  • Although it is not easy to generate EC's (Engineering Characteristics), there have been few studies so far. In this paper, a systematic method for generating EC candidates is proposed. This method use the concept of 'attribute listing'. In the proposed method, customer attributes are divided into their objects and issues. And attributes from their objects and issues are generated by help of guidelines proposed, respectively. Then, measurable metrics are attained from those attributes. Finally, the metrics result in EC candidates by consistency and redundancy check. This paper gives an illustrative example for easy comprehension of the proposed method.

경상남도 천연기념물 노거수의 생육환경 연구 (Growth Conditions of Natural Monument Old Big Trees in Gyeongsangnamdo, Korea)

  • Old big tree transcends the simple meaning of trees as they are the natural monuments that embody the people's history and culture of this land. The Cultural Heritage Administration of Korea(CHA) defines and protects old big tree based not only on the size of the tree but also on its definitive cultural and natural factors such as value, implications, and originality. This research aims to identify and analyze the growth conditions, soil conditions and location character of 20 old big tree in Gyeongsangnamdo korea. The research examined the soundness of the arboreal form, the degree of damage on the bark, as well as the quantity of leafs levels to evaluate the overall condition of growth and development. Also, 9 elements such as soil texture, nitrogen and organic matter content, soil pH, phosphoric acid and EC were further analyzed The research analyzed in correlation of Growth condition and soil. Tree health related positivity that total nitrogen and organic matter. The result which analyzes location character, With natural monument old big trees raising a hand the area where is contiguous appeared with the fact that the farming village style where the rice field and the arable land of field etc. This research aimed at generating some foundational reference data for the analysis of the habitation and management conditions of natural monument old big tree within the Gyeongsangnamdo korea.

The ABA Effect on the Accumulation of an Invertase Inhibitor Transcript that Is Driven by the CAMV35S Promoter in ARABIDOPSIS

  • Invertase (${\beta}$-D-fructofuranosidase; EC 3.2.1.26) catalyzes the conversion of sucrose into glucose and fructose and is involved in an array of important processes, including phloem unloading, carbon partitioning, the response to pathogens, and the control of cell differentiation and development. Its importance may have caused the invertases to evolve into a multigene family whose members are regulated by a variety of different mechanisms, such as pH, sucrose levels, and inhibitor proteins. Although putative invertase inhibitors in the Arabidopsis genome are easy to locate, few studies have been conducted to elucidate their individual functions in vivo in plant growth and development because of their high redundancy. In this study we assessed the functional role of the putative invertase inhibitors in Arabidopsis by generating transgenic plants harboring a putative invertase inhibitor gene under the control of the CaMV35S promoter. A transgenic plant that expressed high levels of the putative invertase inhibitor transcript when grown under normal conditions was chosen for the current study. To our surprise, the stability of the invertase inhibitor transcripts was shown to be down-regulated by the phytohormone ABA (abscisic acid). It is well established that ABA enhances invertase activity in vivo but the underlying mechanisms are still poorly understood. Our results thus suggest that one way ABA regulates invertase activity is by down-regulating its inhibitor.

MPEG-21 방송 디지털 아이템을 위한 생성 및 파싱 시스템에 대한 연구 (A Study on Generation and Parsing System for MPEG-21 Broadcasting Digital Item)

  • 오늘날 인터넷의 보급과 다양한 멀티미디어기술의 개발로 방송 환경은 방송통신 융합환경으로 변화하고 있다. 이러한 새로운 환경에서 서로 다른 기술들간의 상호 호환성과 컨텐츠의 투명한 전달 등은 중요한 문제가 되고있다. 이런 문제를 해결하기 위해 IS0/1EC MPEG 미팅에서 MPEG-21이 설립되었다. MPEG-21은 멀티미디어 컨텐츠의 전달과 소비를 위한 기반구조를 세우는 큰 그림에 대해 활발히 논의되고 있다. 본 논문에서는 MPEG-21에서 디지털 아이템을 방송용 멀티미디어 컨텐츠에 적용하는 연구를 수행하였다. 방송 디지털 아이템은 디지털 아이템 선언과 디지털 아이템 식별 기술뿐만 아니라 멀티미디어 컨텐츠 서술을 위해 국제 표준인 MPEG-7 MDS를 사용하여 발생된다. 또한 방송 디지털 아이템을 생성 및 파싱 시스템을 제시한다. 실험에서 다양한 사용자 환경에서 교육용 컨텐츠를 생성하고 소비에 의해 방송 디지털 아이템의 유용성을 확인하였다.

