Anti-Ulcer Activity of Newly Synthesized Acylquinoline Derivatives

  • Anti-ulcer activity of newly synthesized acylquinoline derivatives was investigated. For the in vitro screening, the effects of compounds on gastric $H^{+}/K^{+}$ ATPase isolated from hog and rabbit were examined. Among them, AU-090, AU-091, AU-254, AU-413 and AU-466 exhibited good in vitro activity on both enzymes. To correlate the in vitro activity with in vivo action, the effects of the compounds on the basal gastric acid secretion were studied. Some derivatives showed considerable anti-secretory activities, and AU-413 was selected for further studies. AU-413 protected gastric damage induced by either ethanol or NaOH dose dependently when given orally. $ED_{50}$ values of 12 mg/kg, p.o. (ethanol) and 41 mg/kg, p.o. (NaOH) were obtained. In addition, histamine-stimulated gastric secretion was reduced upon AU-413 administration. Taken together, newly synthesized acylquinoline derivatives, especially AU-413, is worthy of further investigation to be developed as an anti-ulcer agent.

위장질환 환자의 양도락에 대한 경락학설과 자율신경이론의 연관성에 대한 연구 (위 운동장애형 소화불량증과 위궤양 환자의 증례를 대상으로) (An Association of Kyung-Rak Principle and Autonomic Nerve Theory related with Ryodoraku of Patients with Gastric Dysmotility and Gastric Ulcer)

  • It has been shown that bilateral decrease of point H4,5,6 in Ryodoraku test is related with gastric dysmotility. This suggested that the system of Kyung-Rak related with anatomical gastric disease may not be the Stomach Meridian, in the view that the system of Kyung-Rak is similar to that of Ryodoraku, and which was not consistent with classical contents of Kyung-Rak principle. Therefore, this paper was done to investigate whether bilateral decrease of point H4,5,6 is a general sign of common gastric disease in Ryodoraku test and to study its relation between Kyung-Rak principle and autonomic nervous system as a mediator explaining Ryodoraku response. As shown in the results of this study, different electrical response of Ryodoraku between patients with dyspepsia of gastric dysmotility and gastric ulcer revealed discrepancy of location indicating anatomical stomach between Kyung-Rak principle and Ryodoraku and instability of explanation of autonomic nerve theory to Ryodoraku. Thus, it presented the possibility that artificial application of Kyung-Rak principle against Ryodoraku may destroy its originality in the clinical field. To correctly use Ryodoraku in the diagnosis or evaluation of disease, Ryodoraku test should be used according to Nakatani's suggestion and clinical indication of which is limited to the diseases complicated with dysfunction of the autonomic nervous system.

Effects of Ethylacetate Fraction of Persimmon Leaves on Experimentally-induced Gastric Mucosal Damage and Gastric Ulcers in Rats

  • The protective effects of the ethylacetate fraction of persimmon leaves(PEF) against experimentally induced gastric mucosal damage and gastric ulcers were evaluated in ratss. In prophylatic study, 100 mg/kg ethylacetate fraction of persimmon leaves (PEFH) exhibited a total protection of 73.8% and 65.7% against HCl-ethanol and 0.2N NaOH-induced gastric mucosal membrane lesions, respectively, which was superior to cimetidine 50 mg/kg, a commonly used anti-ulcer drug. PEFH showed excellent anti-ulcer effects against pylorus ligation induced gastric ulcers, compared to the control group, however, 50 mg/kg ethylacetate fraction of persimmon leaves (PEFL) and PEFH did not affect ulcers induced by water immersion stress, and that is inferior to cimetidine 50 mg/kg. In conclusion, the results suggest that the ethylacetate fraction of persimmon leaves can be used both in prevention and treatment of experimentally induced gastric mucosal damage and ulcers.

