Skin Permeation and Crosslinking of a Biological Tissue with Hydrolyzed Product of Gardeniae Fructus

  • For the purpose to treatment of skin disease genipoiside and hydrolyzed product of Gardeniae Fructus were studied on skin permeation and crosslinking of a biologied tissue. Geniposide was hydrolyzed to genipin by ${\beta}$-glucosidase and the rate of hydrolysis was rapid on the condition of high temperature of medium and high concentration of ${\beta}$-glucosidase. The permeation enhancing effects of geniposide and genipin under cream and gel preparations were tested using Franz type diffusion cell and the skin of hairless mouse. (omitted)

한약의 수치에 관한 연구(제 7보) -치자의 수치에 의한 성분변화 및 생리활성- (Studies on the Processing of Crude Drugs(VII) -On the Constituents and Biological Activities of Gardeniae Fructus by Processing-)

  • We have conducted to characterize the physico-chemical change and pharmacological transformation of traditional herbal medicines by means of processing. Processed Gardeniae Fructus was prepared by heating of fruit of Gardenia jasminoides(GF) for $30{\sim}50\;minute$ in the roster designed for herb processing. The contents of drying loss, water extract, diluted ethanol extract, ether extract and geniposide in non-processed GF and processed GF were examined. The contents of drying loss, water extract and geniposide in processed GF showed a decrease as compared with those of non-processed GF, however the contents of dilute ethanol and ether extract showed a increase as compared with those of non-processed GF. The rate of decrease/increase of those index were in proportion to heating time. And, biological activities of methanol extract of non-processed GF and processed GF were investigated. DPPH scavenging effects and inhibitory effect of xanthine oxidate and hemolysis of processed GF exhibited more effective than those of non-processed GF in vitro. Accelerating effect of large intestinal transport and purgative action of non- processed GF were discriminated by processing of GF. Methanol extracts of non-pro- cessed GF and processed GF showed the protective effects against the hepatotoxicity induced by ${\alpha}-naphthylisothiocyanate$ in rats. These results suggested that the transformation of biological activities of GF by means of processing may be due to the physico-chemical change of the constituents in GF by heating.

두중(杜仲)의 수치(修治)에 따른 실험적(實驗的) 연구(硏究) (EXPERIMENTAL STUDIES OF EUCOMMIAE CORTEX ACCORDING TO PROCESSING)

  • In order to estimate the effects of Eucommiae Cortex, which is one of the most important herb medicines used for invigorating the liver and kidney and strengthening ones and muscles, fetus-soothing etc., this experiment was conducted the quantitative analysis of geniposidic acid and geniposide by HPLC, and the analgesic effects of mice, the lipid metabolism of rats, the catecholamine concentration in the brain and the plsma of rats stressed by immobilization. The results obtained are as follows. 1. The contents of geniposidic acid and geniposide were 22.3mg/g, 5.4mg/g in Eucommiae Cortex without processing, and 119.8mg, 10.4mg/g in Eucommiae Cortex fried with salts. 2. In analgesic effects of mice, there were all significant in Eucommiae Cortex without processing and Eucommiae Cortex fried with salts. 3. In the effects of lipid metabolism of rats, there were not significant on the change of organ weight and the level of serum total lipid and total cholesterol, triglyceride, phospholipid, HDL-cholesterol. But, there were significant on the level of free fatty acid. 4. In tile effects of the catecholamine concentration in the brain and the plsma of rats stressed by immobilization, there were all significant in Eucommiae Cortex without processing and Eucommiae Cortex fried with salts, especially in case of norepinephrine. From the results mentioned above, there were significant on the analgesic effects of mice, and the catecholamine concentration in the brain and the plsma of rats stressed by immobilization. And there were different from Eucommiae Cortex without processing and Eucommiae Cortex fried with salts in the contents of geniposidic acid and geniposide. These results are relative to invigorating the liver and kidney and strengthening ones and muscles, and fetus-soothing, therefore when we use Eucommiae Cortex, it is more desirable to use Eucommiae Cortex with processing.

