Population genetic assessment is essential for the conservation and management of endangered and rare plants. Neofinetia falcata is endangered epiphyte orchid and protected by law in Korea. In Korea, this species is only found on islands in the South Sea of Korea (including Jeju-do) and the southern coast of the Korean Peninsula. We developed nine microsatellite makers to assess the genetic diversity and population genetic structure of three populations of N. falcata. The genetic diversity at the species level was low, which can be attributed to inbreeding or fragmentation into small, isolated populations. A recent bottleneck was detected in one population, likely due to overcollection. N. falcata exhibited moderated levels of differentiation among populations, with the three populations were divided into two clusters based on genetic structure. The genetic diversity and structure of N. falcata are affected by restricted gene flow by pollen or seeds due to isolation and geographic distance. Strategies for in situ and ex situ conservation of this species are been proposed based on the results of our study.
The most difficult and time-intensive issue in the successful implementation of genetic algorithms is to find good parameter setting, one of the most popular subjects of current research in genetic algorithms. In this study, we present a new efficient experimental design method for parameter optimization in a genetic algorithm for general job shop scheduling problem using the Taguchi method. Four genetic parameters including the population size, the crossover rate, the mutation rate, and the stopping condition are treated as design factors. For the performance characteristic, makespan is adopted. The number of jobs, the number of operations required to be processed in each job, and the number of machines are considered as noise factors in generating various job shop environments. A robust design experiment with inner and outer orthogonal arrays is conducted by computer simulation, and the optimal parameter setting is presented which consists of a combination of the level of each design factor. The validity of the optimal parameter setting is investigated by comparing its SN ratios with those obtained by an experiment with full factorial designs.
Eleven Italian ryegrass cultivars were examined for their genetic polymorphisms and phylogenetic relationships using randomly amplified polymorphic DNA (RAPD) markers. In RAPD analysis of 34 random primers, 96 of total 162 bands obtained from 16 primers were polymorphic and sizes of polymorphic band ranged between 0.5 and 1.5kb. Number of bands amplified per primer was varied from 3 to 16 and average number was 14.8. Phylogenetic relationship among cultivars based on the RAPD analysis was examined using UPGMA computer program. In pairwise genetic similarity test of 11 Italian ryegrass cultivars, Grazer and Orlando showed highest coefficient of genetic similarity as 0.740, whereas Marshall and Orlando was lowest as 0.438. Eleven Italian ryegrass cultivars were grouped into 3 major clusters and genetic distance of clusters ranged between 0.567 and 0.646, indicating low level of genetic variation.
Six field bean (GI-vcine soza S and Z ) plants were examined for their genetic polymorphisms and intraspecific variations using randomly amplified polymorphic DNA(RAPD) markers. In RAPD analysis of 5 random primers (Rp-1, Rp2, Rp-3, Rp-4, Rp-5), 30 of total 155 bands obtained kom 5 primers were polymorphic and sizes of polymirphic band ranged between 0.5 and 3.0 kb. Number of bands amplyfied per primer was varied from 2 to 11 and average number was 6.0. Genetic variation of intraspecies in the samples of six region was ranged behveen 11 to 25 percent, and genetic similarity among intraspecies was ranged from 0.69 to 0.78. In pairwise genetic similarity test of six field bean plants, Mun and Hoj showed highest coefficient of genetic similarity as 0.67, whereas Sin and Hoj was lowest as 0.45. According to the genetic similarity, the level of intraspecific variation is higher than that of regional distance in GI-vcine soza.
The objective of the present study was to analyze genetic variation and characteristics in rainbow trout(Oncorhynchus mykiss) using amplified fragment length polymorphism(AFLP) method as molecular genetic technique, to evaluate the usefulness of AFLP as genetic markers, and to compared the efficiency of agarose and polyacrylamide sequencing gels. The amplified products were performed by agarose and sequencing gel electrophoresis to detect AFLP band patterns, respectively. Using 9 primer combinations, total of 141 AFLP bands were produced, 108 bands(82.4%) of which were polymorphic in agarose gels. In sequencing gels, total of 288 bands were generated, and 220 bands (76.4%) were polymorphic. The level of bandsharing(BS) ranged from 0.18 to 0.32 for the 9 primer combinations tested, with a mean of 0.24. Consequently, AFLP markers of these rainbow trout could be used as genetic information such as species identification, genetic relationship or analysis of genome structure, and selection aids for genetic improvement of economically importment traits in fish species.
희귀수종인 모감주나무(Koelreuteria paniculata Laxm.) 6개 자생 집단에 대해서 잎의 형태적 특성 및 식생개황을 조사하였으며, 동위효소 분석에 의한 유전변이를 조사하였다. Nested design에 의한 분산분석 결과 8가지 잎의 형태적 특성 모두에서 집단간 및 집단내 개체간에 고도의 통계적 유의성이 인정되었으며, 대부분 형질의 경우 총 분산 가운데 집단간 차지하는 비율이 집단내 개체가 차지하는 비율보다 큰 것으로 나타났다. 식생분석 결과 각 집단별로 출현한 수종 수는 많지 않았으며, 대부분 집단이 인위적 교란을 받고 있는 것으로 나타났다. 다른 수종, 특히 지리적으로 광범위하게 분포하는 수종들과 비교할 때 유전변이는 매우 적고(A/L= 1.1, P=9.5%, $H_o=0.021$, $H_e=0.035$) 집단간 분화($F_{ST}=0.114$)는 다소 큰 것으로 나타났는데, 유전적 부동 및 근친교배의 영향을 받고 있는 것으로 추정되었다. 마지막으로 모감주나무 집단의 유전적 다양성을 보전하기 위한 방법에 대해서 논의하였다.
