Effect of Proton Beam Radiation on Bulbil Yield and Gibberellins of Chinese Yam (Dioscorea opposita Thunb.)

  • The study was carried out to evaluate the affect of proton beam radiation on production of bulbil and tuber including change of endogenous gibberellins, of Dioscorea opposita Thunb. The yield of bulbils and tubers from non- and irradiated D. opposita Thunb at doses of 5, 10, 15 and 20 Gy were determined. Endogenous gibberellins were also quantified by GC/MS analysis. D. opposita tubers irradiated at 15 Gy produced higher bulbil production than non-irradiated plants. Enlarged bulbil (above size diameter 4 mm) was significantly increased at 15 Gy. Bioactive endogenous $GA_4$ was dominant in bulbils and tubers irradiated with proton beam rather than $GA_1$. Major gibberellins biosynthetic pathways in bulbils and tubers of D. opposita plants were non C-13 hydroxylation route. From the results of this study, 15 Gy proton beam radiation was suggested as an optimal dose that can produce high amounts of bulbil for mass production of D. opposita plant.

Proximate Chemical Composition and Endogenous Gibberellins of Chufa (Cyperus esculentus L.) in Upland and Wetland

  • Proximate chemical components (protein, oil, carbohydrate, ash, fiber, and starch) were determined from tubers grown in upland and wetland conditions. The contents of crude protein, oil, carbohydrate, and starch were higher in upland condition than in wetland condition. Eight gibberellins were commonly identified and quantified in leaves and tuber of chufa grown in wetland and upland field during growing season. Gibberellin content was always higher specifically in the leaves and tubers grown in wetland condition than in those grown in upland condition. The current knowledge of gibberellin biosynthesis suggests that the two endogenous bio-active gibberellins both $GA_1$ and $GA_4$ are differently metabolized according to cultural conditions. Major gibberellin biosynthesis route is ascertained dominantly the non C-13 hydroxylation pathway leading $GA_4$ in chufa plants.

Gibberellins-Producing Rhizobacteria Increase Endogenous Gibberellins Content and Promote Growth of Red Peppers

  • The growth of red pepper plants was enhanced by treatment with the rhizobacterium, Bacillus cereus MJ-1. Red pepper shoots showed a 1.38-fold increase in fresh weight (fw) and roots showed a 1.28-fold fw gain. This plant growth-promoting rhizobacterium (PGPR) has been reported to produce gibberellins (GAs). Other GAs-producing rhizobacteria, Bacillus macroides CJ-29 and Bacillus pumilus CJ-69, also enhanced the fw of the plants. They were less effective than B. cereus MJ-1, though. The endogenous GAs content of pepper shoots inoculated with MJ-1 was also higher than in shoots inoculated with CJ-29 or CJ-69. When inoculated with MJ-1, bacterial colonization rate of the roots was higher than that of roots inoculated with CJ-29 or CJ-69. These results support the idea that the plant growth-promoting effect of the bacteria also positively related with the efficiency of root colonization by the bacteria. In addition, we identified the major endogenous GAs of the red pepper as originating from both the early C-13 hydroxylation and the early non C-13 hydroxylation pathways, with the latter being the predominant pathway of GA biosynthesis in red pepper shoots.

Effects of Mepiquat Chloride and Trinexapac-Ethyl on Oil Composition, Seed Yield and Endogenous Gibberellins in Flax

  • The aim of this field experiment was to investigate the possible effects of mepiquat chloride (TE) and trinexapac-ethyl (MC) on oil composition, seed yield and endogenous gibberellins content of flax cultivar. Foliar application of plant growth retardants mepiquat chloride (0.897, 1.794 and 2.691 kg a.i. $ha^{-1}$) and trinexapac-ethyl (0.756, 1.512 and 2.668 kg a.i. $ha^{-1}$) had significantly increased seeds ripening rate and seed yield. In contrast, plant height was decreased by foliar application of MC and TE. The application of MC significantly increased seed oil yield (730 kg $ha^{-1}$ by 27.0%) compared to the control. Seed and oil yield, and unsaturated fatty acids (oleic acid, linoleic acid and linolenic acid) were increased by foliar application of MC.

  • Seed of Codonopsis lanceolata exhibits low germination due to impermeable seed coat. Prolonged seed dormancy can be overcome by the application of gibberellins, as it promotes growth of the embryo and weakens the structures surrounding of embryo. The effects of photoperiod, sugar and gibberellin concentration were investigated at constant temperature for 12 days and 22 days in vitro and invivo conditions respectively. The rate of germination of seeds of Codonopsis lanceolata in wet filter paper in both complete dark and light treatments was significantly lower than that of seed treated with $GA_3$. It clearly indicates that there is significant coat imposed dormancy in the seed of Codonopsis lanceolata. The rate of germination in vivo condition was lower than that of the in vitro condition supplemented with $GA_3$ Thus, the physical dormancy due to impermeable seed coat and low level of endogenous gibberellins in the seed was the cause of poor germination rate in Codonopsis lanceolata.

Gibberellin 산류 생산균주의 분리와 생산성검토 (Productivity test on some screened strains of Gibberella fujikuroi(Saw.))

