Pratical Use of Several Ground Covers on a Slope Revegetation Construction - Miscanthus sinensis var. purpurascens, Festuca arundinacea, Pennisetum alopecuroides, Zoysia japonica - (몇몇 지피식물의 비탈면녹화공사 활용성 연구 - 억새, 톨훼스큐, 수크령, 한국잔디 -)

  • Research was initiated to investigate a vegetation characteristics of the species of ground cover plants mixed with herbeceous and woody plants on a cut slope. 4 different ground cover plants (Miscanthus sinensis var. purpurascens, Pennisetum alopecuroides, Festuca arundinacea and Zoysia japonica) were seeded at the same rate of $10.0g/m^2$ with herbaceous plants (Lotus corniculatus var. japonicus, Dianthus sinensis, Aster yomena, Chrysanthemum zawadskii and Coreopsis drummondii L) at the total seeding rate of $7.5g/m^2$ and native woody plants (Lespedeza bicolor Turcz, Indigofera pseudo-tinctoria and Albizzia julibrissin) at the total seeding rate of $7.5g/m^2$, respectively. This experiment was treated with 3 replications on a randomized block design. Data such as surface coverage rate(%), the germinating number, plant height and visual quality were analyzed. There were no statistic differences observed in the soil hardness and the soil moisture content while a significant difference was observed in the surface coverage rate and the germinating number. The surface coverage rate and the germinating number were significantly high in the Pennisetum alopecuroides plot when compared with the Miscanthus sinensis var. purpurascens plot, the Festuca arundinacea plot and the Zoysia japonica plot, respectively from the viewpoint of ground covers. Especially, the surface coverage rate was considerably low in the Festuca arundinacea plot. However, the surface coverage rate and the germinating number were no statistic differences observed in this treatment from the viewpoint of herbaceous and woody plants. Also, both of the plant height and visual quality were high in the Pennisetum alopecuroides plot when compared with the Miscanthus sinensis var. purpurascens plot, the Festuca arundinacea plot and the Zoysia japonica plot, respectively. We concluded that Pennisetum alopecuroides was the optimum plant from the treated ground covers mixed with native woody and herbaceous plants for the balanced emergence from the viewpoint of the ecological slope vegetation.

Seed Pelletizing of Salvia splendens and Calendula officinalis for the Greening and Re-vegetating (녹화와 식생조성을 위한 샐비어와 금잔화 종자의 펠렛처리)

  • The purpose of this study was to pelletize to calendula and salvia seeds with peat moss as basic material for the efficient greening and vegetating of slopes and damaged areas. Also the pelletizing was compressed by spherical types that mixed basic fertilizer of N.;300 mg/l, P.;200 mg/l, K.;400 mg/g and plant growth regulator of A and NAA each 300PPM. Soil and soil surface seeding methods were researched to find the growing state of germination percent, germination date, germination force, length of leaf, number of leaf, width of leaf, length of plant, and etc. Comparing with controlled pelletizing, peat moss and +GA pelletizing treatments resulted in higher from two to three times as following growing states : length of leaves, number of leaves, width of leaves, length of plants, length of roots, fresh weights, and ratio of germination. Especially the two treatments above showed four more times effects than the +NNA treatment. Also their germinating date germinating force were earlier and stronger. The surface seeding method was superior to soil seeding.

Studies on Protein Profiles and Isozymes in Germinating Seeds (종자발아에 있어서 Protein Profile과 Isozyme에 관한 연구)

  • The purpose of this experiment was to study one side of germination physiology based on that protein profiles and protease relating to protein metabolism, that peroxidase, catalase, $\alpha$-amylase, $\beta$-amylase, and malate dehydrogenase involved in the carbohydrate metabolism of seed germination. All these experiments were divided into the two groups with and without acetone treatment, and were carried out. The protein bands of each germinating stage between the groups treated with and without acetone showed certain basic pattern in polyacrylamide gel disc electrophoresis. However, there was a little difference in the number of protein band, optical density, and migration velocity between two groups. The isozyme bands of peroxidase, and catalase between two groups in polyacrylamide gel disc electrophoresis did not show the numeral difference, but the optical density of certain germinating stage treated with acetone was higher than the group untreated with it and it showed their enzyme activity. The $\alpha$-amylase and $\beta$-amylase activities which involved in starch metabolism of seed germination were higher in the treated group than the other. On one hand, the protease activity of hydrolase occurred in the seeds for germination was also higher, more or less in the treated group than in the other. The isozyme band pattern of malate dehydrogenase in TCA cycle of energy metabolism pathway was very different between two groups growing for 72 hours with and without acetone treatment in cellulose acetate electrophoresis. It indicated that two isozyme bands of malate dehydrogenase was high. Consequently these experimental results mentioned above indicated that acetone treatment before sowing had an effect on dissolving certain complexed lipid substance involved in the seed coats, the activity of carbohydrate hydrolase increased with water absorption which was most comfortable in its germination, dissolved glycerin and fatty acid became certain energy source, and they stimulated the acceleration of respiration metabolism.

