To establish the optimal fermentation periods in the manufacture of mushroom fermentation foods using sugar, changes of quality characteristics of the mushrooms fermentation broth were investigated with changes of enzyme activity and physiological functionality during fermentation. Viscosity, L value (lightness), a value (redness) and b value (yellowness) were significantly decreased after 3 months of fermentation and after that, increased. In sensory evaluation test, unique flavors and tastes of mushrooms in the fermented broth were decreased during fermentation, whereas the other tastes and flavors were gradually increased. Overall acceptability was the highest in the 3 months of fermentation broth. ${\alpha}-Amylase$ activities of the fermented broth were significantly increased to 1 month of fermentation, however invertase and cellulose activities were low or not detected in the fermented broth. Antioxidant activities were the highest in 4 months of fermentation and after that, decreased. Tyrosinase inhibitory activities were high in all samples and they were not changed during fermentation. SOD-like activity was high in the fermentation broth of Flammulina velutipes and it was also not changed during fermentation. In conclusion, optimal fermentation periods in the manufacture of mushroom fermentation foods using sugar was 3 or 4 months.
Most clinicians understand clinical trials as the evaluation process for new medicine before their use. However, clinical trials can also be applied to laboratory diagnostic tests (LDTs) to verify diagnostic accuracy and efficacy before their clinical laboratory implementation for patients. The clinical trial of LDT has two distinctive characteristics that are different from the case of pharmaceuticals and thus worth special consideration. One of them is the level of evidence. The well-designed randomized controlled trials (RCTs) are known to provide the best evidence to prove the clinical efficacy of any pharmaceutical products. However, RCTs lose practicality when applied to LDTs due to various issues including ethical complications. For this reason, comparative study format is considered more feasible approach for LDTs. In addition pharmaceuticals and LDTs are different in that the user's intervention is not required for the former but critical to the latter. Moreover, in the case of pharmaceuticals, end-products are produced by manufacturers before being used by clinicians. However, in LDTs, once reagents and instruments are provided by manufacturers, they are first utilized by clinical laboratories to produce test results in order for clinicians to use them later. In other words, when it comes to LDTs, clinical laboratories play the role of manufacturers, providing reliable test results with improved quality assurance. Considering the distinctive characteristics of LDTs, we would like to offer detailed suggestions to successfully perform clinical trials in LDTs, which include analytical performance measures, clinical test performance measures, diagnostic test accuracy measures, clinical effectiveness measures, and post-implementation surveillance.
The aim of study was to scrutinize the physicochemical and protein profile of milk obtained from local Pakistani breeds of milch animals such as Nilli-Ravi buffalo, Sahiwal cow, Kajli sheep, Beetal goat and Brela camel. Physicochemical analysis unveiled maximum number of total solids and protein found in sheep and minimum in camel. Buffalo milk contains the highest level of fat (7.45%) while camel milk contains minimum (1.94%). Ash was found maximum in buffalo (0.81%) and sheep (0.80%) while minimum in cow's milk (0.71%). Casein and whey proteins were separated by subjecting milk to isoelectric pH and then analyzed through sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE). The results showed heterogeneity among these species. Different fractions including αS1, αS2, κ-casein, β-casein and β-lactoglobulen (β-Lg) were identified and quantitatively compared in all milk samples. Additionally, this electrophoretic method after examining the number and strength of different protein bands (αS1, αS2, β-CN, α-LAC, BSA, and β-Lg, etc.), was helpful to understand the properties of milk for different processing purposes and could be successfully applied in dairy industry. Results revealed that camel milk was best suitable for producing allergen free milk protein products. Furthermore, based on the variability of milk proteins, it is suggested to clarify the phylogenetic relationships between different cattle breeds and to gather the necessary data to preserve the genetic fund and biodiversity of the local breeds. Thus, the study of milk protein from different breed and species has a wide range of scope in producing diverse protein based dairy products like cheese.