The Gene Encoding γ-Glutamyl Transpeptidase II in the Fission Yeast Is Regulated by Oxidative and Metabolic Stress

  • $\gamma$-Glutamyl transpeptidase (GGT, EC 2.3.2.2.) catalyzes the transfer of the $\gamma$-glutamyl moiety from $\gamma$-glutamyl containing ompounds, notably glutathione (GSH), to acceptor amino acids and peptides. A second gene (GGTII) encoding GGT was previously isolated and characterized from the fission yeast Schizosaccharomyces pombe. In the present work, the GGTII-lacZ fusion gene was constructed and used to study the transcriptional regulation of the S. pombe GGTII gene. The synthesis of $\beta$-galactosidase from the GGTII-lacZ fusion gene was significantly enhanced by NO-generating SNP and hydrogen peroxide in the wild type yeast cells. The GGTII mRNA level was increased in the wild-type S. pombe cells treated with SNP. However, the induction by SNP was abolished in the Pap1-negative S. pombe cells, implying that the induction by SNP of GGTII is mediated by Pap1. Fermentable carbon sources, such as glucose (at low concentrations), lactose and sucrose, as a sole carbon source, enhanced the synthesis of $\beta$-galactosidase from the GGTII-lacZ fusion gene in wild type KP1 cells but not in Pap1-negative cells. Glycerol, a non-fermentable carbon source, was also able to induce the synthesis of $\beta$-galactosidase from the fusion gene, but other non-fermentable carbon sources such as acetate and ethanol were not. Transcriptional induction of the GGTII gene by fermentable carbon sources was also confirmed by increased GGTII mRNA levels in the yeast cells grown with them. Nitrogen starvation was also able to induce the synthesis of $\beta$-galactosidase from the GGTII-lacZ fusion gene in a Pap1-dependent manner. On the basis of the results, it is concluded that the S. pombe GGTII gene is regulated by oxidative and metabolic stress.

Binding Mode Analysis of Bacillus subtilis Obg with Ribosomal Protein L13 through Computational Docking Study

  • Introduction: GTPases known as translation factor play a vital role as ribosomal subunit assembly chaperone. The bacterial Obg proteins ($Spo{\underline{0B}}$-associated ${\underline{G}}TP$-binding protein) belong to the subfamily of P-loop GTPase proteins and now it is considered as one of the new target for antibacterial drug. The majority of bacterial Obgs have been commonly found to be associated with ribosome, implying that these proteins may play a fundamental role in ribosome assembly or maturation. In addition, one of the experimental evidences suggested that Bacillus subtilis Obg (BsObg) protein binds to the L13 ribosomal protein (BsL13) which is known to be one of the early assembly proteins of the 50S ribosomal subunit in Escherichia coli. In order to investigate binding mode between the BsObg and the BsL13, protein-protein docking simulation was carried out after generating 3D structure of the BsL13 structure using homology modeling method. Materials and Methods: Homology model structure of BsL13 was generated using the EcL13 crystal structure as a template. Protein-protein docking of BsObg protein with ribosomal protein BsL13 was performed by DOT, a macro-molecular docking software, in order to predict a reasonable binding mode. The solvated energy minimization calculation of the docked conformation was carried out to refine the structure. Results and Discussion: The possible binding conformation of BsL13 along with activated Obg fold in BsObg was predicted by computational docking study. The final structure is obtained from the solvated energy minimization. From the analysis, three important H-bond interactions between the Obg fold and the L13 were detected: Obg:Tyr27-L13:Glu32, Obg:Asn76-L13:Glu139, and Obg:Ala136-L13:Glu142. The interaction between the BsObg and BsL13 structures were also analyzed by electrostatic potential calculations to examine the interface surfaces. From the results, the key residues for hydrogen bonding and hydrophobic interaction between the two proteins were predicted. Conclusion and Prospects: In this study, we have focused on the binding mode of the BsObg protein with the ribosomal BsL13 protein. The interaction between the activated Obg and target protein was investigated with protein-protein docking calculations. The binding pattern can be further used as a base for structure-based drug design to find a novel antibacterial drug.
