Shattering-related Characteristics and Germination Speed Based on Grain Type and Esterase Isozyme Zymogram in Korean Off-rype Rice

  • This study was carried out to investigate the characteristics of shattering habit and germination of offtype rice plants collected in Korea which were classified into seven groups based on grain characteristics. In the long-grain red rice group, the short-grain red rice group, the long-grain normal rice group, and the long-grain waxy rice group, the tensile strength of grain and primary rachis branch was relatively low and the fiber cell of the primary rachis branch was short. Characteristics of shattering-related traits and germination in relation to grain and esterase isozyme zymogram type of off-type rice plants. In most lines of the extremely late and sterile rice group, the tensile strength of grain was low, while that of primary rachis branch was high, and the fiber cell was long. However, in the type 1 esterase isozyme zymogram(EIZ) lines among the long-grain normal rice group and the type 6 and 5+6 EIZ plants among the extremely late and sterile rice group, the tensile strength of grain was relatively higher. The long-grain waxy rice group and the type 3 EIZ lines among the long-grain red rice group, showed higher germinability than did cultivars which are known to germinate well even in low-temperature. The other off-type rice group, except for the short-grain waxy rice group and type 1 and 7 EIZ lines among the long-grain normal rice group, had higher germination speed than that of the cultivars.

Germination Characteristics and Early Growth of Acer pictum subsp. mono (Maxim.) H. Ohashi by Harvesting Dates (고로쇠나무 종자 수확시기에 따른 발아특성 및 초기생육)

  • Background : Acer pictum is a deciduous tree belonging to the family Aceraceae. This study was conducted to collect basic data on mass propagation techniques by surveying the germination and growth characteristics of A. pictum seeds. Methods and Results : A. pictum seeds were harvested in 2014 on August 19, September 5, September 29 and October 31. The seeds were then sown on the same dates they were harvested. A portion of seeds harvested in August 2014 were stored at low temperatures and subsequently sown on March 11, 2015. The germination rate of A. pictum seeds was highest in seeds that were stored harvested on October 31. Mean germination time (MGT) was shortest and germination velocity (Rs) was fastest in seeds harvested on October 31. Root collar diameter, total root length, and dry weight were also highest from seeds collected October 31. Conclusions : A. pictum seeds harvested on October 31 had highest germination and growth rates compared with seeds harvested on earlier dates. It is expected that these results demonstrating optimum harvesting and sowing dates will be applicable to future seedling production for this tree species.

  • Background: Many Taraxacum spp. plants are widely used in medicine, but some of them have propagation problems, such as strong dormancy and poor germination rates. This study investigated the effects of temperature, gibberellic acid ($GA_3$), and potassium nitrate ($KNO_3$) on seed germination in Taraxacum ohwianum. Methods and Results: The seeds (NIBRGR0000135524) were exalbuminous, and their length and width were $4.54{\pm}0.032mm$ and $0.97{\pm}0.029mm$, respectively. Various germination temperatures were tested (15, 20, 25, and $30^{\circ}C$), the optimum temperature for germination was found to be $20^{\circ}C$ (31.3%). High temperature ($30^{\circ}C$) resulted in non-typical seedlings (thickened and crumpled cotyledons, with a restricted root system). To increase the germination capacity, $GA_3$ (200, 500, and $1,000mg/{\ell}$) or $KNO_3$ (20, 50, 100, 200, and $500mg/{\ell}$) solutions were used as pre-soaking solutions instead of water. The $GA_3$ treatments increased the germination percentage and rate, but germination percentage was higher with the $KNO_3$ treatments. Under the $50-200mg/{\ell}\;KNO_3$ treatments, the germination percentage exceeded 80% after 12 days, and $50mg/{\ell}\;KNO_3$ was notably effective (91.2% after 15 days). Conclusions: T. ohwianum seeds showed improved germination at low temperatures. The best method for germination was presoaking in $50mg/{\ell}\;KNO_3$ for 24 h, in the dark at $4^{\circ}C$, and then incubating the germinated seed at $20^{\circ}C$ for 15 days.

