Gas vesicles are intracellular gas-filled protein-shelled nanocompartments. The structures are spindle or cylinder-shaped, and typically $0.1{\sim}2{\mu}m$ in length and 45~250 nm in width. A variety of prokaryotes including photosynthetic bacteria and halophilic archaea form gas vesicles in their cytoplasm. Gas vesicles provide cell buoyancy as flotation devices in aqueous habitats. They are used as nanoscale molecular reporters for ultrasound imaging for biomedical purposes. The structures in halophilic archaea are poorly resolved due to the low signal-to-noise ratio from the high salt concentration in the medium. Such a limitation can be overcome using focused ion beam-thinning or inelastically scattered electrons. As the concentric bodies (~200 nm in diameter) in fungi possess gas-filled cores, it is possible that the concept of gas vesicles could be applied to eukaryotic microbes beyond prokaryotes.
The in vitro effect of bisphenol A (BPA) on ovarian steroidogenesis of the longchin goby (Chasmichthys dolichognathus) was investigated. Oocytes taken during the maturing phase (vitellogenic, fully vitellogenic or germinal vesicle breakdown stage) were incubated with BPA (100 ng/mL) in the presence of exogenous precursor $^{3}H-17\alpha\;hydroxyprogesterone\;(^{3}H-17\alphaOHP).$ Steroids were extracted from the media and the isolated oocytes, and the extracts were separated and identified by thin layer chromatography and gas chromatography-mass spectrometry. The identities of the major metabolites were progestogens $[17{\alpha}-hydroxy,20{\alpha}-dihydroprogesterone\;(17{\alpha}20{\alpha}OHP)\;and\;17{\alpha}-hydrxy,20{\beta}-dihydroprogesterone\;(17{\alpha}20{\beta}OHP),$ androgens [androstenedione (A4) and testosterone (T)] and estrogens [estrone $(E_1)\;and\;estradiol-17{\beta}(E_2)].$ BPA treatment inhibited production of estrogens in all the maturing phases and progestogens in the germinal vesicle migrating stage. Percentage yield of estrogens was decreased with increased yield of androgens. In conclusion, BPA had an inhibitory effect on the conversion of $^3H-17\alphaOHP$ to estrogens and progestogens. These results demonstrate that BPA can act either estrogenic or anti-estrogenic effects.
The genes of Halohacteria. gvpD and gvpE. take part in formation of gas vesicle. These mRNA cause a lot of experimental prohlems due to its eharacteristic instahility in the analysis of transcripts. This study allowed easy cloning and sequencing of RNA hy substituting a stable complementary DNA for the mRNA of the genes for an analysis. The weak 111 RNA was reverse transcribed to DNA using reverse transcriptase. and was amplified using PCR method. The transcripts confirmed in this ~,tudy have not heen round in the northern hybridization covering almost all ranges of ORF of the gene. gvpD.
The rigid gas permeable(RGP) contact lens has nearly side effect on the cornea. So that, this lens has used the clinical reflective correction of the eye. This study have used several methods for research the fine internal structure on the RGP contact lens by scanning electron microscopy. The results have indicated that the postfixation of 1% $OsO_4$ and tannic acid is responsible for a fine structure in the internal plane of RGP contact lens. These internal surface of contact lens appeared the several shape of the hole of the stereo shape form with arrangement of round form. But, on the contact lens with non-postfixation, the stereo shape have not present and the boundary of the vesicle have not clear. Maybe, these results suggest that the fixation methods have effect on the morphological characters of materials on the RGP contact lens.
We studied steroid metabolites produced from red-spotted grouper ovarian follicles during maturation. Oocytes with 350-500 ㎛ diameter were in vitro incubated in the presence of [3H] 17α-hydroxyprogesterone as a precursor. Steroid metabolites were extracted from incubated media and oocytes. The extracts were separated and identified using thin layer chromatography, high performance liquid chromatography and gas chromatography-mass spectrometry. The identified metabolites were androstenedione (A4), testosterone (T) and estrone (E1). The metabolites of A4 was dominant in all size of oocytes and it was the highest in 480 ㎛ diameter oocytes. The metabolites of two progestins, 17α,20β-dihydroxy-4-pregnen-3-one and 17α,20α-dihydroxy-4-pregnen-3-one were detected in the oocytes less than 480 ㎛ diameter although they were not identified definitely. In the oocytes of 480 ㎛ diameter, metabolite of progestin was the highest, and germinal vesicle (GV) was still in the middle of cytoplasm. In the oocytes of 500 ㎛ diameter, GV was began to migrate and the major metabolites were A4 and E1. The metabolite of E1 was detected in all size of oocytes and it was higher than that of E2. These results suggest that oocytes of 480 ㎛ diameter are the transitional stage involving steroidogenic shift to final oocyte maturation and potential function of E1 during maturation process.
