Geographic Variation of Granulilittorina exigua (Littorinidae, Gastropoda) in Korea Based on the Mitochondrial Cytochrome b Gene Sequence

  • Partial sequence of the mitochondrial cytochrome b gene was analyzed to investigate genetic variation from 10 geographic populations of Granulilittorina exigua in Korea. The sequence of 282 base pairs was determined by PCR-directed silver sequencing method. The sequences of two species within the genus Littorina reserved in NIH blast search were utilized to determine geographic variations of species referred. The levels of mtDNA sequence differences were 0.00-2.54% within populations and 0.71-4.43% between populations. There were four amino acid differences between representative species of the genera Granulilittorina and Littorina, but no differences within populations of the genus Granulilittorina. The UPGMA and the N-J trees based on Tamura-Nei genetic distance matrix were constructed, which showed that the genus Granulilittorina was divided into three groups such as eastern (even exception for Tokdo population), southern, and western regional populations. The degrees of genetic divergence within populations of each group were p=0.021, p=0.019, and p=0.018, respectively. The divergence between the eastern and southern populations was p=0.032, showing closer relationship than with the western populations (p=0.052). Based on the diverged time estimation, the eastern and southern populations of Granulilittorina exigua in Korea diverged from the western populations about 2.1 MYBP, and the eastern and southern populations diverged from each other about 1.3 MYBP.

Genetic Variability and Association of Yield Attributing Characters with Grain Yield in Deepwater Rice

  • A study on genetic variability and association of yield attributing characters with grain yield was carried out using 35 deepwater rice genotypes. High genotypic co-efficient of variation (GCV) was observed for plot yield, $EBT/m^2$, plant height and days to $50\%$ flowering (DFF). For all the traits, estimates of the phenotypic co-efficient of variation (PCV) were higher than GCV, indicating presence of environmental influence. High heritability and genetic advance was observed for plot yield, $EBT/m^2$ and plant height. Plot yield had significant positive association with test weight, $EBT/m^2$ and DFF. However, test weight had the maximum direct effect on grain yield

Determination of Phylogenetic Relationships of Turkish Native Cattle Breeds with Other Cattle Breeds Using Mitochondrial DNA D-loop Sequence Polymorphism

  • The aim of this study was to determine the specific polymorphic sites in cattle breeds and inter- and interbreed genetic variation among breeds and to develop a databank of Turkish native cattle mtDNA using sequence analysis. The entire D-loop region was analyzed based on DNA sequences in Turkish Grey, East Anatolian Red, South Anatolian Red, and Anatolian Black native breeds. In total, 68 nucleotide differences were observed at 26 different sites. The variable positions consisted of 22 transitions, two transversions, and two insertions, but no deletions. Haplotype number, haplotype diversity, nucleotide diversity, and mean number of pairwise difference values were found to be 17, 0.993, 0.00478, and 4.275, respectively. In addition, a phylogeny was developed by comparison among cattle populations for which the entire D-loop sequence was available. A high level of genetic variation was observed within and among the native cattle breeds.

Genetic approaches toward understanding the individual variation in cardiac structure, function and responses to exercise training

  • Cardiovascular disease (CVD) accounts for approximately 30% of all deaths worldwide and its prevalence is constantly increasing despite advancements in medical treatments. Cardiac remodeling and dysfunction are independent risk factors for CVD. Recent studies have demonstrated that cardiac structure and function are genetically influenced, suggesting that understanding the genetic basis for cardiac structure and function could provide new insights into developing novel therapeutic targets for CVD. Regular exercise has long been considered a robust nontherapeutic method of treating or preventing CVD. However, recent studies also indicate that there is inter-individual variation in response to exercise. Nevertheless, the genetic basis for cardiac structure and function as well as their responses to exercise training have yet to be fully elucidated. Therefore, this review summarizes accumulated evidence supporting the genetic contribution to these traits, including findings from population-based studies and unbiased large genomic-scale studies in humans.

