Antibacterial activity of Bio-fermented Galla Rhios Extract (오배자 발효추출물의 항세균활성)

  • Objectives : This experimental study was performed in order to investigate the antibacterial effect of bio-fermented Galla Rhois extract. Methods : The Galla Rhois extract was fermented by Streptococcus thermophilus, Saccharomyces cerevisiae and Lactobacillus delbrueckii, and their products was tested for antibacterial activity against six pathogenic microorganisms namely, Bacillus cereus, Bacillus subtilis, Staphylococcus aureus, Vibrio parahaemolyticus, Escherichia coli and Salmonella typhimurium by paper disc diffusion method. Results : The Galla Rhois fermented extract by Lactobacillus delbrueckii and Saccharomyces cerevisiae showed more effective antibacterial activity than not fermented extract against Bacillus subtilis and Vibrio parahaemolyticus. Antibacterial activity of fermented extract using especially Lactobacillus delbrueckii and Saccharomyces cerevisiae was proved that it was good with even 2 percents concentration. Antibacterial activity of Galla Rhois extract within pH 3 to pH 7 had been safe regardless of pH but low over pH 9. The growth of Bacillus cereus, Staphylococcus aureus, and Vibrio parahaemolyticus had a tendency to decrease depend on the increasing concentration of the extract. EtOEt, EtOAc and n-BuOH fractions of the Galla Rhois extract had a high level of antibacterial activity against Bacillus cereus, Bacillus subtilis, Staphylococcus aureus and Vibrio parahaemolyticus, respectively. Surprisingly, EtOAc fractions of the Galla Rhois extract showed higher antibacterial activity against Vibrio parahaemolyticus alone. And antibacterial activity against six pathogenic microorganisms had a tendency to increase depend on the increasing concentration of the fractions of the Galla Rhois extract. Conclusions : Bio-fermented Galla Rhois extract, efficiently inhibited the growth of Bacillus cereus and Vibrio parahaemolyticus.

Comparison of Antioxidant Activity and ${\alpha}$-Glucosidase Inhibiting Activity by Extracts of Galla rhois

  • We studied antioxidant activity and inhibitory effect of ${\alpha}$-glucosidase from aqueous, ethanolic and methanolic fractions of Galla rhois. In FRAP and ORAC assay for measuring antioxidant activity, we confirmed that Galla rhois extracts had strong antioxidant activity and ethanolic and methanolic extracts were relatively stronger than aqueous extract. We used trolox as a positive control. In order to measure the inhibitory effect of ${\alpha}$-glucosidase, we compared acarbose and Galla rhois extracts. As a result of ${\alpha}$-glucosidase inhibitory assay, aqueous, ethanolic and methanolic extracts of Galla rhois showed high inhibitory activitity and ethanolic and methanolic extracts were relatively stronger than aqueous extract. The 50% inhibitory concentrations (IC50s) of acarbose, aqueous, ethanolic and methanolic fractions were 0.45 mM, $0.53{\mu}g/ml$, $0.415{\mu}g/ml$ and $0.37{\mu}g/ml$, respectively. These results suggest that Galla rhois extracts can be a clinically useful anti-diabetic ingredient, indicating that it needs to be fractionated and isolated and should be further investigated.

Studies on the Cytoprotective and Anti-inflammatory Effects of Isolate from Galla Rhois (오배자의 생리활성 연구)

  • Galla Rhois is a nest of parasitic bug, has been traditionally used for the treatment of the therapy of diarrhea, peptic ulcer, hemauria, etc., that showed various anti-inflammatory activity, and other biological properties. We studied the effect of Galla Rhois ethanol extract. we investigated whether compounds isolated from the ethanol extract of Galla Rhois, could modulate iNOS and COX-2 expression in RAW 264.7 macrophages stimulated with lipopolysaccharide (LPS). We found compounds that suppressed LPS-induced iNOS and COX-2 expression. Suppression of the expression of iNOS and COX-2 was in parallel with the comparable inhibition of the production of nitric oxide (NO) and prostaglandin E2 (PGE2). Our results suggest that compounds can inhibit NO and PGE2 productions through suppression of LPS-induced iNOS and COX-2 expression. Because COX-2- or iNOS-dependent mechanisms are involved in inflammation and tumor progression, our findings provide a new uncovering mechanism responsible for anti-inflammatory and antitumor effects of Galla Rhois.

