Comparison of IgE induction in mice by pollens from three pine tree species

  • Background: Over the years, pine pollens have been excluded as an allergen due to its relatively large size, low protein content, and waxy hydrophobic layer, despite their abundance. However, recent studies suggest the possibilities of pine pollens being allergens, and it has been reported that allergy symptoms were highly prevalent in areas with considerably large pine forests and high possibility of exposure to the pollen. Therefore, we conducted a comparative analysis of the allergenicities of the pollens from the dominant species of Korean pines, red pine (Pinus densiflora), black pine (Pinus thunbergii), and pitch pine (Pinus rigida), in mice. Methods: The protein composition of the pollens from the three pine species was compared via sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). The pine pollens and proteins extracted from the pollens were introduced to BALB/c mice by nasal inhalation and application to exposed skin and the IgE produced by the mice were extracted from blood and analyzed via ELISA. Results: SDS-PAGE showed differing protein compositions of the pollens of the three pine species. Analysis of blood IgE compositions showed a similar amount of IgE produced when pollens were applied to skin. In contrast, when mice inhaled the pollens, P. densiflora was shown to induce significantly more IgE production than those of the other two species. Conclusions: The experimental results demonstrate that the pollens of all three South Korean pine species induce IgE production, and this production was more pronounced when the pollens were inhaled than when they were applied to the skin. Of the three species, the pollen of P. densiflora was found to induce the highest level of IgE production.

  • A conductive oxide-based sapphire glass indium tin oxide/metal electrode and the optical coating, through patterning process was studied in excellent optical properties and integrated touch panel has a high strength. Indium tin oxide conductive oxides of the sapphire glass to 0.3 A at DC magnetron sputtering method of 10 min, gas flow Ar 10 Sccm Ar, $O_2$ 1.0 Sccm the formation conditions of the thin film after annealing at $550^{\circ}C$ for 30min was achieved through a 86% transmittance. In addition, the coating 130 nm hollow silica sol-gel was to improve the optical transmittance of the indium tin oxide to 91%. For the measurement by the modeling hollow silica sol by Macleod simulation and calculated the average values of silica part to the presence or absence in analogy to actual. Refractive index value and the actual value of the material on the simulation the transmittance difference is it does not completely match the air region similar to the actual value (transmission) could be confirmed that the measurement is set to a value of between 5 nm and 10 nm.

Anti-Metastatic Activity of Glycoprotein Fractionated from Acanthopanax senticosus, Involvement of NK-cell and Macrophage Activation

  • Previously, we reported that water-extracted Acanthopanax senticasus exhibited anti-meta-static activity by stimulating the immune system. In this study, we fractionated glycoproteins (EN-SP) from the soluble protein layer (GF-AS) of A. senticasus and determined their basic chemical properties. We also investigated the anti-tumor and immunostimulating activities of the fractionated glycoprotein, EN-SP. We found that intravenous (i.v.) administration of GF-AS dramatically inhibited metastasis of colon26-M3.1 carcinoma cells to the lung in a dose-dependent manner. In vitro analysis showed GF-AS to enhance the proliferation of splenocytes. GF-AS also stimulated peritoneal macrophage, which was followed by the production of various cytokines such as IL-1$\beta$, TNF-$\alpha$, IL-12 and IFN-${\gamma}$. Furthermore, the production of these cytokines was partially blocked when peritoneal macrophage was cultured with the polyclonal antibodies against GF-AS. The depletion of NK cells by rabbit anti-asialo GM1 serum partly abolished the inhibitory effect of GF-AS on lung metastasis of colon26-M3.1 cells. Using gel filtration, EN-SP, an active glycoprotein fraction, is isolated from GF-AS. While both GF-AS and EN-SP stimulated the proliferatation of splenocytes of normal mice, EN-SP showed higher anti-metastatic activity and more potently stimulated the proliferation of splenocytes compared to GF-AS. These results suggest the use of EN-SP, the fractionated glycoprotein from A. senticasus, can be used as a therapeutical reagent to prevent or inhibit tumor metastasis.

미세패턴 전사기법을 위한 다양한 몰드 제작법 소개 (The Review for Various Mold Fabrication toward Economical Imprint Lithography)

  • NIL, S-FIL과 같은 각인 기술(Imprinting lithography)를 적용하기 위한 투명하고 단단한 복제 틀(replica hard mold)을 제작하여 고가의 원판(master)와 패턴이 형성되는 기판과의 접촉을 근본적으로 방지해 경제적인 공정이 가능함을 제안한다. 실리콘 웨이퍼(Si wafer)와 같은 원판(master)과 패턴 형성 시 사용되는 기판과 직접적인 접촉을 방지하기 위해 우선 액상 공정을 이용하여 비접착성 표면처리된 고분자 복제(polymer copy)를 매개체(carrier)로 단단한 복제 틀을 제작한다. 이렇게 제작된 단단한 복제 틀(replica hard mold)는 유리와 거의 같은 강도와 투명도를 나타내며, 각인 공정(imprinting process)에서 석영 틀, 실리콘 웨이퍼(quartz mold, Si wafer)과 같이 값비싼 원판(master)의 직접 사용을 대체하여 성공적으로 패턴을 구현할 수 있다.