  • 벼의 키다리병(Bakanae disease)병원균 Gibberella fujikuroi Sawada는 그의 대사 산물로 Gibberellin 산류를 생산하고 있어 작물생장 조절작용이 있는 이 물질을 얻기 위하여는 우수한 균주를 알맞은 조건에서 배양하는 것이다. 이 시험은 우리나라의 야생균 가운데 Gibberellin 산류의 생산성이 우수한 균주를 분리 선발하기 위하여 먼저 평택, 예산, 당진 및 순천지역에서 키다리병에 걸려있는 볏모 뿌리를 수집하여 평택지역 4, 예산지역 7, 당진지역 7, 순천지역 5 균주를 각각 분리하여 형태학적으로 검토하여 동정하였다. 분리 선발된 각 균주와 대조근주(IAM-8048)에 대하여 일정한 배양 조건에서 Gibberellin 산류의 생산성을 배양액중 농도 측청과 볏모에 대한 배양액 추출물의 생산촉진 효과실험을 통하여 우수한 활성 균주 셋을 선정 Gibberella fujikuroi P-105, Y-14 및 T-58이라 각각 기호를 부쳤으며 그 가운데 P-105가 가장 우수하였다. 균주 Y-5, Y-7, T-54 및 S-152 등은 배양액중 Gibberellin 산류의 농도와 볏모에 대한 생장 촉진 효과가 낮거나 약간 억제적이었다. 배양 조건을 추구하기 위하여 조성과 함량이 다른 여섯가지 배양기에 대하여 비교 시험한 결과 Raulin-Thom 배양기 (glucose 16%, $NH_4NO_3$ 0.24%)에서 Gibberellin 산류의 생산이 가장 높았으며 그 밖에 다른것은 대략 Cross, Borrow, West, Stodola, Kurosawa 배양기의 순위로 생산성이 낮았다.

Isolation of a Gibberellin-producing fungus (Penicillium sp. MH7) and Growth Promotion of Crown Daisy (Chrysanthemum coronarium)

  • Plant growth promoting fungi (PGPF) are well known for the production of useful secondary metabolites. However, limited information is available on the gibberellin (GA) production capacity of PGPF of endophytic origin. In the current study, 15 fungal endophytes were isolated from the roots of Crown daisy, and then screened on Waito-c rice, in order to identify plant growth promoting fungi. The fungal isolate MH7 significantly increased the shoot length (12.1 cm) of Waito-c in comparison with control treatment (7.9 cm). In a separate experiment, the culture filtrate (CF) of MH7 significantly promoted the growth attributes of Crown daisy. The MH7 CF was analyzed for gibberellins and it contained all physiologically active gibberellins ($GA_1$, 1.37 ng/ml; $GA_3$, 5.88 ng/ml; $GA_4$, 8.62 ng/ml; and $GA_7$, 2.05 ng/ml) in conjunction with physiologically inactive $GA_9$ (0.83 ng/ml), $GA_{12}$ (0.44 ng/ml), $GA_{15}$ (0.74 ng/ml), $GA_{19}$ (1.16 ng/ml), and $GA_{20}$ (0.98 ng/ml). The CF of MH7 produced higher amounts of $GA_3$, $GA_4$, $GA_7$, $GA_9$, and $GA_{12}$ than wild-type Fusarium fujikuroi, which was used as a control for GA production. The fungal isolate MH7 was later identified as a new strain of Penicillium on the basis of its morphological characteristics and phylogenetic analysis of the 188 rDNA sequence.

Identification of Endogenous Gibberellins by Feeding of [$^{14}\textrm{C}$] $\textrm{GA}_{12}$ in Chinese Yam, Dioscorea opposite

  • The metabolism of [$^{14}\textrm{C}$] $\textrm{GA}_{12}$ in the Chinese yam (Dioscorea opposita Thunb. var. Tsukune) was examined to determine the identification of endogenous gibberellins. [$^{14}\textrm{C}$] $\textrm{GA}_{12}$ was metabolized to $\textrm{GA}_{53}$, $\textrm{GA}_{44}$, $\textrm{GA}_{19}$, $\textrm{GA}_{20}$, $\textrm{GA}_1$, $\textrm{GA}_8$, $\textrm{GA}_{15}$, $\textrm{GA}_{24}$, $\textrm{GA}_9$, $\textrm{GA}_{36}$ and $\textrm{GA}_4$. Radioactivity of GAs in non C-13 hydroxylation route was five-fold higher than that of early C-13 hydroxylation in analyzed GA-metabolites. Radioactivity of $\textrm{GA}_4$ was always four times higher than that of $\textrm{GA}_1$ at every feeding time. $\textrm{GA}_1$ radioactivity has always a lower level to below 200 DPM. The major pathway of endogenous GA metabolism in seedlings of the Chinese yam might be the non C-13 hydroxylation pathway.

Effect of Nitrogen and Silicon Nutrition on Bioactive Gibberellin and Growth of Rice under Field Conditions

  • Gibberellins are growth hormones that play a pivotal role in the growth and development of plants. Present investigations were carried to check the effect of nitrogen(N) and silicon(Si) on bioactive $GA_1$ and its immediate precursor $GA_{20}$ at different growth stages of two rice cultivars with different maturity traits. It was observed that the endogenous bioactive $GA_1$ level gradually increased during vegetative stage and anthesis stage of both Junghwabyeo(early flowering cultivar) and Daesanbyeo(late flowering cultivar). However, the $GA_1$ and $GA_{20}$ content start decreasing during the seed filling stage in both rice cultivars, which indicated a possible relationship of bioactive $GA_1$ and floral development. Our results also confirmed that early 13-hydroxylation pathway was operated at all developmental stages of rice plant. Variation in the levels of the endogenous gibberellins in rice shoots were measured by GCMS-SIM using $^2H_2$-labeled gibberellins as internal standards. Combined application of N and Si enhanced growth parameters and reduced lodging index of both rice cultivars. It was thus concluded that the level of physiologically active $GA_1$ increased during vegetative and early reproductive stage, but starts declining at seed filling stage.
