Headspace 가스크로마토그라피를 이용한 식용유의 산패도 측정 (Evaluation of vegetable oil rancidity by headspace gas chromatographic analysis)

  • 면실유, 옥배유 및 채종유의 산패도를 신속하고 재현성 있게 측정하고자 $60^{\circ}C$$70^{\circ}C$로 저장한 식용유의 pentanal과 hexanal을 headspace 가스크로마토그라피 방법으로 분석하였고, 그 결과를 관능검사 결과와 상관분석을 실시하였다. Headspace 가스크로마토그라피 방법에서 휘발성 산화생성물의 포집 및 용출은 cold trap-thermal desorption 방법으로 실시하였으며, GC 주입부에 들어가는 splitless mode glass liner insert에 Tenax GC를 충전하여 사용하였다. 23종의 휘발성 산화생성물이 gas chromatography 및 gas chromatography-mass spectrometry에 의하여 검출되었으며, 저장 기간에 따른 pentanal과 hexanal 함량 변화와 관능검사에 의한 산패취 측정 결과의 상관분석을 실시한 결과, pentana(PE), hexanal(HE) 및 log{PE) log(HE) $log(PE){\times}log(HE)$의 세 가지를 변수로 한 회귀식의 상관계수의 값이 각각 0.831, 0.866 및 0.896으로 다같이 높을 값을 보였다. 따라서 headspace 가스크로마토그라피를 이용하여 면실유, 옥배유 및 채종유의 휘발성 산화생성물 중 pentanal과 hexanal을 분석하면 각각의 식용유의 산패도를 객관적으로 평가할 수 있음을 알 수 있었다.

  • Salsolinol (1-methyl-6,7-dihydroxy-1,2,3,4-tetrahydroisoquinoline, SAL), a dopaminergic isoquinoline neurotoxin, has been implicated to contribute the etiology of Parkinson's disease and neuropathology of chronic alcoholism. In our previous results, SAL was reported to have the mutagenicity and clastogenicity not in bacteria but in mammalian cells, and its genotoxic potential was known to be potentiated in the presence of rat liver S-9 fraction. This may indicate that some metabolite(s) of SAL was involved in the mutagenic potentials. To investigate the SAL metabolites, the metabolism studies of SAL were conducted in vitro rat liver S-9 fraction and in vivo using rats by high performance liquid chromatography and gas chromatography/mass spectrometry. The methylated metabolite of SAL was found in urine of rats, while the same methylating form of metabolite was not produced from the in vitro metabolism system using rat liver S-9 fraction.

Extraction and Bioassay of Allelochemicals in Jerusalem Artichoke

  • Helianthus tuberosus has been known to inhibit the growth of weeds and other plants sharing its habitat. This study was conducted to identify the allelochemicals of Helianthus tuberosus which were extracted with water and solvents. Aqueous extracts of leaf, stem, root, tuber and tuber peel of Helianthus tuberosus except tuber did not show significant differences in phytotoxicity to alfalfa seedlings. It was considered that Helianthus tuberosus contained fewer or less potential water-soluble substances that were toxic to alfalfa. Methanol extract of leaves of Helianthus tuberosus was sequentially partitioned in increasing polarity with n-hexane, ethylacetate and n-butanol. Each extract had a yield of 148, 12, 15.7 and 9.5g, respectively. Inhibitory effects on germination of alfalfa seeds treated with four fractions were not significantly different. But the significant reductions on hypocotyl length were observed for all the solvent extracts. Among the four fractions, the ethylacetate fraction showed the most significant inhibition effect on bioassay with alfalfa. Further separation of the active ethylacetate fraction by open column chromatography led to the 25 subfractions. In bioassay of each sub-fraction with alfalfa seeds, sub-fraction No. 13 showed the most inhibitory effect on seedling growth. $^1$H NMR and gas chromatography-mass spectrometry analysis revealed that sub-fraction No. 13 was the mixture of straight-chain saturated fatty acids.
