Anti-obesity Activities of Cirsium setidens Nakai Ethanolic Extract (고려엉겅퀴 주정 추출물을 함유하는 임상시험제품의 항비만 활성 평가)

  • The purpose of this study was to evaluate anti-obesity activity of Cirsium setidens Nakai test material (CNTM) in 3T3-L1 adipocytes and obese C57BL/6J mice fed with a high-fat diet using various obesity-related in vitro experiments. During adipocyte differentiation, CNTM significantly inhibited lipid accumulation and ROS production compared to controls. To evaluate whether CNTM could exert glycerol release effects on mature 3T3-L1 adipocytes, we treated cells with various concentrations of CNTM for 1 h. Treatment of mature adipocytes with $160-320{\mu}g/mL$ of CNTM increased the release of glycerol, but not in a significant dose-dependent manner. Anti-adipogenic and anti-lipogenic effects of CNTM seemed to be mediated by the inhibition of $PPAR{\gamma}$ and $C/EBP{\alpha}$. Moreover, CNTM stimulated fatty acid oxidation in an AMPK-dependent manner. CNTM-treated groups of C57BL/6J mice showed reduced body weights and adipose tissue weight with improving serum lipid profiles and adiponectin protein expression in obese C57BL/6J mice fed with a high-fat diet. These results suggest that CNTM might have anti-obesity effect on adipogenesis and lipid metabolism in vitro and in vivo. This presents the possibility of developing a treatment for obesity using nontoxic natural resources.

Molecular and functional characterization of the adiponectin (AdipoQ) gene in goat skeletal muscle satellite cells

  • Objective: It is commonly accepted that adiponectin binds to its two receptors to regulate fatty acid metabolism in adipocytes. To better understand their functions in the regulation of intramuscular adipogenesis in goats, we cloned the three genes (adiponectin [AdipoQ], adiponectin receptor 1 [AdipoR1], and AdipoR2) encoding these proteins and detected their mRNA distribution in different tissues. We also determined the role of AdipoQ in the adipogenic differentiation of goat skeletal muscle satellite cells (SMSCs). Methods: SMSCs were isolated using 1 mg/mL Pronase E from the longissimus dorsi muscles of 3-day-old female Nanjiang brown goats. Adipogenic differentiation was induced in satellite cells by transferring the cells to Dulbecco's modified Eagle's medium supplemented with an isobutylmethylxanthine, dexamethasone and insulin cocktail. The pEGFP-N1-AD plasmid was transfected into SMSCs using Lipofectamine 2000. Expression of adiponectin in tissues and SMSCs was detected by quantitative polymerase chain reaction and immunocytochemical staining. Results: The three genes were predominantly expressed in adipose and skeletal muscle tissues. According to fluorescence and immunocytochemical analyses, adiponectin protein expression was only observed in the cytoplasm, suggesting that adiponectin is localized to the cytoplasm of goat SMSCs. In SMSCs overexpressing the AdipoQ gene, adiponectin promoted SMSC differentiation into adipocytes and significantly (p<0.05) up-regulated expression of AdipoR2, acetyl-CoA carboxylase, fatty-acid synthase, and sterol regulatory element-binding protein-1, though expression of CCAAT/enhancer-binding $protein-{\alpha}$, peroxisome proliferator-activated receptor ${\gamma}$, and AdipoR1 did not change significantly. Conclusion: Adiponectin induced SMSC differentiation into adipocytes, indicating that adiponectin may promote intramuscular adipogenesis in goat SMSC.

Physicochemical Properties of a Biopolymer Flocculant Produced from Bacillus subtilis PUL-A (Bacillus subtilis PUL-A로부터 생산된 Biopolymer 응집제의 물리화학적 특성)

  • Soybean milk cake (SMC) was used for the solid-state fermentation by Bacillus subtilis PUL-A isolated from soybean milk cake. In the presence of 5% glutamate the maximum production of biopolymer (59.9 g/kg) was performed by fermentation at $42^{\circ}C$ for 24 hr. The recovered biopolymer was consisted of 87% $\gamma$-polyglutamic acid with molecular weight of $1.3{\times}10^6$ dalton and other biopolymer. The biopolymer solution showed the great decrease in consistency below pH 6.0, regardless of the molecular weight of PGA. Biopolymer solution has a typical pseudoplastic flow behavior and yield stress. The consistency of biopolymer solution was greatly decreased by increasing heating time and temperature in acidic condition compared to the alkaline condition. In kaolin clay suspension, the flocculating activity of biopolymer was the highest value with 15 mg/L biopolymer and 4.5 mM $CaCl_2$, but decreased greatly with $FeCl_3$. The flocculating activity of biopolymer was maximum at pH5, but decreased drastically by heating at $60{\sim}100^{\circ}C$. In particular, biopolymer with native PGA showed the efficient flocculating activity compared to that of modified biopolymer containing low molecular weight of PGA.

