Evaluation of the taxonomic rank of the terrestrial orchid Cephalanthera subaphylla based on allozymes

  • The taxonomic rank of the tiny-leaved terrestrial orchid Cephalanthera subaphylla Miyabe & $Kud{\hat{o}}$ has been somewhat controversial, as it has been treated as a species or as an infraspecific taxon, under C. erecta (Thunb.) Blume [C. erecta var. subaphylla (Miyabe & $Kud{\hat{o}}$) Ohwi and C. erecta f. subaphylla (Miyabe & $Kud{\hat{o}}$) M. Hiro]. Allozyme markers, traditionally employed for delimiting species boundaries, are used here to gain information for determining the taxonomic status of C. subaphylla. To do this, we sampled three populations of five taxa (a total of 15 populations) of Cephalanthera native to the Korean Peninsula [C. erecta, C. falcata (Thunb.) Blume, C. longibracteata Blume, C. longifolia (L.) Fritsch, and C. subaphylla]. Among 20 putative loci resolved, three were monomorphic (Dia-2, Pgi-1, and Tpi-1) across the five species. Apart from C. longibracteata, there was no allozyme variation within the remaining four species. Of the 51 alleles harbored by these 17 polymorphic loci, each of the 27 alleles at 14 loci was unique to a single species. Accordingly, we found low average values of Nei's genetic identities (I) between ten species pairs (from I = 0.250 for C. erecta versus C. longifolia to I = 0.603 for C. falcata vs. C. longibracteata), with C. subaphylla being genetically clearly differentiated from the other species (from I = 0.349 for C. subaphylla vs. C. longifolia to 0.400 for C. subaphylla vs. C. falcata). These results clearly indicate that C. subaphylla is not genetically related to any of the other taxa of Cephalanthera that are native to the Korean Peninsula, including C. erecta. In a principal coordinate analysis (PCoA), C. subaphylla was positioned distant not only from C. falcata, C. longibracteata, and C. longifolia, but also from C. erecta. Finally, K = 5 was the best clustering scheme using a Bayesian approach, with five clusters precisely corresponding to the five taxa. Thus, our allozyme results strongly suggest that C. subaphylla merits the rank of species.

  • The buffer is a key component of an engineered barrier system that safeguards the disposal of high-level radioactive waste. Buffers are located between disposal canisters and host rock, and they can restrain the release of radionuclides and protect canisters from the inflow of ground water. Since considerable heat is released from a disposal canister to the surrounding buffer, the thermal conductivity of the buffer is a very important parameter in the entire disposal safety. For this reason, a lot of research has been conducted on thermal conductivity prediction models that consider various factors. In this study, the thermal conductivity of a buffer is estimated using the machine learning methods of: linear regression, decision tree, support vector machine (SVM), ensemble, Gaussian process regression (GPR), neural network, deep belief network, and genetic programming. In the results, the machine learning methods such as ensemble, genetic programming, SVM with cubic parameter, and GPR showed better performance compared with the regression model, with the ensemble with XGBoost and Gaussian process regression models showing best performance.

Phylogenetic Analysis on Wild Cordyceps Collected from Miryang Region of South Korea (밀양근교에서 채집한 야생 동충하초 계통의 PCR 산물에 근거한 계통 유전학적 연구)

  • The phylogenetic relationships among thirty-two strains (P1~P32; including Cordyceps sp., Paecilomyces sp., Beauveria sp., Aranthomyces sp., Isaria sp. and Himenostilbe sp.) in Miryang region located in the southern part of Korea, were investigated based on internal transcribed spacer (ITS) sequences of ribosomal DNA. A fragment of ITS region was amplified by polymerase chain reaction (PCR) using the specific primer pairs ITS1 and ITS4. After obtained same size of PCR products from various strains, we cloned them into a pGEM-T easy vector to determine their sequences. BLAST analyses of the nucleotide sequence ITS1, 5.8S and ITS2 gene fragments revealed the identity and their phylogenetic relationship. Among 32 strains isolated from Miryang region, Cordyceps militaris was shared 100% sequences with Genbank (AY49191, EU825999, AY491992), while some species are not shared perfectly with reported sequences. For example, strain P17 (P. tenuipes in Ulju-gun Gaji Mountain) has some differences among the other strains of P. tenuipes (Miryang-si Jocheon-eup, Miryang-si Gaji Mountain) and those of gene bank. We conclude that ITS analyses with strains in the suburbs of Miryang in this study can be effectively used as a tool for classification, evaluation and collection of the natural eco-type genetic resources.

