상압조건에서 1-부탄올 젤 연료액적의 연소특성 (Combustion Characteristics of a 1-Butanol Gel Fuel Droplet in Atmospheric Pressure Condition)

  • Combustion characteristics of a 1-butanol gel fuel were studied in atmospheric pressure condition. The butanol gel fuel was manufactured by adding hydroxypropyl-methyl cellulose (HPMC) as a gellant and the effect of the gellant concentration was observed. The combustion process of a single butanol gel droplet was divided into 3 stages including droplet heating, microexplosion, and gellant combustion. The flame was distorted compared to butanol + water mixture because of micro-explosion during the combustion. Increase of gellant concentration delayed the droplet ignition, but the combustion rate was improved due to the mass ejection during the micro-explosion.

Candida tropicalis 포괄고정 담체를 적용한 Airlift Loop Bioreactor에서의 복합 휘발성유기화합물 제거 (Removal of Volatile Organic Compounds using Candida tropicalis Immobilized on Polymer Gel Media in an Airlift Loop Bioreactor)

  • 본 연구는 휘발성유기화합물질의 분해능력을 가진 yeast인 Candida tropicalis를 이용하여, 대표적인 휘발성 유기화합물질인 톨루엔과 MEK의 제거효율을 향상시키기 위하여 수행되었다. 반응기는 가스상으로 유입되는 톨루엔과 MEK의 물질전달 능력을 향상시키기 위하여 airlift loop 형태로 선택하였고, yeast 미생물의 효과적인 포괄고정화를 위해 분말활성탄(PAC)과 알지네이트(Alginate), PEG로 고분자 담체를 형성하였다. Airlift loop bioreactor의 물질전달성능을 평가하기 위한 실험을 수행하였으며, 기체체류시간 15초에서 담체를 첨가하지 않은 액상의 톨루엔 물질전달계수($K_La$) 값이 1.29 $min^{-1}$이었으나, 고분자 담체를 첨가한 경우 톨루엔의 $K_La$는 4.07 $min^{-1}$로 증가하였다. 따라서 고분자담체를 적용하는 것이 기상으로 유입되는 휘발성유기화합물의 물질전달을 향상시키는 것으로 확인되었다. 이러한 airlift loop bioreactor와 yeast 포괄고정 담체를 적용하여 체류시간을 60초, 30초, 15초에서 유입부하에 변화를 주며 실험을 진행한 결과, 톨루엔 5, 10, 19, 37 g/$m^3$/hr, MEK 4.5, 8.9, 17.8, 35.1 g/$m^3$/hr의 유입부하 변화에도 전체 80% 이상의 안정적인 처리효율을 나타내었다. 또한 airlift loop bioreactor의 분해능을 확인하기 위하여 유입부하를 단기간 변화시켜 주며 실험한 결과, 톨루엔과 MEK의 최대분해능은 각각 70.4 g/$m^3$/hr, 56.4 g/$m^3$/hr로 확인되었다.

Purification and Characterization of Acetolactate Synthase from Barley

  • Acetolactate synthase (ALS) catalyzes the first common step in the biosynthesis of branchedchain amino acids, valine, leucine, and isoleucine. ALS is the target site for several structually diverse classes of herbicides including sulfonylureas, imidazolinones. and triazolopyrimidines. We have purified ALS from etiolated barley shoots to homogeneity. The five major purification steps are ammonium sulfate fractionation, DEAE anion exchange, hydroxylapatite, Bio-Gel A gel filtration, and low pressure Mono-Q chrornatoqraphy. Approximately 170-fold purification was achieved and the yield was 0.45% of initial activity in the crude extract. Both SDS-PAGE and Western blot analysis showed a single polypeptide of ALS with an apparent molecular mass of 64 kDa. The result of nondenaturing gel electrophoresis with activity staining indicated that the molecular mass of its native form is approximately 225 to 250 kDa. The values of $K_m$ for pyruvate. pl. and optimum pH of ALS were determined to be 2.0 mM, 5.2. and 7.0. respectively Feedback inhibition studies showed that ALS is more susceptible to leucine than valine. And $IC_{50}$ value of Cadre, a class of irnidazolinones, is about $1.5\mu{M}$ for ALS.

Capillary Size-exclusion Chromatography as a Gel-free Strategy in Plasma Proteomics

  • Although 2D-PAGE has been widely used as the primary method for protein separation, difficulties in displaying proteins with an extreme values of isoelectric paint (pI), molecular size and hydrophobicity limit the technique. In addition, time consuming steps involving protein transfer and extraction from the gel-pieces can result in sample loss. Here, we describe a novel protein separation technique with capillary size-exclusion chromatography (CSEC) for rapid protein identification from human plasma. The method includes protein fractionation along with molecular size followed by in-solution tryptic digestion and peptide analysis through reversed phase liquid chromatography (RPLC) coupled to nanoflow electrospray-tandem mass spectrometry (ESI-MS/MS). Tryptic peptides are applied an a $100\;{\mu}m\;i.d.{\times}10mm$ length pre-column and then separated on a $75\;{\mu}m{\times}200mm$ analytical column at -100 nL/min flaw rate. Proteins were identified over the wide ranges of pI (3.7-12.3) when this technique was applied to the analysis of $1-2\;{\mu}L$ of human plasma. This gel-free system provides fast fractionation and may be considered a complementary technique to SDS-PAGE in proteomics.

