Studies on the Insecticide Resistance of the German Cockroach (Blattella germanica L.) I. Comparisons of Toxicity by Bioassay (바퀴(Blattella germanica L.)의 살충제 저항성에 관한 연구. I. 생물검정 방법에 따른 살충력 비교)

  • Experiments were conducted to study effects of different bioassay methods on toxicity to the German cockroach (Blattella germanica L.) ith six insecticides. For male adults, by dry film method, the {TEX}$LC_{50}${/TEX} value of chlorpyrifos. fenvalerate, propoxur, permethin, DDVP, and hydramethylnon was 1.79, 1.87, 3.04, 4.37, 32.72, and 270.81 ppm, respectively and the same tendency was shown in the female. For male adults, by topical micro-application method, the {TEX}$LD_{50}${/TEX} value of DDVP and fenvalerate, chlorpyrifls, permethrin, propoxur, hydramethylnon was 2.63, 3.79, 4.51, 6.73, 44.99 $\mu\textrm{g}$/g, respectively and the same tendency was shown in the female. From the diet toxicity method against male adults, the CT value (Concentration%$\times${TEX}$LT_{50}${/TEX}) showed that chlorpyrifos was rapid effective and hydramethylnon was residual effective.

Field trial on the control effect of fipronil bait against German cockroaches

  • A field trial on the control effect of fipronil poison bait against German cockroaches (Blatella germanica) was carried out at different restaurant types in Sinchon, Seoul, Republic of Korea. Monitoring was performed applying food baited traps for 2 days per week. Reduction rates of German cockroaches by applying fipronil baits were 90.9% at Korean restaurants, 96.4% at Chinese restaurants, and 89.4% in beer hall kitchens after 4 weeks of the treatment. Overall average of the reduction rate was 93.9%. As the natural reduction rate at untreated restaurants was 11.5% after 4 weeks, a correction of the average reduction rate by applying the Abbot formula was 93.1 %.

Effective Control in Managing German Cockroach, Blattella germanica (Orthoptera: Blatellidae) Using a Push-Pull Strategy (기피-유인을 이용한 바퀴의 효율적 방제)

  • To enhance the control efficacy against cockroaches in dwelling space, repellent ("Push") was set down on one comer and a poison bait including attractant ("Pull") was on the opposite side. And we tested the control efficacy of Push-Pull strategy by behavior-stimuli of cockroaches. In a mini-field test, German cockroach males primarily chose shelters nearest to the attractant-treated surfaces and farthest from the repellent-treated surfaces. The consumption of food or bait was also highest from food nearest to the preferred shelters by push-pull effect. We evaluated the push-pull insecticidal efficacies of five essential oils belong to Citrus plant, namely, grapefruit, lemon, lime, orange, and petitgrain. The combined push-pull treatments appeared to be faster and higher insecticidal effects than single (pull) treatment, and the repellent efficacy of Citrus oils was in the order of grapefruit > lemon > lime > orange > petitgrain. Therefore, we propose the effective control of the german cockroach using Citrus oils as repellents and push-pull method into a limited area.

  • In this study, the trypsin gene (bgtryp-1) from the German cockroach, Blattella germanica, was cloned via the immunoscreening of patients with allergies to cockroaches. Nucleotide sequence analysis predicted an 863 bp open reading frame which encodes for 257 amino acids. The deduced amino acid sequence exhibited $42-57\%$ homology with the serine protease from dust mites, and consisted of a conserved catalytic domain (GOSGGPLV). bgtryp-1 was determined by both Northern and Southern analysis to be a 0.9 kb, single-copy gene. SDS-PAGE and Western blotting analyses of the recombinant protein (Bgtryp-1) over-expressed in Escherichia coli revealed that the molecular mass of the expressed protein was 35 kDa, and the expressed protein was capable of reacting with the sera of cock-roach allergy patients. We also discussed the possibility that trypsin excreted by the digestive system of the German cockroach not only functions as an allergen, but also may perform a vital role in the activation of PAR-2.

  • Bla g 2 is a cockroach allergen of great importance, This study was conducted to identify IgE-binding epitope(s) of Bla g 2 using the recombinant protein technique, Approximately 50% of tested sera showed IgE reactivity to Pichia-expressed Bla g 2 (PrBla g 2) and E. coli-expressed Bla g 2 (ErBla g 2), Only 5,3% of serum samples showed stronger reactivity to PrBla g 2 than ErBla g 2, indicating that serum was reactive to conformational or carbohydrate epitopes. The full-length and 5 peptide fragments of Bla g 2 were produced in E. coli. All fragments showed IgE-binding activity to the cockroach-allergy patients' sera. Specifically, peptide fragments of amino acid residue 1-75 and 146-225 appeared to be important for IgE-binding. The information about the IgE-binding epitope of Bla g 2 can aid in the diagnosis and treatment for cockroach allergies.

Insecticidal Effect of Cockroach Baits and their Persistent Efficacy Against the German Cockroach, Blattella germanica (바퀴 독먹이제의 살충 효과 및 지속성 검증)

  • Cockroaches are a not only nuisance insects but also have medical importance as they mechanically propagate various pathogens. To date, baits have been widely suggested for use in cockroach control. In this study, we investigated the insecticidal effects of three Korean baits-Korea Combat Ultra Slim (K-CUS), Korea Combat Power (K-CP), and Korea Zaps Dual Bait (K-ZDB)-and three US baits-US Combat Source Kill Max (US-CSKM), US Hot Shot MaxAttrax Roach Bait (US-HSM), and US Raid Double Control Small Roach Bait (US-RDC)-on the German cockroach (Blattella germanica). Among the six baits, three (K-CUS, K-CP, and US-CSKM), the main active ingredients of fipronil, showed rapid and strong insecticidal efficacy. In addition, we compared the persistence of the insecticidal effects of K-CUS and K-CP 6 months and 12 months after initial opening. Insecticidal effects of both baits decreased over time, but the mortality rates of cockroaches exposed to 6-month- and 12-month-aged baits were over 90%, suggesting that these baits can maintain their insecticidal effect for at least one year after indoor installation.

  • The ultrastructure of freeze-substituted Malpighian tubule cells of German cockroach after rapid freezing was compared to that fixed with conventional chemical fixation. As results, freeze-substitution proved to be the superior technique revealing much more structural details of the tissue cells. Especially, all kinds of membrane structures including infoldings, microfilament, microtubules and ribosomes were best preserved in this material.

Studies on the Insecticide Resistance of the German Cockroach (Blattella germanica L.) II. Resistant Developments and Cross resistance (바퀴(Blattella germanica L.)의 살충제 저항성에 관한 연구. 2. 저항성 발현 및 교차저항성)

  • The German cockroach (Blattella germanical L.) populations were successively selected with chlorpyrifos and permethrin for xis generations. The resulting resistant strains, $R_{chlorpyrifos}$(Rc) and $R_{permethrin}$(Rp) were observed resistant development and the cross-resistance. The result indicated that the values of $LC_{50}$ were increased 3.32 and 2.10 times from the susceptible strain, respectively. In the Rc strains, the values of the $LC_{50}$ of fenvalerate, propoxur, permethrin were to 3.89, 2.97, and 1.31 times higher than that of the suseptible, respectively. In the Rp strains, the values of the $LC_{50}$ of fenvalerate, chlorpyrifos, and propoxur showed 5.23, 1.42, and 1.80 times cross-resistance.
