한약의 수치에 관한 연구(제 7보) -치자의 수치에 의한 성분변화 및 생리활성- (Studies on the Processing of Crude Drugs(VII) -On the Constituents and Biological Activities of Gardeniae Fructus by Processing-)

  • We have conducted to characterize the physico-chemical change and pharmacological transformation of traditional herbal medicines by means of processing. Processed Gardeniae Fructus was prepared by heating of fruit of Gardenia jasminoides(GF) for $30{\sim}50\;minute$ in the roster designed for herb processing. The contents of drying loss, water extract, diluted ethanol extract, ether extract and geniposide in non-processed GF and processed GF were examined. The contents of drying loss, water extract and geniposide in processed GF showed a decrease as compared with those of non-processed GF, however the contents of dilute ethanol and ether extract showed a increase as compared with those of non-processed GF. The rate of decrease/increase of those index were in proportion to heating time. And, biological activities of methanol extract of non-processed GF and processed GF were investigated. DPPH scavenging effects and inhibitory effect of xanthine oxidate and hemolysis of processed GF exhibited more effective than those of non-processed GF in vitro. Accelerating effect of large intestinal transport and purgative action of non- processed GF were discriminated by processing of GF. Methanol extracts of non-pro- cessed GF and processed GF showed the protective effects against the hepatotoxicity induced by ${\alpha}-naphthylisothiocyanate$ in rats. These results suggested that the transformation of biological activities of GF by means of processing may be due to the physico-chemical change of the constituents in GF by heating.

약선식품소재의 유산균 증식 효과 (Bifidogenic Effects of Yaksun (functional herbal) Food Materials)

  • 1. 약선식품소재의 물추출물 중 pH 저하효과가 가장 큰 것은 B. breve를 배양하였을 경우 Citrus aurantium (진피), Liriipe platyphylla (맥문동), Platycodon grandiflorum (길경), Schizandra chinensis (오미자)였고, 사람의 장내균총을 배양했을 경우는 Angelica gigas (당귀), Hordeum vulgare (맥아), Liriipe platyphylla (맥문동), Platycodon grandiflorum (길경), schizandra chinensis (오미자) 등이었다. 2. 약선식품소재의 $\beta$-glucosidase 효소활성억제효과는 B. breve를 배양했을 경우 Citrus aurantium(진피), Eugenia caryophyllata (정향), Liriipe platyphylla (맥문동), Platycodon grandiflorum (길경), Schizandra chinensis (오미자)가 90%. 이상의 억제효과를 나타내었고 사람의 장내균총에 대해서는 Eugenia caryophyllata (정향), Liriipe platyphylla (맥문동)가 40% 이상의 억제효과를 나타내었다. 3. $\beta$-Glucuronidase 활성을 억제하는 것으로 나타난 약선식품소재는 사람의 장내균총에 대해서는 Cinnamomum cassia(계지), Gardenia jasminoides(치자), Platycodon grandiflorum (길경) 등이였다. 4. 길경으로부터 in vitro에서 유산균증식효과를 나타내는 compound B를 분리하였으며 이 화합물은 기기분석을 한 결과 sucrose임을 밝혀졌으며, 사람의 장내균총에 대한 효과를 측정한 결과 $\beta$-glucosidase활성은 크게 억제하지 못했지만 $\beta$-glucuronidase 및 tryptophnase활성은 각각 49%, 67%의 억제효과를 나타내었다. 또한 사람의 장내우세균인 B. breve를 증식시키는 효과가 있었다.

천연색소 추출공정에서 추출특성에 따른 목표색의 재현 (Presentation of Target Color with Extraction Variables in Natural Dyes Extraction)

  • 천연색소를 이용한 컬러의 재현은 현대의 웰빙 생활과 밀접한 연관성을 갖게 된다. 이 연구는 일상생활에서 자주 쓰이는 녹색 계, 황색 계, 그리고 적색 계의 컬러를 자연 상의 녹차, 치자, 흑미를 색소원으로 하여 목표 색을 설정하였다. 목표색의 명도(L), 색 좌표(a, b)의 정량적 수치 값을 지정하고 추출용매의 물성을 조절함으로써 목표 색에 가장 접근하는 색을 추출하여 표현하였다. 색소추출 공정에서 용매의 pH, 추출온도, 추출시간이 변수로 설정되었다. 치자의 경우 추출시간이 길수록 진한색이 추출되었다. 추출색소는 색차계를 이용해 색의 L-a-b좌표를 측정하여 목표 색과 비교하였다. 실험조건을 최적화함으로써 목표 색과 가장 색차 값(${\Delta}E$)이 적은 녹색, 황색, 적색의 CIE 기준 값을 제시하고 이 좌표에 맞는 색을 표현하였다.

