Raw material of gelidioid red algae yielding high-quality agar has been in short supply due to overharvesting, but in situ farming of gelidioids has not been practical due to their slow growth. To produce vegetative seedstock of a cosmopolitan species, Pterocladiella capillacea, we investigated the number and length of regenerated branches arising from sectioned fragments during 3 weeks of laboratory culture at 10, 15, 20, and 25℃. All sectioned fragments formed axis-like branches mostly from the upper cut edge and stolon-like branches mostly from the lower cut edge, showing a high capacity of regeneration and intrinsic bipolarity. At 20℃, the number of regenerated branches increased to 2.74 ± 1.29 on the upper cut edge and 4.26 ± 2.66 on the lower cut edge. Our study reveals that the use of fragments bearing regenerated branches as seedstock can be a simple method to initiate fast propagation for mass cultivation in the sea or outdoor tank.
Porphyroglossum is the last one of nine genera within the family Gelidiaceae that has yet to be analyzed by molecular markers. We analyzed rbcL and cox1 genes from P. zollingeri specimens collected near the type locality in Indonesia and compared them with other gelidioid algae. Thalli are cartilaginous, complanate, and up to 15 cm high. Abundant rhizoidal filaments are concentrated in the medullary layer. Tetrasporangial sori are on small, determinate ramuli. In all gene analyses, P. zollingeri consistently nested within Gelidium. The sister relationship of P. zollingeri to G. floridanum was well resolved. Because Gelidium has priority over Porphyroglossum, a new combination is proposed, viz. Gelidium zollingeri. Network analysis of the four cox1 haplotypes revealed many missing haplotypes, indicating high genetic diversity in the species.
Two species of the agar-yielding genus Gelidium, G. galapagense and G. isabelae, have previously been reported from Korea but their occurrence has not been confirmed with molecular data. We intensively collected samples of Gelidium from Jeju Island, where the two species were reported, and the southern coast of Korea. Phylogenetic analyses based on cox1 and rbcL sequences revealed that only a single species occurred in Korea. The Korean species was distantly related to G. galapagense and G. isabelae from the Galápagos Islands, and formed a clade with G. microdonticum, G. millarianum, and G. pakistanicum. A new species, G. palmatum, is described for those specimens that were previously recognized as either G. galapagense or G. isabelae from Korea. G. palmatum is small in size (up to 0.7 cm), with compressed, lanceolate axes, irregular, digitate to palmate branches, abundant rhizines in the medulla, tetrasporangial sori without sterile margins, and rounded bilocular cystocarps borne subapically on palmate branchlets.

Recently seaweed polysaccharides have been extensively studied for alternative energy application. Because their producing cost is high and efficiency low, their industrial applications have been limited. The main component of cell wall of red algae represented by Gelidiales and Gracilariales is agar. Red-algae agar or galactan, consisting of D-galactose and 3, 6-anhydro-L-galactose, is suitable for bio-product application if hydrolyzed to monomer unit. For the hydrolysis of algae, chemical or enzymatic treatment can be used. A chemical process using a strong acid is simple and efficient, but it generates together with target sugar and toxic compounds. In an enzymatic hydrolysis process, target sugar without toxic compounds generation. The objective of this review is to summary the recent data of saccharification by chemical and enzymatic means from red seaweed for especially focused on automobile industry.
The taxonomic distinctiveness and cosmopolitan distributions of the red algae $Gelidium$$crinale$ and $G.$$pusillum$ remain unclear. Both species were first described in Devon in southwestern England; namely in Ilfracome for $G.$$crinale$ and Sidmouth for $G.$$pusillum$. We analyzed mitochondrial $cox$1 and plastid $rbc$L sequences from specimens collected in East Asia, Australia, Europe and North America. In all phylogenetic analyses of $cox$1 and $rbc$L sequences, $G.$$crinale$ was distinct from congeners of the genus. The analyses also revealed a sister relationship with the $G.$$coulteri$ and $G.$$capense$ clade. Nineteen $cox$1 haplotypes were identified for $G.$$crinale$, and they were likely geographically structured. Despite the distinctiveness in both $cox$1 and $rbc$L datasets, the sister relationship of $G.$$pusillum$ in the genus was not resolved. Our $cox$1 and $rbc$L datasets indicate that $G.$$crinale$ is a cosmopolitan species, found in East Asia, Australia, Europe and North America, while the distribution of $G.$$pusillum$ is restricted to Europe and Atlantic North America. Our results suggest that infraspecific classification of $G.$$pusillum$ may be abandoned.

The number of species of non-native and invasive marine algae is growing, with concomitant public concern about native ecosystems and coastlines. Gelidium vagum, recently introduced from northeast Asia to Europe and North America, commonly occurs from the intertidal to subtidal zones in Korea, China, and Japan. To investigate the level of genetic diversity of native populations, we analyzed mitochondrial cox1 and cob from 108 specimens of G. vagum from Korea, China, eastern Russia, including from the Netherlands and USA. The haplotype network of individual and cox1 + cob datasets revealed no genetic structure in local populations, suggesting genetic flow between Korean populations. Our results corroborate a typical pattern of genetic diversity for introduced species, with low levels in introduced populations and high levels in native populations. All haplotypes were shared between the Netherlands and USA, but not between Korea and the Netherlands / USA except cox1. Additional sampling will identify donor populations in native northeast Asian waters. This is the first report of the utility of the mitochondrial coding cob sequences in red algae.
Although Pterocladiella is a well-known agar-producing red algal genus, its taxonomy in Korea appears to be confused. In the present study, we demonstrate the occurrence of Pterocladiella nana and P. tenuis, as well as P. capillacea in Korea on the basis of morphological and plastid rbcL sequence comparisons. P. capillacea commonly occurs along the coasts of Korea and has regular pinnate branches with abundant second-order branches, growing up to 10 cm in length. P. nana occurs in the intertidal zone of Jeju and is distinguished by the small size of its thalli (up to 5 cm) and regular branches with up to third-order branches. P. tenuis is distributed mostly subtidally on the southern coast of Jeju and has irregular branches with rarely second-order branches, growing up to 19 cm in length. We determined rbcL sequences from 19 specimens (15 from Korea and four from France) and downloaded 28 sequences from GenBank. Analyses of all 47 rbcL sequences revealed that each of three species was consistently resolved. P. capillacea and P. tenuis always formed a sister clade with P. nana at the base. Given that 12 rbcL haplotypes from 28 specimens of P. capillacea have been found to date, analysis of a fast-evolving gene from across the range of the species should highlight its genetic diversity.
