대두 종자 발아세 검정방법 비교 (The Use of Multiple Tests in Predicting the Vigor of Soybean Seeds)

  • 대두종자의 퇴화과정에서 일어나는 생리ㆍ화학적인 변화를 탐색하여 종자세의 차이를 비교하고 종자세 예측에 적합한 방법을 구명하기 위하여 대두 5품종의 종자를 인위노화처리시켰다. 대두종자세의 지표로서 발아율$\times$배축의 길이로 나타낸 값을 매개변수로 하여 각종 종자세 검정성적과 비교검토하였다. 종자세검정의 가장 적합한 방법으로는 발아시험에 의한 방법임을 알 수 있었다. ATP와 GADA 검정법을 제외하고는 모든 검정법이 종자세 지표와 고도의 유의성이 있었다. 대두종자세 검정을 위하여는 배축의 길이. 전기전도도에 의한 검정 및 저온발아 검정법이 적합한 방법임을 알 수 있었다.

Effect of light, ultrasonication and liquid smoke on germination of proso millet (Panicum miliaceum L.) seeds

  • High quality seed of proso millet that has high germination percentage, germination speed, and uniformity demanded to increases rates of mechanization in cereal crop cultivation. In order to improve germination characteristics, proso millet seeds were treated with red light, ultrasonication and liquid smoke (LS) solution that generated from hickory wood. All treatments were performed in seed priming solution with 100mM $NH_4NO_3$ at $15^{\circ}C$ for 24hrs under aeration condition. Seeds were exposed under light intensity of 2000 lux for 15m, 30m, 60m, and 120m in priming solution. Ultrasonic treatment was performed at 60%, 80%, and 100% intensity of 21.6 KHz for 5m, 10m, and 20m in priming solution. For LS treatment seed were soaked in 0%, 0.5%, 1.0%, 5.0% and 10.0% of diluted solution with $dH_2O$ or 100mM $NH_4NO_3$ solution. The effect of seed treatment was evaluated with germination percentage (GP), mean germination time (MGT), germination index (GI), germination rate (GR), Germination uniformity (GU) and heath seed percentage (HS). Our results demonstrate that red light (15min) or ultrasonication (21.6kHz, 5min) treatment improved MGT, GI, GR, and GU comparing to untreated control. Importantly, we show that LS treatments have significant effect on the health seedling and germination characteristics. Proso millet seeds that treated with 5% LS solution for 24hrs improves HS up to 97% that similar results obtained in 100mM $NH_4NO_3$ priming for 24hrs. The combined treatment with LS solution and 100mM $NH_4NO_3$ priming were not effective in all treatments. Our results demonstrate that treating seeds with LS or 100mM $NH_4NO_3$ priming or ultrasonication improves germination characteristics. The methods described here will help advance research using this species by increasing the germination performance at which successive seed pellet process.

Germination and Growth Performance of A Native Threatened Tree Species Quercus gomeziana A. Camus in Nursery Stage: Case of Bangladesh

  • This study was conducted to investigate the effect of different pre-sowing treatments of seeds on germination and growth performance of native threatened tree species Quercus gomeziana A. Camus at the nursery of Chittagong University, Bangladesh. Furthermore, seedling growth attributes under different doses of fertilizer (urea) was also experimented to find the best dose of fertilizer on this tree species at the nursery stage for better field level growth. Seeds were placed to six pre-sowing treatments e.g. control (PT0), treated with sand paper rubbing (PT1), nicking (PT2), seeds immersed in cold water for 48 hours (PT3), seeds immersed in cold water for 7 days (PT4) and seeds sown at propagator house with increased temperature (PT5). It was found from the study that germination was started earlier (at 31 days) in treatments sand paper rubbing (PT1) and nicking (PT2). The highest germination percentage (93%) was in PT1 followed by 86% in seeds immersed in cold water for 7 days (PT4) and 80% in PT0 (control). Germination percentage was observed least (63%) in PT2 even though germination started earlier. For fertilizer dose experiment to seedlings at the nursery level, treatment FT1: 100 kg/ha (0.33679 g urea/pot/seedling) comparing with other treatments FT0: 0 kg/ha (Control), FT2: 200 kg/ha (0.67358 g urea/pot/seedling), FT3: 300 kg/ha (1.01037 g urea/pot/seedling) showed better performance in case of collar diameter (6.74 mm), number of leaves, shoot dry weight (19.74), total dry weight (28.16 g), total fresh weight (67.96 g), volume index (3904.82), sturdiness (127.69). Finally, it can be concluded that Quercus gomeziana seedlings revealed better performances under the treatment FT1 in growth and biomass production. Findings of this study will be helpful to take decision on organic fertilizer dose application to seedlings of Q. gomeziana for large scale plantation and conservation of this species.

