Free radical scavenging activity and characterization of the extrcts from Alpinia katsumadai and Areca catechu (초두구와 빈량 추출물의 자유라디칼 소거 활성 효과와 특성에 관한 연구)

  • 자유라디칼은 반응성이 커서 세포의 구성 물질인 단백질, 지질, 당, DNA등과 반응하여 세포 및 조직을 손상시킴으로서 노화를 가져온다. 이러한 자유라디칼들은 자외선, 일반 대사, 스트레스, 질병, 흡연 등에 의해 생성되고 특히, 활성 산소 종인 hydroxyl radical, superoxide radical, lipid peroxide radical등은 피부 노화를 발생시키는 원인이 된다. 따라서 본 연구에서는 한방 식물로부터 유해한 활성 산소종을 소거하는 활성을 가진 물질을 기 위해 1차 스크리닝 하였고, 이중 육두구, 관중, 초두구, 빈랑, 금은화, 포공영, 우롱차, 황금, 녹차 추출물이 효과가 높게 나타났으며, 그중 초두구와 빈랑 90% 메탄올 추출물에서 가장 우수한 소거 활성을 나타냄으로써 이 추출물에 대하여 자유라디칼 소거 활성에 대하여 여러 가지 생물학적 활성을 조사하였다. 초두구와 빈랑추출물에 대한 hydroxyl radical, superoxide radical, lipid peroxide radical을 소거하는 활성을 핵산, 클로로포름, 에틸아세테이트, 물층으로 분획하여 조사한 결과 에틸아세테이트층에서 가장 우수한 활성을 보였다. 에틸아세테이트 분획에 대한 lipid peroxide radical 소거에 있어서 빈랑 추출물이 $IC_{50}$/ 값 50 $\mu\textrm{g}$/ml, 초두구 추출물은 $IC_{50}$/ 값 59 $\mu\textrm{g}$/ml로서 기준물질로 사용되는 vitamic C (120 $\mu\textrm{g}$/ml)나 butylated hydroxyl toluene (80 $\mu\textrm{g}$/ml) 보다 더 우수한 소거 효과를 보여주었고, hydroxyl radical을 소거하는 능력은 25 $\mu\textrm{g}$/ml와 150 $\mu\textrm{g}$/ml로 hydroxyl radical의 소거 능이 좋은 vitamin C (180 $\mu\textrm{g}$/ml) 보다 뛰어난 소거 활성을 나타내었다. Superoxide radical을 소거하는 효과는 초두구 추출물의 $IC_{50}$/ 값 10 $\mu\textrm{g}$/ml, 빈랑 추출물이 15 $\mu\textrm{g}$/ml을 나타냈고, 이는 기준 물질인 vitamin C (35 $\mu\textrm{g}$/ml)보다 좋은 소거 활성을 보여주었으며, gallic acid 9 $\mu\textrm{g}$/ml과 유사한 효과를 나타내었다. 사람 섬유아세포를 배양하여 hydroxyl radical과 superoxide radical를 발생시킨 후 초두구와 빈랑 추출물의 세포 보호 효과를 실험한 결과 25 $\mu\textrm{g}$/ml의 농도로 처리하였을 때 각각 85% 이상의 우수한 세포 보호 효과를 나타내었다. 초두구로부터는 자유라디칼 소거 활성이 있는 물질을 분리하기 위하여 분획한 후 가장 높은 소거 활성을 보인 에틸아세테이트 층에 대하여 silica column chromatography, preparative TLC를 수행하였다. 초두구로부터 분리된 물질은 HPLC를 이용한 분리에서 phenol성 물질인 gallic acid와 동일한 retention time을 보여줌으로써 초두구로부터 분리된 물질은 gallic acid와 유사한 phenol성 물질이거나 그의 유도체일 것으로 추측된다. 따라서, 초두구와 빈랑 추출물은 피부 노화의 주요인이 되고 있는 lipid radical, hydroxyl radical, superoxide radical을 소거하는 활성이 뛰어나 자유라디칼에 의하여 발생되는 피부노화를 방지할수 있는 물질로서의 효과가 기대된다.가 기대된다.