이부프로펜에 의해 유발된 급성 위궤양에 있어 Platycodin D의 보호효과 (Protective Effect of Platycodin D in the Acute Gastric Ulcer Induced by Ibuprofen in Rats)

  • 급성 위궤양은 위점막에서 세포증식과 세포사멸의 불균형으로 발병되어진다. 현재 Platycodin D (PD)는 항산화 및 항염증 등의 다양한 약리효능을 가진다고 보고되고 있다. 본 실험은 ibuprofen에 의해 유발된 급성 위궤양이 전처치한 PD에 의해 위궤양 보호효과를 가지는 가를 알아보기 위해 실시하였다. PD의 효능은 위점막에서의 COX-2의 발현과 더불어 위점막상피세포의 증생과 세포사멸정도에 의해서 평가하였다. 실험군은 정상대조군, ibuprofen 유발 위궤양군, 2.5 mg/kg PD 전처치군, 5 mg/kg PD 전처치군으로 분류하였다. 급성위궤양은 200 mg/kg의 ibuprofen을 하루에 3번 8시간 간격으로 경구 투여하여 유발하였다. PD는 5일간 경구로 하루에 한 번씩 전처치하였다. PD의 전처치가 ibuprofen에 의해 유발된 위궤양 병변을 유의적으로 감소시켰으며 과도한 위점액 분비로 인한 점액질의 소실을 억제하였다. 또한 PD 전처치가 위점막의 상피세포증식층에서 Ki-67 양성세포의 감소 및 세포사멸을 억제하였다. 추가적으로 PD의 전처치가 위궤양에 의해 증가된 COX-2 발현을 감소시켰다. 이상의 연구결과는 PD의 전처치가 ibuprofen에 의해 유발된 위점막손상에 있어 COX-2의 발현조절을 통하여 위점막세포의 증식과 사멸에 관여할 것으로 보여진다.

Diversity of the Gastric Microbiota in Thoroughbred Racehorses Having Gastric Ulcer

  • Equine gastric ulcer syndrome is one of the most frequently reported diseases in thoroughbred racehorses. Although several risk factors for the development of gastric ulcers have been widely studied, investigation of microbiological factors has been limited. In this study, the presence of Helicobacter spp. and the gastric microbial communities of thoroughbred racehorses having mild to severe gastric ulcers were investigated. Although Helicobacter spp. were not detected using culture and PCR techniques from 52 gastric biopsies and 52 fecal samples, the genomic sequences of H. pylori and H. ganmani were detected using nextgeneration sequencing techniques from 2 out of 10 representative gastric samples. The gastric microbiota of horses was mainly composed of Firmicutes (50.0%), Proteobacteria (18.7%), Bacteroidetes (14.4%), and Actinobacteria (9.7%), but the proportion of each phylum varied among samples. There was no major difference in microbial composition among samples having mild to severe gastric ulcers. Using phylogenetic analysis, three distinct clusters were observed, and one cluster differed from the other two clusters in the frequency of feeding, amount of water consumption, and type of bedding. To the best of our knowledge, this is the first study to investigate the gastric microbiota of thoroughbred racehorses having gastric ulcer and to evaluate the microbial diversity in relation to the severity of gastric ulcer and management factors. This study is important for further exploration of the gastric microbiota in racehorses and is ultimately applicable to improving animal and human health.

괴화산(槐花散)이 항소화성궤양(抗消化性潰瘍) 및 혈액(血液) 응고작용(凝固作用)에 미치는 실험적(實驗的) 연구(硏究) (A Study of Goiwhasan's Antigastric ulcer and Blood Hemostasis)