The Anti-wrinkle Effects and Whitening Effects of Galla Rhois (오배자의 항 주름 효과 및 미백 효과에 미치는 실험적 연구)

  • Recently, the demands for the effective and safe depigmentating and anti-aging agents of the skin have increased due to the medical, pharmaceutical and cosmetic reasons. The aim of this study is to search Galla Rhois (GR) inhibitory activity against aging process of skin in vitro. GR was screened for their inhibitory activity against elastase. GR (final concentration 1 mg/ml) appeared over 40% of inhibition of elastase activity. So we investigated anti wrinkle effects of GR look through MMP-1 inhibition activity. Also Extracts of GR showed higher anti-tyrosinase activity than arbutin (final concentration 1 mg/ml). These results suggest that GR could be potential sources of anti-aging and whitening effects for the skin. Objective : Galla Rhois extracts, Elastase (pancreatic from porcine pancreas), N-Succinyl-(Ala)3-p-nitroanilide, Ursolic acid, mushroom tyrosinase, L-tyrosine, arbutin, Amicon Ultra, Hand Hold UV lamp, MMP-1 primary antibody, MMP-1 secondary antibody Result : 1. GR appeared over 40% of elastase inhibitory activity. 2. GR had MMP-1 inhibition activity. 3. GR showed higher anti-tyrosinase activity than arbutin. Conclusion : According to above results, it is supposed that Galla Rhois could be strong potential sources of anti-aging and whitening effects for the skin.

${\beta}-Glucuronidase-inhibitory\;Activity$ and Hepatoprotective Effect of Herbal Medicines (생약의 ${\beta}-Glucuronidase$ 저해와 간장보호효과)

  • Inhibitory effect of water and ethyl acetate extract of 60 kinds of herbal medicines was investigated on ${\beta}-glucuronidase$. Among water extract of them, Galla Rhois had the most potent ${\beta}-glucuronidase-inhibitory\;activity$. Termaliae Fructus, Amomi Tsa-ko Fructus and Arecae Semen were also potent inhibitors. Among ethyl acetate extract of them, Galla Rhois had the most potent ${\beta}-glucuronidase-inhibitory\;activity$. Nelumbinis Semen. Ephedrae Radix and Termaliae Fructus were also potent inhibitors. The extract of Galla Rhois had potent hepatoprotective effect on $CCl_4-induced$ hepatotoxicity of rats. These results suggest that the ${\beta}-glucuronidase$ seems to be closely related to the liver injury, which could be prevented by the inhibitor of ${\beta}-glucuronidase$.

Development of Anti-Obesity Agent from Resource Plants

  • Obesity is a physical condition that results from excessive storage of fat in the body. The present study examined the anti-obesity effects of the selected natural medicine, Galla rhois extract (GRE) and solvent fractions on 3T3-L1 preadipocytes and in vivo studies. Here, we show that EtOAc fraction of Galla rhois inhibits the differentiation of the 3T3-L1 preadipocytes induced by differentiated medium in a dose-dependent manner. To investigate the effect of the GRE-EtOAc fraction on obesity in high fat diet-fed C57BL/6 mice, which included a normal diet (ND), high-fat diet (HFD) and HFD+GRE concentration-dependent, were fed to the mice for 6 weeks. The GRE-EtOAc fraction was inhibited the highest adipocyte differentiation in vitro, the GRE supplement significantly decreased body weight and visceral fat mass compared to the HFD group. The total cholesterol and triglyceride levels in the plasma were significantly decreased by GRE supplementation compared with those of the HFD group. Also, we aimed to determine the differentiation inhibition and the modulation of differentiation genes brought about by the Galla rhois in adipocyte. A cDNA microarray-based method was introduced for the high contents screening (HCS) of gene expressions. This technology has revolutionized gene expression studies by providing the means to measure mRNA levels in thousands of genes simultaneously in simple and complex biological samples. 13 genes were founded to be affected in their expression levels by more than 5-fold up-regulation after 4 days treatment with the EtOAc fraction from Galla rhois. Otherwise, 21 genes were founded to be affected in their expression levels by more than 5-fold down-regulation treated with the EtOAc fraction. Therefore, Galla rhois extract may be considered for use in a therapeutic agent to control obesity.