두충(杜沖) 추출물의 암세포증식 저해효과 (Inhibitory Effects of the Stem Bark Extract of Eucommia ulmoides on the Proliferation of Human Tumor Cell Lines)

  • A bioassay-guided fractionation of the stem bark extract of Eucommia ulmoides Oliver (Eucommiaceae) led to the isolation of three iridoid constituents, genipin (1), geniposide (3), geniposidic acid (4) as well as $({\pm})-guaiacylglycerol$ (2) and fatty acid mixtures as active ingredients of the extract responsible for the antitumoral property. The EtOAc soluble part and BuOH soluble part of the extract demonstrated a potent inhibition on the proliferation of cultured human tumor cells such as A549 (non small cell lung), SK-OV-3 (ovary), SK-MEL-2 (melanoma), XF498 (central nerve system) and HCT-15 (colon) in vitro, whereas the remaining water soluble part exhibited a poor inhibition. The intensive investigation of the EtOAc soluble part and BuOH soluble part of the extract yielded genipin, guaiacylglycerol, geniposide, geniposidic acid and large amounts of fatty acid mixtures as active components.

치자(梔子)의 규격 기준 모니터링 및 HPLC 표준크로마토그램 연구 (Quality Monitoring of Specification Standard of Gardeniae Fructus in the Korean Pharmacopoeia and Studies HPLC Standard Chromatogram)

  • Objectives : Gardeniae Fructus is a ripe fruit of Gardenia jasminoides Ellis, which has been used as traditional medicines for anti-inflammatory, diuretic, antipyretic, and antibacterial activity. The aim of this study was to compare of Gardeniae Fructus in South Korea collected during three years according to the standards in monographs of the Korean Pharmacopoeia Eleventh edition (KP11). Methods : 30 items of Gardeniae Fructus from two cultivation regions were classified into dried(n=15) & steamed (n=15) and tested according to the standards in monographs of the KP11. Gardeniae Fructus was carried out identification(comparison of colors, thin layer chromatography), heavy metals, residual pesticides, total ash, and assay registered at KP11. Add to we tested loss on dry, contents of ethanol-soluble extracts, and HPLC profiling. Results : In TLC chromatogram of identification test, the spot of gardenoside and geniposide were observed at $R_f$ value of about 0.3 and 0.5. Heavy metals and residual pesticides met the requirements of the standards for all samples. The results of total ash of each samples are measured maximum 4.87 %. According to HPLC for assay, the samples contain 4.80~6.10 % of geniposide and 0.45~1.83 % of gardenoside. Conclusion : We have verified the current specification standard of Gardeniae Fructus and standard that is not set. By the results, it is proposed a new draft of loss on drying and confirmed the content of gardenoside revised. HPLC standard chromatogram of Gardeniae Fructus is proposed. We hope that it will help the standardization of Gardeniae Fructus.

천연 치자 색소의 연구개발 동향 (A Trend in Research and Development of Natural Gardenia Pigments)