Effect of Ferulic Acid on Polyamine Titers and Enzyme Activities during the Radicle Growth of Glycine max (대두 종자의 유근생장시 Ferulic Acid 가 Polyamine 함량과 효소활성에 미치는 영향)

  • Changes in polyamine titers and enzyme activities during radicle growth of Glycine max were studied in order to investigate the effect of ferulic acid in regulation of polyamine biosynthesis. Among eight compounds used, gallic acid stimulated the radicle growth and ferulic acid inhibited it significantly. During the radicle growth of Glycine max, the content of putrescine was shown the highest level at the second day, while at the fourth day spermidine was the highest and spermine followed. Ornithine decarboxylass (ODC, EC 4.1.1.17) seems to be responsible for biosynthesis of putrescine. As the concentration of ferulic acid (0.001, 0.01, 0.1 mM) treated increased, the content of spermine was gradually enhanced and putrescine was increased at 0.001~0.01 mM, decreased after 0.1 mM concentration but spermidine was not affected. Ferulic acid elevated ODC and S-adenosylmethionine decarboxylass (SAMDC, EC 4.1.1.50) activity. ODC activity was increased more than 120% and SAMDC activity was increased about 50% more than that of the control. Diamine oxidase (DAO EC, 1.4.3.6) activity was enhanced about 20% at low concentration, decreased after then.

Optimization of Ascorbic Acid Extraction from Rugosa Rose (Rosa rugosa Thunb.) Fruit Using Response Surface Methodology and Validation of the Analytical Method

  • In this study, ultrasound-assisted extraction was performed to extract ascorbic acid from rugosa rose (Rosa rugosa Thunb.) fruit. The optimal conditions were investigated by response surface methodology, using two variable including reaction time (16-44 min) and temperature (16-44℃). The ascorbic acid extraction was sensitive to the reaction time rather than the reaction temperature, and the optimal conditions for ascorbic acid extraction were 25℃ and 30 min. Ascorbic acid and gallic acid in the rugosa rose fruit extract were completely separated by HPLC, with a resolution factor of over 1.5 between the two. The correlation coefficient of the ascorbic acid was 0.999 in a linearity test for 50-150 ㎍/mL concentration of extract. The limit of detection and limit of quantification values were 0.16 ㎍/mL and 29.89 ㎍/mL, respectively. The relative standard deviations (RSD) for repeatability and reproducibility were determined, and each RSD showed good precision at less than 5% (N=6).

Effects of High Pressure Treatment on Antioxidant Compounds and Activity of Germinated Rough Rice (Oryza sativa L.) (고압처리가 발아벼의 항산화 성분과 활성에 미치는 영향)

  • This study is investigated to evaluate the enhancement of antioxidant compound and activity of rough rice with different germination periods and high pressure treatment. The subject was germinated at $37^{\circ}C$ for 6 days (HP0), and then the germinated rough rice were subjected to 30 MPa for 24 hr (HP24) and 48 hr (HP48), respectively. HP0, HP24 and HP48 samples were prepared and extracted with 80% ethanol. The highest total polyphenol contents (5.15 mg/g) occurred in treating at HP48 after germination for 5 days. The total phenolic acid contents including gallic acid, chlorogenic acid, catechin, ${\rho}$-coumaric acid, ferulic acid, salicylic acid, naringin, myricetin, trans-cinnamic acid, naringenin and kaempferol increased from $37.26{\sim}204.32{\mu}g/g$ at HP0 to $77.29{\sim}283.05{\mu}g/g$ at HP48. In antioxidant activity analyses, HP48 extracts showed higher values in ABTS and DPPH radical scavenging activity, reducing power, and $Fe^{2+}$ iron chelating effect than those of the HP24 and HP0 extracts. These results suggest that the combined treatment of high pressure treatment and germination process efficiently enhanced antioxidant compound and activity of rough rice.

Phenolic compounds of must and wine supplemented with Muscat Bailey A grape fruit stem (송이줄기 첨가에 따른 Muscat Bailey A 포도의 발효 중 발효액 및 포도주의 생리활성 물질 함량)

  • This study investigated the phenolic compounds of must and wine supplemented with different concentrations (0% (no added stems), 1%, 2%, 3%, and 5%) of fruit stems during winemaking using Muscat Bailey A (MBA) grapes. The red color, and total anthocyanin, total polyphenol, and tannin contents of the must and wine significantly (p<0.05) increased with increasing added amounts of grape fruit stems, while the volatile acid content decreased with increasing added amounts of grape fruit stems. Catechin (8.16~23.08 mg/L), gallic acid (2.32~3.28 mg/L), trans-resveratrol (1.38~3.27 mg/L), and ferulic acid (1.51~1.59 mg/L) were detected in the must and wine via HPLC. The bioactive substance contents increased with increasing added amounts of grape fruit stems, except for ferulic acid. The DPPH $IC_{50}$ activity was higher in the wine (12 mg/L) with 5% grape fruit stems than in ascorbic acid (67 mg/L). These results suggest that the fruit stems of MBA grapes can be used as functional materials for winemaking.

