A Splice Variant of the C2H2-Type Zinc Finger Protein, ZNF268s, Regulates NF-κB Activation by TNF-α

  • $I{\kappa}B$ kinase (IKK), the pivotal kinase in signal-dependent activation of nuclear factor-${\kappa}B$ (NF-${\kappa}B$), is composed of multiple protein components, including IKK ${\alpha}/{\beta}/{\gamma}$ core subunits. To investigate the regulation of the IKK complex, we immunoaffinity purified the IKK complex, and by MALDI-TOF mass spectrometry identified a splice variant of zinc finger protein 268 (ZNF268) as a novel IKKinteracting protein. Both the full-length and the spliced form of the ZNF268 protein were detected in a variety of mammalian tissues and cell lines. The genes were cloned and expressed by in vitro transcription/translation. Several deletion derivatives, such as KRAB domain (KRAB) on its own, the KRAB/spacer/4-zinc fingers (zF4), and the spacer/4-zinc fingers (zS4), were ectopically expressed in mammalian cells and exhibited had different subcellular locations. The KRAB-containing mutants were restricted to the nucleus, while zS4 was localized in the cytosol. TNF-${\alpha}$-induced NF-${\kappa}B$ activation was examined using these mutants and only zS4 was found to stimulate activation. Collectively, the results indicate that a spliced form of ZNF268 lacking the KRAB domain is located in the cytosol, where it seems to play a role in TNF-${\alpha}$-induced NF-${\kappa}B$ activation by interacting with the IKK complex.

Comparison of Mathematical Models Applied to F1 Dairy Sheep Lactations in Organic Farm and Environmental Factors Affecting Lactation Curve Parameter

  • The objective of this study was to compare the goodness of fit of four lactation curve models: Wood's Gamma model (WD), Wilmink (WL), and Pollott's multiplicative two (POL2) and three parameters (POL3) and to determine the environmental factors affecting the complete lactation curve of F1 dairy sheep under organic management. A total of 5,382 weekly milk yields records from 150 ewes, under organic management were used. Residual mean square (RMS), determination coefficients ($R^2$), and correlation (r) analysis were used as an indicator of goodness of fit for each model. WL model best fitted the lactation curves as indicated by the lower RMS values (0.019), followed by WD (0.023), POL2 (0.025) and POL3 (0.029). The four models provided total milk yield (TMY) estimations that were highly correlated (0.93 to 0.97) with observed TMY (89.9 kg). The four models under estimated peak yield (PY), whereas POL2 and POL3 gave nearer peak time lactation estimations. Ewes lambing in autumn had higher TMY and showed a typical curve shape. Higher TMY were recorded in second and third lambing. Season of lambing, number of lambing and type of lambing had a great influenced over TMY shaping the complete lactation curve of F1 dairy sheep. In general terms WL model showed the best fit to the F1 dairy sheep lactation curve under organic management.

삽목에 의한 갯버들 근계의 토양전단강도 보강효과 (The Effect of Reinforcing Soil Shear Strength by a Root System Developed from Direct Sticking of Salix gracilistyla Miq)

  • The purpose of this study was to verify the shore margin protection effect of a root system developed from direct sticking of Salix gracilistyla Miq., focusing on the reinforcement of soil shear strength. The materials were 20cm long sticks whose average diameter and weight were 7.52mm and 14.58g respectively, and sandy loam(Sand 60.36%, Silt 28%, Clay 11.64%), whose maximum dry weight(${\gamma}$$_{dmax}$) was 1.59gf/㎤ at the water ratio( $W_{opt}$) 13.8%. The direct shearing test(KS F 2343) was applied to cylindric columms(diameter 132mm) of pure soil and two years old root reinforced soil. At each condition of vertical stress, 10N/$ extrm{cm}^2$, 14.41N/$\textrm{cm}^2$ and 18.82 N/$\textrm{cm}^2$, five soil+root columns were sheared. After shear tests, the root area ratio and soil moisture on the shear plane were measured. The results of this research were as follows: 1. The average of root area ratio was 1.86% and the soil moisture 14.67%. 2. Two years old root system was found to increase the soil shear strength of pure soil in terms of Cohesion(C) and Inner friction Angle($\phi$) as follows. 3. 베타아밀로이드로 유도된 신경소교세포 사멸에 대한 루테올린의 보호효과 연구 (Protective Effect of Luteolin against β-Amyloid-induced Cell Death and Damage in BV-2 Microglial Cells)