Effects of Herbal Extracts on the Inflammatory Reactions Which Use the Makeup Preparations (색조화장품에 적용될 수 있는 17종의 천연한약재가 피부 염증억제에 미치는 임상적 연구)

  • This study was carried out to investigate the effects of herbal extracts on the skin inflammatory reactions which use the makeup preparations. In experiment 1, among the herbal ingredients of herbal extracts, ethanol extracts and 1,3-BG(Butylene Glycol) extracts of Galla Rhois showed potent radical scavenging activity, more than 91% at all concentrations, tested by DPPH (1,1-diphenyl-2-picryl-hyrazyl) method. In experiment 2, ethanol extracts of Chrysanthemi Flos, Gardenias Flos, Galla Rhois showed potent inhibitory activity of the lipopolysaccharide-induced nitric oxide(NO) production, more than 87% at $10{\mu}g/m{\ell}$, by the macrophage RAW 246.7 cells. And 1,3-BG extracts of Taraxaci Herbs, Corm Fructus, Galla Rhois showed potent inhibitory activity of nitric oxide production, more than 89% at $10{\mu}g/m{\ell}$. In experiment 3, ethanol extracts of Chrysanthemi Flos, Gardeniae Flos, Galla Rhois showed potent inhibitory effects of cyclooxygenase-II activity, more than 78% at $10{\mu}g/m{\ell}$, by using ELISA kit. And 1,3-BG extracts of Galla Rhois, Carthami Flos, Chrysanthemi Flos, Taraxaci Herba, Corm Fructus showed potent inhibitory effects of cyclooxygenase-II(COX-II) activity, more than 80% at $10{\mu}g/m{\ell}$. Therefore, 1 expect that herbal extracts, especially Galla Rhois may be used as a drug for treatment on skin inflammation and a material of the makeup preparations.

  • This study investigated the antioxidant and anti-inflammatory effects of Galla Rhois (Rhus verniciflua) on LPS (lipopolysaccharide)-stimulated human osteoblastic cells (MG-63). The aim of this study was to evaluate the LPS induced nitric oxide (NO) production and antioxidant radical in human osteoblast-like MG-63 cells. Therefore, the present work indicate that Galla Rhois extracts may be an ideal candidate for further research into their use for dental caries prevention component as well as, natural plant-based products. This suggested that 80% methanol and hexane extracts of Galla Rhois were inhibited NO generation and 1,1-diphenyl-2-pirylhydrazyl (DPPH) radical. Therefore, 80% methanol and hexane extracts of Galla Rhois may be utilized as a good source of protection against inflammation and oxidative stress. This study is intended to pursue wellness convergence in quality of life in the excavations in natural ingredients to create a variety of oral products with fewer side effects, cheap oral products for the dental treatment.

Constituents of Antioxidative Activity and Free Radical Scavenging Effect from Galla Rhois (Rhus javanica Linne) (오배자의 항산화 활성성분 및 자유라디칼 소거효과)

  • This study was carried out to investigate the structure of antioxidative constituents and the free radical scavenging effect of the main ingredients from Galla Rhois(Rhus javanica Linne). Antioxidative activities of n-hexane, EtOAc and BuOH extracts of Galla Rhois were similar or even higher than that of natural (tocopherol) or synthetic antioxidant (BHA). It is suggested that major fractions for the antioxidative activity of Galla Rhois were the n-hexane, EtOAc and BuOH extract compartments. In the subsequently experiment, one active compound from n-hexane extract, three active compounds from EtOAc extract and one active compound from BuOH extract were isolated. Their chemical structures were identified as syringic acid, protocatechuic acid, gallic acid methylester, gallic acid and $1,2,3,4,6- penta-O-galloyl-{\beta}-D-glucose$ on the basis of the speculation of spectral data and chemical reaction. Among the compounds, protocatechuic acid, gallic acid methylester and $1,2,3,4,6- penta-O-galloyl-{\beta}-D-glucose$ showed most potent radical scavenging effect using DPPH method.

Effect of Hexane Extract of Galla Rhois on Inflammatory Alveolar Bone Loss

  • One of functions of Galla Rhois (GR) is reportedly an anti-inflammatory effect on the several inflammatory diseases. However, an effect of GR related to periodontitis has not been investigated. In the present study, we examined the effect of the hexane extract of Galla Rhois (GR-H) on periodontitis. Cytotoxicity was assessed by MTS analysis using human gingival fibroblast (hGF) cells. Experimental periodontitis was induced by injecting E.coli LPS into the palatal gingiva maxillary molar thrice weekly for 3 weeks (LPS group). GR-H diluted in 1xPBS was orally administrated using a syringe at 30 mg/kg body weight and 100 mg/kg body weight once a day (GR-H group). GR-H effect on the alveolar bone loss (ABL) was digitized with a micro-CT. GR-H treatment at concentrations exceeding 0.5 mg/ml showed cytotoxic effect in hGF cells. The micro-CT among groups were presented for the different distances from cemento-enamel junction (CEJ) to alveolar bone crest (ABC). The results indicated an inhibitory effect on alveolar bone loss for orally administered GR-H in a model of LPS-induced periodontitis.