Isolation and Determination of Phenolic Compounds from Tuber of Color-fleshed and White-fleshed Potatoes

  • Color-fleshed potatoes 'Hong-young' and 'Ja-young' were developed by RDA, and it has reported that they have high content of anthocyanin. Additionally they show higher radical scavenging activity compared to white or yellow fleshed potatoes. So it will be expected that the consumption of color-fleshed potatoes gradually increase by pre-peeled potatoes and color potato chips. This study was conducted to enhance the utilization of the tuber of color-fleshed potatoes. At first, we isolated four compounds from the organic solvents soluble layer in ethanol extract of tuber, and their structures were characterized by spectroscopic methods and by comparing their data to those in the literature. Their structures were characterized to be caffeic acid (1), chlorogenic acid (2), gallic acid (3) and protocatechuic acid (4) for the first time reported from this source. These compounds were already reported ingredients but considered to exhibit a high physiological activity. The quantitative determination on the four compounds in tuber of color-fleshed [Hong-young (HY) & Ja-young (JY)] and white-fleshed [Superior (SP)] potatoes samples were measured using HPLC. The concentration of caffeic acid in each total fractionations of HY (184.4 g/g) and JY (435.1 g/g) were higher than in total fractionation of SP (31.1). The concentration of gallic acid in each total fractionations of HY (282.1 g/g) and JY (315.2 g/g) were higher than in total fractionation of SP (143.3 g/g). The concentration of chlorogenic acid contents appeared to be highest in total fractionation of SP (954.2 g/g). The concentration of protocatechuic acid in total fractionation of HY (120.3 g/g) was higher than in each total fractionationss of JY (74.4 g/g) and SP (102.7 g/g). Overall, color-fleshed potatoes had higher amount of physicochemical properties than 'Superior'. Therefore, color-fleshed potatoes are expected to be highly valuable items for the development and applications of a functional food. In addition, these results will provide fundamental information for improving sitological value, and breeding of new cultivar.

Evaluation of the antioxidant potential and enzyme activities in species of Rhus (옻나무류의 항산화력 및 항산화 효소 활성)

  • Comparison of the antioxidant potential and enzyme activities of the extracts of Rhus showed considerable differences. The antioxidative activities of Rhus javanica were the highest of three species tried (7 $\mu\textrm{g}$/$m\ell$). The highest activities showed in fraction No.3 of 12 fractions by Sephadex LH-20 column chromatography and the antioxidative activity showed, in purified extract of each stem,1.5 $\mu\textrm{g}$/$m\ell$ (in Rhus verniciflua), 1.9 $\mu\textrm{g}$/$m\ell$ (in Rhus javanica) and 2.3 $\mu\textrm{g}$/$m\ell$ (in Rhus tricocarpa) respectively. These were identificated as phenolic compounds which are well known antioxidant compounds such as 2-propenoic acid (Caffeic acid), Benzoic acid (Gallic acid), 7-hydroxy- 6methoxy-2H-1-Benzopyran-2-one (Scopoletin). POD activities of stem were higher than leaf. Especially, POD activity in stem of Rhus javanica was 193 times higher than leaf. Rhus tricocarpa, however, showed very low POD activity. SOD activities of stem were higher than that of leaf in Rhus javanica and Rhus verniciflua but in Rhus tricocarpa, the activity of leaf was 25 times higher than that of stem.

  • The repeated column chromatographic separation of the EtOH extract of Portulaca oleracea afforded seven compounds. The structures of these isolates were identified as bergapten (1), umbelliferone (2), daidzein (3), genistein (4), protocatechuic acid (5), ferulic acid (6), and gallic acid (7) by the analysis of physico-chemical and spectral data. Their antioxidant effect on free radical scavenging was evaluated in the DPPH assay.

Optimization of Tannase Production by Aureobasidium pullulans DBS66

  • Tannase production by Aureobasidium pullulans DBS66 was optimized. The organism produced maximum tannase in the presence of 1% tannic acid after 36 h. Maximum gallic acid accumulation was observed within 36 h and tannic acid in the fermented broth was completely degraded after 42 h of growth. Glucose had a stimulatory effect on tannase synthesis at 0.1% (w/v) concentration. The organism showed maximum tannase production with $(NH_4)_2HPO_4$ as nitrogen source. Shaking speed of 120 rpm and 50-ml broth volume have been found to be suitable for maximum tannase production.

  • This study aimed to search for separate and identify of Quercus acutissima CARRUTHERS tannins. Tannins were extracted with methanol and ethylacetate from acorn powder and identified TLC, UV spectrum, HPLC, IR, GC/MS, and $^{1}H$ NMR. Three spots($R_{f}$ 0.94, 0.84 and 0.29) detected on TLC. These spots gave dark blue color fairly on spraying with 0.3% potassium ferricyanid${\cdot}$0.3% ferricchloride reagent, and these tannins identified as gallic acid, caffeic acid and ellagic acid by UV spectrum, HPLC, IR, GC/ MS, and $^{1}H$ NMR.
