Cordyceps militaris has shown to have various pharmacological activities including an immune-modulatory effect. Previously, we reported that anti-influenza effect of C. militaris in DBA/2 mice was mediated by increased IL-12 and the activation of NK cells. In this study, we investigated the effect of C. militaris on adaptive immune responses using DBA2 mice immunized with influenza vaccine. To determine the effect of C. militaris on antigen presentation capability, we treated RAW 264.7 cells with various concentrations of ethanol extract of C. militaris, which showed a significant upregulation of CD86 (B7.2), CD284 (TLR4), CD40, H-2k (MHC I) and I-Ad (MHC II). To examine the direct effect of C. militaris on adaptive immune responses, we immunized DBA2 mice with influenza vaccine in presence or absence of C. militaris. After 2 or 4 weeks, influenza-specific T cell proliferation, HAI titers and IFN-${\gamma}$ production were measured in plasma or PBMCs isolated from animals. Influenza-specific T cell proliferation and HAI titers were not considerably increased in immunized mice in presence of C. militaris. However, the production of IFN-${\gamma}$ was much greater in immunized mice with C. militaris as adjuvant than only immunized mice.
본 연구는 김치의 발효 말기에 비타민 C의 증가 현상을 조사하였으며, 이는 김치 발효 후기에 생육하는 효모에 의한 현상으로 미생물인 효모에서 비타민 C 생합성 경로를 밝히기 위해 수행된 것으로 그 결과는 다음과 같았다. 김치의 숙성에 따른 pH와 총 산함량의 변화는 김치가 숙성되면서 pH는 담금 초기 5.29에서 숙성 33일에 pH 3.50으로 감소되었고, 총 산함량은 담금 초기 0.24%에서 숙성 33일에 2.48%로 증가되었으며, 비타민 C의 함량을 측정한 결과 발효 3일째까지 점점 상승하다 감소되었으나, 발효 15일째는 19.58mg%로 초기 함량보다 다소 높게 측정되었다. 김치의 PG 활성은 김치 숙성초기에는 낮았으나, 김치 숙성 10일 이후부터 증가되어 발효 13일에는 15.4unit/mg protein으로 PG활성이 증가하는 경향을 보였다. 김치에서 분리된 효모 중 배양액의 비타민C의 함량이 높게 측정된 효모를 선별하였으며, 동정결과 Pichia onychis, Pichia rabaulenis, Pichia jadinii, Candida humilis로 각각 동정되었다. 분리, 동정된 효모 중 비타민 C생성량이 가장 높았던 Pichia onychis 16-4 균주로부터 비타민 C 합성 조효소액을 추출하였으며, specific activity는 7.26unit/mg protein로 나타났다. 최종 정제 시 효소활성은 4,698unit/mg protein으로 나타나 처음보다 647.10배로 농축되었음을 알 수 있었다. 정제된 효소액의 분자량은 31,000, 39,000, 50,000 KD으로 3개의 subunits를 갖는 것이 확인되었다. 정제된 효소는 $L-galactono-{\gamma}-lactone$를 최적 기질로 사용하였으며 $35^{\circ}C$에서 pH 7.8일때 최적활성을 나타내었다. 본 연구에서 정제된 효소는 산소를 전자수용체로 하는 $L-galactono-{\gamma}-lactone$ oxidase임을 알 수 있었다.