현장시험을 통한 알칼리성 실리카졸 주입재의 적용성 및 주입효과에 대한 연구 (A Study on the Application and Grouting Effects of Akali-Silicasol Grout Material from Field Test)

  • 본 연구는 최근 개발된 알칼리성 실리카졸 그라우팅재의 적용성 및 주입효과를 규명한 것으로서 연구에 사용된 그라우트재는 3차원겔 형성재인 알칼리성 실리카졸 그라우팅재와 규산나트륨 그라우팅재에 대한 공학적 특성을 비교하기 위하여 실내시험을 실시하여 강도특성 및 환경영향성을 검토하였으며, 현장시험 등을 통해 차수성과 주입효과를 검토하여 현장적용성을 규명하고자 하였다. 실내시험결과 알칼리성 실리카졸의 일축압축강도는 규산나트륨에 비해 약 5배 정도 증가하는 현상이 나타났고 용탈시험결과 용탈되는 원소의 총량도 극히 작아 친환경적인 재료임이 입증되었다. 현장시험결과 동수층에서도 상당한 투수계수 저감효과를 확인하였고, 시방기준인 $10^{-5}cm/sec$보다 낮은 투수계수를 확보하였다. 주입확인시험결과 페놀프탈레인 반응시험, 전기비저항탐사에서 주입효과가 양호한 것으로 나타났다.

Zn 농도변화에 따른 ZnO 박막의 구조, 광학 및 전기적 특성 연구 (Structural, Optical and Electrical Properties of ZnO Thin Films with Zn Concentration)

  • 저온 박막 공정을 위해 비등점이 낮은 용매인 isopropanol을 사용하였고, 용질로 zinc acetate의 몰 농도를 0.3∼1.3 mol/l까지 변화시켜 sol을 합성하였다. Zn 농도 변화에 따른 ZnO 박막의 구조 및 광학, 전기적 특성을 분석하였다. XRD 측정에서 Zn의 농도가 0.7 mol/l 일 때 c-축으로 결정 배향성이 뚜렷하였다. SEM으로 박막의 표면 morphology를 관찰한 결과 0.7 mol/l 에서 균일한 표면층을 갖는 나노구조를 이루고 있었다. UV-vis. 측정을 통한 ZnO 박막의 광투과도는 Zn의 농도가 0.7 mol/l 이하에서 87%였으나, 1.0 mol/l 이상의 농도에서는 급격히 감소하였다. 이때 광 밴드갭 에너지는 3.07∼3.22 eV의 값을 나타내며, 벌크 ZnO의 특성과 유사하였다. 박막의 전기 비저항 값은 150 $\Omega$-cm로 Zn의 농도변화에 따라 큰 변화를 보이지 않았으며, I-V 특성분석에서 전형적인 ohmic contact 특성을 보였다.

Core-shell 구조의 Au/TiO2 나노 미립자의 합성 및 특성 평가 (Synthesis and Characterization of Au/TiO2 Nanoparticles with Core-shell Structure)

  • Au/TiO$_2$ core-shell 구조 나노 미립자가 졸-겔법에 의해서 제조되었고, TiO$_2$ shell의 형상과 결정성이 TEM과 UV-Vis. absorption spectrometer에 의해서 조사되었다. Au/TiO$_2$ core-shell 나노 미립자는 Au 콜로이드 에탄올 수용액 중에서 TOAA(Titanium Oxide Acethylacetonate)의 가수분해에 의해 합성될 수 있었다. Au 나노 미립자의 표면에 형성된 TiO$_2$ shell의 두께는 약 1 nm이었다. TiO$_2$ shell의 결정성을 조사하기 위하여. TiO$_2$가 피복된 Au 콜로이드 에탄올 용액에 254 nm의 자외선과 $^{60}$Co의 방사선을 조사하였다. Au 나노 미립자의 surface plasmon 현상은 방사선이 조사되었을 때만 나타났고, 이 결과로부터 TiO$_2$ shell은 비정질 상태임을 알 수 있었으며, Au의 분산성 향상을 위해 표면에 처리된 MUA(Mercaptoundecanoic Acid)층은 전자의 이동을 방해하는 장애물로 작용하지 않음을 확인할 수 있었다.

