Population genetic structure based on mitochondrial DNA analysis of Ikonnikov's whiskered bat (Myotis ikonnikovi-Chiroptera: Vespertilionidae) from Korea

  • Background: Ikonnikov's whiskered bat (Myotis ikonnikovi) is found throughout the Korean Peninsula, as well as in Kazakhstan, Russia, Mongolia, China, and Japan. It is small-sized and primarily inhabits old-growth forests. The decrease and fragmentation of habitats due to increased human activity may influence the genetic structure of bat populations. This study was designed to elucidate the population genetic structure of M. ikonnikovi using mitochondrial genes (cytochrome oxidase I and cytochrome b). Results: The results showed that M. ikonnikovi populations from Korea have high genetic diversity. Although genetic differentiation was not detected for the COI gene, strong genetic differentiation of the Cytb gene between Mt. Jeombong and Mt. Jiri populations was observed. Moreover, the results indicated that the gene flow of the maternal lineage may be limited. Conclusions: This study is the first to identify the genetic population structure of M. ikonnikovi. We suggest that conservation of local populations is important for sustaining the genetic diversity of the bat, and comprehensive studies on factors causing habitat fragmentation are required.

A pheromone mutant of Schizosaccharomyces pombe displays nucleolar fragmentation

  • Stresses and nutritional starvation are two main external signals for the induction of sex pheromones in the fission yeast Schizosaccharomyces pombe. In an attempt to identify the components involved in transduction of starvation signals, we screened 135 temperature-sensitive (ts) mutants and isolated 6 mutants that induced the pheromone even in the presence of a nitrogen source. These mutants exhibited two distict induction phenotypes: pheromone induction at restrictive but not at permissive temperatures; and pheromone induction at both permissive and restrictive temperatures. The times required for the maximum pheromone induction at the restrictive temperature differed slightly in each mutant. In addition to the pheromone induction phenotype, the ts243 and ts304 mutants exhibited cell-division-cycle defects. The ts304 mutant cells showed an abnormal cytoplasmic DAPI staining pattern. The nucleolus of this mutant seemed to be fragmented, a phenomenon which is typically observed in aged yeast cells. The result of our genetic analysis indicated that the pheromone induction mutants belonged to 6 separate complementation groups. We designated these mutants pws1 to pws6.

  • 본 논문에서는 인터넷상의 대용량 자료에서 원하는 정보를 검색하기 위한 지원 기능으로서 분산 객체지향 데이타베이스에서 클래스를 분할하여 여러 사이트에 분산시키는 기법에 관하여 설계 및 구현하였다 제안된 분산 기법은 클래스의 분할 과정과 할당 과정으로 구성된다. 클래스의 분할 과정에서는 메소드, 계승 및 복합 객체와 같은 객체지항 데이터베이스의 특성을 반영하여 클래스를 분할하였으며 할당 과정에서는 저장, 질의 처리 및 전송비용을 고려하여 할당수식을 정의하였으며 이를 유전자 알고리즘을 이용하여 구현하였다.

Trichoplusia ni 세포의 apoptosis 메커니즘 규명을 위한 기초연구 (Basic Studies on the Apoptosis Mechanism of Trichoplusia ni Cell Line)

  • 본 연구에서는 Ttichoplusia ni 세포의 apoptosis 유도 및 억제 현상의 기초연구를 수행하였다. Apoptosis 유도제로 알려진 hygromycin B에 의한 세포 성장 저해는 $200\;{\mu}/ml$의 수준에서부터 나타났고, $400\;{\mu}/ml$ hygromycin B를 처리한 세포에서는 배양 후 2일부터 DNA가 분절되어지는 것을 확인할 수 있었다. 그러나 dexamethasone과 sodium butyrate를 첨가시 세포성장은 저해되었지만 DNA 분절현상이 보이지 않아 apoptosi의 유발여부를 확인할 수 없었다. 그리고 caspase 기능억제제의 apoptosis 지연효과를 보기 위해 $200\;{\mu}/ml$ hygromycin B로 apoptosis를 유발한 상태에서 Ac-DEVD-CHO를 첨가하여 세포성장을 비교해 본 결과 이 저해제에 의해 약 36%정도 apoptosis가 억제되었음을 확인하였다. N-acetylcysteine의 경우도 apoptosis지연 효과가 있었다. Bcl_계에 속하는 anti-apoptotic 유전자의 발현연구로서 apoptosis 저해 단백질인 bcl-2 유전자를 곤충세포에 형질전환시킨 후 이 단백질이 한시적으로 발현되는 것을 western blot분석법으로 확인하였으며 apoptosis가 지연된 곤충세포주의 개발이 가능하다는 결론을 보였다.

Genetic diversity and population structure of endangered Neofinetia falcata (Orchidaceae) in South Korea based on microsatellite analysis

  • Population genetic assessment is essential for the conservation and management of endangered and rare plants. Neofinetia falcata is endangered epiphyte orchid and protected by law in Korea. In Korea, this species is only found on islands in the South Sea of Korea (including Jeju-do) and the southern coast of the Korean Peninsula. We developed nine microsatellite makers to assess the genetic diversity and population genetic structure of three populations of N. falcata. The genetic diversity at the species level was low, which can be attributed to inbreeding or fragmentation into small, isolated populations. A recent bottleneck was detected in one population, likely due to overcollection. N. falcata exhibited moderated levels of differentiation among populations, with the three populations were divided into two clusters based on genetic structure. The genetic diversity and structure of N. falcata are affected by restricted gene flow by pollen or seeds due to isolation and geographic distance. Strategies for in situ and ex situ conservation of this species are been proposed based on the results of our study.