  • Longevity in medicine can be defined as a long life without mental or physical deficits. This can be prevented by Alzheimer's disease (AD). Current conventional AD treatments only alleviate the symptoms without reversing AD progression. Recent studies demonstrated that Panax ginseng extract improves AD symptoms in patients with AD, and the two main components of ginseng might contribute to AD amelioration. Ginsenosides show various AD-related neuroprotective effects. Gintonin is a newly identified ginseng constituent that contains lysophosphatidic acids and attenuates AD-related brain neuropathies. Ginsenosides decrease amyloid ${\beta}$-protein ($A{\beta}$) formation by inhibiting ${\beta}$- and ${\gamma}$-secretase activity or by activating the nonamyloidogenic pathway, inhibit acetylcholinesterase activity and $A{\beta}$-induced neurotoxicity, and decrease $A{\beta}$-induced production of reactive oxygen species and neuro-inflammatory reactions. Oral administration of ginsenosides increases the expression levels of enzymes involved in acetylcholine synthesis in the brain and alleviates $A{\beta}$-induced cholinergic deficits in AD models. Similarly, gintonin inhibits $A{\beta}$-induced neurotoxicity and activates the nonamyloidogenic pathway to reduce $A{\beta}$ formation and to increase acetylcholine and choline acetyltransferase expression in the brain through lysophosphatidic acid receptors. Oral administration of gintonin attenuates brain amyloid plaque deposits, boosting hippocampal cholinergic systems and neurogenesis, thereby ameliorating learning and memory impairments. It also improves cognitive functions in patients with AD. Ginsenosides and gintonin attenuate AD-related neuropathology through multiple routes. This review focuses research demonstrating that ginseng constituents could be a candidate as an adjuvant for AD treatment. However, clinical investigations including efficacy and tolerability analyses may be necessary for the clinical acceptance of ginseng components in combination with conventional AD drugs.

Enhancement of Immune Activities of Canavalia gladiata & Arctium lappa complexes in immobilization stress mouse model. (부동화 스트레스 유도 마우스 모델에서 도두(刀豆), 우방근(牛蒡根) 복합물의 면역증진 작용)

  • Objective : Soybeans of Canavalia gladiata(CG) and root of Arctium lappa(AL) have been reported to have anti-inflammatory, antioxidant effect. However, the immunoregulatory mechanisms of its combinational prescription remain a matter of considerable debate. In the current study, we investigated whether CG and AL and its combinational prescription(CG+AL) regulate immune system using chronic immobilization-stress mouse model. Methods : C57BL/6J mice fixed for 2 hours into immobilization tube after CG, AL, CG+AL oral administration after 2 hours daily for 21 days. After every experiment has ended the C57BL/6J mice were sacrificed on 22 days. The production of Serotonin and Cortisol, lgA were observed by ELISA method, The proportion of immune cells such as T/B cell and macrophage, NK cell were measured by FACS. Then, Real-time PCR was performed to measure the mRNA expression of Inflammatory cytokines(IL-1beta, IL-6, TNF-a) and T cell activation cytokines(IL-2, IL-10, IFN-gamma, IL-12p35 / p40). Result : When chronic immobilization-stress mouse model were treated with CG+AL(1:4), the expression of mRNA were significantly decreased at the Inflammatory cytokines(IL-1beta, IL-6, TNF-a). While, the levels of mRNA were significantly increased at immune T cell activation cytokines. Additionally, CG+AL(1:4) combinational prescription group enhanced immune cells such as T/B cell and macrophage, NK cell. Furthermore, the Immuno-fluorescence result of brain tissue can confirm that CG+AL(1:4) group significantly increased the BDNF expression. Conclusion : These result suggest that CG+AL(1:4) combinational prescription has Immune System enhancement via stress-mediated immunocyte.