Evaluation of Regional Adaptability in Introduced Super Sweet Corn Hybrids and Heritability of Agronomic Traits (도입 초당옥수수 교잡종의 지역 적응성 및 농업 형질의 유전력 평가)

  • This study evaluated newly introduced, commercial super sweet corn hybrids (Zea mays L.) for their potential as breeding materials. Agronomic traits were measured and variance components were obtained using a linear mixed model to estimate the heritability. The trials were carried out in 2018 at two locations (Haenam and Oksan in South Korea). All traits had low heritability, except for mid tasseling and silking days. These traits with low heritability mostly had low genetic variance component estimate. In case of ear height ratio, significant genotype by location appeared to be responsible for low genetic variance, which in turn led to low heritability. Low heritability estimates from the trials with commercial hybrids were perhaps because those hybrids were highly improved for commercial success. Hence, this does not necessarily point to them having poor potential as breeding materials. To overcome low heritability, significant genotype by environment interaction, and achieve high selection efficiency, intermating among hybrids is recommended to create new recombinants before inbred line development.

Improved Method for Learning Context-Free Grammar using Tabular representation

  • In this paper, we suggest the method to improve the existing method leaning context-free grammar(CFG) using tabular representation(TBL) as a chromosome of genetic algorithm in grammatical inference and show the more efficient experimental result. We have two improvements. The first is to improve the formula to reflect the learning evaluation of positive and negative examples at the same time for the fitness function. The second is to classify partitions corresponding to TBLs generated from positive learning examples according to the size of the learning string, proceed with the evolution process by class, and adjust the composition ratio according to the success rate to apply the learning method linked to survival in the next generation. These improvements provide better efficiency than the existing method by solving the complexity and difficulty in the crossover and generalization steps between several individuals according to the size of the learning examples. We experiment with the languages proposed in the existing method, and the results show a rather fast generation rate that takes fewer generations to complete learning with the same success rate than the existing method. In the future, this method can be tried for extended CYK, and furthermore, it suggests the possibility of being applied to more complex parsing tables.

Evaluation of the Diversity of Cyclodextrin-Producing Paenibacillus graminis Strains Isolated from Roots and Rhizospheres of Different Plants by Molecular Methods

  • To address the diversity of cyclodextrin-producing P. graminis strains isolated from wheat roots and rhizospheres of maize and sorghum sown in Australia, Brazil, and France, restriction fragment length polymorphism analysis of part of genes encoding RNA polymerase (rpoB-RFLP) and DNA gyrase subunit B (gyrB-RFLP) was used to produce genetic fingerprints. A phylogenetic tree based on rpoB gene sequences was also constructed. The isolates originated from Brazil could be separated from those from Australia and France, when data from the rpoB-based phylogenetic tree or gyrB-RFLP were considered. These analyses also allowed the separation of all P. graminis strains studied here into four clusters; one group formed by the strains GJK201 and $RSA19^T$, second group formed by the strains MC22.02 and MC04.21, third group formed by the strains TOD61, TOD 221, TOD302, and TOD111, and forth group formed by all strains isolated from plants sown in Cerrado soil, Brazil. As this last group was formed by strains isolated from sorghum and maize sown in the same soil (Cerrado) in Brazil, our results suggest that the diversity of these P. graminis strains is more affected by the soil type than the plant from where they have been isolated.

Equivalent Circuit Modeling of Aperture-Coupled Microstrip-to-Vertically Mounted Slotline Coupler (개구면을 통한 마이크로스트립-수직 슬롯 라인 결합 구조의 회로망 해석과 모델링)

  • A general analysis of a microstrip-to-vertically mounted slotline(VMS) coupler is presented with a view to developing an equivalent circuit, and the efficient evaluation of the related circuit element values. Based on this theory, the effects of frequency and structure parameters such as aperture length and VMS width on the characteristics of the coupler are studied. In order to check the validity of the proposed analysis and design theory, a C-band linearly tapered slot antenna fed by an aperture-coupled back-to-back microstripline-to- VMS coupling structure is optimally designed using a hybrid genetic algorithm. Moreover, the computed characteristics from the network analysis is compared to the measurement and simulation results. The obtained results fully validate the efficiency and accuracy of the proposed network model.