  • Ethanol extracts of Mung Bean seeds and seedings were analysed by 2-dimensional and circular paper partion chromatography for Nitrogen compounds as a part of the study on the Amino acids metabolism. In the seeds, 18 ninhydrin positive substances were present, before germination, but the number increased to 21 after germination. There were 3 unknown substances and one of it formed newly after germination. After 2-days germination, the amount of amides, such as Asparagine and Glutamine. where increased very large which were very small amount before it. Those were accumulated more in dark place than in light and the amount of Asparigine were more than that of Glutamine. Through the germination, there were large amount of Glutarmic acid, Aspartic acid and Alanine which seems to be concerned in transamination reaction in seedings. Valine, Leucine, and Phenylalanine increased to considerable amount after germination. This is very remarkable fact as those Amino acids were reported to be concerned in transamination reaction recently. ${\gamma}$-amino butyric acid was detected in both Cotyledon and Embroy through the germination. It seemed that there is no any Nitrogen Metabolism in the unbroken seed even if it is preserved very long period.

Effect of Shifting Planting - time and Different Nitrogen Level on the Yield and Characteristics of Plant Growth in Safflower, Carthamus Tinctorius L. (파종기 추동 및 질소비료수준차이가 홍화의 생육, 수량에 미치는 영향)

  • Effect of shifting planting-time and different nitrogen on the yield and characteristics of plant growth in safflower, Chrthamus Tinctorius L. This study was carried out to investigate the optimum sowing-date and the ecological variations due to differerent amount of nitrogen for safflower on the Experiment Farm of Sang Ji college from March 17 to may 16, 1980, sowing seeds with an interval of 10 days and the amount of nitrogen were applied in 5 levels (Non, half-standard, standard, one and half-ordinary amount-No and twice amount). The triple super phosphate and pottassium chloride were applied only in standard amount. The results obtained are summarized as follows; 1. As sowing-date was delayed, the germination-ratio decreased, germinating speed increased, and the time required to attain the most vigorous germinating stage and days required for germination shortened. 2. Plant height, number of branch and stem diameter showed a decrease as sowing-date was delayed. And at the same time, plant height and number of branch showed a tendency to increase as amount of nitrogen was increased. 3. As sowing-date was delayed, the number of pods and the weight of 1000-grains were decreased. Moreover, the earlier sowing-date was, the more it increased. And as amount of nitrogen was increased, number of pods and yield also were strikingly increased. 4. The fresh-weight of flower with orange and orange-red colour, as influenced by the different sowing-date, was found out to be largest at the begining of April. But increasing amount of nitrogen did not show influence upon the promotion of flowering. 5. Judging from the results reported above, the optimum sowing-date of safflower seemed to be the begining of April; also the culture of safflower seemed to be the effects of much amount of nitrogen.

Application of in vitro ovary culture for cottonwood (Populus deltoides) breeding

  • Five different poplar hybrids were tested for rescuing embryo to elongate in vitro plantiets after hybridization. Ovaries and ovules were cultured on Woody Plant Medium (WPM) supplemented with cytokinins, 6-benzylamine (BA) and zeatin. Multiple shoots were initiated from half section of capsule with immature embryos after 21 days from pollination and tiny shoots were formed after the expansion of cotyledons in ovule cultures. Germinating response was better in intraspecific hybrids $(6.53\pm1.66)$ than interspecific crosses $(0.93\pm0.54)$ from half section of capsules on WPM medium. In general, zeatin was better than BA in inducing multiple shoots from isolated ovules. The highest average number $(19.40\pm4.53)$ of shoots was produced from immature ovules of 21 days post-pollination of WPM medium supplemented with 5.0 mg/L zeatin. The highest percentage of germination was 93% from the half section of in vitro ovary cultures. Soil acclimation was successfully conducted in cell tray containing artificially mixed soil with 96% survival rate.