Interpretation of Geophysical Well Logs from Deep Geothermal Borehole in Pohang (포항 심부 지열 시추공에 대한 물리검층 자료해석)

  • Various geophysical well logs have been made along the four deep wells in Pohang, Gyeongbuk. The primary focus of geophysical well loggings was to improve understanding the subsurface geologic structure, to evaluate in situ physical properties, and to estimate aquifer production zones using fluid temperature and conductivity gradient logs. Especially natural gamma logs interpreted with core logs of borehole BH-1 were useful to discriminate the lithology and to determine the lithologic sequences and boundaries consisting of semi-consolidated Tertiary sediments and intrusive rocks such as basic dyke and Cretaceous sediments. Cross-plot of physical properties inferred from geophysical well logs were used to identify rock types such as Cretaceous sandstone and mudstone, Tertiary sediments, rhyolite, and basic dyke. The temperature log indicated $82.51^{\circ}C$ at the depth of 1,981.3 meters in borehole BH-4. However, considering the temperature of borehole BH-2 measured under stable condition, we expect the temperature at the depth in borehole BH-4, if it is measured in stable condition, to be about 5 or $6^{\circ}C$ higher. Several permeable fractures also have been identified from temperature and conductivity gradient logs, and cutting logs.

The Effects of BGG on Various Immunological Factors Related to Pathogenesis of Allergic Dermatitis in NC/Nga Mice Induced by Der-f (보음거풍지양탕(補陰祛風止痒湯)이 아토피 피부염을 유발(誘發)한 NC/Nga 생쥐의 면역(免疫) 조절작용(調節作用)에 미치는 영향(影響))

  • To evaluate the therapeutic effects of BGG on atopic dermatitis, we investigated the composition of immune cells of lymph node, PBMC and skin of Dermatophagoides farinae-induced NC/Nga mice. The levels of immunoglobulins in serum were analyzed at the protein level and the amount of pathologic cytokines were investigated using CD3/CD28 stimulated splenocytes. The results are summarized below; 1. BGG showed no cytotoxic effect up to $200\;{\mu}g/m{\ell}$ on mLFC in vitro. 2. BGG showed no hepatotoxicity in vivo based on the levels of ALT and AST. 3. Atopic dermatitis was improved through naked eye examination. BGG reduced the skin clinical index from 2.9 to 1.3 (p<0.01). 4. H&E and toluidine blue staining of tissue biopsies revealed that BGG inhibited the infiltration of lymphocytes and mast cells to skin. 5. BGG reduced the number of CD19 positive B cells in PBMCs by 16% (p<0.01), whereas cells were increased by 26% (p<0.05) in lymph nodes. 6. BGG reduced the numbers of B220+/CD23+ cells by 15% (p<0.01) and 33% in PBMCs and lymph node, respectively. 7. BGG reduced the numbers of B220+/IgE+ cells in PBMCs and lymph node by 21% and 33% (p<0.01), respectively. 8. BGG suppressed the levels of IgE (13%, p<0.001) as well as IgM (34%, p<0.001), IgG2a (40%, p<0.001) and IgG2b (26%, p<0.05). 9. BGG reduced the levels of IL-4 and IFN-$\gamma$ by 7% (p<0.05) and 13% (p<0.001) in anti-CD3 and anti-CD28-activated splenocytes, respectively. 10. BGG considerably inhibited the production of TNF-$\alpha$ and IL-6 by 42% (p<0.01) and 15% in the serum, respectively. Based on the results above, we concluded that BGG has therapeutic effects on atopic dermatitis by regulating the differentiation of B cells and isotype switching of IgE. Further investigations on the molecular mechanisms of BGG on atopic dermatitis are anticipated.