구형 SiO2@Y2O3: Eu 코어-쉘 복합체 형광체 합성 및 특성 (Synthesis and Characterization of Spherical SiO2@Y2O3 : Eu Core-Shell Composite Phosphors)

  • The monodisperse spherical $SiO_2$ particles were overcoated with $Y_2O_3:Eu^{3+}$ phosphor layers via a Pechini sol-gel process and the resulting $SiO_2@Y_2O_3:Eu^{3+}$ core-shell phosphors were subsequently annealed at $800^{\circ}C$ at an ambient atmosphere. The crystallographic structure, morphology, and luminescent property of core-shell structured $SiO_2@Y_2O_3:Eu^{3+}$ phosphors were characterized by X-ray diffraction (XRD), field emission scanning electron microscopy (FE-SEM), and photoluminescence (PL). The spherical, nonagglomerated $SiO_2$ particles prepared by a Stober method exhibited a relatively narrow size distribution in the range of 260-300 nm. The thickness of phosphor shell layer in the core-shell particles can be facilely controlled by varying the coating number of $Y_2O_3:Eu^{3+}$ phosphors. The core-shell structured $SiO_2@Y_2O_3:Eu^{3+}$ phosphors showed a strong red emission, which was dominated by the $^5D_0-^7F_2$ transition (610 nm) of $Eu^{3+}$ ion under the ultraviolet excitation (263 nm). The PL emission properties of $SiO_2@Y_2O_3:Eu^{3+}$ phosphors were also compared with pure $Y_2O_3:Eu^{3+}$ nanophosphors.

티탄산바륨 분말과 박막의 제조 및 특성 연구 (Preparation and Characterization of BaTiO3 Powders and Thin films)

  • Ethylene glycol의 polymerization-complex route를 통한 졸-겔 합성법으로 안정하고 균일한 barium titanate 분말 및 박막을 제조하였다. 출발 용액으로 킬레이팅 리간드인 acetylacetone을 barium과 titanium 용액에 치환시켜 합성한 복합 산화물 졸 용액을 사용했을 때 박막을 만들 수 있었다. 졸 용액의 입자 분포도는 안정한 gaussian 분포를 보였으며, $1100^{\circ}C$에서 열처리한 겔 분말의 입자 크기는 40~77 nm이었다. 열분석 및 FT-IR, $^{13}C$ CP/MAS NMR 스펙트라와 XRD 결과로부터 (Ba-Ti)-oxycarbonate 중간상을 거쳐 $BaTiO_3$ 분말이 형성됨을 알 수 있었다. Quartz에 스핀 코팅으로 제조한 박막은 치밀하고 균열 없는 미세 조직을 보였다. $1100^{\circ}C$에서 열처리한 박막 표면의 입자 크기는 220 nm였으며 치밀한 입자 성장을 관찰할 수 있었다.

유류오염의 미생물학적 제어 (Bioremedation of petrolium pollution)

  • As basic study for purpose bioremedation in oil-contaminated environment, Primarily, we isolated biosurfactant producer- strains utilized of oil-agar plate, and measured surface tension and emulsifying activity. We investigated in oil-contaminated soil and sea water. In this laboratory, Pseudomonas sp. EL-012S strain isolated from oil-contaminated soil was able to product novel biosurfactant under the optimal culture condition. Its condition was n-hexadecane 2.0%, NH$_4$NO$_3$0.4%, Na$_2$HPO$_4$0.6%, KH$_2$PO$_4$0.4%, MgSO$_4$.7$H_2O$ 0.02%, CaCl$_2$.2$H_2O$ 0.001%, FeSO.7$H_2O$ 0.001%, initial pH 7.0 and aeration at 3$0^{\circ}C$, respectively. This biosurfactant was produced in both late-exponential and early-stationary phase. The biosurfactant from Pseudomonas sp. EL-012S was composed of carbohydrate, lipid and protein. The purified-biosurfactant was examined two (biosurfactant type I, II) with the silica gel G60 column chromatography and the purified biosurfactant confirmed thin layer chromatography, high performed liquid chromatography and gas chromatography. The biosurfactant type I involved in carbohydrate-lipid-protein characteristics lowered surface tension of water to 27dyne/cm and interfacial tension 4.5dyne/cm aginst to n-hexadecane and the biosurfactant type B involved in carbohydrate lipid characteristics lowered surface tension of water to 30dyne/cm and interfacial tension 8dyne/cm against to n-hexadecane. Specially type I had the properties such as strong emulsifying activity, emulsion stability, pH-stability, thermo-stability, high cleaning activity and forming ability.