  • 괭이갈매기는 동북아시아에서 가장 흔한 갈매기류 중 하나이며, IUCN에 의하면 LC(Least Concern)로 분류되어 있다. 인간의 활동이 해양환경에 영향을 미친 이래 괭이갈매기는 해양 환경오염과 서식지 교란의 중요한 지표종이 되었다. 본 연구에서 괭이갈매기가 6개의 미토콘드리아 CO1 하플로타입(haplotype)이 있고, 이중 일부 희귀 haplotype은 지리적으로 제한되어 나타난다는 사실을 확인하였으며, 이러한 현상은 본 종이 멀리 분산하지 않음을 암시한다. 본 논문은 한국에서 괭이갈매기를 대상으로 한 첫번째 개체군 유전적 연구결과이며, 환경지표종으로서 모니터링과 유전학적 연구의 기초자료가 될 것이다.

Pyrolysis of Lignin Obtained from Cinnamyl Alcohol Dehydrogenase (CAD) Downregulated Arabidopsis Thaliana

  • Despite its potential as a renewable source for fuels and chemicals, lignin valorization still faces technical challenges in many aspects. Overcoming such challenges associated with the chemical recalcitrance of lignin can provide many opportunities to innovate existing and emerging biorefineries. In this work, we leveraged a biomass genetic engineering technology to produce phenolic aldehyde-rich lignin structure via downregulation of cinnamyl alcohol dehydrogenase (CAD). The structurally altered lignin obtained from the Arabidopsis thaliana CAD mutant was pyrolyzed to understand the effect of structural alteration on thermal behavior of lignin. The pyrolysis was conducted at 400 and $500^{\circ}C$ using an analytical pyrolyzer connected with GC/MS and the products were systematically analyzed. The results indicate that aldehyde-rich lignin undergoes fragmentation reaction during pyrolysis forming a considerable amount of C6 units. Also, it was speculated that highly reactive phenolic aldehydes facilitate secondary repolymerization reaction as described by the lower yield of overall phenolic compounds compared to wild type (WT) lignin. Quantum mechanical calculation clearly shows the higher electrophilicity of transgenic lignin than that of WT, which could promote both fragmentation and recondensation reactions. This work provides mechanistic insights toward biomass genetic engineering and its application to the pyrolysis allowing to establish sustainable biorefinery in the future.

Evaluation of Genetic Differentiation of Albizia lucida Populations from Eastern Region of the Indian Sub-continent by ISSR Markers

  • Level and distribution of genetic diversity in seven populations of Albizia lucida Benth. in eastern region of the Indian sub-continent were estimated using ISSR markers. Relatively higher level of genetic diversity within populations was observed in seven populations of A. lucida (mean of 0.38). From the result of AMOVA, majority of genetic diversity was allocated within populations (96.2%) resulting in a moderate degree of population differentiation. The observed distribution pattern of I-SSR variant among the populations was coincided with the typical pattern of long-lived woody tree species. Genetic relationships among the populations, reconstructed by UPGMA method, revealed two genetic groups. The population of Anugul and Bargarh turned out to be the most closely related despite a distance location between them. These formations will be of great value in the development of conservation plans for species exhibiting high levels of genetic differentiation due to fragmentation, such as indication of conservation unit size, which populations should be chosen as priority in conservation plans and which samples should be introduced in areas with a low number of individuals of A. lucida.

TES 보존액이 미니돼지 동결 융해 정자의 생존성 및 성상에 미치는 영향 (Effect of TES Extender on Sperm Characteristics and Viability of Frozen Semen in Miniature Pig)

  • The objective of this study is to estimate the effect of adding TES to LEY and FGE freezing extender for the sperm viability, acrosomal morphology and DNA fragmentation from miniature pig sperm, we evaluated sperm characteristics in TFGE, TLE and LEY with various thawing condition ($37^{\circ}C$ for 20 sec, 45 sec and $75^{\circ}C$ for 5 sec, respectively), and in different concentration of glycerol at 1%, 1.5%, 3%. The sperm viability and normal acrosome intact(NAI) in TFGE (Viability : $60.3{\pm}2.4$, NAI : $58.6{\pm}2.2%$), TLE ($61.3{\pm}2.4$, $62.2{\pm}2.2%$) extender significantly(p<0.05) increased than that in LEY ($50.2{\pm}2.4$, $54.5{\pm}2.2%$) extender thawed at $75^{\circ}C$ for 5 sec. According to the results from glycerol concentration, the viability and NAI of miniature pig sperm in 1.5% glycerol TLE ($66.1{\pm}3.2$, $66.2{\pm}1.0%$) was highest among the experimental groups. In accordance with this, DNA fragmentation rates was the lowest in TLE ($43.3{\pm}0.5%$) while that in LEY ($63.5{\pm}2.3%$) is the highest. Therefore, these results suggest that TLE extender method for freezing- thawing of miniature pig sperm increased the viability after thawing.

A Preliminary Population Genetic Study of an Overlooked Endemic ash, Fraxinus chiisanensis in Korea Using Allozyme Variation

  • We used enzyme electrophoresis to evaluate genetic diversity in five populations of endemic ash, Fraxinus chiisanensis in Korea. Of 15 putative allozyme loci examined 26.7% were polymorphic and expected heterozygosity for the species was low (0.082). Within the range, population were highly differentiated ($F_{ST}$=0.356) and little genetic variation was explained by geography. The pattern of distribution of variation showed low genetic variation within populations and pronounced divergence among populations, which was consistent with the prediction for the effects of limited gene flow and local genetic erosion. Although the frequencies of male plants were dominant ranging from 79.3% to 89.4%, most mating events seems to be inevitable mating between relatives in small populations based on heterozygote deficiency of this species. Small effective population size and the limited dispersal contributed to the low rates of gene flow within as well as between populations.