  • This study was investigated to evaluate germination rate of Astragalus membranaceus B. in Korea as affected by storage temperature, germination temperature and storage period of seed. The highest germination rate was obtained from condition of $25^{\circ}C$ in germination temperature. Seeds were stored at $-20^{\circ}C$ and $5^{\circ}C$ for 8 weeks has showed higher germination rate than one at room temperatures. The germination rates showed significantly difference by harvested year of 2010, 2011 and 2012. The seed of A. membranaceus in harvested year of 2011 and 2012 had germinated well. On the other hand, seeds in harvested year of 2010 were not nearly germinated. Consequently, the longer storage period after seed harvest lower germination rate and seed vigor as well.

  • In this study, we investigated the effect of seed collection stage and alternating temperature on the germination of Deutzia paniculata (Hydrangeaceae). The seeds were collected from naturally growing D. paniculata populations in Mt. Palgong, Gyeonsangbuk-do, Korea every two weeks over a three month period from August through October, 2014, totaling five collections. We examined seed characteristics such as seed size (length and width) and weight, and germination percentages for each collection stage under four different germination temperature regimes. We also examined germination after 30 and 60 days of cold moist stratification. Each collection phase had a different germination percentage and the lowest germination percentage was observed in the seeds from phase I (average GP of 15.7%). The germination percentage was similar at the later collection phases and in the higher temperatures suggesting that later collection times and higher temperatures are optimal for D. paniculata germination. We found that the optimum collection stage with high viability and germination percentage of for D. paniculata was during the middle of the September.

Effects of Plug Cell Trays, Soil and Shading Rates on Seed Germination and Seedling Growth Characteristics of Hippophae rhamnoides L.

  • In this study, basic data with respect to the introduction of Hipphophae rhamnoides L. and its cultivation in Korea could be obtained. According to the size of the plug cell tray, Chinese origin's rate of seed germination was relatively high in 128 plug cell tray, and growth was vibrant in 50 plug cell tray. The germination and growth of Russian origin seeds showed that they were relatively effective in 50 plug cell tray and with respect to soil environment, TKS-2 soil with untreated shading relatively promoted both germination and growth for Chinese origin, the rate of germination was high in bed soil for horticulture and growth result was good in TKS-2 in the case of Russian origin. It was confirmed that the germination rate of Chinese origin H. rahmnoides L. was highest in untreated shading and the shoot growth was vibrant in 70% shading while the growth in roots was vibrant in the untreated shading. In the Russian origin, H. rhamnoides L. the germination rate in 30% and 70% shading was about 50% which was higher than that in the untreated shading and general growth was vibrant in 30% shading.

Optimal Condition for Pollen Germination of Rare and Endangered Forsythia saxatilis (희귀.멸종위기 산개나리(Forsythia saxatilis) 화분의 최적 발아 조건)

  • Optimal condition for pollen germination was suggested as a basic research of flowering physiology in order to identify the characteristics of flowering and seed setting of rare and endangered Forsythia saxatilis Nakai. Pollen samples were collected during flowering time from the end of March to the beginning of April. First, a suitable germination temperature, medium sucrose concentration and germination time were determined for pollen germination and pollen tube elongation in vitro, and then an optimal pH of culture medium. Pollen germination and tube elongation were significantly different among the levels of germination temperature, sucrose concentration and germination time. Interactive effects were observed between germination temperature and time, germination temperature and sucrose concentration, germination time and sucrose concentration. Pollen germination was the highest at $10^{\circ}C$ and increased with the increase of sucrose concentration, whereas it had no relation with germination time. In addition, pollen germination and tube elongation did not increase at more than 15% of sucrose concentration and 24 hours later. Pollen germination was the highest at pH 5 (20.8%) and the lowest at pH 6 (3.8%). In conclusion, $10^{\circ}C$, 15% sucrose and pH 5 were proposed as the optimal condition for pollen germination 24 hours later of pollen culture.

