Molecular and cultivation techniques were used to characterize the bacterial communities of biobead reactor biofilms in a sewage treatment plant to which an Aerated Up-Flow Biobead process was applied. With this biobead process, the monthly average values of various chemical parameters in the effluent were generally kept under the regulation limits of the effluent quality of the sewage treatment plant during the operation period. Most probable number (MPN) analysis revealed that the population of denitrifying bacteria was abundant in the biobead #1 reactor, denitrifying and nitrifying bacteria coexisted in the biobead #2 reactor, and nitrifying bacteria prevailed over denitrifying bacteria in the biobead #3 reactor. The results of the MPN test suggested that the biobead #2 reactor was a transition zone leading to acclimated nitrifying biofilms in the biobead #3 reactor. Phylogenetic analysis of 16S rDNA sequences cloned from biofilms showed that the biobead #1 reactor, which received a high organic loading rate, had much diverse microorganisms, whereas the biobead #2 and #3 reactors were dominated by the members of Proteobacteria. DGGE analysis with the ammonia monooxygenase (amoA) gene supported the observation from the MPN test that the biofilms of September were fully developed and specialized for nitrification in the biobead reactor #3. All of the DNA sequences of the amoA DGGE bands were very similar to the sequence of the amoA gene of Nitrosomonas species, the presence of which is typical in the biological aerated filters. The results of this study showed that organic and inorganic nutrients were efficiently removed by both denitrifying microbial populations in the anaerobic tank and heterotrophic and nitrifying bacterial biofilms well-formed in the three functional biobead reactors in the Aerated Up-Flow Biobead process.
Variation at 25 allozyme loci in Korean wateralder (Alnus hirsuta Rupr.) from nine distinct populations was measured to estimate the amount and pattern of genetic diversity and population structure. The mean genetic diversity within population was 0.166. Korean alder populations have slightly high levels of genetic diversity compared to those present in associated temperature-zone species and two Canadian alder species. Among population s genetic differentiation accounted for an significant 9% of the total variation. High gene flow(Nm=2.63) was observed. Analysis of fixation indices, calculated for all polymorphic loci in each population, showed a substantial deficiency of heterozygotes relative to Hardy-Weinberg expectations. The mean GST value A. hirsuta in Korea (GST = 0.087) is similar to those of A. rogosa in Canada (GST = 0.052). These low values of GST in two countries. reflecting little spatial genetic differentiation, may indicate extensive gene flow (via pollen and/or seeds) and/or recent colonization. These factors reduce the effect of geographic isolation of breeding and the chance for genetic divergence. A pattern of increasing is observed with increasing rainfall per year. Regression analysis indicates that 54% (F = 4.67) of the variability observed can be explained by this relationship.
The effect of breeding length of sire on genetic progress was examined in the Holstein dairy cattle population in Japan. Genetic progress was extimated by gene flow method. Breeding length of sires directly influences the replacement rates of sires and the selection intensity of sires because there are a fixed number of progeny tested young bulls per year. As breeding length of sires increased, rate of gene flow decreased and average proportions of genes deriving from selected animals had lower asymptotic values. When breeding length was short, average proportions of genes required a longer period to converge to asymptotic values. Changes of Rcow-sire's(sire to breed recorded cows) and Ncow-sire's(sire to breed non recorded cows) breeding length influenced not only transmission of their genes but also that of genes derived from all other selected animals. Irrespective of whether the discount rate was assumed to be 0 or 6%, longer term (${\geq}$ 20 years) expected total genetic improvement was maximized by a sire breeding length of five years. For shorter term assessment(10 years), genetic improvement was maximized by a sire breeding length of three years. There was a linear increase in the contribution of the sire to bulls pathway to the total genetic improvement, with increase in the term of assessment.
Plant-derived triterpenoids commonly possesses biological properties such as anti-inflammatory, antimicrobial, anti-viral and anti-cancer. Luvunga scandens is one of the plant that produced triterpenoids. The aims of the study was to analyze cell cycle profile and to determine the expression of p53 unregulated modulator of apoptosis (PUMA), caspase-8 and caspase-9 genes at mRNA level in MCF-7 cell line treated with two triterpenoids, flindissol (1) and 3-oxotirucalla-7,24-dien-21-oic-acid (2) isolated from L. scandens. The compounds were tested for cell cycle analysis using flow cytometer and mRNA expression level using quantitative RT-PCR. The number of MCF-7 cells population which distributed in Sub G1 phase after treated with compound 1 and 2 were 7.7 and 9.3% respectively. The evaluation of the expression of genes showed that both compounds exhibited high level of expression of PUMA, caspase-8 and caspase-9 as normalized to ${\beta}-actin$ via activation of those genes. In summary, the isolated compounds of L. scandens plant showed promising anticancer properties in MCF-7 cell lines.
Introgression from genetically modified plants (GMPs) may be dependent on the genetic similarity to wild relative plants. In Korea, many wild plant species are botanically related to the cultivated plants that have a potential to be genetically transformed. The controversy for hazards of GMPs is continuing because the studies on gene flow or introgression are little. Based on the systematic criteria, we have surveyed Korean wild plant species that showed the similarity to cultivating crops. The consideration for feasibility of genetic pollution (introgression of transgene) is necessary for the successful accomplishment in the practical use of GMPs. Although the detrimental effects of GMPs on wild relatives have not been clearly verified, Korean wild plant species related to crop plant (potential GMP) have to be investigated with respect to the introgression. Korean flora consists of ca. 5,500 species. Among them, 1,448 species are classified as weed species (966 native, 325 naturalized, and 167 escaped ones), which is vulnerable to GMPs in term of introgression. We suggested the principal Korean wild plants related to major crops that might be affected by GMPs via introgression. The investigated species herein are selected based on the morphological and phenological relationship. It is necessary to verify the genetic relationship between cultivated plants and wild relatives sing more precise molecular techniques, which provide the information of likelihood for the introgression of transgene.