다양한 씨앗의 발아 및 발아지수에 근거한 나노입자 생물학적 독성평가 (Bioassessment of Nanoparticle Toxicity based on Seed Germination and Germination Index of Various Seeds)

  • 본 연구에서는 다섯 종의 씨앗(춘채, 아욱, 양배추, 배추, 당근)을 이용하여 금속산화물 나노입자(CuO, NiO, Fe2O3, Co3O4, TiO2, ZnO)들이 씨앗발아와 발아지수에 미치는 영향을 조사하였다. CuO, ZnO, NiO 나노입자는 씨앗 활성에 뚜렷한 독성 영향을 나타내었으며, 나노입자종류에 따라 상이한 민감도를 나타내었다. 각 나노입자에 대한 독성순서(EC50 범위)는 다음과 같다: CuO 6~27 mg/L > ZnO 16~86 mg/L > NiO 48~112 mg/L. 나머지 조사 대상 나노입자인 Co3O4, TiO2, Fe2O3은 최대 1,000 mg/L 높은 농도 노출에도 뚜렷한 영향을 나타내지 않았다. 씨앗별 상이한 민감도를 나타내었으며, 가장 민감한 종인 아욱의 씨앗발아 EC50은 CuO 5.5 mg/L ZnO 16.4 mg/L, NiO 53.4 mg/L로 조사되었다. 씨앗별 나노입자에 대한 독성 영향은 CuO > ZnO > NiO > Fe2O3 ≈ Co3O4 ≈ TiO2 나타났으나, 당근씨앗은 NiO [EC50 80.4(71.41~90.54) mg/L]와 ZnO [EC50 85.8(69.31~106.29) mg/L]가 유사한 독성을 나타내었다.

벼 건답직파재배(乾畓直播栽培)시 저온발아성(低溫發芽性)과 파종심(播種深)에 따른 출아특성(出芽特性) (Influence of low temperature and sowing depth on emergence in dry direct seeded rice)

  • 본 시험은 벼 건답직파재배(乾畓直播裁培)시 적응품종선정(適應品種選定)을 위한 기초자료를 얻고자 16개 기존장려품종(旣存奬勵品種)과 미국 6품종을 가지고 저온(低溫)하에서 발아력(發芽力) 검정(檢定)과 금오벼, 안중벼 2품종을 공시하여 세조파기에 의한 건답직파시 적정파종심(適正播種深)을 구명(究明)하기 위해 수행하였던 바 그 결과가 다음과 같이 요약(要約)되었다. 1. $13^{\circ}C$ 저온 하에서의 평균발아율은 81.2%로 그 범위는 64.3~94.84%로서 품종간 차이를 보였으며 평균발아일수는 17.4일 정도이고 그 범위는 14~24일로서 품종간 차이가 있었다. 2. 저온발아력(低溫發芽力)과 출아소요일수(出芽所要日數)간에는 부의 높은 유의상관(r= -0.922) 관계가 있었고 발아율(87% 이상) 및 발아소요일수(1 7일 이하)로 본 저온발아성이 양호한 품종들은 자광도, 낙동벼, 청명벼, 카롤로, 안중벼, 계화벼, 대관벼, 동진벼, 서해벼 이었다. 3. 벼 건답직파시 도복지수와 유의상관관계가 있었던 형질은 파종심, 간직경, 중심고, 좌절중이었으며 도복에 직접적으로 연관되는 좌절중은 파종심, 간직경, 간벽두께와 유의상관(有意相關) 관계가 있었다. 4 파종심 1~3cm에서 입모율(立毛率)이 높았고 출아소요일수(出芽所要日數)가 짧았다. 도복지수는 파종심이 깊을수록 낮았는데 파종심 1cm에서는 출아율은 높았으나 도복지수가 높았고, 7cm에서는 도복지수는 낮았으나 출아율(出芽率)이 저조하였는데, 출아율과 도복 안정성 및 수량성을 고려한 적정 파종심은 3cm 내외였다.

Priming 처리가 팥배나무의 종자 발아 및 유묘 생장에 미치는 영향 (Effect of Priming Treatments on Seed Germination and Seedling Growth of Sorbus alnifolia)

  • 종자 priming은 발아율을 향상시킬 뿐만 아니라 빠르고 균일하게 하는 유용한 종자처리기술로서 조경적 가치가 뛰어난 팥배나무 종자의 번식 효율성을 위해 다양한 발아촉진법을 개발하기 위해 실시하였다. Priming 처리를 위해 NaCl, $CaCl_2$, $KNO_3$, PEG(polyethylene glycol)을 이용하여 농도에 따라 각각 2일과 3일간 처리하였으며, 실험종료 후 발아특성 및 유묘생장특성을 분석하였다. 발아율은 $KNO_3$ 100 mM 2일 처리구에서 24%로 가장 높게 나타났으나, 평균발아일수는 $KNO_3$ 200 mM 2일 처리에서 가장 높았다. 발아속도와 발아균일지수 역시 $KNO_3$ 100 mM 2일 처리구에서 가장 높은 값을 보여 발아특성 증진에는 $KNO_3$가 적정한 처리제임을 짐작하였다. Priming 처리된 종자로부터 생장한 유묘들을 대상으로 상대생장율과 유묘활력지수를 조사한 결과 무처리구와 차이를 나타내었는데(p<0.05), 상대생장율(수고 및 근원경)의 경우 NaCl 400 mM 2일 처리구에서 가장 높은 값을 보였으며, 유묘활력지수는 발아율에서 가장 높았던 $KNO_3$ 100 mM 2일 처리구가 가장 높게 나타났다. 결과적으로, priming 처리를 위한 염류(NaCl, $CaCl_2$, $KNO_3$, PEG) 중 팥배나무의 종자 발아 및 유묘의 생장에 영향을 미치는 적정 처리제는 $KNO_3$로 판단되었으며, 적정처리조건은 100 mM 2일처리였다.