Studies on Seed Germination Characteristics and Patterns of Protein Expression of Lithospermum erythrorhizon by Plant Growth Regulators and Seed Primings (생장조절제와 프라이밍 처리에 의한 지치종자의 발아특성과 단백질 발현 양상에 관한 연구)

  • This study was conducted to investigate the quality of seeds, the germination rates and the days required for germination, to examine the patterns of protein expressions during the germination and to improve the techniques of managing and storing seeds and viability of the seeds of Lithospermum erythrorhizon Sieb. et Zucc. After collecting and harvesting seeds, they were classified to white and brown colors of seed coat through testing their seed size, weight, and quality. The germination rates, the days required for germination, and the protein expressions were examined with different colors of seed coats, storing temperatures and durations by treating the different plant growth regulators and primings. One hundred seed weight of white color was heavier about 1.17 g than those of brown one about 0.81 g. The germination rates in white color of seed coat was higher, 3.05 ~ 5.75%, than those in brown one. Its rates were decreased with getting longer in storage durations. There was no big differences on germination rates between storage temperatures. The plant growth regulator of $GA_3$ and Kinetin was affected to improve the seed germination. $GA_3$ increased the seed germination clearly at 25 ppm level, while kinetin increased it gradually from 25 to 100 ppm levels. In germination by seed primings, PEG6000 made higher germination rate with increasing their levels, whereas $KNO_3$ increased the germination until 100 mM level and then decreased it with 200 mM unlike PEG6000. The protein expressed during the seed germination were appeared more and clearer bands in the seed after germination, especially 20 ~ 30 kDa, compared to those in the seed before germination. These results showing more and clearer bands were positively related to the germination rates which were different by seed colors, storage temperatures and durations, and plant growth regulators and primings.

  • The germination characteristics of Diplachne fusca (L.) P. Beauv. seeds were investigated under different seed storage conditions to find out reliable system for maintaining the seeds with high and uniform germination rate and thus for possible use of the seeds in herbicide screening with a continuous seed supply. When the seeds were stored under wet-low temperature($4^{\circ}C$) condition, the germination rate was 88% after 4-week-storage. The germination rate slightly declined after the storage for longer than 3 months. Dry seeds stored at room temperature exhibited very low germination rate. The wet-low temperature treatment was effective for inducing the germination of the seeds which had been stored under dry-room temperature condition for 4 months. The germination rate was 70% after 2-week- storage under the wet-low temperature condition. The germination rate was much higher under an alternate temperature condition than under a continuous temperature condition. The optimum temperature was 35/$25^{\circ}C$(14/10hrs). The seeds had a capability to germinate under NaCl-treated condition even at a concentration of 1.0%, whereas the germination of Echinochloa crus-galli seeds was completely inhibited by 0.5% NaCl. This result indicates that D. fusca has an advantage over E. crus-galli to survive in reclaimed 1and from the sea.

Effect of Water Uptake Rate on Germination Characteristics of Waxy Rice Seeds and Guaiacol Peroxidase Activity during Early Imbibition (종자의 수분흡수속도가 찰벼 품종별 발아특성과 침윤초기 Guaiacol Peroxidase활성에 미치는 영향)

  • Germination is controlled by internal factors of seeds and external factors such as water, temperature and light. We investigated the relationship between germination characteristics of four waxy rice cultivars and patterns of water uptake, antioxidant enzymes and free soluble sugars during early imbibition. Seed viabilities by tetrazolium test of four different rice cultivars were higher than 95% and germination rates of the hulled rice seeds were on 95% average. However, germination rate of intact rice among four cultivars showed a big difference depending on temperature. Water uptake of hulled and intact rice seeds during imbibition reached a stationary phase at around 30% moisture content. Although rates of water uptake were faster in hulled rice and high temperature than intact rice and low temperature condition, difference of those among cultivars was greater under low temperature than high temperature. The time required for rice seeds to uptake 30% water was negatively correlated with percentage of germination, germination energy, germination speed and mean germination time. Guaiacol peroxidase activity at 24h of imbibition was correlated with germination energy and germination speed but not percentage of germination. Catalase activity, soluble protein and maltose concentration at 24h of imbibition were not correlated with characteristics of germination. These results suggest that a time required for rice seeds to uptake 30% of water significantly correlated with germination and guaiacol peroxidase activity during early imbibition plays an important role in initiation of germination.