To investigate the $C_{21}$-steroids produced from maturating oocytes in the longchin goby, Chasmichthys dolichognathus, the oocytes ($0.74{\sim}0.97\;mm$) were incubated with radiolabeled $17{\alpha}$-hydroxyprogesterone ($^3H-17{\alpha}OHP$) for 24 hours. The resulting metabolites were analyzed by thin layer chromatography and identified by gas chromatography-mass spectrometry. Two $C_{21}$-steroids, $17{\alpha}$-hydroxy, $20{\alpha}$-dihydroprogesterone ($17{\alpha}20{\alpha}P$) and $17{\alpha}$-hydroxy, $20{\beta}$-dihydroprogesterone ($17{\alpha}20{\beta}P$), were converted from $^3H-17{\alpha}OHP$ in the maturing oocytes. These two main metabolites were detected at 0.80 mm diameter oocytes or greater. In addition, the effects of these metabolites on in vitro germinal vesicle breakdown (GVBD) were tested. The sensitivity of oocytes to the induction of GVBD was greater at $17{\alpha}20{\beta}P$ than $17{\alpha}20{\alpha}P$. This result showed that $17{\alpha}20{\beta}P$ is a major maturation inducing steroid (MIS) in longchin goby, suggesting $17{\alpha}20{\alpha}P$ may play a role in regulating the oocyte maturation process.
We studied oocyte steroidogenesis in the blackfin flounder Glyptocephalus stelleri as a region-specific species, in the East Sea of Korea during the spawning season. Maturing oocytes (0.76, 0.82, 0.88, and 0.91 mm in oocyte diameter) were incubated in vitro in the presence of [$^3H$] $17{\alpha}$-hydroxyprogesterone ($[^3H]17{\alpha}$-OHP) as a precursor. Steroid metabolites were extracted from the incubated medium and oocytes, and the extracts were separated and identified by thin-layer chromatography (TLC), high-performance liquid chromatography (HPLC) and gas chromatographymass spectrometry (GC/MS). The major metabolites produced from $[^3H]17{\alpha}$-OHP were androgens [androstenedione (A4) and testosterone (T)] and estrogens [$17{\beta}$-estradiol (E2) and estrone (E1)] and progestins [$17{\alpha},20{\alpha}$-dihydroxy-4-pregen-3-one ($17{\alpha}20{\alpha}P$) and $17{\alpha}20{\beta}$-dihydroxy-4-pregnen-3-one ($17{\alpha}20{\beta}P$)] in maturing oocytes. The metabolic rate of $17{\alpha}20{\beta}$ was elevated (29.04%) in oocytes measuring 0.88 mm (nucleus migration stage following the induction of germinal vesicle breakdown), but was very low in oocytes measuring 0.76, 0.82, and 0.91 mm (0.42, 0.67, and 2.62%, respectively). From these results, we suggest that $17{\alpha}20{\beta}P$ acts as a maturation-inducing steroid in the blackfin flounder.
Morphological development from egg to juvenile stages of the trident goby, Tridentigertrigonocephalu5 were observed in the laboratory at Pusan, Korea. The ripe eggs were spherical in shape, measuring 0.49-0.62 mm in diameter. The eggs became ellipsoid shape after the insemination and measured about 1.40-1.58 mm on the long axis. Hatching began about 158 hours after insemination at water temperature of $20.5-24^{\circ}C$. The newly hatched larvae were 2.88-3.14 mm in total length(TL), with 27-28(10+ 17-18) myomeres. Many melanophore and guanophores are distributed on eye cups, gas bladder, optic vesicle, intestine and the caudal region. Three days after hatching the yolk and oil-globule was completely absorbed and the larvae attained a total length 3.26-3.62 mm. The larvae swam actively in the aquarium and fed on the rotifer. Ten days after hatching, the larvae averaged 5.20 mm in TL and the caudal notochord flexed at $45^{\circ}$. Twenty days after hatching, the larvae averaged 7.47 mm in TL and rudimental anal, second dorsal, caudal and pectoral fins were formed. The larvae attained 12.05-12.65 mm in TL thirty five days after hatching and are found to transit the bottom-life, and first dorsal and ventral fins are completely formed. The larvae reached the juvenile stage at 45-50 days after hatching and attained 15.85-16.95 mm in TL, and all scales appeared on the body.
Spawning behavior and development of eggs and larvae of Mugilogobius abei were observed in the laboratory at Pusan, Korea. The adult male of Mugilogobius abei was observed making nest-like spawning-bed to lay eggs and showing territorial and courtship behaviors. The eggs were transparent and spherical in shape, measuring 0.40~0.50 mm in diameter. They have a bundle of adhesive filaments at their basal end and a cluster of small oil globules. The eggs became ellipsoid shape after the insemination and measured about 0.93~0.96 mm on the long axis. Hatching began about 110 hours after fertilization at water temperature of $24.5{\sim}25.5^{\circ}C$. The newly hatched larvae were 2.04~2.10 mm in total length, with 24~25(8~9+16) myomeres. Many melanophore and guanophore are distributed on eye cups, gas bladder, optic vesicle and the caudal region. Four days after hatching the yolk and oil-globule were completely absorbed and the larvae attained a total length 2.20~2.35 mm. The larvae swam actively in the aquarium and start to practice feeding on the rotifer. Twelve days after hatching, the larvae averaged 3.20 mm in TL and the caudal notochord flex at $45^{\circ}$. Rudimental second dorsal, anal, caudal and ventral fins are also formed. The larvae attained 10.40~10.80 mm in TL, 35 days after hatching, are found to start the bottom-life after having completely formed first dorsal and ventral fins. The larvae reached the juvenile stage at 50~60 days after hatching and attained 15.37~20.25 mm in TL. At this period all scales appeared on the body.