Objectives: Lawsone (1,4 naphthoquinone) is a non redox cycling compound that can be catalyzed by DT diaphorase (DTD) into 1,2,4-trihydroxynaphthalene (THN), which can generate reactive oxygen species by auto oxidation. The purpose of this study was to evaluate the toxicity of the phytomarker 1,4 naphthoquinone and its metabolite THN by using the molecular docking program AutoDock 4. Methods: The 3D structure of ligands such as hydrogen peroxide ($H_2O_2$), nitric oxide synthase (NOS), catalase (CAT), glutathione (GSH), glutathione reductase (GR), glucose 6-phosphate dehydrogenase (G6PDH) and nicotinamide adenine dinucleotide phosphate hydrogen (NADPH) were drawn using hyperchem drawing tools and minimizing the energy of all pdb files with the help of hyperchem by $MM^+$ followed by a semi-empirical (PM3) method. The docking process was studied with ligand molecules to identify suitable dockings at protein binding sites through annealing and genetic simulation algorithms. The program auto dock tools (ADT) was released as an extension suite to the python molecular viewer used to prepare proteins and ligands. Grids centered on active sites were obtained with spacings of $54{\times}55{\times}56$, and a grid spacing of 0.503 was calculated. Comparisons of Global and Local Search Methods in Drug Docking were adopted to determine parameters; a maximum number of 250,000 energy evaluations, a maximum number of generations of 27,000, and mutation and crossover rates of 0.02 and 0.8 were used. The number of docking runs was set to 10. Results: Lawsone and THN can be considered to efficiently bind with NOS, CAT, GSH, GR, G6PDH and NADPH, which has been confirmed through hydrogen bond affinity with the respective amino acids. Conclusion: Naphthoquinone derivatives of lawsone, which can be metabolized into THN by a catalyst DTD, were examined. Lawsone and THN were found to be identically potent molecules for their affinities for selected proteins.
The gold-silver mineralizations in Korea are closely related to Jurassic Daebo igneous activity (121 and 183 Ma) and Cretaceous Bulgugsa igneous activity (60 and 110 Ma). A compilation and re-evaluation of chemical data in arsenopyrite suggest that the As contents vary, reflecting different genetic environments or mineral assemblages. The gold-silver vein deposits from various mineralized area were investigated using arsenopyrite geothermometer. Arsenopyrites from the Jurassic Au-dominant deposits are distinct by high As contents (29.68~33.46 atomic %) with narrow variations, equivalent to a temperature range of $370{\sim}450^{\circ}C$ and a sulfur fugacity of about $10^{18}-10^{-6}$ atm. On the contrary, arsenopyrites from the Cretaceous Au-Ag and Ag-dominant deposits show a wider range in atomic % As composition of 27.47-32.74. They may have formed at temperatures of $250{\sim}350^{\circ}C$ and about $f_{S_2}=10^{-12}-10^{-10}$ atm. The data of arsenopyrite geothermometer, electrum-sphalerite geothermometer, fluid inclusions, vein morphology and emplacement depth of igneous rocks indicate that the gold mineralizations of Group IIA occurred at temperatures between 300 and $500^{\circ}C$ at depth of several tens km or more (about 4-5 kbar), and the gold-silver deposits of Groups III, IV and V were formed at a temperature range of about $170{\sim}370^{\circ}C$ under the shallow environment (<1 kbar).

World is experiencing abnormal weather caused by urbanization and industrialization increasing greenhouse gas and one of these phenomenon domestically happening is flood and drought. The increase of green-house gases is due to urbanization and industrialization acceleration which are causing abnormal climate changes such as the El Nino and a La Nina phenomenon. It is expected that there will be many difficulties in water management, especially considering the topography and seasonal circumstances in Korea. Unlike in the past, a variety of water conservation initiatives have been undertaken like the river-management flow and water capacity expansion projects. To meet the increasing demand for water resources, new environmentally-friendly small and medium-sized dams have been built. Therefore, the development of a new paradigm for water resources management is essential. This study shows that additional security is needed for potential water resources through diversion tunnels and is very important to consider for future water supplies and situations. Using RCP 6.0 and RCP 8.5 in representative concentration pathway climate change scenario, specific hydrologic data of study basin was produced to analyze past observed basin rainfall tendency which showed both scenario 5%~9% range increase in rainfall. Through sensitivity analysis using objective function, population in highest goodness was 1000 and cross rate was 80%. In conclusion, it is expected that the results from this study will help to make long-term and stable water supply plans by using the potential water resource evaluation model which was applied in this study.

Resistance of pepper (Capsicum spp.) to anthracnose (Colletotrichum acutatum) was evaluated during regeneration of Capsicum spp. in National Agrobiodiversity Center. Disease severity of 896 pepper accessions (430 accessions of C. annuum, 219 accessions of C. baccatum, 14 accessions of C. chacoense, 153 accessions of C. chinense, 70 accessions of C. frutescens, 2 accessions of C. pubescens, and unidentified 8 accessions) was investigated at 14 days after inoculation in $28^{\circ}C$ humid chamber. Forty nine accessions of pepper germplasm were resistant to C. acutatum. Among them, nine accessions were highly resistant to C. acutatum without wounding spray inoculation. Four accessions belonged to the species C. baccatum, one accession to C. chacoense, and four accessions to C. frutescens. Forty two resistant candidate accessions were inoculated with pin-prick wounding using a syringe needle. Five accessions were resistant as a less than 3% of disease severity to C. acutatum with wounding inoculation 5 days after inoculation. All resistant accessions were C. baccatum. These five pepper germplasm might be used as breeding resources for the anthracnose resistance breeding program.