Antifungal Activity of a Synthetic Cationic Peptide against the Plant Pathogens Colletotrichum graminicola and Three Fusarium Species

  • A small cationic peptide (JH8944) was tested for activity against a number of pathogens of agricultural crops. JH8944 inhibited conidium growth in most of the tested plant pathogens with a dose of $50{\mu}g/ml$, although one isolate of Fusarium oxysporum was inhibited at $5{\mu}g/ml$ of JH8944. Most conidia of Fusarium graminearum were killed within 6 hours of treatment with $50{\mu}g/ml$ of JH8944. Germinating F. graminearum conidia required $238{\mu}g/ml$ of JH8944 for 90% growth inhibition. The peptide did not cause any damage to tissues surrounding maize leaf punctures when tested at a higher concentration of $250{\mu}g/ml$ even after 3 days. Liposomes consisting of phosphatidylglycerol were susceptible to leakage after treatment with 25 and $50{\mu}g/ml$ of JH8944. These experiments suggest this peptide destroys fungal membrane integrity and could be utilized for control of crop fungal pathogens.

Kinds, Abundance and Pathogenicity of Pythium Species Isolated from Maize Rhizosphere of Various Habitats in El-Minia Governorate, Egypt

  • A total of 374 Pythium isolates were isolated and identified from the rhizosphere soils of maize plants grown in 100 different agricultural fields in nine provinces at El-Minia Governorate, Egypt. Five Pythium spp. of P. deliense, P. graminicola, P. irregulare, P. oligandrum and P. splendens were obtained P. deliense and P. oligandrum were predominant in all of the locations with 48.1% and 41.4% of total counts, respectively. P. graminicola, P. irregulare and P. splendens were not isolated in 4, 2 and 4 provinces out of 9 provinces with 5.3%, 3.5%, and 1.6% of isolation percentage from total counts, respectively. Number of Pythium isolates in each were 34, 31, 34, 33, 34, 96, 37, 37 and 38 out of 374, for locations of El-Edwa, Maghagha, Beni-Mazar, Matai, Samalout, El-Minia city, Abou-Querquas, Mallawi and Der Mawas, respectively. Pre- and post-emergence pathogenicity tests indicated that only P. deliense was highly pathogenic to germinating grains and seedlings of maize whereas P. oligandrum was non-pathogenic.

Isolation and Characterization of Four Carboxypeptidases in Canavalia lineata Cotyledons

  • Four carboxypeptidases, CP1, CP2, CP3, and CP4 were isolated from the cotyledons of germinating seedlings of Canavalia lineata by sequential chromatography on the following four columns: 1) CM-cellulose, 2) Sephacryl 5-300, 3) Procion red dye, and 4) Sephacryl S-200. A number of properties of the enzymes, such as substrate specificity, molecular weight, optimum pH, thermal stability, have been determined. Enzyme activities were measured using the Cbz(carbobenzoxy)-dipeptides containing phenylalanine at the penultimate position. The $K_m$ values of four carboxypeptidases for Cbz-Phe-Ala were 0.50, 0.65, 1.30, and 1.35 mM, respectively. The inhibition studies indicated that the four carboxypeptidases were all serine type. Each of the carboxypeptidases with molecular weights of 145, 114, 105, and 104 kDa, respectively, had the optimum enzyme activity at pH 5.0~6.0. And they were sensitive to high temperature.

Analysis of germinating seed stage expressed sequence tags in Oryza sativa L. (벼 발아종자 발현유전자의 발현특성분석)

  • Seed germination is the important stage to express many genes for regulation of energy metabolism, starch degradation and cell division from seed dormancy state. For the functional analysis of seed germination mechanisms, we were analyzed the rice cDNA clones (Oryzasativa cultivar Ilpum) obtained from seed imbibition during 48 hours. Total number of 18,101 Expressed Sequence Tags (ESTs) were clustered using SeqMan program. Among them, 8,836 clones were identified as unique clones. We identified the chitinase gene specifically expressed in seed germination and amylase gene involved to starch degradation from the full length cDNA analysis, and several genes were registered to NCBI GeneBank. To analyzed the commonly expressed genes between inmature seed and germinated seed, 25,66 inmature ESTs and 18,101 germinated ESTs were clustered using SeqMan program and identified 2,514 clones as commonly expressed unigene. Among them, alpha-glubulin and alcohol dehydrogenase I were supposed to LEA genes only expressed in the immature and germinated seed stages. For the clustering of orthologous group genes, we further analyzed the 8,836 EST clones from germinating seeds using NCBI clusters of orthologous groups database. Among the clones, 5,076 clones were categorized into information storage and processing, cellular processes and signaling, metabolism and poorly characterized genes, proportioning 783 (14.29%), 1,484 (27%), 1,363 (24.8%) and 1,869 (34%) clones to the previous four categories, respectively.