  • 치자나무 (Gardenia jasminoides)는 우리나라 남부 지방에 널리 분포하는 이리도이드 (iridoid)계 꼭두서니과의 수목으로서 그 열매인 치자는 예로부터 옷감의 염료, 식용 색소 및 한방 약재로 널리 사용되어 왔다. 치자에 함유된 주요성분으로는 치자 과실에 이리도이드 배당체인 제니핀 (genipin: $C_{11}H_{14}O_5$)과 제니포사이드 (geniposide: $C_{17}H_{24}O_{10}$) 및 겐티오비오시드 (gentiobioside), 크로신 (crocin), 노나코산, 기르네노시드, 만니톨 등이 있고 잎에는 가데오시드 (gardeoside)가 포함되어 있다. 치자색소중 가장 대표적인 치자황색소는 매염처리에 따라 색상이 거의 변하지 않는 단색성 염료이면서 매염처리를 하지 않고도 면직물 등의 식물섬유에 염색이 되는 직접 염료로서 그 시장가치가 높다. 현재 치자황색소는 가격경쟁의 격화에 따라 금액 기준으로 약간 감소하고 있으나 수요 그 자체는 라면 등 식품을 중심으로 광범위한 시장을 형성하고 있다. 다음으로 치자청색소는 치자추출물을 효소반응이나 미생물 배양을 통한 가수분해에 의해 다량의 제니핀으로 전환하여 여러 아미노산과 함께 생산이 이루어지게 된다. 치자청색소는 단백질 섬유에 염색한 결과 매염제 없이도 우수한 착색을 보였으며, 견뢰도도 우수하게 나타났다. 또한 냉과류의 합성착색료 대체품으로 신장하고 있는 스피룰리나 청색소와 가격경쟁력이 있어서 앞으로의 수요는 더 커질 수 있을 것으로 사료된다. 치자적색소는 520-540 nm에서 최대흡광도를 나타내며, 이는 청색소와 마찬가지로 이리도이드 배당체를 효소 및 미생물로 가수분해한 구성물과 일차 아미노 그룹을 포함한 기질과 함께 반응함으로써 형성된다. 치자 색소는 식품 뿐만 아니라 섬유의 염색에도 우수한 천연색소로서 가치가 있다고 사료된다. 따라서 앞으로 식품에 있어서 천연색소로의 대체가 당면한 과제이며 이를 위해 식용색소로서 조건들을 만족시킬 수 있는 기술개발은 계속 이루어져야 할 것이다. 천연색소의 안정성 문제는 식품 뿐만 아니라 화장품, 섬유 염색 등 거의 모든 부분에서 제약이 되므로 추후 천연염료 (특히 치자 염료)의 안정성을 증가시킬 수 있는 품질 개발과 생산 코스트 다운을 위한 연구는 전체 천연색소 시장의 성장에 필수불가결하다.

Separation and Characterization of Water Soluble Blue Pigments Formed from Geniposide of Gardenia Fruits

  • Genipin, aglycone of geniposide isolated from fruits of Gardenia jasminoides, was transformed into blue pigments through reaction with glycine and methylamine. The blue pigments formed from glycine-reacted genipin were passed through Bio-Gel P-2 resin yielding fractions GG1 and GG2, and those from methylamine-reacted genipin were separated into fractions GM1-GM4. The first eluted higher molecular-weight fractions, GG1 and GM1, had higher tinctorial strength than the later eluted lower molecular-weight fractions, GG2 and GM2-GM4, respectively. $^1H-NMR$ spectra of GG1 and GM1 showed very broad peaks indicating that structures of the pigments were highly polymeric. $^1H-NMR$ spectra of GG2, GM3, and GM4 showed several sharp peaks at aliphatic and aromatic regions with accompanying broad peaks, although the spectrum of GM2 was rather simple. Determination of the structural and physical nature of the isolated pigments is in progress.

치자의 항산화 활성성분에 관한 연구 (Antioxidant Components of Gardenia Fruit)

  • An antioxidant activity of Gardenia Fruit (Gardenia jasminoides Ellis) which has been used for food coloring was studied. The antioxidant activity was determined by measuring lipid peroxide produced when a mouse liver homogenate was exposed to the air at $37^{\circ}C$, using 2-thiobarbituric acid (TBA). Both water and methanol extracts of Gardenia Fruit showed the antioxidant activity. On solvent fractionation, the antioxidant activity was removed into the ethyl acetate and butanol soluble fractions. And the final water soluble fraction also showed the antioxidant activity in the low concentration, but it promoted the lipid peroxidation in the high concentration. Two compounds (I and II) having the antioxidant activity were isolated from the butanol fraction, and compound I also occurred in the ethyl acetate fraction. The antioxidant activity of compound II was more potent than that of I. By analyzing data for UV, IR and $^1H-NMR$, compounds I and II were identified as geniposide and crocin, respectively.

