.0 log CFU/g (or mL, hand, and $100cm^2$) in all samples, but other pathogen bacterias were not detected in any samples from all farms. Although E. coli were detected in irrigation water, the level of microbial for the three farms was lower than the regulation limit. According to the results, the ginsengs produced from the 3 farms were comparatively safe with respect to microbiological hazard. However, cross-contamination of bacteria from environments and workers to ginseng has been considered as potential risks. Therefore, to minimize microbial contamination in ginseng, GAP model should be applied for ensuring the safety of ginsengs.

연초 버어리종 웅성불임 일대잡종 KB 111의 육성경과 및 농경적 특성

  • The vein necrotic strain of Potato Virus Y (PVY - VN) and black shank (Phyto-phthora parasitica var. nicotianae) are the two major diseases causing severe damages especially in burley tobacco (N. tabacum L.) area in Korea. A new tobacco variety, KB 111, resistant to PVY and black shank disease, was developed by Korea Ginseng & Tobacco Research Institute in 1997. It is a male sterile(MS) F$_1$ hybrid of the cross between MS TC 613 and KB 108. KB 111 was compared to Burley 21 on the agronomic characteristics and disease resistances in performance tests: It possessed upright growth habit and flowered two days later than Burley 21. It was resistant to both PVY and black shank and yielded about 3% more cured leaf than Burley 21, but other characteristics are very simiar to those of Burley 21. The chemical composition and physical properties of the cured leaf of KB 111 were as much acceptable as those of Burley 21 while it produced average yield of good quality leaf and appeared to resistant to PVY and black shank disease on regional farm test in 1998.

Growth Characteristics and Ginsenoside Contents of 6-Year-Old Ginseng (Panax ginseng C. A. Meyer) by Shade Materials in Paddy Field (논토양에서 해가림 피복물 종류별 6년생 인삼의 생육과 진세노사이드 함량)

  • This study was carried out to select optimal shade materials among four-layered polyethylene (PE) net (FLPN), aluminium-coated PE sheet (APSS), and blue PE sheet (BPSS) in condition of paddy field cultivated 6-year-old ginseng. The order of light-penetrated ratio and air temperature by shade materials was BPSS > APSS > FLPN. Light-penetrated ratio of BPSS before two fold shade was more 3 times and 2 times than that of FLPN and APSS, respectively. Air temperature of BPSS was also higher $1.6^{\circ}C$ and $1.4^{\circ}C$ than that of FLPN and APSS, respectively. BPSS showed good cultural environment because all of light-penetrated ratio and air temperature were become higher in spring and fall season but lower in summer season by additional shade with two-layered PE net. Survived-leaf ratio was highest in BPSS and lowest in FLPN causing a little water leak on a rainy day. Rusty-root ratio was also highest in FLPN because soil moisture content was increased by water leak. The order of root yield was BPSS > APSS > FLPN, and the cause of highest yield in BPSS was higher light-penetrated ratio during spring and fall season, higher survived-leaf ratio, and lower rusty-root ratio than that of APSS and FLPN. BPSS showed highest total ginsenoside content because of high light-penetrated ratio, blue light effect, and the difference in dry matter partitioning ratio such as low taproot ratio, and high lateral root ratio.