Genetic Polymorphism among Korean Salmonids Determined by RAPD (Randomly Amplified Polymorphic DNA) Analysis

  • RAPD analyses using 60 OPERON primers and 13 URPs were performed in order to assess the genetic variation and frequency of polymorphisms in Korean salmonids. RAPDS were very reproducible and most useful at the sub-species level. In RAPD analysis, 138 polymorphic bands were detected between Oncorhynchus masou subspecies and 99 bands were generated in two types of rainbow trout. Estimated genetic distances between O. masou subspecies were 0.28794, and between wild rainbow trout and an albino mutant was 0.22786. Each species of salmonid was well characterized using URP 4R, the obtained bands could be useful as a species specific RAPD markers.

I-SSR 표지자에 의한 눈측백나무 남한 잔존집단의 유전변이와 구조 (Genetic Variation and Structure of the Relict Populations of Korean Arborvitae (Thuja koraiensis Nakai) in South Korea, Employing I-SSR Markers)

  • 본 연구에서는 눈측백나무(Thuja koraiensis Nakai) 4개 천연집단을 대상으로 84개체를 선발한 뒤 다형성을 보인 29개의 I-SSR amplicons를 이용하여 유전변이를 조사하였다. 6개의 I-SSR primer로 유전다양성을 추정한 결과 평균 유효대립유전자의 수($A_e$)는 1.44개, 이형접합도의 기대치($H_e$)는 0.258, Shannon의 다양성 지수(S.I.)는 0.385로 크게 높지는 않은 것으로 추정되었다. 이를 다양한 표지자를 이용하여 측백나무과(Cupressaceae)에 속하는 지금까지 연구된 수종들과 비교하여보면 국내의 눈측백나무는 유사하거나 다소 높은 유전변이량을 보유하고 있는 것으로 나타났다. 조사된 4개의 집단을 대상으로 AMOVA를 수행한 결과 전체 유전변이 가운데 13%가 집단간 차이로부터 기인하는 것으로 나타났고, 나머지 87%는 집단내 개체간 차이로부터 기인한 것으로 나타났다. 유전적 거리에 의한 UPGMA 유집분석을 실시한 결과 지리적인 경향은 나타나지 않았다. 유전변이량이 가장 높은 장산집단과 유전적 조성에서 가장 이질적인 방태산집단이 보전 가치가 큰 것으로 판단된다.

Estimation of Effective Population Size in a Clonal Seed Orchard of Chamaecyparis obtusa

  • Clonal differences in fertility (expressed as the number of female and male strobili) were determined for five consecutive years (2002-2006) in a clonal seed orchard of Chamaecyparis obtusa. Fertility varied among clones and among years with producing five-year averages of 378.8 and 871.2 for female and male strobili per ramet, respectively. Correlation between female and male strobilus production was positive over the five years and statistically significant. Based on the observed fertility variation, the effective population sizes (estimated by status numbers, $N_e$) were calculated and varied from 24.3 to 47.9 (48.6% to 95.8% of census number, N) among the five studied years. On average (pooled), the relative effective population size was 82% of the N. Variation in female fertility was higher than that in male fertility, and this variation was reflected on female and male parents' status numbers. Pooled $N_e$ estimated from the five years was higher than that from poor seed production years. From our results, it was concluded that genetic diversity collected from good flowering years would be higher than that from poor flowering years.

在來種마늘의 量的形質에 대한 遺傳變異와 相關 (Genitic Variability and Correlation of Quantitative Characters in Local Garlic Cultivars)

  • This study was conducted to investigate morphological characteristics of Korean local garlic varieties and to define the genetic variability and the correlations amongthe characters associated with yield. The plant height, the leaf sheath diameter, the nubmer of leaves, and bulb weight were significantly increased in the garlic plants from heavier seed bulbs. The proportation of six clove bulbs was more than 60% in Seosan variety and this was not realted with the number of cloves in seed bulb. While more than 70% bulbs in Jeoksung and Danyang varieties cultivated in paddy field were composed of 6 or 7 cloves, majority of the bulbs of Euisung and Danyang varieties cultivated in upland were composed of bulbs with 7 cloves or more. The highest genotypic coefficient of variation (GCV) and phenotypic coefficient of variation (PCV) were observed in bulb weight. GCV and PCV also showed high value in leaf sheath diameter. Thus, there is a greater scope for selection of these characters. The small difference between PCV and GCV values in plant height indicates that the environmental influence would be limited in this character. High heritability was observed of plant height (56.76%) and bulb weight (45.95%). And the weight per bulb (34.24%) exhibited highest genitic advance followed by leaf sheath diameter (18.8%) and plant height (9.61%), and those would be the ideal characters for selelction.