초임계 추출을 적용한 식물추출물의 항산화 및 피부장벽 효과 (Anti-oxidative and skin barrier effects of natural plants with a supercritical extract)

  • 본 연구에서는 다양한 식물추출물의 항산화성 및 미용관련 기능성을 알아보기 위하여 기존의 용매추출에 비해 안전성이 높은 것으로 알려진 초임계 추출방식을 적용하여 구절초(Chrysanthemum zawadskii), 수세미(Lufa cylindrica), 작약(Paeonia lactiflora), 치자나무(Gardenia jasminoides) 및 황금(Scutellaria baicalensis)의 천연식물 초임계 추출물을 확보하고, 이들의 항산화력 및 피부장벽 향상 효과를 조사하였다. 초임계 식물추출물의 피부장벽기능 향상 효과를 조사하기 위하여 항산화 활성 외 proliferator-activated receptor(PPAR)-${\alpha}$ 활성, cornified envelope(CE)에 관련된 단백질인 involucrin의 발현량을 측정하였다. 이들 추출물 중 황금추출물이 가장 높은 항산화 활성을 보였으며, 모든 추출물에서 대조군과 비교하여 유의적으로 높은 수준의 PPAR-${\alpha}$ 활성을 나타내었다. 또한, 피부장벽 기능 지표 단백질인 involucrin 발현량도 모든 추출물에서 높게 나타났으며, 황금추출물이 가장 높은 단백질 발현 양상을 보였다. 따라서 본 연구에서 조사한 5개 식물의 초임계 추출물은 항산화 및 피부장벽 기능개선과 같은 기능성 생물활성 소재로 활용 될 수 있을 것으로 판단된다.

대두와 면 혼방직물의 염색에 따른 태의 비교 (Hand Value Assessment of Soybean/Cotton Blended Fabric after Natural and Synthetic Dyeing)

  • The purpose of this study is to investigate the dyeability, and the hand of soybean/cotton blended fabric after dyeing with natural and synthetic dyes. Soybean/cotton blended fabrics dyed with extracted solution from turmeric, sappan wood, gardenia and synthetic dyes(weak-acid dyes, reactive dyes, direct dyes). The hand value of soybean/cotton blended fabrics dyed with 6 different dyes was measured using Kawabata Evaluation System. The results are as follows; 1. The K/S value of the soybean/cotton blended fabric dyed with sappan wood extracts was the highest. 2. The color fastness of the soybean/cotton blended fabric dyed with reactive dyes was excellent. The fastness to the light of the same fabric was much better than the other fabrics. 3. Linearity of load-extension and tensile energy of the soybean/cotton blended fabric dyed with sappan wood extracts showed very high. Shear stiffness of the soybean/cotton blended fabric dyed with weak-acid dyes was much higher than that of fabric of dyed with reactive dyes. Bending property of the fabrics dyed with natural colorants(sappan wood, gardenia) showed very high, but that the fabric dyed with reactive dyes was very poor. 4. In the primary hand value, stiffness and anti-drape stiffness of the fabric dyed with weak-acid dyes showed the highest. Fullness and softness of the fabric dyed with weak-acid dyes showed very low.

염료첨가 인공사료에 의한 유색 고치 생산 (Production of Colored cocoons from Silkworm by Feeding Artificial Diet mixed Dye)

  • 인공사료로 먹인 누에에 천연염료와 화학 염료를 각각 혼합하여 5령 3일 째부터 먹인 누에의 유색고치 생산을 검토한 결과 다음과 같다. 1. 천연염료와 인공사료를 혼합 할 경우에 치자 색소만 청자색으로 변환하여 유색 견을 생산하였고 고치의 중량은 대조 보다 낮았다. 2. 화학 염료 혼합 인공사료는 Orang II, Methyl Orang, Acid Blue R의 유색고치 생산은 주황색 노란색, 파란색 순서로 유색 고치를 생산하였다.

만성적인 에탄올 섭취와 천연물 투여가 흰쥐의 항산화계와 에탄올 산화계에 미치는 영향 (Effect of Chronic Ethanol Consumption and Herbal Extracts Administration on the Antioxidant System and Ethanol Oxidation System in Rats)

  • This study had been done for the investigation of the effect of Vitis vinifera extract(V), Schisandra chinensis extract (S), Taraxacum officinale extract (T), Gardenia jasminoides extract (G), Angelica acutiloba extract (A) and Paeonia japonica extract (P), and their mixtures on the antioxidant and ethanol oxidation system which was induced by Lieber-DeCarli ethanol liquid diet. Male Sprague-Dawley rats were randomly divided into eight groups: ethanol diet (ED), normal diet (ND), ED+V (100 mg/kg/day), ED+S, ED+T, ED+G, ED+A and ED+P (300 mg/kg/day). We studied the effect on alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) after herbal extracts administration for 6 weeks in rats induced by Lieber-DeCarli ethanol liquid diet. The differences in ADH and ALDH activity of the rats treated with herbal extracts and ED group were not significant. Phase I enzyme activity was found to be significantly higher in the ED+V than the ED group. Phase II enzymes (glutathione-S-transferase, phenol sulfatransferase) activities were found to be higher in the herbal extracts than the ED group. Herbal extracts not only reduced ethanol-induced elevation of level malondialdehyde but also protected against ethanol-induced decrease of reduced glutathione, gluthione reducatse, glutathione peroxidase, catalase and superoxide dismutase activities. Therefore, they can be utilized as a health functional food or new drug candidate for fatty liver and hepatotoxicity which was induced by chronic alcohol consumption.