  • Objectives : The purpose of this study is to investigate neuroprotective effects and molecular mechanisms of luteolin against ${\beta}$-amyloid ($A{\beta}_{25-35}$)-induced oxidative cell death in BV-2 cells. Methods : The protective effects of luteolin against $A{\beta}_{25-35}$-induced cytotoxicity and apoptotic cell death were determined by MTT dye reduction assay and TUNEL staining, respectively. The apoptotic cell death was further analyzed by measuring mitochondrial transmembrane potential and expression of pro- and/or anti-apoptotic proteins. To elucidate the molecular mechanisms underlying the protective effects of luteolin, intracellular accumulation of reactive oxygen species, oxidative damages, and expression of antioxidant enzymes were examined. Results : Luteolin pretreatment effectively attenuated $A{\beta}_{25-35}$-induced apoptotic cell death indices such as DNA fragmentation, dissipation of mitochondrial transmembrane potential, increased Bax/Bcl-2 ratio, and activation of c-Jun N-terminal kinase and caspase-3 in BV-2 cells. Furthermore, $A{\beta}_{25-35}$-induced intracellular formation of reactive oxygen species and subsequent oxidative damages such as lipid peroxidation and depletion of endogenous antioxidant glutathione were suppressed by luteolin treatment. The neuroprotective effects of luteolin might be mediated by up-regulation of cellular antioxidant defense system via up-regulation of ${\gamma}$-glutamylcysteine ligase, a rate-limiting enzyme in the glutathione biosynthesis and superoxide dismutase, an enzyme involved in dismutation of superoxide anion into oxygen and hydrogen peroxide. Conclusions : These findings suggest that luteolin has a potential to protect against $A{\beta}_{25-35}$-induced neuronal cell death and damages thereby exhibiting therapeutic utilization for the prevention and/or treatment of Alzheimer's disease.

고연소도 사용후 핵연료의 가열산화와 고온가열을 통한 미세조직 변화고찰 (Study of morphology on the Oxidation and the Annealing of High Burn-hp $UO_2$ Spent Fuel)

  • 조사후 핵연료 가열(PIA장비)를 이용한 고연소도 UO2 사용후 핵연료의 산화 및 가열후 미세조직의 변화를 관찰하였다. 울진 2호기에서 한국원자력연구소 조사후시험시설로 이송된 국산 경수로용 고연소도 사용후 핵연료는 봉평균 연소도가 57,000 MWd/tU-rod avg.이였다. 본 시험에 사용된 시편은 국부연소도 65,000 MWd/tU UO2 소결체의 고형체 200 mg을 사용하였다. 본 시편을 사용후 핵 연료 가열(PIA) 시험장비를 이용하여 핫셀 내에서 3시간의 산화시험과 연속적으로 $1,400^{\circ}C$ 까지 가열하였다. 결정립경계까지의 산화를 위하여 $500^{\circ}C$에서 헬륨 50 ml, 표준공기 100 ml를 흔합한 산화분위기로 3시간을 유지하였다. 핵분열기체 방출거동을 알기위해 시험 전과정중에 85Kr의 방출량을 베타 측정기와 감마 측정기를 이용하여 실시간으로 측정 하였다. 가열시험이 종료된 후 전자주사현미경을 이용하여 미세구조의 변화를 관찰하였다. 시험결과 가열하는 동안 핵분열생성물은 UO2기지의 결정립경계와 표면으로 이동된 것을 관찰하였다. 이 시편은 환원과정을 통하여 재구조화 되었고, $5\~10\;{\mu}m$ 정도의 결정립크기를 가진 것으로 나타났다.

  • Mercury is a widespread metal and consequently there are large populations that currently exposed to low levels of mercury. Endotoxin is a component of the gram-negative bacteria and promotes inflammatory responses. The present study was designed to determine the impact of mercury on lymphocytes phenotype populations and endotoxin-induced inflammatory cytokine expressions in immune organ, spleen and thymus. Male BALB/c mice were exposed continuously to 0, 0.3, 1.5, 7.5, or 37.5 ppm of mercuric chloride in drinking water for 14 days and at the end of the treatment period, lipopolysaccharide (LPS, 0.5 mg/kg) was injected intraperitoneally 2 h prior to euthanasia. The dose-range of mercury used did not cause hepatotoxicity. Mercury at 7.5 and 37.5 ppm dose-dependently decreased CD3$^{+}$ T lymphocytes in spleen; both CD4$^{+}$ and CD8$^{+}$ single positive lymphocyte populations were decreased. Exposure to 7.5 and 37.5 ppm of mercury decreased the CD8$^{+}$ T lymphocyte population in the thymus, whereas double positive CD4$^{+}$ / CD8$^{+}$ and CD4$^{+}$ thymocytes were not altered. Mercury altered LPS-induced inflammatory cytokine gene expressions such as, tumor necrosis factor $\alpha$, interferon ${\gamma}$, and interleukin-12 in spleen and thymus. Results indicated that decreases in T lymphocyte populations in immune organs and altered cytokine gene expression may contribute to the immune-modulative effects of inorganic mercury.ganic mercury.