Linoleic acid methylester와 토코페놀류$({\alpha}-,\;{\beta}-,\;{\gamma}-,\;{\delta}-tocopherol$과 ${\alpha}-,\;{\gamma}-,\;{\delta}-tocotrienol)$를 사용한 model system에서의 자동산화과정에서 온도$(40,\;60,\;80^{\circ}C)$와 산소의 양 $(0,\;10,\;20%\;O_2)$에 따르는 항산화효과를 고찰하였다. Linoleic acid methylester에서 생성된 주요 산화생성물인 13-cis/trans-, 13-trans/trans-, 9-trans/cis-, 9-trans/trans-hydroperoxide를 HPLC를 이용하여 분석하였으며, 그 양적 변화로 항산화정도를 비교하였다. 모든 토코페롤류$({\alpha}-tocotrienol$제외)는 $60^{\circ}C$ 범위에서 항산화효과가 있었다. ${\alpha}-tocopherol$, ${\alpha}-tocotrienol$ and ${\delta}-tocotrienol$의 경우 $80^{\circ}C$ 범위에서 산화촉진효과가 관찰되었다. $40^{\circ}C$ 범위에서는 주로 cis/trans-hydroperoxide가 많이 생성되었고 $80^{\circ}C$ 범위에서는 trans/trans-hydroperoxide가 많이 생성되었다. $80^{\circ}C$ 범위의 hydroperoxide 생성량간의 오차가 심한 구간을 제외한 $40^{\circ}C/10%,\;O_2$에서부터 $60^{\circ}C/20%,\;O_2$까지의 산화구간에서 cis/trans-: trans/trans-hydroperoxides의 비율은 ${\alpha}-T>{\alpha}-T_3>{\gamma}-T>{\beta}-T>{\gamma}-T_3>{\delta}-T>{\delta}-T_3$의 순으로 고찰되어, 토코페롤류 중 ${\alpha}-tocopherol$이 가장 낮은 항산화효과가 있는 것으로 나타났다.

Phospholipase $C-{\gamma}1\;(PLC-{\gamma}1)$ is an important signaling molecule for cell proliferation and differentiation. $PLC-{\gamma}1$ contains two pleckstrin homology (PH) domains, which are responsible for protein-protein interaction and protein-lipid interaction. $PLC-{\gamma}1$ also has two Src homology (SH)2 domains and a SH3 domain, which are responsible for protein- protein interaction. To identity proteins that specifically binds to PH domain of $PLC-{\gamma}1$, we prepared and incubated the glutathione S-transferase(GST)-fused PH domains of $PLC-{\gamma}1$ with COS7 cell lysate. We found that 90 kDa protein specifically binds to PH domain of $PLC-{\gamma}1$. By matrix-assisted laser desorption ionization time of flight-mass spectrometry, the 90 kDa protein revealed to be heat shock protein (Hsp) $90{\beta}$. Hsp $90{\beta}$ is a molecular chaperone that stabilizes and facilitates the folding of proteins that are involved in cell signaling, including receptors for steroids hormones and a variety of protein kinases. To know whether Hsp $90{\beta}$ affects on $PLC-{\gamma}1$ activity, we performed $PIP_2$ hydrolyzing activity of $PLC-{\gamma}1$ in the presence of purified Hsp $90{\beta}$ in vitro. Our results show that the Hsp $90{\beta}$ dose-dependently inhibits the enzymatic activity of $PLC-{\gamma}1$ and further suggest that Hsp $90{\beta}$ regulates cell growth and differentiation via regulation of $PLC-{\gamma}1$ activity.
The secondary electron emission coefficient y of MgO single crystal according to the gas mix-ture ratio of Xe, $N_2$ to Ne have been investigated by $\gamma$-focused ion beam system. It is found that the MgO single crystals of (111) crystallinity has the highest $\gamma$ for operating Ne(Xe) ions ranging from 50eV to 200eV throughout this experiment. And it is found that the $\gamma$ for gas mixtures are much smaller than pure Ne ions.