A Second Thioltransferase of Schizosaccharomyces pombe Contains Glutathione S-transferase Activity

  • Two types of the thioltransferase (also called glutaredoxin) have been previously detected in the cytosolic extract of Schizosaccharomyces pombe, a fission yeast. Previously, the one with a smaller molecular mass (14kDa) was purified and characterized. In the present study, the second thioltransferase was purified. The purification procedure included ammonium sulfate fractionation (40-80%), Sephadex G-200 gel filtration, DEAE-cellulose ion-exchange chromatography, Sephadex G-50 gel filtration, and glutathione-agarose affinity chromatography. The purified enzyme showed a single band on SDS-PAGE, and its molecular mass was determined to be 23 kDa. It utilizes various compounds as substrates, including 2-hydroxyethyl disulfide. Interestingly, we found that the purified thioltransferase also contains significant glutathione S-transferase activity.

한국산 초피의 어독성분에 관한 연구 (A Study on the Ichthyotoxic Constituents of Chopi(Zanthoxylum piperitum DC))

  • 한국산 초피나무의 물고기에 대한 독성력을 확인하기 위하여 물질을 용매추출, silica gel column chromatography와 재결정을 통하여 분리 정제하였다. 추출물의 물고기에 대한 독성실험 결과, methanol 추출물에서 가장 강한 독성이 나타났으며, 그 다음으로는 물추출물, 초피의 건조분말 그리고 chloroform추출물 순서로 독성을 보였다. 이 추출물의 구조를 기기분석을 통해 해석한 결과 L-asarinin 임을 확인하였다.

  • 버섯의 cap에 갈변병을 유발하는 독성물질을 P. tolaasii 배양액에서 추출, 정제하였다. 독성물질의 정제는 silica gel chromatography, mass spectrum과 NMR을 사용하였다. 정제된 독성물질은 amylamine group의 aminobenzene인 것으로 밝혀졌으며, UV spectrum으로 분석한 결과 234 nm에서 확인되었고, 융점은 $76^{\circ}C$ 이었다.

  • A fast and matrix-assisted laser desorption/ionization-mass spectrometry compatible protein staining method in one- and two-dimensional sodium dodecyl sulfate-polyacrylamide gel electrophoresis is described. It is based on the counterion dye staining method that employs oppositely charged two dyes, Zincon and Ethyl Violet to form an ion-pair complex. It is safe to use since the methanol used previously in staining solution was replaced with ethanol, which is not toxic. The protocol including fixing, staining and quick washing steps can be completed in 1 to 1.5 h depending upon gel thickness. (omitted)

Mass Transport Properties and Influence of Natural Convection for Voltammetry at the Agarose Hydrogel Interface

  • Agarose hydrogel, a solid electrolyte, was investigated voltammetrically in terms of transport properties and natural convection effects using a ferrocenyl compound as a redox probe. To confirm the diffusion properties of solute on the agarose interface, the diffusion coefficients (D) of ferrocenemethanol in agarose hydrogel were determined by cyclic voltammetry (CV) according to the concentration of agarose hydrogel. While the value of D on the agarose interface is smaller than that in the bulk solution, the square root of the scan rate-dependent peak current reveals that the mass transport behavior of the solute on the agarose surface shows negligible convection or migration effects. In order to confirm the reduced natural convection on the gel interface, scan rate-dependent CV was performed in the solution phase and on the agarose surface, respectively. Slow scan voltammetry at the gel interface can determine a conventional and reproducible diffusion-controlled current down to a scan rate of 0.3 mV/s without any complicated equipment.

Isolation of Antimicrobial Substance from the Korean Traditional Leaf Mustard, Brassica juncea Coss.

  • The antimicrobial effect of each fraction after fractionation of an ethanol extract of leaf mustard was examined in terms of nucleic acid, chloroform, ethylacetate, and butanol. The ethylacetate fraction, which showed the strongest level of antimicrobial effect among the different ethanol extract fractions of leaf mustard, was isolated and purified using silica gel column chromatography and HPLC, respectively, to obtain a single antimicrobial substance called KLM-1. The antimicrobial effect of this substance was 10 times higher than that of the ethylacetate fraction. A further study is on the way to confirm the structure of the antimicrobial substance KLM-1 through LC/Mass and NMR.