A whole-cell biocatalyst was constructed by immobilizing an enzyme on the surface of the yeast Saccharomyces cerevisiae. The gene encoding Bacillus macerans cyclodextrin glucanotransferase(CGTase) was fused with the AGA2 gene encoding a small peptide disulfide-linked to the aga1, a cell wall protein of a-agglutinin. The plasmid was introduced S. cerevisiae and expressed in the medium consisting of 10g/L yeast extract, 20g/L peptone, and 20g/L galactose. The activity was detected with the formation of cyclodextrin(CD) from 10g/L soluble starch. Surface display of CGTase was also verified with the halo-test, flow cytometry, and immunofluorescence microscopy. The recombinant S. cerevisiae produced ${\alpha}-cyclodextrin$ more efficiently than the free CGTase by simultaneous fermentation and cyclization as yeast consumes glucose and maltose which are inhibitors for CD synthesis.

Nine microsatellite loci were analyzed in 84 random individuals to characterize the genetic variability of three domestic goat breeds found in Korea and China: Korean goat, Chinese goat and Saanen. Allele diversity, heterozygosity, polymorphism information content, F-statistics, indirect estimates of gene flow (Nm) and Nei's standard distances were calculated. Based on the expected mean heterozygosity, the lowest genetic diversity was exhibited in Korean goat ($H_E$=0.381), and the highest in Chinese goat ($H_E$=0.669). After corrections for multiple significance tests, deviations from Hardy-Weinberg equilibrium were statistically significant over all populations and loci, reflecting the deficiencies of heterozygotes (global $F_{IS}$=0.053). Based on pairwise FST and Nm between different breeds, there was a great genetic differentiation between Korean goat and the other two breeds, indicating that these breeds have been genetically subdivided. Similarly, individual clustering based on the proportion of shared alleles showed that Korean goat individuals formed a single cluster separated from the other two goat breeds.

In agriculture, the rice is the one of important things. Many farmers and scientists have long tried to increase the yield of rice. So many technologies have been developed these days. One type of technology has given rise to a host of concerns and questions, namely Genetically Modified Organisms (GMOs). The increasing cultivation of GM crops has raised a wide range of concerns with respect to food safety, environmental effects and socio-economic issues and now commercially planted on about 100 million hectares in some 22 developed and developing countries. The scientific evidence concerning the environmental and health impacts of GMOs is still emerging, but so far there is no conclusive information on the definitive negative impacts of GMOs on health or the environment. Nevertheless, public perceptions about GMOs in food and agriculture are divided with a tendency toward avoiding GM food and products in many developed and developing countries. Also Korea is one of that country and is not allow the GMOs now. So I studied whether these GMOs are actually dangerous for environment and whether there are differences in cultivar characteristics such as germination test with TTC tetrazolium, germination test in frozen soil and gene-flow test with glufosinate and strip-bar test. With these experiments, we evaluated the agricultural safety of GM rice and to identify and assess environmental risks.
We investigated the genetic structure of Korean and Japanese ayu Plecoglossus altivelis populations by examining 669 individuals from 14 populations using three microsatellite loci. Genetic variation did not differ significantly among the populations examined in terms of allelic number and heterozygosity. Korean populations were genetically close to each other, implying that persistent gene flow has occurred in these populations. This suggests that eastern populations in Korea form a single large population and all of the Korean populations are distinct from the Japanese populations. Pairwise population $F_{ST}$ estimates, principal component analyses, and a neighbor-joining tree showed that genetic separation between the southern and pooled eastern coast populations was probably influenced by restricted gene flow. Hierarchical analysis of molecular variance (AMOVA) revealed a weak but significant genetic structure among three ayu groups (eastern and southern coasts of Korea and the Japan coast), and no genetic variation within groups. The estimated genetic population structure and potential applications of microsatellite markers may aid in the proper management of ayu populations.