CLINICAL STUDY OF POLYPOID VOCAL CORDS (REINKE'S EDEMA) (폴립양 성대의 임상적 고찰)

  • 성대의 점막하 조직의 부종을 특징으로 하는 폴립양 성대는 음성의 남용 및 상기도에 대한 다양한 자극 요인들에 의하여 발생된다고 보고되어 있으며, 현재까지 부종의 기전 및 병태에 대하여도 혈괴의 유리화 현상(hyalinization), 또는 단순한 혈관의 투과력 증대 등의 논란이 많은 실정에 있어 치료방법 역시 학자에 따라 견해를 달리하고 있다. 이에 저자들은 폴립양 성대의 치료에 도움을 얻고자 최근 5년간 본 교실에서 경험한 폴립양 성대 34례(남자 18예, 여자 16예, 평균연령 53.7세)에 대한 임상 소견 및 저자들의 치료 성적을 분석 검토하여 다음과 같은 결과를 얻었다. 1. 측별로는 총 34예중 양측 23예(67.6%), 편측 11예(32.4%)이었다. 2. 동반질환으로 성대에 타 질환이 있었던 예가 9예(후두용 5예, 과각화증 3예, 성대마비 1예 ; 26.5%), 수면무호흡증 5예(14.7 %)이었으며 전신질환이 있었던 예가 4예(기관지 천식 2예, 폐결핵 2예 ; 11.8%)이었다. 3. 유발요인으로는 음성남용 7예(20.6%), 상기도감염 3예(8.8%), 흡연 26 예 (76.5%)이었다. 4. 공기역학검사 및 청각심리검사가 가능하였던 14예에서 최대발성지속시간이 정상이하이었던 예가 10예(71.4%), 발성율이 정상이상이었던 예가 9예 (64.3%)이었으며, 애성의 특징도 조호성(rough)이 10예(71.4%)로 가장 많았다. 5. 총 34예를 sucking technique를 적용하여 수술적 치료를 하였는데 음성이 호전된 경우는 32예(정상 15예, 호전 17예 : 94.1%)이었으며 호전되는데 걸린 평균 기간은 2.8개월이었다. 이상의 성적으로 보아 폴립양 성대는 국소 또는 전신적으로 동반질환이 많고 흡연 등의 만성 자극 요인이 있으며 술후 음성 호전에 걸리는 기간이 길어 보다 복합적인 측면에서 치료에 임하여야 할 것으로 사료된다. with such configuration.trap with 2.88[eV] deep of injected space charge from the chathode in the crystaline regions.   • Makgeolli is made from rice or flour, yeast, and nuruk, a fermentation starter. The flavor of makgeolli is affected by sugars, amino acids, organic acids and volatile flavor compounds produced by various microorganisms. In this study, lactic acid bacteria (LAB) were isolated from unsterilized makgeolli samples collected from several provinces in Korea, and then later identified. Under anaerobic conditions, LAB density ranged from $5.0{\times}10^6$ to $1.5{\times}10^8CFU/mL$; yeast density ranged from $2.5{\times}10^7$ to $1.5{\times}10^8CFU/mL$. Of the LAB isolated from makgeolli, 1,126 were analyzed using restriction fragment length polymorphism (PCR-RFLP) analysis of 16S rRNA, which allowed for classification into five groups. Of the 1,126 LABs tested, 130 produced ${\gamma}$-aminobutyric acid (GABA).

Multiple Alternating Immunizations with DNA Vaccine and Replication-incompetent Adenovirus Expressing gB of Pseudorabies Virus Protect Animals Against Lethal Virus Challenge

  • The prime-boost vaccination with DNA vaccine and recombinant viral vector has emerged as an effective prophylactic strategy to control infectious diseases. Here, we compared the protective immunities induced by multiple alternating immunizations with DNA vaccine (pCIgB) and replication-incompetent adenovirus (Ad-gB) expressing glycoprotein gB of pseudorabies virus (PrV). The platform of pCIgB-prime and Ad-gB-boost induced the most effective immune responses and provided protection against virulent PrV infection. However, priming with pCIgB prior to vaccinating animals by the DNA vaccine-prime and Ad-boost protocol provided neither effective immune responses nor protection against PrV. Similarly, boosting with Ad-gB following immunization with DNA vaccine-prime and Ad-boost showed no significant responses. Moreover, whereas the administration of Ad-gB for primary immunization induced Th2-type-biased immunity, priming with pCIgB induced Th1-type-biased immunity, as judged by the production of PrV-specific IgG isotypes and cytokine IFN-$\gamma$. These results indicate that the order and injection frequency of vaccine vehicles used for heterologous prime-boost vaccination affect the magnitude and nature of the immunity. Therefore, our demonstration implies that the prime-boost protocol should be carefully considered and selected to induce the desired immune responses.