치자 Genipin과 아미노산의 청색소변환반응에 관한 물리화학적 연구 (Physicochemical Characteristics for the Transformation of Blue Pigments from Genipin of Gardenia jasminoides with Amino Acids)

  • 우리 나라에서 오랫동안 식용 및 황색 색소원으로 이용되어 온 치자(Gardenia jasminoides)열매로부터 iridoid glycoside인 geniposide를 분리, 정제한 후 ${\beta}-glucosidase$로 가수분해하여 얻은 genipin을 glycine, alanine, histidine, lysine, phenylalanine, glutamate 등 여섯 종류의 아미노산과 반응시켜 수용성 치자청색소로 전환되는 과정을 규명하였다. Genipin이 아미노산과 반응하여 청색소가 되는 과정에서 pH의 영향을 알아보기 위하여 여러 pH에서 반응을 시켜본 결과 청색소 생성의 최적조건은 pH 7.0 이었고, pH 3.0 조건에서는 청색소가 전혀 생성되지 않았으며, pH 12.0 조건에서는 미량의 청색소만 생성되었다. 아미노산의 종류에 따라서도 청색소 생성량 및 색감에 차이가 있었는데 $Iysine({\lambda}_{max}=573\;nm),\;glycine({\lambda}_{max}=595 \;nm),\;phenylalanine({\lambda}_{max}=602\;nm),\;alanine({\lambda}_{max}=595\;nm)$에 비해 $histidine({\lambda}_{max}=601\;nm)$$glutamate({\lambda}_{max}=601\;nm)$의 경우에는 비교적 적은 양의 청색소가 생성되었다. 청색소 생성 속도상수를 여러 온도$(60,\;70,\;80,\;90^{\circ}C,\;pH\;7.0\;phosphate$ 완충용액)에서 구하였는데, 염기성 아미노산이 중성 및 산성 아미노산에 비해 생성속도가 빨랐다. 이들 값으로부터 Arrhenius 활성화에너지를 계산한 결과 $glycine(E_A=9.8\;kcal/mol)$이 다른 아미노산$(E_A=13.3{\sim}15.4\;kcal/mol)$에 비해 특히 작은 값의 활성화에너지를 나타내었다.

황련해독탕의 항균활성 및 표준화 연구 (Anti-bacterial Activity and Standardization on Hwangryunhaedok-tang)

  • 황련해독탕은 한방산업에서 염증관련 질환에 다빈도로 사용되는 처방이다. 본 연구의 목적은 전통적 한약인 황련해독탕의 항균활성을 확인하고 지표성분을 선정하는 것이다. 황련해독탕의 항균활성을 측정하기 위하여 디스크 확산법과 최소억제농도법을 사용하였다. 사용균주는 Staphylococcus aureus ATCC 25923, Escherichia coli ATCC 23724였으며 대조군으로 oxacillin과 ciprofloxacin을 이용하여 황련해독탕의 항균활성을 확인하였다. 결과적으로 디스크 확산법에서는 S.aureus ATCC 25923에서 황련해독탕 20 mg과 10 mg은 $11.7{\pm}1.3mm$, $8.7{\pm}0.7mm$의 억제환을 형성하였으며 E.coli ATCC 23724에서는 황련해독탕 20 mg이 $8.0{\pm}0.0mm$의 억제환을 형성하였다. 최소억제농도는 S.aureus ATCC 25923가 31.25 mg/mL, E.coli ATCC 23724 에서는 125.0 mg/mL였다. 표준화를 위한 HPLC 정량분석 결과, 황련해독탕은 berberine을 16.55 mg/g 함유하고 있었으며 geniposide를 81.85 mg/g 함유하고 있었다. 천연물을 이용하여 항생제를 개발할 경우에는 내성세균을 억제하고 부작용을 줄일 수 있다는 장점이 있다. 이러한 점을 착안하여 지속적으로 연구를 진행한다면 한방산업과 더불어 여러 산업에서의 발전을 기대할 수 있을 것이다.