기능성 증진을 위한 치자색소와 창이자에 의한 견직물의 복합염색 (Mixture Dyeing of Silk Fabrics with Gardenia and Xanthium Strumarium L. to Improve the Functionality)

  • This study had a purpose of further investigating and developing the usage of the eco-friendly functional dye that has physiological and pharmacological functions. This study was implemented complex dyeing method which used both natural dyes such as Gardenia jasminoides J.Ellis colorant, which are the yellow dyes that have high brightness and low chroma, and Xanthium strumarium L. extract. The silk fabrics were used to dye in various methods. They were also used to investigate functionalities dyed fabrics by various treating method, such as their color fastness, antibacterial activities and deodorization, the following were obtained. The fabrics that were dyed with herbs had decreasing brightness as the herbs extract concentrations were increased and had subtly changing chroma. But, because their K/S values were under 0.5, they were not properly dyed. Color fastness differed according to the natural dyes and herbs' complex dyeing order. The Color fastness was good when the fabrics were dyed with the colorant first, and then with the herb. When the fabrics were dyed with mixture dyeing solution, the natural dye and the herb extract, the herb didn't affect the dyeabilities of fabrics as much. When the fabrics were dyed with the mixture of the colorant and the herb extract, the dyeabilities didn't have much changes. However, their color fastnesses were improved a little bit due to the complex dyeing condition. When the fabrics were dyed with the mixture of the colorant and the herbs extract, their antibacterial activities and deodorization were increased.

Natural Blue Pigment from Gardenia jasminoides Ellis (Rubiaceae) as a Dental Plaque Disclosant

  • Background: Dental caries and periodontal disease are bacterial infectious disease, mainly caused by plaque, a bacterial colony deposited on the tooth surface and gum tissue. Dental plaque disclosants easily stain the dental plaque, making them effective for scaling and tooth brushing education. As the erythrosine typically contained in dental plaque disclosants is highly cytotoxic, a low toxicity additive is needed. In this study, we aimed to examine the natural pigments with negligible cytotoxicity but can effectively stain the dental plaques for use in dental plaque disclosants. Methods: The pigmentation of eight types of natural pigments was tested on bovine tongue and teeth, as well as on head and neck tissue sections of experimental ICR mice. The cytotoxicity of gingival epithelial cells was measured via MTT assay. Pigmentation was performed on the bovine tongue and tooth surface. Pigmentation in the oral environment was observed in four mandibular incisors. A 2 Tone was used as a control. Results: Of the eight types of natural pigments, purple and blue pigments were effective in coloring dental plaques on the enamel surface as well as in the head and neck tissue sections. Additionally, purple and blue pigments were visible on the surface of the bovine tongue. Red, pink, orange, green, purple, and yellow pigments showed strong cytotoxicity, whereas brown and blue pigments had relatively low cytotoxicity. Blue pigment was effective in staining the dental plaque of four mandibular incisors. Conclusion: We suggest that the blue pigment derived from Gardenia jasminoides Ellis (Rubiaceae), which is effective for coloring dental plaques and has low cytotoxicity, is useful as a naturally derived dental disclosant.

지속 가능한 해초 함유 섬유의 천연염료 염색성에 관한 연구 (A Study on the Dyeability of Natural Dyes of Sustainable Seaweed-Containing Fiber)

  • This study investigated the dyeability and color change of the natural dyes of SeaCell, a biodegradable functional fiber that is permanently added to cellulose fibers with natural additives extracted from seaweeds. The natural dyes used in the study are five dyes. Gardenia and turmeric, which are yellow-based natural dyes, Sappan wood and Lac, which are red-based natural dyes, and Indigo, a blue-based natural dye, were selected. The dyeability and color change according to the change of the mordant conditions and the number of times of dyeing were investigated. In addition, the dyeing properties and colors of cotton and silk fibers were compared under the same dyeing conditions as SeaCell. The study results are as follows. It was found that SeaCell had lower dyeing properties than silk, a protein fiber, in gardenia, sappan wood, and lac dyes, but had higher dyeing properties than cotton with the same cellulose component as SeaCell fibers. In the case of turmeric, it showed higher dyeing properties than cotton except for the no mordant condition. In the case of Indigo dye, SeaCell shows the best dyeability, indicating that it is a very suitable fiber for Indigo dyeing. As sustainable functional fibers are continuously developed in the future, natural dyes that are environmentally friendly and human-friendly are actively introduced and commercialized, and it is expected that they can be used as reference materials.