  • The purpose of this research was to investigate the efficacy of Goiwhasan extract powder on the gastric injuries, antiulcer, gastrointestinal tract and blood hemostasis. Animals were used through this studies mice and rats. All animals were divided into 3 groups, contol group(no treatment), sample Ⅰ group(375mg/kg administration), sample Ⅱ group(750mg/kg administration). The gastric injuries and ulcer have been made by using pyloric ligation, indomethacin, HCI-ethanol, acetic acid and then The histological observation was followed. In the gastrointestinal tract, gastric juice secretion, gastric acidity, pepsin output, blood gastrin and secretin level, transport potentials in the small and large intestine were checked. And studies on blood hemostasis were performed on normal hemostatic activities and plasma prothrombin time, plasma recalcification time, plasma fibrinogen levels in the hypoprothrombinemic mice induced by warfarin. The results were as follows: 1. The antigastric ulcer effects on the pyloric ligation, indomethacin, HCl-ethanol, acetic acid induced gastric injuries were shown in Sample Ⅱ group(p<0.05). 2. Through the morphologic examination on the acetic acid induced ulcer, Sample Ⅰ group showed mild regeneration of epithelium and slight decrease of periulcer edema then that of Control group, while Sample Ⅱ group showed more retraction of round ulcer site, remarkable loss of swelling and edema then that of Control group, and revealing the regenerated epithelium in the surrounding ulcer site. Thus it was noted that both Sample groups have antigastriculcer effects on the experimentally induced gastric ulcer. 3. The inhibitory effects on gastric juice were noted in both Sample Ⅰ group(p<0.05) and Sample Ⅱ group(p<0.01). However, only Sample Ⅱ group showed the inhibitory effects on total acidity and pepsin output(p<0.05). 4. The significant inhibition of blood gastrin level showed at 30 min.(P<0.05) and 90 min.(P<0.05) after starting medication in only Sample Ⅱ group, but significance of blood secretin level in both groups was not recognized. 5. Any significant changes in barium sulfate transport in the small intestine of mice was not recognized in both groups, but the significantly inhibitory effect in large intestine was recognized in both Sample Ⅰ group(p<0.05) and Sample Ⅱ group(p<0.001). 6. In hemostatic effect on both normal mice and hypoprothrombinemic mice induced by warfarin, the significantly shortening effect on coagulation time was seen in only Sample Ⅱ group(p<0.01). 7. On plasma prothrombin time in hypoprothrombinemic rat induced by warfarin, Sample Ⅱ group have shortened the prothrombin time significantly(p<0.001). 8. On plasma reclcification time in hypoprothrombinemic rat induced by warfarin, the recalcification time have been shortened significantly in both Sample Ⅰ group(p<0.05) and Sample Ⅱ group(p<0.01). 9. On plasma fibrinogen levels in hypoprothrombinemic rat induced by warfarin, the fibrinogen contents in Sample Ⅱ have been decreased significantly(p<0.01). Overall the above results suggest that Goiwhasan has an therapeutic efficacy on antigastric ulcer and blood hemostasis. Further studies would be needed on the interaction of its herbal medicine and its mechanism in the future.

Ameliorating Effects of Banhasasim-tang Extrat on the HCl/EtOH-induced Gastric Mucosa Damages in Mice

  • The object of this study was to observe the effect of Banhasasim-tang (BHSST) on the HCl/EtOH-induced gastric ulcer in mice. Three different dosages of BHSST extract (400, 200 and 100 mg/kg) were once orally administered 1 hr before HCl/EtOH mixture treatment. One hour after HCl/EtOH mixture single oral treatment, the changes on the gross hemorrhagic lesion scores, fundic histopathology, gastric nitrate/nitrite contents, lipid peroxidation and antioxidant defense system (catalase and superoxide dismutase (SOD) activities) were observed, and compared with that of ranitidine 100 mg/kg. As results of all three different dosages of BHSST extract treatment in the HCl/EtOH-induced gastric ulcer mice, significant and dose-dependent decreased gastric damages. BHSST extracts also increased gastric nitrate/nitrite contents and strengthened the antioxidant defense systems, and increased the activities of catalase and SOD, respectively. BHSST extracts 200 mg/kg showed similar anti-ulcerative effect as compared with ranitidine 100 mg/kg, in this experiment. BHSST has favorable protective effects against to the HCl/EtOH-induced gastric damages, through the strengthening of the body antioxidant defense systems. These gastroprotective effects of BHSST against HCl/EtOH-induced gastric ulcer considered as results of complicated synergic effects of their 8 kinds of herbal components, but exact synergic or individual herbal effects are difficult to discuss in this study. Therefore, more detail synergic effects between 8 kinds of individual herbal component of BHSST should be tested with screening of active anti-inflammatory chemical ingredients, in future.