Selection of Sahiwal Cattle Bulls on Pedigree and Progeny

  • The objective of the study was to compare ranking of Sahiwal bulls selected on the basis of highest lactation milk yield of their dams with their estimated breeding values (EBVs) using an animal model. Data on 23,761 lactation milk yield records of 5,936 cows from five main Livestock Experiment Stations in Punjab province of Pakistan (1964-2004) were used for the study. At present the young A.I bulls are required to be from A-category bull-dams. Dams were categorized as A, B, C and D if they had highest lactation milk yield of ${\geq}$2,700, 2,250-2,699, 1,800-2,249 and <1,800 litres, respectively. The EBVs for lactation milk yield were estimated for all the animals using an individual animal model having fixed effect of herd-year and season of calving and random effect of animal. Fixed effect of parity and random effect of permanent environment were incorporated when multiple lactation were used. There were 396 young bulls used for semen collection and A.I during 1973-2004. However, progeny with lactation yields recorded, were available only for 91 bulls and dams could be traced for only 63 bulls. Overall lactation milk yield averaged 1,440.8 kg. Milk yield was 10% heritable with repeatability of 39%. Ranking bulls on highest lactation milk yield of their dams, the in-vogue criteria of selecting bulls, had a rank correlation of 0.167 (p<0.190) with ranking based on EBVs from animal model analysis. Bulls' EBVs for all lactations had rank correlation of 0.716 (p<0.001) with EBVs based on first lactation milk yield and 0.766 (p<0.001) with average EBVs of dam and sire (pedigree index). Ranking of bulls on highest lactation yield of their dams has no association with their ranking based on animal model evaluation. Young Sahiwal bulls should be selected on the basis of pedigree index instead of highest lactation yield of dams. This can help improve the genetic potential of the breed accruing to conservation and development efforts.

Evaluation of Effects of Metformin in Primary Ovarian Cancer Cells

  • Background: Ovarian cancer is the third most common cause of cancer in Indian women. Despite an initial 70-80% response rate, most patients relapse within 1-2 years and develop chemoresistance. Hence, identification or repositioning of drugs to resensitise ovarian cancer cells to existing chemotherapy is needed. Traditionally immortalized cell lines have been used in research, but these may contain genetic aberrations and chromosomal abnormalities serving as poor indicators of normal cell phenotype and progression of early-stage disease. The use of primary cells, maintained for only short periods of time in vitro, may serve as the best representative for studying in vivo conditions of the tissues from which they are derived. In this study we have attempted to evaluate the effect of metformin (an antidiabetic drug) in primary ovarian cancer cells because of its promising effect in other solid tumours. Materials and Methods: Primary cultures of epithelial ovarian cancer cells established from ascitic fluid of untreated ovarian cancer patients were used. The cells were treated with metformin at doses standardized by MTT assay and its ability to induce apoptosis was studied. The cells were analysed for apoptosis and apoptosis related proteins by flow cytometry and western blotting respectively. Results: Metformin induced apoptosis in ovarian cancer cells, provoking cell cycle arrest in the G0/G1 and S phase. It induced apoptosis in ovarian cancer cells by, down-regulating Bcl-2 and up-regulating Bax expression. Conclusions: Metformin was able to induce apoptosis in primary ovarian cancer cells by modulating the expression of Bcl-2 family proteins. These data are relevant to ongoing translational research efforts exploring the chemotherapeutic potential of metformin.

Regression Equations for Estimating the TANK Model Parameters (TANK 모형 매개변수 추정을 위한 회귀식 개발)

  • The TANK model has been widely used in rainfall-runoff modeling due to its simplicity of concept and computation while achieving forecast accuracy. A major barrier to the model application is to determine parameter values for ungauged watersheds, leading to the need of a method for the parameter estimation. The objective of this study was to develop regression equations for estimating the 3th TANK model parameters considering their variations for the ungauged watersheds. Thirty watersheds of dam sites and stream stations were selected for this study. A genetic algorithm was used to optimize TANK model parameters. Watershed characteristics used in this study include land use percent, watershed area, watershed length, and watershed average slope. Generalized equations were derived by correlating to the optimized parameters and the watershed characteristics. The results showed that the TANK model, with the parameters determined by the developed regression equations, performed reasonably with 0.60 to 0.85 of Nash-Sutcliffe efficiency for daily runoff. The developed regression equations for the TANK model can be applied for the runoff simulation particularly for the ungauged watersheds, which is common for upstream of agricultural reservoirs in Korea.