Genetic Analysis of SCN5A in Korean Patients Associated with Atrioventricular Conduction Block

  • Recent several studies have shown that the genetic variation of SCN5A is related with atrioventricular conduction block (AVB); no study has yet been published in Koreans. Therefore, to determine the AVB-associated genetic variation in Korean patients, we investigated the genetic variation of SCN5A in Korean patients with AVB and compared with normal control subjects. We enrolled 113 patients with AVB and 80 normal controls with no cardiac symptoms. DNA was isolated from the peripheral blood, and all exons (exon 2-exon 28) except the untranslated region and exon-intron boundaries of the SCN5A gene were amplified by multiplex PCR and directly sequenced using an ABI PRISM 3100 Genetic Analyzer. When a variation was discovered in genomic DNA from AVB patients, we confirmed whether the same variation existed in the control genomic DNA. In the present study, a total of 7 genetic variations were detected in 113 AVB patients. Of the 7 variations, 5 (G87A-A29A, intervening sequence 9-3C>A, A1673G-H558R, G3578A-R1193Q, and T5457C-D1819D) have been reported in previous studies, and 2 (C48G-F16L and G3048A-T1016T) were novel variations that have not been reported. The 2 newly discovered variations were not found in the 80 normal controls. In addition, G298S, G514C, P1008S, G1406R, and D1595N, identified in other ethnic populations, were not detected in this study. We found 2 novel genetic variations in the SCN5A gene in Korean patients with AVB. However, further functional study might be needed.

RAPD를 이용한 한국 김 집단의 유전적 다양성과 표현형 관계 (Studying the Genetic Diversity and Phenetic Relationships of Porphyra yezoensis Populations in Korea Using Random Amplified Polymorphic DNA (RAPD))

  • 김(Porphyra yezoensis)은 김속의 홍조류이다. RAPD (random amplified polymorphic DNA) 마커를 이용하여 한국 내 네 집단의 표현형과 유전적 다양성을 조사하였다. 전체적으로 20 시발체로 김에서 55분절이 관찰되었다. 이들 밴드 중 30개(54.5%)는 다형성을 나타내었다. OPA-18-02 밴드는 낙동 김 집단에서만 증폭되었다. OPA-20-02 밴드는 서천 김 집단에서만 증폭되었다. 이 두 밴드는 특별한 집단을 구별해주는 특이밴드로 판정되었다. 대립유전자좌위의 수(Ae)는 1.161에서 1.293로 평균은 1.366였다. 서천 김 집단이 가장 높은 다형성을 나타내었다(0.163). 다른 집단과 격리되고 조간대에 위치한 낙동 김 집단은 가장 낮은 다형성을 나타내었다(0.092). 샤논의 표현형 다양성(I)은 서천 김 집단이 가장 높았다(0.238). 전체 유전적 다양도($H_T$)는 0.132(OPA-02)에서 0.420(OPA-19)로 나타났다. 대립유전자좌위에서 유전적 다양성($H_S$)은 0.059(OPA-18)에서 0.339(OPA-19)였다. 대립유전자좌위에 근거에서 전체 유전적 다양도에서 집단 간 차이($G_{ST}$)는 0.012(OPA-11)에서 0.762(OPA-18)이였으며 평균은 0.415였다. 이는 전체 변이의 약 42%는 집단 간에서 발견된다는 것을 의미한다. 종 내 다양도의 58.5%는 집단 내에 있었다. 유전자 흐름(Nm)은 0.705로 낮았다.