Anti-ulcer and antioxidant activity of leaves of Madhuca indica in rats

  • The leaves of Madhuca (M.) indica J.f.Gmel. (Sapotaceae) have been used traditionally in folk medicine due to its astringent properties and are effective in treatment of eczema and snake bites. Methanolic extract of M. indica is rich in tannins and has been proven experimentally to possess antibacterial activity. The present study was undertaken to evaluate the anti-ulcer and antioxidant activity of M. indica in rats. The methanolic extract of leaves of M. indica was tested at various doses (75, 150 and 300 mg/kg, p.o.) for its effect on gastric secretion and gastric ulcers in pylorus-ligation and on ethanol- induced gastric mucosal injury in rats. The significant reduction in ulcer index in both the models along with an increase in the pH of the gastric fluid and mucin content of stomach, and the acid secretory parameters such as total acidity and volume of gastric fluid were also significantly reduced along with reduction in the pepsin activity in pylorusligated rats proved the anti-ulcer activity of M. indica. The increase in the levels of superoxide dismutase, catalase and reduced glutathione and decrease in lipid peroxidation in both the models proved the antioxidant activity of M. indica. Thus it can be concluded that M. indica possesses anti-ulcer activity, which can be attributed to its antioxidant mechanism of action.

흰쥐의 급만성궤양모델에서 제산제와 Aceglutamide aluminium의 병용효과 (Effect of Antacids, Aceglutamide Aluminium or Their Combination on Acute and Chronic Ulcer Models in Rats)

  • The combined products of antacids(AM) composed of aluminium hydroxide, magnesium hydroxide, and simethicone with a ratio of 1 : 1 : 0.1 and aceglutamide aluminium(AGA) were assayed for the antiulcer activity. The effect of the antacids(AM) in concurrent treatment with AGA was studied in acute gastric lesion induced by Shay's method, stress, ethanol, and indomethacin, in chronic gastric ulcers induced by acetic acid, and in duodenal ulcer induced by mepirizole. In all experimental models, the combined treatment of AM and AGA in the ratio of 2.3:1 showed significant potentiation in inhibition against acute gastric and duodenal ulcer and revealed a significant potentiation of the healing of chronic gastric ulcer.

영지(靈芝)버섯(Ganoderma lucidum)의 소화기계(消化器系)에 대한 약효연구(藥硏究) (Studies on the Efficacy of Ganoderma lucidum in Digestive System)

  • This study was attempted to investigate the effect of Ganoderma lucidum extract on digestive system in experimental animals. Ganoderma lucidum water extract (GWE) was found to be promoted the charcol transport rate in the small intestine of mice. GWE exhibited the augmentation of spontaneus movement(motility) and contractile response(tension) in the ileum and colon strips of rabbit, and these action were inhibited by atropine. GWE given intraduodenaly(i.d.) exhibited the significant increase of gastric acid secretion in pylorus-ligated rats. GWE inhibited the formation of some experimental gastric ulcers(pylorus ligation ulcer i.d., indomethacin-induced ulcer p.o., i.d. and aspirin-induced ulcer p.o.) in rats, which are considered to relate to a protective action. GWE and EtOH extract(water soluble phase) were remarkably increase of bile excretion, when administration of i.d., intravenation(i.v.) and per os (p.o.) compared with normal-control group. GWE was observed antibacterial activity aginst several intestinal microoganisms and others bacteria in vitro test.