Although pneumococcus is one of the most frequently encountered opportunistic pathogen in the world, the mechanisms responsible for its infectiveness have not yet been fully understood. In this paper, we have attempted to characterize the effects of pneumococcal transformation on the pathogenesis of the organism. We constructed three transformation-deficient pneumococcal strains, which were designated as Nos. 1d, 2d, and 22d. The construction of these altered strains was achieved via the insertion of the inactivated gene, comE, to strains 1, 2 and 22. We then conducted a comparison between the virulence of the transformation-deficient strains and that of the wild-type strains, via an evaluation of the ability of each strain to adhere to endothelial cells, and also assessed psaA mRNA expression, and the survival of hosts after bacterial challenge. Compared to what was observed with the wild-type strains, our results indicated that the ability of all of the transformation-deficient strains to adhere to the ECV304 cells had been significantly reduced (p < 0.05), the expression of psaA mRNA was reduced significantly (p < 0.05) in strains 2d and 22d, and the median survival time of mice infected with strains Id and 2d was increased significantly after intraperitoneal bacterial challenge (p < 0.05). The results of our study also clearly indicated that transformation exerts significant effects on the virulence characteristics of S. pneumoniae, although the degree to which this effect is noted appears to depend primarily on the genetic background of the bacteria.

Background: The emerging mobile colistin resistance gene, mcr-1, is an ongoing worldwide concern and an evaluation of clinical isolates harboring this gene is required in Korea. We investigated mcr-1-possessing Enterobacteriaceae among Enterobacteriaceae strains isolated in Korea, and compared the genetic details of the plasmids with those in Escherichia coli isolates from livestock. Methods: Among 9,396 Enterobacteriaceae clinical isolates collected between 2010 and 2015, 1,347 (14.3%) strains were resistant to colistin and those were screened for mcr-1 by PCR. Colistin minimum inhibitory concentrations (MICs) were determined by microdilution, and conjugal transfer of the mcr-1-harboring plasmids was assessed by direct mating. Whole genomes of three mcr-1-positive Enterobacteriaceae clinical isolates and 11 livestock-origin mcr-1-positive E. coli isolates were sequenced. Results: Two E. coli and one Enterobacter aerogenes clinical isolates carried carried IncI2 plasmids harboring mcr-1, which conferred colistin resistance (E. coli MIC, 4 mg/L; E. aerogenes MIC, 32 mg/L). The strains possessed the complete conjugal machinery except for E. aerogenes harboring a truncated prepilin peptidase. The E. coli plasmid transferred more efficiently to E. coli than to Klebsiella pneumoniae or Enterobacter cloacae recipients. Among the three bacterial hosts, the colistin MIC was the highest for E. coli owing to the higher mcr-1-plasmid copy number and mcr-1 expression levels. Ten mcr-1-positive chicken-origin E. coli strains also possessed mcr-1-harboring IncI2 plasmids closely related to that in the clinical E. aerogenes isolate, and the remaining one porcine-origin E. coli possessed an mcr-1-harboring IncX4 plasmid. Conclusions: mcr-1-harboring IncI2 plasmids were identified in clinical Enterobacteriaceae isolates. These plasmids were closely associated with those in chicken-origin E. coli strains in Korea, supporting the concept of mcr-1 dissemination between humans and livestock.
The objective of this study was to investigate the effects of environments (farms born, testing groups, age at the tests, date at slaughter, ages at slaughter) on body weights at 6, 12, 18 and 24 months of ages, body type measurements at 18 months of age and carcass characteristics in Hanwoo steer populations that were collected from commercial farms and reared in a progeny testing station. Performances of a total of 1,838 steer calves set for tests from 2004 to 2008 were recorded. Carcass characteristics were the carcass grading results evaluated and data collected slaughter scores at 24 months of age. For growth traits of all age classes and body type traits measured at 18 months of age, farms born, test group and linear covariate of age at test were fit in the models. For carcass traits, date at slaughter and linear covariate of ages at slaughter were fit in the models. Effect of farm at birth was not significant for body weight at 24 months of age. Carcass weight, eye muscle area, yield score and back fat thickness were affected by dates at slaughter but not by the ages at slaughter. Marbling score, however, was affected by these two effects. Farms at birth did not seem to affect body type measures greatly. This study will be utilized for Hanwoo Steers genetic evaluation.