There is considerable evidence that ionizing radiation (IR) mediates checkpoint control, repair and cell death. In this study, we have used a high density microarray hybridization approach to characterize the transcriptional response of human breast carcinoma MCF-7 cell line to ${\gamma}$-radiation, such as 4 Gy 4 hr, 8 Gy 4 hr, and 8 Gy 12 hr. We found that exposure to ${\gamma}$-ray alters by at least a $log_2$ factor of 1.0 the expression of 115 known genes. Of the 66 genes affected by ${\gamma}$-radiation, 49 are down-regulated. High Temperature deformation behavior of newly developed beta-gamma TiAl alloy was investigated in this study. The optimum processing condition was investigated with the aid of Dynamic Materials Model (DMM). Processing maps representing the efficiency of power dissipation for microstructural evolution and instability were constructed utilizing the results of hot compression test at temperatures ranging from $1000^{\circ}C$ to $1200^{\circ}C$ and strain rate ranging from $10^{-4}/s$ to $10^2/s$. The Artificial Neural Network (ANN) simulation was adopted to consider the deformation heating. With the help of processing map and microstructural analysis, the optimum processing condition was presented and the role of $\beta$ phase was also discussed in this study.
The high temperature deformation behavior of two-phase gamma TiAl alloy has been investigated with the variation of temperature and ${\gamma}/{\alpha}_2$ volume fraction. For this purpose, a series of load relaxation tests and tensile tests have been conducted at temperature ranging from 800 to $1050^{\circ}C$. In the early stage of the deformation as in the load relaxation test experimental flow curves of the fine-grained TiAl alloy are well fitted with the combined curves of two processes (grain matrix deformation and dislocation climb) in the inelastic deformation theory. The evidence of grain boundary sliding has not been observed at this stage. However, when the amount of deformation is large (${\epsilon}{\approx}$ 0.8), flow curves significantly changes its shape indicating that grain boundary sliding also operates at this stage, which has been attributed to the occurrence of dynamic recrystallization during the deformation. With the increase in the volume fraction of ${\alpha}_2$-phase, the flow stress for grain matrix deformation increases since ${\alpha}_2$-Phase is considered as hard phase acting as barrier for dislocation movement. It is considered that cavity initiation is more probable to occur at ${\alpha}_2/{\gamma}$ interface rather than at ${\gamma}/{\gamma}$ interface.
This study was carried out to discover the effects of eating habits and health-related life style on blood pressure, $\gamma$-Glutamic acid Peptide Transferase($\gamma$-GPT), blood glucose and High Density Lipoprotein-Cholesterol(HDL-C). 185 subjects(85 male, 100 female) were selected, who were living in the Cheonju area aged 40#s to 60#s. The mean systollic blood pressure(SBP), diastollic blood pressure (DBP), $\gamma$-GPT, fasting blood sugar(FBS) and HDL-C for all the subjects were 118mmHg, 77mmHg, 281U/l, 90mg/dl and 45mg/dl, respectively. The SBP and DBP for subuects over 60 years old were 126mmHg and 81mmHg and were significantly higher than subjects in their 40#s and 50#s(p<0.001, p<0.005). The HDL-C of the group that rarely ate breakfast was 57mg/dl and that was significantly higher than the 44mg/dl scored by those who ate breakfast everyday(p<0.05). The SBP for subjects having a snack 2-3 times/week was 125mmHg and that was significantly higher than the 114mmHg of those having a snack everyday(p<0.05). The $\gamma$-GPT for subjects consuming alcohol everyday was 44IU/L and that was significantly higher than 18IU/I of the non-drinking group(p<0.001). The $\gamma$-GPT of light smokers was 53IU/I and that was significantly higher than the 22IU/I for non-smoking participants(p<0.001). The DBP, SBP, $\gamma$-GPT, FBS and HDL-C related to exercise not significantly different. The SBP(p<0.001) and DBP(p-0.01) between age group was positively correlated. The $\gamma$-GPT between drinking frequency(p<0.001), drinking quantity(p<0.05), and smoking(p<0.05) was also positively correlated. The FBS between exercises had a negative correlation(p<0.05), and the HDL-C between breakfast had a negative correlation(p<0.05). These results indicate that decreasing drinking and smoking, when combined with appropriate exercise, will decrease the $\gamma$-GPT and fasting blood sugar level, and help preventing adult diseases.