해수농도가 큰비쑥의 종자발아 및 유식물 생장에 미치는 영향 (Effect of Seawater Concentration on Seed Germination and Seedling Growth of Artemisia fukudo)

  • Halophytes are plants of saline habitats that grow under conditions that may vary in extremes of temperatures (freezing to very hot), water availability (drought to water logging) and salinity (mild to almost saturation). Halophytes may also face sudden micro-environmental variations within their habitats. In this study, we examine some of the factors that determine the ability of seeds of Artemisia fukudo to germinate when conditions are optimal for seedling growth and survival. Germination percentage was the highest at 77.5% when treated in 0% seawater and seedling growth was best in 0% seawater plug cell tray. Physiological quality (chlorophyll contents) and mean germination time were best in the condition in which seeds were treated with 0% seawater (control). Germination performance index in the control group showed the highest value to 3.8. Seedling growth, seedling vigor index in the length of seedling growth andthe relative growth rate were the highest values of 0.76 cm, 1.43, 0.0099, respectively.

The effect of hydropriming and explants on shoot initiation and tetraploids in small watermelons

  • Hydropriming had positive effects on the time for germination to reach 50%, the germination index, the time to final germination percentage, and the number of uniform seedlings with enlarged cotyledons in in vitro germination of small watermelon. In addition, the highest shoot initiation was obtained from hydroprimed cotyledonary nodes ($95{\pm}6%$), followed by non-primed cotyledonary nodes ($78{\pm}6%$), hydroprimed cotyledons ($72{\pm}4%$), and non-primed cotyledons ($48{\pm}4%$). Meanwhile, no shoots were initiated from hypocotyls. The total number of shoots that initiated from cotyledonary nodes and cotyledon explants was insignificant, indicating that both cotyledons and cotyledonary node were good sources for the in vitro culture. Choosing explant sources that favor tetraploidy should be the key for producing higher polyploidy plants; a total of 10.5% of tetraploid regenerants were entirely identified from cotyledon explants. Cotyledons with highly differentiated cells might show higher variations than cotyledonary nodes with more preexisting meristematic cells. Cells of cotyledon tissue might undergo changes in ploidy level during differentiation of the culture, or it might be that some of the variations were already present in the tissues of the donor plant. Morphological changes in fruit length of tetraploid regenerants are genotype-dependent.

교란지 복구를 위한 표토의 매토종자 발아특성 - 예산군의 나지와 묵밭 토양- (Seed Germination of Surface Soil for Restoration of Disturbance Place - Bare Land and abandoned Field, Yesan-gun, Korea -)

  • To offer the basic data of vegetation restoration by buried seed of soil, this research was conducted at bare land and abandoned field in Yesan-gun. Germination plants of buried seed were consisted of 40 taxa (37 species, 3 varieties) in bare land, and 41 taxa (37 species, 4 varieties) in abandoned field. Classification of germination plants by family was the most in Gramineae, and emergent frequency of plots was the highest of Digitaria ciliaris. The soil depth of the most plants appearance was 2~5cm in bare land and 5~10cm in abandoned field, and the soil depth of the most population appearance was 0~2cm both in bare land and in abandoned field. Population number of buried seed germination was decreased according to soil depth. Crepidiastrum sonchifolium was a plant that population number of buried seed germination is the most. Similarity index was 0.33 in aerial part plants and buried seed plants, and 0.55 in bare land and abandoned field.

봄파종을 위한 인삼 종자 저장 적정 온도 연구 (Optimum Storage Temperature for Spring Sowing of Panax ginseng Seeds)

  • Background: Usually ginseng seeds are sown during autumn and spring. Sowing in spring often results in poorer seedling establishment than in autumn. One of the reasons for poor germination could be cold-treatment condition for breakage of physiological dormancy during winter. Here we tested the effects of storage temperature used during cold treatment on germination. Methods and Results: Germination properties were observed after dehiscent seeds were stored as wet and dry at 2℃, -2℃, -3.5℃ and alternating temperature (AT). Seed dryness and storage temperature affected germination properties (p < 0.01). Wet and AT condition germinated highest, and wet and -3.5℃ condition germinated lowest, which was 91.2% and 1.4% respectively. Mean germination time (MGT) of the wet and AT condition was faster than other treatments at 2.4 days, and the dry and -2℃ condition was the longest. Germination performance index (GPI) was highest for wet and AT condition (37.7%) and the lowest for wet and -3.5℃ condition (0.5%). The growth of above-ground and below-ground were the best for wet and 2℃ condition, and wet seeds showed better growth than dry seeds (p < 0.01). Conclusions: For cold treatment, ginseng seeds may not be stored below -2℃ for successful germination during spring sowing.
