Anti-inflammatory effect of ganodermanondiol from Ganoderma lucidumon RAW 264.7 cells (영지 유래 가노더마논디올의 RAW 264.7 세포주에 대한 항염 효과)

  • Owing to its diverse range of bioactive compounds, Ganoderma lucidumhas garnered significant research attention for health promotion and disease prevention. Ganodermanondiol, which has a triterpenoid structure, is one of the major active compounds of G. lucidum. In the present study, the anti-inflammatory effects of ganodermanondiol were investigated to evaluate its usefulness as a functional ingredient. Ganodermanondiol (0.5-2 ㎍/mL) significantly inhibited the production of nitric oxide (NO), the expression of the cytokines tumor necrosis factor (TNF)??and interleukin 6 (IL-6), and the expression of cyclooxygenase (COX)-2 and inducible nitric oxide synthase (iNOS) in lipopolysaccharide-induced RAW 264.7 (murine macrophage) cells. Ganodermanondiol (0.5-2 ㎍/mL) also inhibited the phosphorylation of mitogen-activated protein kinase (MAPK) signal molecules, including p38 and c-Jun N-terminal protein kinase (JNK) in RAW 264.7 cells. Ganodermanondiol significantly inhibited the essential factors involved in the inflammatory responses of RAW 264.7 cells and would, therefore, serve as a potential prophylactic and therapeutic agent for immune-related diseases.

Enhancement of Ganodermanondiol and Anti-melanogenesis Effect of Ganoderma lucidum by Rhus verniciflua Extract Supplementation (옻나무 추출물 첨가에 따른 영지버섯의 가나도마난디올 생합성 증대 및 멜라닌 생성 저해효과)

  • This study was carried out to investigate the effect of lacquer tree (Rhus verniciflua) extract on ganodermanondiol (GN), tyrosinase and melanin biosynthesis inhibitor, biosynthesis in Ganoderma lucidum mycelia. In HPLC analysis, GN contents were significantly increased in G. lucidum mycelial extracts supplemented with of 1, 5, 10, and 15% lacquer tree extracts (LTE). In addition, G. lucidum mycelial extracts supplemented with LTEs which had no cytotoxicity activity against B16F10 cells, significantly inhibited melanogenesis in B16F10 cells. GN biosynthesis was facilitated by LTE. Taken together, we propose that G. lucidum mycelial extracts supplemented with LTE can be used as an effective ingredient of skin care products in the future.

Anti-melanogenesis Effect of Ganoderma lucidum Mycelial Extract Supplemented with Oriental Raisin Tree (Hovenia dulcis) Extract (헛개나무 추출물이 첨가된 영지버섯균사 추출물의 멜라닌 생성 억제효과)

  • The aim of this study is to investigate the effect of Hovenia dulcis (oriental raisin tree) extract on ganodermanondiol (GN) contents in Ganoderma lucidum (G. lucidum) mycelia. GN has a triterpenoid structure and is one of the major active components of G. lucidum. Furthermore, we previously proved its inhibitory effects on tyrosinase activity and melanin biosynthesis in B16F10 melanoma cells. In this study, we observed significantly increased GN contents in G. lucidum mycelial extracts supplemented with 15% (v/v) oriental raisin tree extract (ORTE) by HPLC analysis. In addition, melanogenesis was significantly inhibited by G. lucidum extract supplemented with 15% ORTE when compared to G. lucidum extract without ORTE supplementation. Furthermore, mycelial growth of G. lucidum was increased by ORTE supplementation in both solid and liquid cultivation. These results suggest that the oriental raisin tree is useful as natural ingredient for increasing GN biosynthesis as well as whitening effect of G. lucidum.

Cytotoxic Triterpenoids from the Fruiting Bodies of Ganoderma lucidum

  • Twelve triterpenoids (1 - 12) were isolated from $CHCl_3$-soluble fraction of fruiting bodies of Ganoderma lucidum. Extensive spectroscopic and chemical studies established the structures of these compounds as butyl lucidenate P (1), butyl lucidenate $E_2$ (2), butyl lucidenate $D_2$ (3), butyl lucidenate Q (4), ganoderiol F (5), methyl ganoderate H (6), methyl ganoderate J (7), lucidumol B (8), ganodermanondiol (9), methyl lucidenate N (10), methyl lucidenate A (11) and butyl lucidenate N (12). All of the compounds were examined for their cytotoxic activity against HL-60, HeLa, and MCF-7 cancer cell lines. Among them, compounds 4 and 8 showed cytotoxic activity with $IC_{50}$ values of 6.6 and 1.6 ${\mu}M$ against HL-60, respectively. In addition, compound 8 also showed cytotoxic activity with $IC_{50}$ values of 2.0 ${\mu}M$ against HeLa cancer cell line, other compounds were moderate or inactive.