  • An analysis method for the electromagnetic scattering of a tE polarized plane wave from a periodic strip grating over a grounded dielectric slab is consisered from the viewpoint of reflectio ngraing problem. The strip gratings showing bragg and off-bragg balzing phenomena at the frequency of 10GHz are designed, respectively. The strip grating structure is implemented using aluminum plate (hround conductor), paraffin(dielectric material ; .xi.$_{\gamma}$=2.24) and copper (strip conductor ; 0.08mm thickness). The experimental results (reflection power) for bragg as well as off-bragg blazing phenomenon have been compared with the theoretical results and fairly good agreements between theory and experiment have been observed.ed.

The Effects of Sodium Chloride on the Physiological Characteristics of Listeria monocytogenes

  • Sodium chloride is used to improve various properties of processed meat products, e.g., taste, preservation, water binding capacity, texture, meat batter viscosity, safety, and flavor; however, many studies have shown that sodium chloride increases the resistance of many foodborne pathogens to heat and acid. Listeria monocytogenes has been isolated from various readyto- eat (RTE) meat and dairy products formulated with sodium chloride; therefore, the objective of this paper was to review the effects of sodium chloride on the physiological characteristics of L. monocytogenes. The exposure of L. monocytogenes to sodium chloride may increase biofilm formation on foods or food contact surfaces, virulence gene transcription, invasion of Caco-2 cells, and bacteriocin production, depending on L. monocytogenes strain and serotype as well as sodium chloride concentration. When L. monocytogenes cells were exposed to sodium chloride, their resistance to UV-C irradiation and freezing temperatures increased, but sodium chloride had no effect on their resistance to gamma irradiation. The morphological properties of L. monocytogenes, especially cell elongation and filament formation, also change in response to sodium chloride. These findings indicate that sodium chloride affects various physiological responses of L. monocytogenes and thus, the effect of sodium chloride on L. monocytogenes in RTE meat and dairy products needs to be considered with respect to food safety. Moreover, further studies of microbial risk assessment should be conducted to suggest an appropriate sodium chloride concentration in animal origin foods.

The optimal system for series systems with warm standby components and a repairable service station

  • This paper deals with the reliability and availability characteristics of three different series system configurations with warm standby components and a repairable service station. The failure time of the primary and warm standby are assumed to be exponentially distributed with parameters ${\lambda}$ and ${\alpha}$ respectively. The repair time distribution of each server is also exponentially distributed with parameter ${\mu}$. The breakdown time and the repair time of the service station are also assumed exponentially distributed with parameters ${\gamma}$ and ${\beta}$ respectively. We derive the reliability dependent on time, availability dependent on time, the mean time to failure, $MTTF_i$, and the steady-state availability $A_i$(${\infty}$) for three configurations and perform comparisons. Comparisons are made for specific values of distribution parameters and of the cost of the components. The three configurations are ranked based on: $MTTF_i$, $A_i$(${\infty}$), and $C_i/B_i$ where $B_i$ is either $MTTF_i$ or $A_i$(${\infty}$).

내식성 및 표면경도 향상을 위한 AISI 304L 스테인리스강의 저온 플라즈마질화 프로세스 (Low Temperature Plasma Nitriding Process of AISI 304L Austenitic Stainless Steels for Improving Surface Hardness and Corrosion Resistance)

  • The effects of processing parameters on the surface properties of the hardened layers processed by the low temperature plasma nitrocarburizing and the low temperature two-step plama treatment (carburizing+nitriding) were investigated. The nitrogen-enriched expanded austenite structure (${\gamma}_N$) or S phase was formed on all of the treated surface. The surface hardness reached up to 1200 $HV_{0.025}$, which is about 5 times higher than that of untreated sample (250 $HV_{0.1}$). The thickness of hardened layer of the low temperature plasma nitrocarburized layer treated at $400^{\circ}C$ for 40 hour was only $15{\mu}m$, while the layer thicknesss in the two-step plama treatment for the 30 hour treatment increased up to about $30{\mu}m$. The surface thickness and hardness increased with increasing treatment temperature and time. In addition, the corrosion resistance was enhanced than untreated samples due to a high concentration of N on the surface. However, higher treatment temperature and longer treatment time resulted in the formation of $Cr_2N$ precipitates, which causes the degradation of corrosion resistance.