Background: Development of effective educational strategies should accompany increases in public awareness and the availability of genetic testing for breast cancer (BC). These educational strategies should be designed to fulfill the knowledge gap while considering factors that influence women's interest in order to facilitate decision making. Objective: To determine the possible correlates of Saudi women's interest in BC genes testing including socio-demographics, the level of awareness towards BC genes, the family history of BC and the perceived personal risk among adult Saudi women in Al Hassa, Saudi Arabia. Subjects and methods: This cross-sectional study was carried out during the second BC community-based campaign in Al Hassa, Saudi Arabia. All Saudi women aged ${\geq}18$ years (n=781) attending the educational components of the campaign were invited to a personal interview. Data collection included gathering information about sociodemographics, family history of BC, the perceived personal risk for BC, awareness and attitude towards BC genes and the women's interest in BC genes testing. Results: Of the included women (n=599), 19.5% perceived higher risk for BC development, significantly more among < 40 years of age, and with positive family history of BC before 50 years of age. The participants demonstrated a poor level of awareness regarding the inheritance, risk, and availability of BC genetic testing. The median summated knowledge score was 1.0 (out of 7 points) with a knowledge deficit of 87.8%. The level of knowledge showed significant decline with age (> 40 years). Of the included women 54.7% expressed an interest in BC genetic testing for assessing their BC risk. Multivariate regression model showed that being middle aged (Odds Ratio 'OR'=1.88, confidence intervals 'C.I'=1.14-3.11), with higher knowledge level (OR=1.67, C.I=1.08-2.57) and perceiving higher risk for BC (OR=2.11, C.I=1.61-2.76) were the significant positive correlates for Saudi women interest in BC genetic testing. Conclusion: Saudi women express high interest in genetic testing for BC risk despite their poor awareness. This great interest may reflect the presence of inappropriate information regarding BC genetic testing and its role in risk analysis.
Comprehensive information on genetic diversity and introgression is desirable for the design of rational breed improvement and conservation programs. Despite the concerns regarding the genetic introgression of Western pig breeds into the gene pool of the Korean native pig (KNP), the level of this admixture has not yet been quantified. In the present study, we genotyped 93 animals, representing four Western pig breeds and KNP, using the porcine SNP 60K BeadChip to assess their genetic diversity and to estimate the level of admixture among the breeds. Expected heterozygosity was the lowest in Berkshire (0.31) and highest in Landrace (0.42). Population differentiation ($F_{ST}$) estimates were significantly different (p<0.000), accounting for 27% of the variability among the breeds. The evidence of inbreeding observed in KNP (0.029) and Yorkshire (0.031) may result in deficient heterozygosity. Principal components one (PC1) and two (PC2) explained approximately 35.06% and 25.20% of the variation, respectively, and placed KNP somewhat proximal to the Western pig breeds (Berkshire and Landrace). When K = 2, KNP shared a substantial proportion of ancestry with Western breeds. Similarly, when K = 3, over 86% of the KNP individuals were in the same cluster with Berkshire and Landrace. The linkage disquilbrium (LD) values at $r^2_{0.3}$, the physical distance at which LD decays below a threshold of 0.3, ranged from 72.40 kb in Landrace to 85.86 kb in Yorkshire. Based on our structure analysis, a substantial level of admixture between Western and Korean native pig breeds was observed.
한국 내 국부적으로 분포하는 지리사초(Carex okamotoi)의 14집단에 대해 유전적 다양성과 집단구조를 실시하였다. 이 식물은 세 산맥(태백산맥, 소백산맥 노령산맥)의 고지대(700~1,500 m)에 제한적으로 자생한다. 다형성을 나타내는 대립유전자좌위는 40.0%였다. 종 수준과 집단 수준에서 유전적 다양성은 낮았으며($H_{ES}$=0.106; $H_{EP}$=0.094), 집단간 분화도 낮았다($G_{ST}$=0.082). 14집단에 대한 임의 교배로부터 산출한 편차는 0.238이였다. 간접적으로 산출한 세대간 이주하는 개체들의 수는 2.78이였다(Nm=2.78). 고정지수 분석에서 이형접합체의 실질적 결핍이 일부 집단과 대립유전자좌위에서 나타났다. 집단간 유전적 동질성은 평균 0.986이였다.
The relationship between second messenger cGMP and human cytomegalovirus (HCMV) replication was investigated. First, the intracellular level of cGMP ([cGMP]i) in HCMV-infected cells was measured. The [cGMP]i increased at early times after HCMV infection, reached maximum level at 12 hr and returned to basal level at 24 hr after virus infection, while [cGMP]i in mock-infected cells remained relatively unchanged. Increasing [cGMP]i resulted in enhanced transcription of HCMV major immediate early gene. For early gene expression, cGMP had varying effect. Expression of 1.2 kb RNA decreased and 2.2 kb RNA increased with increasing cGMP, while 2.7 kb RNA gene expression was not affected. HCMV early genes are regulated by immediate early gene, and the effect of cGMP on the regulatory effect of major immediate early gene on early genes was investigated. In the absence of cGMP, major immediate early gene repressed 2.7 kb RNA gene expression, while 1.2 kb RNA and 2.2 kb RNA early genes were not significantly affected. In the presence of $1\;{\mu}M$ cGMP, however, major immediate early gene stimulated the expression of three early genes.
